Diversity of Culturable Bacteria Associated with Hard Coral from the Antarctic Ross Sea

  • The bacterial diversity of an Antarctic hard coral, Errina fissurata, was examined by isolating bacterial colonies from crushed coral tissue and by sequencing their 16S rRNA gene. From the analyzed results, the bacteria were classified as Actinobacteria (56%), Firmicutes (35%) and Proteobacteria (9%). The thirty-four isolates were cultured in liquid media at different temperatures and their growth was assessed over time. The majority of the isolates displayed their highest growth rate at 25℃ during the first three days of cultivation, even though the coral was from a cold environment. Nevertheless, strains showing their highest growth rate at low temperatures (15℃ and 4℃) were also found. This study reports the composition of an Antarctic hard coral-associated culturable bacterial community and their growth behavior at different temperatures.

Artificial seed production and cultivation of Sargassum macrocarpum (Fucales, Phaeophyta)

  • Sargassum macrocarpum is a rich source of anti-inflammatory compounds. Recently, one of the compounds, tuberatolide B, has been reported as a functional anti-inflammatory additive for foods and nutraceuticals. The artificial seeding, growth and maturation of S. macrocarpum were investigated from May 2018 to September 2019. Indoor culture experiments for induction of egg release were conducted at temperatures of 17, 20, 23, and 26℃ and irradiances of 0, 10, 20, 40, and 80 μmol photons m-2 s-1 under 14 : 10 h (L : D) photoperiod. Within a given treatment combination, higher temperatures and irradiance levels favoured the maturation of receptacles in S. macrocarpum. Using artificial temperature and irradiance control, thalli matured one month earlier than thalli in nature. Under natural condition, receptacle formation began in April, and the eggs were released in June and July. The release of eggs from the receptacles was promoted at 17-20℃ and 40-80 μmol photons m-2 s-1, and the fastest growth of germlings occuring at 15-17℃ and 40 μmol photons m-2 s-1. For mature thalli, 300 g wet-weight was sufficient to seed 100 m of seed string. Thalli grew to 10.5 ± 2.6 cm in length at a density of 6.7 ± 3.3 individuals m-1 after 1 year of cultivation, from germination. This study demonstrates that it is possible to cultivate S. macrocarpum for the production of anti-inflammatory products.

  • For successful microalgal biodiesel production, the strain should be selected carefully. Fast growth rate and high fatty acid contents are desired traits for algal biodiesel production. In ocean cultivation of microalgae, seawater temperature slowly changes over seasons, and rotating algal strains in accordance with their optimal temperature could improve overall productivity. Additionally, use of indigenous strain is preferred to alleviate potential impacts on the environment. In this study, five strains of Tetraselmis sp. from nearshore of Youngheung Island, Incheon, Korea, were isolated during winter and characterized for their growth patterns and fatty acid compositions in the low temperatures ($5-15^{\circ}C$). The five strains showed various characteristics in optimal growth temperature, fatty acid contents, and compositions. Compared with a strain of Tetraselmis sp., isolated from Ganghwa island in a previous study, a rapid-growing strain with 237% higher biomass productivity and an oleaginous strain with twice higher fatty acid contents at $10^{\circ}C$ were isolated. The oleaginous Tetraselmis strain showed the highest fatty acid productivity among the strains, having 438% higher productivity than the previous strain. Using the new isolates in the seasons with low seawater temperature would improve microalgal fatty acid productivity in ocean cultivation.

  • Poria cocos is an edible and pharmaceutical mushroom with a long history of medicinal use in Korea. For the last 30 years, the domestic cultivated supply of Poria cocos has been unable to meet consumer demand, so Poria cocos is collected in mountainous areas and also imported from China. Thus, to increase the supply of Poria cocos, many artificial cultivation methods have been studied. In this study, Poria cocos is cultivated under different environmental conditions using plastic bags and the results compared. When cultivating Poria cocos at different temperatures (20, 25, 30 and $35^{\circ}C$) and using different numbers of plastic bag layers (1, 2), the most efficient cultivation conditions were a temperature of $25-30^{\circ}C$ and 2 plastic bag layers. The fastest growth was at $25-30^{\circ}C$, and the Poria cocos exhibited no weight change when cultivated using layers of plastic bags (1, 2).

