Effect of Molecular Chaperones on the Soluble Expression of Alginate Lyase in E. coli

  • When the alginate lyase gene (aly) from Pseudoalteromonas elyakovii was expressed in E. coli, most of the gene product was organized as aggregated insoluble particles known as inclusion bodies. To examine the effects of chaperones on soluble and nonaggregated form of alginate lyase in E. coli, we constructed plasm ids designed to permit the coexpression of aly and the DnaK/DnaJ/GrpE or GroEL/ES chaperones. The results indicate that coexpression of aly with the DnaK/DnaJ/GrpE chaperone together had a marked effect on the yield alginate lyase as a soluble and active form of the enzyme. It is speculated this result occurs through facilitation of the correct folding of the protein. The optimal concentration of L-arabinose required for the induction of the DnaK/DnaJ/GrpE chaperone was found to be 0.05mg/mL. An analysis of the protein bands on SDS-PAGE gel indicated that at least 37% of total alginate lyase was produced in the soluble fraction when the DnaK/DnaJ/GrpE chaperone was coexpressed.

  • Let R be a commutative ring, M be a Noetherian R-module, and N a 2-absorbing submodule of M such that r(N :R M) = 𝖕 is a prime ideal of R. The main result of the paper states that if N = Q1 ∩ ⋯ ∩ Qn with r(Qi :R M) = 𝖕i, for i = 1, . . . , n, is a minimal primary decomposition of N, then the following statements are true. (i) 𝖕 = 𝖕k for some 1 ≤ k ≤ n. (ii) For each j = 1, . . . , n there exists mj ∈ M such that 𝖕j = (N :R mj). (iii) For each i, j = 1, . . . , n either 𝖕i ⊆ 𝖕j or 𝖕j ⊆ 𝖕i. Let ΓE(M) denote the zero-divisor graph of equivalence classes of zero divisors of M. It is shown that {Q1∩ ⋯ ∩Qn-1, Q1∩ ⋯ ∩Qn-2, . . . , Q1} is an independent subset of V (ΓE(M)), whenever the zero submodule of M is a 2-absorbing submodule and Q1 ∩ ⋯ ∩ Qn = 0 is its minimal primary decomposition. Furthermore, it is proved that ΓE(M)[(0 :R M)], the induced subgraph of ΓE(M) by (0 :R M), is complete.

We define the sationary Hilbert space process $X_k={\sum}^{\infty}_{j=0}a_j{\xi}_{k-j}$ and prove that $n^{-1}{\sum}^n_{k=1}X_k$ converges to zero.

Evidence of Interaction of Phage P22 Tailspike Protein with DnaJ During Translational Folding

  • Phage P22 tailspike is a thermostable homotrimeric protein, and temperature-sensitive folding (tsf) and global suppressor mutations affect its folding yields at elevated temperatures. We earlier suggested that the folding of the tailspike protein in Escherichia coli requires an unidentified molecular chaperone. Accordingly, in the present study, the interactions of purified DnaK, DnaJ, and GrpE heat-shock proteins with the tailspike protein were investigated during the translation and folding of the protein. The cotranslational addition of DnaJ to the tailspike protein resulted in the arrest of folding, when Dnak and GrpE were missing. However, the presence of DnaK, DnaJ, and GrpE had no effect on the folding yield of the tails pike protein, thus, providing evidence for the binding of the nascent tailspike protein with DnaJ protein, a member of DnaK chaperoning cycle.

  • In this paper, we study the existence of finite order meromorphic solutions of the following non-linear difference equation fn(z) + Pd(z, f) = p1eα1z + p2eα2z + p3eα3z, where n ≥ 2 is an integer, Pd(z, f) is a difference polynomial in f of degree d ≤ n - 2 with small functions of f as its coefficients, pj (j = 1, 2, 3) are small meromorphic functions of f and αj (j = 1, 2, 3) are three distinct non-zero constants. We give the expressions of finite order meromorphic solutions of the above equation under some restrictions on αj (j = 1, 2, 3). Some examples are given to illustrate the accuracy of the conditions.

  • 정보통신의 비약적인 발전과 더불어 보건의료에 대한 사회적 인식 변화로 인해 의료 서비스에 대한 개선의 요구가 점차 증대되고 있다. 의료서비스와 정보통신의 결합은 e-Health라는 보건의료분야의 새로운 비즈니스 영역을 탄생시켰다. 늦은 감이 있지만, 국내에서도 e-Health 산업의 성장이 본격화됨에 따라 정책적으로도 이를 적극 육성하려는 움직임을 보이고 있다. 이에 본 고에서는 영국, 일본, 미국의 e-Health 현황을 살펴보고 국내 e-Health 산업에 대한 시사점을 제시하였다.
