Control of Ginseng Damping-Off Disease Using Chitinolytic Bacterial Mixtures (키틴분해미생물을 이용한 인삼 잘록병 방제)

  • An effective bioformulation of mixtures of chitin-degrading bacteria has been used successfully to control plant diseases and nematodes. In this study, the bioformulation approach was assessed to control damping-off disease of ginseng. In pot experiments with soils infested with dapming-off pathogens of ginseng, root-drenchings of Chrobacterium sp. C-61, Lysobacterium enzymogenes C-3, and mixture of two bacterial strains grown in chitin minimal medium were signficantly increased emergence of seeds and reduced damping-off disease incidence of seedlings. Efficacy of the bioformulated product depended on the dose and timing of application. In two-year-old ginseng field, the high control efficacies were achieved by soil drenching of two times with an undiluted product or three times with a 10-fold diluted product. In a To-jik nursery (self soil nursery), biocontrol efficacy of the undiluted product against damping-off disease were similar to that of a seed dressing with fungicide, Tolclofos-methyl WP. These results suggest that the bioformulated product containing Chromobacterium sp. C-61 and L. enzymogenes C-3 could be an effective approach to control of ginseng damping-off disease.

Biological Control of Pythium Root Rot by Radiation Induced Mutant, Bacillus lentimorbus WJ5a17 (방사선유도 돌연변이체 Bacillus lentimorbus WJ5a17에 의한 Pythium Root rot의 방제)

  • To control pythium root rot, Bacillus lentimorbus WJ5a17 with high anti-fungal activity against Pythium ultimum was induced from B. lentimorbus WJ5 by gamma radiation ($^{60}Co$). The biocontrollers of FWJ5 and FWJ5a17 were formulated ($1.0\times 10^{11}$) with B. lentimorbus WJ5 and WJ5a17, respectively, The population density of FWJ5 and FWJ5a17 maintained highly up to $1.0\times 10^{9}$ CFU $g^{-1}$ in nursery and field soils until 30 days after treatment. P. ultimum spores germination were inhibited 71.0% and 81.4% by FWJ5 and FWJ5a17, respectively. Pythium root rot of yea pepper, Chinese cabbage and radish were significantly (p < 0.05) controled by one time treatment of FWJ5 and FWJ5a17.

Biological Control of Rice Bakanae by an Endophytic Bacillus oryzicola YC7007

  • In our previous study, we reported that a novel endophytic bacterium Bacillus oryzicola YC7007 has suppressed bacterial diseases of rice via induced systemic resistance and antibiotic production. This endophytic strain, B. oryzicola YC7007 was used as a biological control agent against bakanae disease of rice caused by Fusarium fujikuroi, and its mechanism of interaction with the pathogen and the rice was further elucidated. Root drenching with B. oryzicola YC7007 suspension reduced the disease severity of bakanae significantly when compared with the untreated controls. The treatments of B. oryzicola YC7007 suspension ($2.0{\times}10^7cfu/ml$) to the rice rhizosphere reduced bakanae severity by 46-78% in pots and nursery box tests containing autoclaved and non-autoclaved soils. Moreover, in the detached rice leaves bioassay, the development of necrotic lesion and mycelial expansion of F. fujikuroi were inhibited significantly by spraying the culture filtrate of B. oryzicola YC7007. Drenching of ethyl acetate extracts of the culture filtrate to the rhizosphere of rice seedlings also reduced the bakanae disease severity in the plant culture dish tests. With the root drenching of B. oryzicola YC7007 suspension, the accumulation of hydrogen peroxide was observed at an early stage of rice seedlings, and a hormonal defense was elicited with and without pathogen inoculation. Our results showed that the strain B. oryzicola YC7007 had a good biocontrol activity against the bakanae disease of rice by direct inhibition, and was also capable of inducing systemic resistance against the pathogen via primed induction of the jasmonic acid pathway.

Effect of Artificial Soils and Aqueous Solutions for Plantlet Acclimatization of Somatic Embryos of Aralia elata (두릅나무 체세포배 유래 소식물체의 순화에 미치는 배양토 및 공급액의 효과)

  • In order to develop effective acclimatization methods for Aralia elata plantlets regenerated from somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. The plantlets were transplanted into plastic boxes containing artificial soil in the presence of either several levels of MS liquid media, distilled water, 2% sucrose or 0.1% hyponex solution. They were then cultured by spraying of distilled water twice a week and maintained in the normal tissue culture room. Perlite was proved to be better than vermiculite on survival rate and growth of the plantlets. As the size of perlite (larger than 0.2 cm in diameter) increased, both the survival rate and growth of the plantlets improved. Among the various MS liquid media and different aqueous solutions tested, distilled water appeared to result in the best survival rate and growth. MS media were also effective in increasing survival rate and supporting growth when diluted to 1/4 and/or 1/8. The acclimatized plantlets could be transplanted directly onto the nursery bed and grown normally. The above results suggest that plantlets regenerated from somatic embryos of Aralia elata be effectively acclimatized using a plastic box containing perlite with distilled water treatment.

