  • This experiment was carried out to study in stomatal behaviour of wheat (Chokwang) upon the different soil water potentials, lodging and two different nitrogen treatment. Stomatal aperture was measured at flowering stage under diurnal course with two hour intervals, Stomatal apertures were differently changed according to soil water potentials and leaf position on the stem, showing the rapid closure of stomata at low soil water potential and the narrow opening at low leaf position of stem. Wider stomatal opening appeared at the plot of optimal nitrogen application than that of low nitrogen. It was greatly decreased in opening of stomata at plants lodged, appearing that plant investigated of six days after lodging showed less than 50% than that normal plants, which indicated peculiar decrease of low leaf position of stem at afternoon of diurnal course.

Establishment of Sesame Cultivation Practices as Succeeding Cropping System of Watermelon in the Greenhouse Condition

  • This experiment was conducted to develop optimum sesame cultivation practices as succeeding cropping system of watermelon in the greenhouse condition. We also compared major components in sesame seed cultivated in the greenhouse and open field condition. The adaptable varieties under the greenhouse condition were white sesame 'Pyoungan' and black sesame 'Jinki' which showed higher yield and disease resistance. We also conducted several experiments to determine optimum sowing date, planting density and pinching time. Grain yields were statistically different according to the several sowing dates. In general, sesame sowing on June 30th showed higher grain yields than July 10th, July 20th and July 30th in the greenhouse. We also found out sesame cultivation practice with the row spacing of 40 cm and interplant spacing of 30 cm (a few branch type) or 40 cm (many branch type) showed higher yield than other treatments. Optimum pinching time was 25 days after first flowering in main stem when thousand seed weight and grain yield were highest. In the comparison of major components of sesame at the different cultivation conditions, sesame seeds cultivated in the greenhouse contained 4% much more oil content than open field condition. The lignan compounds, sesamin and sesamolin were also higher by 6% in the greenhouse than open field condition. It was concluded that sesame cultivation practices as succeeding cropping system of watermelon in the greenhouse condition guaranteed higher grain yield with less labor input which is now emerging alternative farming practice system in present aging rural society and will also give sesame cultivation farmers to increase net income in Korea.

  • Changes in activity of gibberellin C20-hydroxylase which converts $[^{14}C]GA_{12}\;to\;GA_{l5}$ were studied during seed maturation using the partially purified enzyme preparations of two cultivars, Kentucky Wonder (normal) and Masterpiece (dwarf) of Phaseolus vulgaris. The preparations obtained by methanol precipitation and hydrophobic interaction chromatography efficiently converted; $GA_{l2}\;to\;GA_{4},\;via\;GA_{15},\;GA_{24}\;and\;GA_{9};\;GA_{20}\;to\;GA_{1},\;GA_{5}\;and\;GA_{6}$. The activities of C20-hydroxylase which converts $GA_{l2}\;to\;GA_{l5}$ were almost the same in both cultivars. The C20-hydroxylase activity per protein reached a maximum at the very early immature seed stage, followed by a subsequent rapid decrease during seed maturation, whereas the enzyme specific activity per seed reached a maximum at 21 days after flowering, and showed a similar fiuctuation to that of the 313-hydroxylase which converts $GA_{20}\;to\;GA_{l}$ during seed maturation.ration.

Pre-adaptation to Cu during Plant Tissue Culture Enhances Cu Tolerance and Accumulation in Begonia (Begonia evansiana Andr.)