  • ICT 기반 스마트 표고재배시설을 활용하여 표고재배의 과정을 후숙관리, 발이관리, 생육관리 및 휴양관리 4과정으로 나누어 6월부터 10월까지 시험을 진행하였다. 여름철 재배사 외부 $20{\sim}35^{\circ}C$의 고온환경에서도 재배사 내부의 환경은 설정한 방향으로 제어되었다. 여름철 대표적인 재배용 품종 '산조701호'와 신품종 '산조717호'를 이용한 재배시험 결과 '산조701호'의 경우 353.7 g/봉, '산조717호'는 270.4 g/봉으로 '산조701호'가 높은 버섯생산성을 나타내었다. ICT 기반으로한 스마트 표고재배시설을 활용한 표고재배는 재배과정별 환경조건 설정으로 재배가 어려운 고온의 환경에서도 편리하게 재배환경 관리가 가능하였으며, 재배품종에 적합한 환경제어 범위를 연구한다면 버섯생산 안정화 및 생산성 향상에 크게 기여할 것으로 판단되었다.

조명나방(나비목: 포충나방과) 비휴면태의 생존과 발육, 생식에 미치는 온도의 영향 (Effects of Temperature on Survival, Development, and Reproduction of the Non-diapause Asian Corn Borer, Ostrinia furnacalis (Lepidoptera: Crambidae))

  • 옥수수 주 해충인 조명나방(Ostrinia furnacalis)(나비목: 포충나방과)의 비휴면태 단계의 생존과 발육, 생식에 미치는 온도 영향을 분석하였다. 비휴면태 단계는 16:8 h (명:암)의 광주기 조건에서 유지하였다. 미성숙태를 15~35℃ 범위의 7개 항온조건에서, 성충을 13~33℃ 범위의 8개 항온조건에서 인공사료로 사육하였다. 알은 적용된 모든 온도에서 생존율이 70% 이상이었으나, 유충은 15℃에서 7.4%의 낮은 생존율을 보였다. 온도가 증가함에 따라 미성숙태의 발육기간은 짧아졌으나, 유충기간은 35℃에서 더 짧아지지 않았다. 번데기 몸무게는 온도 증가에 따라 증가하였는데, 암컷의 무게는 35℃에서 다시 감소하였다. 25℃를 제외한 다른 6개 온도 각각에서 마지막 영기가 다른 개체변이가 관찰되었다. 성충은 적용된 모든 온도에서 자손을 생산하였다. 성충 수명과 산란전 기간, 산란기간은 온도가 증가함에 따라 감소하는 경향이었고, 산란전 기간은 33℃에서 다시 길어졌다. 총산란수는 22℃와 31℃에서 400개 이상이었다. 산란기간 중 일산란수와 산란일당 일산란수는 온도가 증가함에 따라 많아졌는데, 33℃에서 다시 감소하였다. 성충 나이에 따른 일일 산란수는 우화 초기 급격히 증가하였고 이후 완만히 감소하는 경향이었다. 산란횟수는 22℃에서 가장 많았다고 모의 추정되었다. 선형방정식으로 추정된 최저발육온도는 1령 유충이 9.7℃로 가장 낮았고, 5령~말령 단계가 14.7℃로 가장 높았다.

자실체 발생이 안정적인 꽃송이버섯 신품종 '한여울' (Characteristics of a New Sparassis latifolia Variety, "Hanyeoul", with Stable Pinheading)

  • 버섯의 편중재배 현상을 줄이고 경쟁력있는 품목을 육성하기 위해 재배기간이 짧고, 발이가 안정적이며, 수량이 높은 꽃송이버섯을 개발하고자 하였다. 2016년부터 국내외에서 유전자원을 수집하고 교배하여, 특성검정 및 생산력 검정, 농가실증을 통해 육성된 꽃송이버섯 신품종 '한여울'의 주요 특성은 다음과 같다. 균사생장적온은 22~25℃, 발이 및 생육온도는 19~21℃로 대조품종(너울)과 유사하였다. 재배일수는 생산력 검정(병재배)에서 125일로 대조품종 대비 39일, 농가실증(봉지재배)에서 98~102일로 대조품종 대비 9~12일 짧게 나타났다. 갓 색은 연황백색으로 대조품종과 유사하였고, 갓 끝부분의 형태는 수국형으로 갓의 굴곡 간격이 대조품종 대비 넓었다. 발이율은 생산력 검정(병재배)에서 93.7%, 농가실증(봉지재배)에서 90.2~90.9%로 대조품종 대비 안정적이었다. 수량은 생산력 검정(병재배)에서 1,100 cc(Ø75 mm)병 기준으로 79.1 g 대조품종 대비 22.6%, 농가실증(봉지재배)에서 2.6 kg봉지 기준으로 641.1~689.5 g로 대조품종 대비 5.8~10.4% 높았다. 모본 및 대조품종과 대치배양 시 대치선이 뚜렷하고, 균사체 DNA PCR반응 결과 밴드패턴이 모본 및 대조품종과 다른 양상을 보여 서로 다른 품종임을 확인하였다. 본 연구를 통해 발이율이 높고 재배기간을 단축할 수 있는 꽃송이버섯 품종을 육성하였으며, 맞춤형 재배기술 개발에 대한 지속적인 연구와 관심, 그리고 농가보급이 이어지길 기대한다.