Resistance Analysis of Cultivars and Occurrence Survey of Fusarium Wilt on Strawberry (국내 딸기 시들음병 발생실태와 품종별 저항성 분석)

  • The occurrence of Fusarium wilt in strawberry fields in Korea was assessed from 2001 to 2003. Fusarium wilt was found from June to August in nursery beds, from September to October after planting in production beds, and from January to March during harvesting period. The symptoms were root rots, discolored vascular tissue in the crown and deformation and yellowing of central leaflets. The disease occurred in up to 30% of plants in 37 of 214 fields surveyed. Fusarium wilt occurred from cvs. ‘Dochiodome’, ‘Maehyang’, ‘Redpearl’, ‘Samaberry ’ and ‘Akihime’ and more severe from cvs. ‘Samaberry’, ‘Maehyang’ and ‘Dochiodome’. Infested soils had high salt concentrations, high nitrogen, phosphate concentrations and low pH. The results of pathogenicity test showed that Fo47 and Fo79 isolated from cvs. ‘Dochiodome’ and ‘Samaberry’ were strong pathogenic to all of four cultivars, and ‘Dochiodome’, ‘Redpearl’, ‘Maehyang’ were relatively susceptible to the all isolates.

Effects of Simulated Acid Rain on Chemical Properties of the Experimental Soil of pinus densiflora S. et Z. and Forsythia Koreana Nak. Seedlings (人工酸性雨가 소나무 및 개나리 盆植苗土壤의 化學的 性質에 미치는 影響)

  • For the purpose of examining the effects of simulated acid rain on the chemical properties of the experimental soil in Pinus densiflora seedlings and Forsythia koreana rooted cuttings, the experimental design of randomized block arrangement with three replications was implemented in the experimental field of Yesan National Agricultural Junior College. One-year-old Pinus densiflora seedlings and Forsythia koreana cuttings were planted in the pots filled the mixed soils (nursery soil: forest soil of siliceous sandy loam = 1 : V/V)in the early spring of 1985. The regime of artificil acid rain, in terms of spray frequency per monthly and spray amount at single treatment per plot, was simulated on the basis of climatological data averaged from 30 years records. Simulated acid rain (pH 2.0, pH4.0, and pH 5.5 as control) containing sulfuric and nitric acid in the ratio of 3:2 (chemical equivalant basis) diluted ground water, were treated on the experimental plants under condition of cutting off the natural precipitation with vinyl tunnel, during the growing season (May 1 to August 31) in 1985. THe results obtained in this study were as follows; 1. Soil acidity was dropped, and exchangeable aluminum contents in the soil was dramatically increased in both species, with decreasing pH levels of acid rain. 2. Exchangeable potassium, clacium, magnesium contents, and base saturation degree of the soil were highly drcreased in two species as the pH levels of acid rain decreased. 3. In two species, sulfate concentrations in the soil were decreased of pH 4.0 treatment, and remarkably increased at pH 2.0 treatment of acid rain in comparison with control. 4. Total nitrogen and available phosphate contents of the soil were not affected by acid rain treatment in the both species, and Fe contents at pH 2.0 treatment were highest among three acid rain treatments.

  • Half-sib seedlings of Ginkgo biloba (one-year-old) were treated with various simulated acid rains(pH2.0, pH3.0, pH4.0 and pH5.0) to examine the effects of acid rain on leaf surface area, leaf injury, leaf chlorophyll content and photosynthetic ability of the leaf tissue. The seedlings were grown in a pot($4500cm^3$) containing one of three different soils(nurseryy soil, mixed soil and sandy soil). Simulated acid rain was made by diluting sulfuric and nitric acid solution($H_2SO_4:HNO_3=3:1$, V/V) with tap water and tap water(pH6.4), and treated by 5mm each time for three minutes during the growing seasons(April to October 1985). Acid rain treatments were done three times per week to potted seedlings by spraying the solutions. The results obtained in this study were as follows : 1. Leaf surface area per seedling at pH2.0 level was the lowest among the levels of pH, but those at other pH levels were not significantly different. 2. Leaf injury(injured leaf rate and injured leaf area) increased with decreasing pH levels of acid rain. 3. Leaf chlorophyll content measured during the period June through October was significantly different among the soil types, and that of the seedling in nursery soil was the highest. The lower pH levels of simulated acid rain was treated ; more leaf chlorophyll content was measured at the beginning of treatment, and the more it severely decreased at the late growing period. 4. Photosyntetic abilities, and the highest value was shown in nursery soil. Significand difference in photosynthetic ability among the levels of pH was observed only in August. Photosynthetic ability increased with decreasing pH levels at the beginning of treatment, but decreased rapidly after July.