  • A simple and efficient protocol was developed for culturing Cu-tolerant and Cu-accumulating plants via pre-adaptation to Cu during plant tissue culture. We induced multiple shoots from begonia (Begonia evansiana Andr.) leaf explants on MS medium supplemented with naphtaieneacetic acid and benzyladenine. After 3 months, small plantlets were transferred to MS medium supplemented with $100{\mu}M\;CuCl_2$ for pre-adaptation to Cu and cultured for 5 months. Then, these plantlets were individually planted in pots containing artificial soil. An additional 500 mg of Cu dissolved in 1/4 strength MS solution was applied to each pot during irrigation over the course of 2 months. We planted pre-adapted and control begonias in soil from the II-Kwang Mine, an abandoned Cu mine in Pusan, Korea, to examine their ability to tolerate and accumulate Cu for phytore-mediation. Pre-adapted begonias accumulated $1,200{\mu}g$ Cu/g dry root tissue over the course of 45 days. On the other hand, non-Cu-adapted controls accumulated only $85{\mu}g$ Cu/g dry root tissue. To enhance Cu extraction, chelating agents, ethylenediamine tetraacetic acid (EDTA)-dipotassiun and pyridine-2,6-dicarboxylic acid (PDA), were applied. While the chelating agents did not enhance accumulation of Cu in the roots of control begonias, EDTA application increased the level of Cu in the roots of pre-adapted begonias twofold (to $2,500{\mu}g$ Cu/g dry root tissue). Because pre-adapted begonias accumulated a large amount of Cu, mainly in their roots, they could be used for phytostabilization of Cu-contaminated soils. In addition, as a flowering plant, begonias can be used to create aesthetically pleasing remediation sites.

New Early Maturing Variety with Large Fruits of Korean Native Cornus kousa BUERGER (조생종 대과형 산딸나무 신품종 선발)

  • This study was carried out to develop new varieties with early maturing and large fruit size in the Korean native dogwood (Cornus kousa) for woody landscape plants from native sites or cultivated area. The results are obtained as follows; We selected strains with early maturing and large fruit. size from the habitats from 2000 to 2003. ‘SKK 2-1’ had 4.0cm of fruit size while 2.0 cm of native variety. In addition, ‘SKK 2-2’, ‘SKK 2-3’, ‘SKK 2-4’, and ‘SKK 2-5’ with large fruit and early maturing were preliminary selected from the habitats. In the preliminary selections, ‘SKK 2-1’ was finally selected as new early maturing variety with large fruits in 2003. New early maturing variety with large fruit size, C. kousa var. ‘SKK 2-1’ was selected from 200 plants of 5-year-old seedlings in C. kousa in 5-12 Nung wean ri Goo sung myun Yong In, Kyung gi do, Korea in 2000. Their fruits were colorized in September 18, 20∼25 days earlier than October 7 of native varieties. Its fruit diameter is larger than 4cm. Its fruit weight was 5 times as heavy as native cultivars grown in Korea. This useful characters were still inherited after grafting. However, characteristics of growth, leaf, flower, and flowering period were not different from those of native varieties. Therefore, it was very promising landscape woody plant, having large fruits in early fall season. The rooting rate of Korean dogwood was high by the IBA treatment. The optimum date for softwood cutting was on June 25 ∼ July 25 when the rooting rate was more than 80%. In addition. Korean dogwood was easily propagated by grafting. Therefore, new early maturing variety with large fruits, C. kousa var. ‘SKK 2-1’ is easily propagated by softwood cutting or grafting and then is a promising cultivar as a woody landscape plant.

Effects of Various Anions on Absorption and Toxicity of Lead in Plants (식물체의 연(Pb) 흡수 및 유해성에 미치는 음이온의 영향)

  • The seeds of bean(Glycine max M.) and corn(Zea mays L.) soaked in 5000ppm lead solution for 24 hours were sowed in the flowerpots being placed sandy-clay soil under the field condition. The fixed concentrations of various anions and 2000 ppm lead were supplied alternately in the sandy-clay soi of the flowerpots at two days interval from May to July in 1976. After the plants were harvested prior to the flowering stage, the lead contents of plnat and soil were analyzed by atomic absorption spectrophotometer. The lead contents absorbed by the plant roots showed the highest in an weak acid soil of the best suitable condition of plant growth. The absorption of lead by the plant roots was inhibited by the various anions, especially divalent anions of the soil. Some phosphate anions inhibited lead absorption more than otehr various anions in the soil. The more various anions were in the soil, the more plants could be protected from the lead toxicity. In the case of lead supply in the soil, 99.5% of lead was accumulated in the upper layer of the soil(0-10cm), and 0.5% of lead accumulated in the lower layer (10-20cm). Therefore, the yellow-brown and white symptoms on the leaves and the inhibition of root growth by lead toxicity was increased in the early stage of the germination, however decreased in accordance with the progress of the growing stage becuase of the root growth toward beneath the lower layer of the soil. In spite of the contents of 3773ppm lead in the soil, the symptoms of lead toxicity was not found in the grown plants. At that time the lead contents of the plants absorbed from the soil were minimum 0.78ppm and maximum 3.64ppm through the growing stage.

