Antioxidative Activity of Extracts from Sambucus williamsii var. coreana (딱총나무(Sambucus williamsii var. coreana) 추출물의 항산화효과)

  • Sambucus williamsii var. coreana have been used as a traditional medical food. This research was conducted to investigate the antioxidants of S. williamsii var. coreana leave and stem extracts. Total phenolic content of S. williamsii var. coreana leaves and stem water extracts were 6.6 and 2.0 mg/mL. The EDA by DPPH free radical scavenging test of S. williamsii var. coreana leaves extracts were 99.5 and 89.7% in water and ethanol extracts contained phenolic 200 ${\mu}g/mL$. The stem extracts were 92.2 and 94.3% in water and ethanol extracts contained phenolic 200 ${\mu}g/mL$. The ABTS radical decolorization activity of water and ethanol extracts from leaves were 79.8 and 99.1% at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 90.8 and 97.2% at phenolic 200 ${\mu}g/mL$. The antioxidant protection factor of water and ethanol extracts from leaves were 1.1 PF and 1.1 PF at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 1.4 PF and 1.0 PF at phenolic 200 ${\mu}g/mL$. The TBARs of water and ethanol extracts from leaves were 88.7 and 98.1% at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 93.6 and 90.6% at phenolic 200 ${\mu}g/mL$. The antioxidative activities of extracts from S. williamsii var. coreana leaves and stem were higher than BHT as positive control. These results suggests that S. williamsii var. coreana extracts have the greatest property as a natural antioxidative source.

The Patterns of CH4 and N2O fluxes from used Litter Stockpile from Korean Native Cattle (Hanwoo) (사용한 한우 깔짚에서 배출되는 CH4 및 N2O의 배출 특성)

  • This study was conducted to measure methane ($CH_4$) and nitrous oxide ($N_2O$) emissions from the 6 month old litter stockpile used for korean native cattle (Hanwoo) from August 3, 2007 to October 4, 2007. Daily mean $CH_4$ emissions was peaked to 273.013 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}1.047{\mu}g\;m^{-2}\;s^{-1}$) on first day and then gradually decreased to 2.309 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.061{\mu}g\;m^{-2}\;s^{-1}$) at the end of this experiment. Daily mean $N_2O$ emissions was as little as 0.269 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.018{\mu}g\;m^{-2}\;s^{-1}$) on first day, but exponentially increased up to 3.569 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.454{\mu}g\;m^{-2}\;s^{-1}$) on 43rd day and then slowly decreased to 1.888 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.012{\mu}g\;m^{-2}\;s^{-1}$) at the end of this experiment. Carbon dioxide equivalent ($CO_2$-eq), calculated by global warming potentials of $CH_4$ or $N_2O$, of $CH_4$ on first day occupied approximately 99% of sum of $CO_2$-eq of $CH_4$ and $N_2O$. Methane emissions decreased and $N_2O$ emissions increased so that $CO_2$-eq ratio of $CH_4$ to $N_2O$ was 50:50 on 34th day. The effect of $N_2O$ on the ratio was increase thereafter. The ratio of daily mean $CH_4$ and $N_2O$ emissions to daily error of the mean was calculated to find daily fluctuation of $CH_4$ and $N_2O$ emissions. The ratio of $CH_4$ was less than 1.0% till 11th day but increased to 10.9% on 57th day. The ratio of $N_2O$ (0.4%~51.0%) was higher than that of $CH_4$, showing high in early stage and then gradually decrease, which was different from the pattern of $CH_4$. The ratio of daily mean emissions to daily error of the mean was little in case of active $CH_4$ or $N_2O$ generation period, which would be caused by the temporal and spatial heterogeneity of composting process. Hence more air supply on early stage to decrease $CH_4$ generation and proper turning to reduce spatial heterogeneity are needed to mitigate greenhouse gas emissions.

Inhibitory Efficacy of Smilax china L. on Pro-collagen Type-1 Activity and MMP-1 Gene Expression in Fibroblasts (CCD-986sk) (섬유아세포(CCD-986sk)에서 청미래덩굴의 pro-collagen type-1 활성 및 MMP-1의 유전자 발현 저해능)