Nematodes Associated with Forest with Forest Trees in Korea I. Studies on Nematodes Associated with Saplings (한국에 있어서 임목에 기생하는 선충조사 I. 묘목기생선충에 관한 연구)

  • The results of survey on the nematodes associated with sapling from 8 Forestry Experiment Stations(F.E.S.) in Korea are reported herein. A total of 35 species under 20 genera, 11 families, and 2 orders were identified from the soils around the roots of 119 tree species and amongst them, Xiphinema insigne is unrecorded species for Korea and Geocenamus sp. is a new species. Tylenchorhynchus claytoni, which showed the highest detection rate (D.R.) from all F.E.S., except Nambu F.E.S, was the dominant species among the nematodes associated with saplings. The D.R. of T. claytoni was the highest in Ch’ungnam F.E.S. by 100% from 15 saplings sampled, followed by 87.5% in Kangwon, 66.6% in Kyongbuk, 58.3% in Cheonbuk, 48.1% in Ch’ungbuk, 41.3% in Kyongki, and 40% in Cheonnam F.E.S. Other important nematode species which showed very high densities by stations are; Criconemella morgensis (D.R.: 37.5%), Helicotylenchus digonicus (D.R. : 35%), Pararotylenchus pini (D.R.: 37%), Tylenchorhynchus nudus (D.R.: 37.9%), Criconemella informis (D.R.: 27.5%), and Meloidogyne sp. (D.R.: 12.5), from Chunbuk, Chunnam, Ch’ungbuk, Kyongki, and Kangwon F.E.S., respectively.

Growth of One-Year-Old Pot-Cultivated 'Fuji'/M.9 Apple Trees under Different Concentrations of Nitrogen Fertilization (질소시비농도에 따른 1년생 사과 'Fuji'/M.9 포트묘목의 수체 생장)

  • The study was carried out to investigate growth of 48.6-L pot-cultivated 1-year-old 'Fuji'/M.9 apple trees depending on different levels of nitrogen concentration. While rise in tree growth was paralleled with increase of nitrogen concentration, more than 32 mM of nitrogen rather restrained tree growth. In particular, growth of 16 mM of nitrogen treated trees was satisfied with criteria for production of high-quality pot-cultivated nursery stocks. Although mineral contents of leaves were higher in 8 and 16 mM nitrogen treatments than commonly recommended mineral contents in apple orchards, such somewhat surplus minerals could be helpful for tree growth after transplanting to apple orchards. In addition, our result indicated that soils of 8 and 16 mM of nitrogen treated pots met appropriate criteria for soil chemical property of apple orchards. Thus, in the light of tree growth, mineral contents of leaves, and soil chemical property in the pots, 16 mM of nitrogen treatment is considered to be suitable for production of 1-year-old 'Fuji'/M.9 apple potted trees.

Study on the Application of Miwon Organic Fertilizer (Byproduct of Amino Acid Fermentation) to the Ginseng Cultivation I.   • In order to find out the application effect of commercial organic fertilizer on the germination rate of ginseng seed, growth of ginseng plant and changes of the physicochemical properties of soils, a Miwon organic fertilizer, byproduct of amino acid fermentation, was applied to the ginseng nursery bed. The application rates of Miwon organic fertilizer were 0kg-N from organic fertilizer and 30kg-N from wild grass compost per 10a as control(namely 0kg-N/10a), 2.5kg-N and 27.5kg-N/ 10a(2.5kg-N/10a) 5.0kg-N and 25kg-N/10a(5.0kg-N/10a), 10kg-N and 20kg-N/10a(10kg-N/10a), 15kg-N and 15kg-N/10a(15kg-N/10a) respectively, The obtained results are as follows: 1. The germination rate of ginseng seed were better in the plots that received 2.5kg, 5.0kg and 10kg-N/10a as compared with control plot. However no difference was found on germination rate between control and 15kg-N/10a, 2. The higher rate of missing plant was found in the 15kg-N/10a plot that showed poor growth. However control and 10kg-N/10a plot showed similar rate of missing plant. The best result, lower rate of missing plant, was obtained in the order of 5kg-N and 2.5kg-N/10a respectively. 3. The significant heavier ginseng root weight was obtained in 5kg-N/10a and 10kg-N/10a plot. However the root weight was decreased in the order of 2.5kg-N/10a, control, 15kg-N/10a. 4. In the physicochemical changes of soils between control and organic fertilizer treated soil before and after harvesting the increment of pH, C.E.C. and organic matter content along with the rate of organic fertilizer application were found. There were significant correlation between nitrate nitrogen and the rate of organic fertilizer application. 5. In the view points of germination rate, missing rate of plant and yield of ginseng root, it can be stated that the best result could be obtained from the plot that received 10kg-N/10a from Miwon commercial organic fertilizer and 10kg-N/10a from wild grass compost.