메기(Silurus asotus) 양어 순환식 아쿠아포닉스와 수경재배 엽채류의 생육 및 수질의 비교 (Comparison of Water Quality and Growth on Leafy Vegetables in Far Eastern Catfish (Silurus asotus) Aquaponics and Hydroponic Culture)

  • This study aimed to investigate the effects of an aquaponics system utilizing catfish recirculating water on the growth and water quality in lettuce and chicory cultivation. After 60 d of catfish cultivation in the BFT-AP, the survival rate was 96.0% and average weight gain was 178.5 g. The WGR grew by 241.4% and the SGR was 3.0%. A comparison between lettuce and chicory cultivation using BFT-AP and hydroponics revealed significant increases in shoot height, leaf count per week, and biomass of the cultivated lettuce and chicory. The SPAD values also increased significantly by 15.4-43.3%. Water quality analysis of the BFT-AP systems and crop cultivation beds showed significant differences, except for K and Na. The average NO3- concentration in the sediment pond was significantly higher at 45.3±1.0. Comparison of water temperature, pH, EC, DO, and ORP in the BFT-AP system and crop cultivation beds revealed similar monthly water temperatures from July to September. The pH in the catfish culture pond was 0.3-1.2 lower than that in the crop cultivation beds, and the EC was approximately 0.01- 0.03 dS/m lower in the catfish culture pond than the crop cultivation beds. The monthly average ORP in the catfish culture bed steadily increased to 479.9 mV in October. Bacillus velezensis (AFB2-2) and Chlorella fusca (CF) were applied to the biological activation bed; the population density of CF showed a continuous increase, whereas that of AFB2-2 decreased after 15 d and gradually increased thereafter. Thus, the application of the recirculating aquaponics cultivation technology with catfish (BFT) enhances catfish cultivation and promotes the growth of lettuce and chicory. Therefore, it is considered suitable for the environment-friendly cultivation of lettuce and chicory.

Effect of the root-zone temperature grown in the greenhouse on the growth of chives

  • This study aimed to determine an optimal temperature of root zone for the chive cultivation in a greenhouse during the winter season that may raise the possibility of chive (Allium schoenoprasum L.) harvest any time year-round by reducing energy consumption. The maximum and minimum temperatures of root zone were 26.8 and $19.8^{\circ}C$ for the R-Z20, 28.3 and $23.6^{\circ}C$ for the R-Z25 and 22.4 and $14.3^{\circ}C$ for the control. The highest fresh weights of shoot and root, plant height, root length and stem diameter were observed in the R-Z20 treatment. There was no significant difference in the growth between the R-Z25 and control treatment. These results suggest that the optimal temperature of root zone is $20^{\circ}C$ for the chive cultivation in the greenhouse during winter season.

충남지방의 시설채소 재배동향과 앞으로의 방향 (Suggestion for the Future of Protected Vegetable Crops Culture in Chungnam)

  • The above results indicate that vegetable crop production using vinyl houses continously increases. However, technical support and facility improvement are still insufficent to establish a full production system. In order to improve the cultural environment in facilities for better vegetable culture, several facters should be considered. 1. Facility design and arrangement should be made to improve micro-environments for crop growth and development because direction of facility and covering materials can affect the amount of light trasmitted into vinyl houses. 2. Cultivation of several leading varieties in each crop may not provide stable production and profit especially under undesirable environmental conditions. Thus, it is required to grow not only leading varieties but also other cultivars to decrease the economic losses. 3. Crops should be carefully managed when planted in early season not to experience low temperatures which induce growth retards. Early season cultivation requires proper selection of cultivars which are resistant to low temperture. 4. Active control of micro-environment in facilities should be made via improving the ventilation and irrigation systems.