A New Yellow and Semi-double Gerbera Cultivar 'Sunny Lemon' with Early Flowering and High Yielding (조기개화성 및 다수성 황색 반겹꽃 거베라 신품종 'Sunny Lemon')

  • A new gerbera cultivar 'Sunny Lemon' was released by the National Institute of Horticultural and Herbal Science (NIHHS) in 2009. A cross was made between 'Pavlov' and 'Patrick' with pink and semi-double in 2006. After investigation of the characteristics for four years (from 2006 to 2009), it was selected for the use of cut flower. 'Sunny Lemon' is a vigorous cultivar with yellow flower color (RHS Y9C), brown center and semi-double. The average flower diameter is 11.0 cm. And inner ray floret and disc diameter are 7.63 cm and 2.59 cm, respectively. Days to first blooming are 84.5, approximately 2 weeks earlier than the standard cultivar. It produces a large yield with continuous supply of cut flowers and has adequate peduncle height. We believe that 'Sunny Lemon' has a great potential for expanding cultivated area of domestic gerbera cultivars and increasing farmers' income.

Breeding of Pink Color Spray Carnation 'Lyra' for Cut Flower (분홍색의 절화용 스프레이 카네이션 '리라' 육성)

  • A new spray carnation cultivar 'Lyra' was released by the National Institute of Horticultural and Herbal Science in 2003. A cross was made between spray cultivar 'Ballantyne' and red spray cultivar 'Chateau' in 1999. After investigation of the characteristics during four years (from 2000 to 2003), the cultivar 'Lyra' was finally selected in 2003. 'Lyra' is a spray cultivar with bright pink color for a cut flower. The main characteristics are early flowering, multi branching, fragrance and long flower stalk. Also, it has medium resistance to Fusarium wilt and 7 days vase life. To avoid calyx splitting caused by high temperature, it is recommended that this cultivar be grown over $8^{\circ}C$ at night and under $25^{\circ}C$ at day time during a flower bud differentiation period and applied with additional nitrate fertilization ($30.0kg{\cdot}10^{-1}$).

  • Germinating ability of green seed in maturing period was investigated to develop the technics on generation shortening. Rapeseed did not germinate until 40 days after flowering (=DAF) in green seed and 50 DAF in dried seed. Effective germination to be utilized for generation shortening came on 55 DAF for dried seed by heating and on 75 DAF for green seed. The periodic time of seed dormancy in maturing period had two cycles and rest period was considered that it was different according to the ecological types of early, middle and late maturity.

  • The experiment was dealt with an investigations on the agronomic potential of primary octraploid triticale in comparion with Chokwang, a major common wheat, local rye cultivar and new triticale selection Suweon #1 and Suweon #2. This octaploid triticale was originated from the cross of Jukoku #81 x Local rye cultivar. The results obtained were summarized as follows; Cold tolerance of the P-Tcl was better than those of the wheat cultivar Chokwang, triticale varieties Suweon #1 and Suweon #2, and comparable to local rye. Culm length of P-Tel was 113cm that was intermediate between the wheat and rye, and its culm thickness was thiner than the checks except the rye. Lateness of heading and maturing date of the P-Tel seemed to be over-dominant. However, flowering date after heading was 3 to 7 days earlier than the Suweon #1 and Suweon #2. Yield potential of the P-Tel was poor in comparision with the triticale cultivar Suweon #1 and Suweon #2 which was attributed to the low fertility and less number of the heads per squ are meter. If it was . improved the low fertility and less number of the valid tillers through the cross with the good secondary triticale it seemed to be an useful material for triticale breeding, expecially for improving cold tolerant winter triticale varieties in Korea.
