Biological Control of Pythium Root Rot by Radiation Induced Mutant, Bacillus lentimorbus WJ5a17

Abstract

To control pythium root rot, Bacillus lentimorbus WJ5a17 with high anti-fungal activity against Pythium ultimum was induced from B. lentimorbus WJ5 by gamma radiation ($^{60}Co$). The biocontrollers of FWJ5 and FWJ5a17 were formulated ($1.0\times 10^{11}$) with B. lentimorbus WJ5 and WJ5a17, respectively, The population density of FWJ5 and FWJ5a17 maintained highly up to $1.0\times 10^{9}$ CFU $g^{-1}$ in nursery and field soils until 30 days after treatment. P. ultimum spores germination were inhibited 71.0% and 81.4% by FWJ5 and FWJ5a17, respectively. Pythium root rot of yea pepper, Chinese cabbage and radish were significantly (p < 0.05) controled by one time treatment of FWJ5 and FWJ5a17.

Pythium ultimum에 의한 뿌리썩음병을 생물학적으로 방제하기 위하여 이에 대한 항진균 활성이 있는 세균 Bacillus lentimorbus WJ5를 방사선 ($^{60}Co$)을 조사하여 보다 더 강한 항진균 활성을 가지는 돌연변이체 B. lentimorbus WJ5a17을 유도하였다. B. lentimorbus WJ5와 WJ5a17을 각각 활용한 미생물제제 FWJ5와 FWJ5a17을 조제하였으며 균체수는 $1.0\times 10^{11}$ CFU $g^{-1}$ 이였다. 미생물제제는 저장 30일째에도 항진균 세균이 $50.\times 10^{10}$ $g^{-1}$ 이상 생존하였고, 상토와 밭 토양에서는 $1.0\times 10^{7}$ CFU $g^{-1}$이상의 생존세포수를 유지하였다. 또한 P. ultimum의 포자발아를 B. lentimorbus WJ5 제제 (FWJ5)가 71.0% 억제하였고, B. lentimorbus WJ5a17 제제(FWJ5a17)는 81.4%억제하였다. 미생물제제인 FWJ5와 FWJ5a17은 고추, 배추와 무의 종자 발아와 초기생장에 유해한 영향을 주지 않을 뿐만 아니라, 작물 생육초기와 중기에 1회 처리하였을 경우에도 P ultimum에 의한 뿌리썩음병징의 발생을 유의하게 (p<0.05) 억제하였기에 매우 효과적인 생물학적 제어제임을 알 수 있었다.