  • The purpose of this study was to research the biological activity of ethanol extract from Smilax china L. which is a vine shrub belonging to the lily family. For antiwrinkle effects, elastase inhibition effect of ethanol and water extracts from S. china L. showed 41.1% and 16.3% at $1,000{\mu}g/ml$ concentration. The collagenase inhibition effect of ethanol and water extracts from S. china L. showed more than 96.6% and 60.0% at $1,000{\mu}g/ml$ concentration. As a result of having fibroblast measured cell viability on fibroblast cell of ethanol extract from S. china L., it showed 71.7% with cell viability at $100{\mu}g/ml$ concentration. At $50{\mu}g/ml$ concentration, the procollagen biosynthesis effect of ethanol extract from S. china L. was 139.86%. At the same concentration, the matrix metalloprotease (MMP)-1 inhibition effect of the ethanol extract was 74.9%. According to the results of Western blot of ethanol extract from S. china L., the expression of the MMP-1 protein was decreased by 35% at $50{\mu}g/ml$ concentration. Reverse transcription-polymerase chain reaction (PCR) of ethanol extract from S. china L. showed that the expression of MMP-1 mRNA was decreased by 45% at $50{\mu}g/ml$ concentration. The findings suggest that 70% ethanol extract from S. china L. (SC) has great potential as a cosmeceutical ingredient with antiwrinkle effects.

  • This paper describes the combination surge generator for applying performance tests of surge protective devices. The $8/20{\mu}s$ waveform applies to low impedance circuits and components and is commonly used to determine the characteristics of surge protective devices. And the $1.2/50{\mu}s$ waveform applies to high impedance circuits and components and is used for testing dielectric behavior. Therefore, the combination surge generator, which generates $1.2/50{\mu}s$ voltage waveform under open-circuit conditions and $8/20{\mu}s$ current waveform under short-circuit condition, was proposed. Also this generator can produce $10/1000{\mu}s$ as well as $0.5{\mu}s/100kHz$ ring waveform.

Survival Strategy of Dominant Diatom Chaetoceros debilis and Leptocylindrus danicus as Southwestern parts of East Sea - The availability of Dissolved Organic Nitrogen under Dissolved Inorganic Nitrogen-limited Environments (동해 남서해역에서 우점 규조류 Chaetoceros debilis와 Leptocylindrus danicus의 생존전략 - 용존 무기 질소 제한 환경에서 용존 유기 질소의 이용가능성)

  • The bioavailability of dissolved organic nitrogen (DON) by dominant species Chaetoceros debilis and Leptocylindrus danicus under dissolved inorganic nitrogen (DIN)-limited condition in the southwestern East Sea was conducted to assess the quantitative evaluation using growth kinetic experiment. Nitrogen sources were nitrate and ammonium as DIN, glycine and urea, which is portion component of DON in East Sea. Maximum specific growth rate (${\mu}_{max}$) and half-saturation constant ($K_s$) of C. debilis calculated from Monod equations were estimated to be $1.50day^{-1}$ and $1.62{\mu}M$ in nitrate, $1.13day^{-1}$ and $6.97{\mu}M$ in ammonium, $1.46day^{-1}$ and $3.36{\mu}M$ in glycine, $0.93day^{-1}$ and $0.55{\mu}M$ in urea, respectively. Also, L. danics was estimated to be $1.55day^{-1}$ and $5.21{\mu}M$ in nitrate, $1.57day^{-1}$ and $4.57{\mu}M$ in ammonium, $1.47day^{-1}$ and $3.80{\mu}M$ in glycine, $1.42day^{-1}$ and $1.94{\mu}M$ in urea, respectively. Both C. debilis and L. dancius have higher affinity of urea than DIN. The high affinity of urea was indicated that the dominant species were able to growth using urea under DIN-limited conditions. Thus, DON utilization of phytoplankton may be one of the important dominant strategy under DIN-limited environments such as southwestern East Sea.

  • A new parameter W/R for the class I test have been introduced in the IEC 61643-1 published in 2005. One of the possible test impulses which meets the parameters for class I test is the 10/350 [${\mu}s$] waveshape proposed in IEC61312-1. 10/350 [${\mu}s$] waveshape meets parameters for class I test in the IEC 61643-1. The impulse generator for making the 10/350 [${\mu}s$] waveshape is consist of IVG, ICG, crowbar coil and crowbar switch. The electric change Q and the specific energy W/R were measured and calculated using the exclusive use measuring software according to the formula of IEC 611643-1. We verified the correspondence between measured results using impulse generator and recommended parameters by IEC 61643-1.

  • This study was made on the physiological reactions of photosynthesis. stomatal transpiration. stomatal conductance of Acanthopanax senticosus of leaves. The results obtained are as follows 1. The light compensation points were approximately $30{\mu}molm^{-2}S^{-1}$ in sun leaves and $15{\mu}molm^{-2}S^{-1}$ in shade leaves. 2. The light saturation points were approximately $1,000{\mu}molm^{-2}S^{-1}$ in sun leaves and $300{\mu}molm^{-2}S^{-1}$ in shade lea ves. 3. There was no significant between various mountains distributed. net photosynthetic rates were approximately 8.0 to $8.8{\mu}molm^{-2}S^{-1}$ in upper leaves. However. net photosynthetic rate in upper leaves of Acanthopanax koreanum in Jeju island was slightly low about $6.9{\mu}molm^{-2}S^{-1}$. 4. Net photosynthetic rate in middle leaves of all mountains was ranged from 40% to 65% of upper leaves. and that in lower leaves was approximately 30% of upper leaves. But the net photosynthesis in lower leaves of Acanthopanax koreanum in Jeju island was slightly high about 71 % of upper leaves. 5. There was no significant between various mountains distributed, stomatal transpirations were ranged from 1.1 to $1.4mmolH_2Om^{-2}S^{-1}$ in upper leaves, 0.7 to $1.0mmolH_2Om^{-2}S^{-1}$ in middle leaves, and 0.5 to $0.6mmolH_2Om^{-2}S^{-1}$ in lower leaves. respectively. 6. There was no significant between various mountains distributed. the stomatal conductance in upper 1eaves was ranged from 70 to $90mmolH_2Om^{-2}S^{-1}$. However, stomatal conductance in upper leaves of Acanthopanax koreanum in Jeju island was approximately $380mmolH_2Om^{-2}S^{-1}$, its remarkedly higher than any other mountain.

  • This study was carried out to know the effect of light intensity, temperature and $CO_2$ concentration on photosynthesis and transpiration in yacon(Polymnia sonchifolia Poepp.& Endl.). Light compensation point was ${58\;{\mu}mol\;m^{-2}\;s^{-1}}$and light saturation point was ${1708\;{\mu}mol\;m^{-2}\;s^{-1}}$. Transpiration rate was increased to about 4 mmol${m^{-2}\;s^{-1}}$ with increasing of light intensity to ${2193\;{\mu}mol\;m^{-2}\;s^{-1}}$. The optimum temperature for photosynthesis was ${24^{\circ}C}$ in air. Photosynthesis was gradually reduced as transpiration rate increased from 4 to 8 mmol ${m^{-2}\;s^{-1}}$ in different air temperature. $CO_2$ compensation point was 63 vpm and $CO_2$ saturation point was 1155 vpm and light saturation point was enhanced with increasing of $CO_2$ concentration from 350 vpm to 1300 vpm.

Regeneration and the Maturation Induction of Free-Living Gametophytes of a Kelp Saccharina sculpera (Phaeophyceae) (갈조류 개다시마 유리배우체의 재생 및 성숙 유도)

  • Investigation of optimal temperature, irradiance and photoperiod conditions for free-living gametophytes of Saccharina sculpera for natural resources conservation and mass cultivation of endangered species in the eastern coast of Korea. Induction of regeneration and maturation of the free-living gametophytes of S. sculpera were cultured at temperatures (5, 10, 15, 20 and $25^{\circ}C$), irradiance (10, 20, 40, 60 and $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and photoperiods (14:10, 12:12 and 10:14 h L:D). The female gametophyte were actively regenerated without reaching sexual maturity under $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D) conditions. In contrast, the conditions for male gametophytes were slightly different at $15^{\circ}C$, $10{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D). The sexual maturation of female and male gametophytes took place under $15^{\circ}C$, $40{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 14:10 h (L:D) and $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 10:14 h (L:D), respectively. These results provide basic information for controlling the regeneration and maturation of free-living gametophytes for conservation and utilization of S. sculpera.

  • Quantitative assessment of edge blunting of saw-teeth was carried out by TALYSURF. 1. Using the following equation, the real shape of a saw-tooth can be traced on the graph of TALYSURF. ${\frac{{\Delta}h}{h}}={\frac{V{\Delta}_x}{V_x}}$ {${\Delta}h$: vertical distance of stylus h: vertical distance in chart $V{\Delta}_x$: Velocity of stylus $V_x$: velocity of chart} 2. As shown on Fig 2, the error from stylus itself can be calculated by following equation. i) 13.8${\mu}{\leqq}$x<20.4${\mu}$ y=-0.2246x+4.59${\mu}$ ii) 0${\leqq}$x<13.8${\mu}$ y=${\sqrt{(-18{\mu})^2-x^2}}-1.42x+32.7{\mu}}$ 3.
