A thermophilic Bacillus sp. strain KK-1 isolated from soil produced an extracellular xylanase. From the culture supernatant of Bacillus sp., the xylanase was purified to homogeneity by ammonium sulfate precipitation and DEAE-Sephadex A-50 chromatography. The molecular weight of the purified xylanase was estimated to be 45 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel permeation chromatography. The apparent $K_m$ values for xylanase, using oat spelt xylan and birchwood xylan as substrates, were 7.1 mg/ml and 3.2 mg/ml, and $V_{max}$ values were $27.0\;{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$ and $29.0\;{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$, respectively. The xylanase hydrolyzed oat spelt xylan to mostly xylobiose, xylotriose, and xylose. The amino acid composition indicated that the xylanase contained high amounts of amino add residues of glutamic acid and glutamine (Glx) and aspartic acid and asparagine (Asx).
As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.
An investigation was conducted to elucidate the effect of sucrose, germicides and abscisic acid (ABA) in the preservatives on postharvest quality, such as fresh weight, solution uptake, vase life, diameter and thickness of flower, and chlorolhyll of leaf in cut chrysanthemum 'Kyoungsubang`. Fresh weight of cut chrysanthemum was highest in 250mg/L aluminum sulfate [{TEX}$Al_{2}(SO_{4})_{3}${/TEX}]+3% sucrose and 250 mg/L 8-hydroxyquinoline sulfate (8-HQS)+ 3% sucrose. Addition of sucrose and germicides inhibited microorganisms growth in solution, and apparently promoted solution uptake. The increased fresh weight seems to attribute to the elevated uptake. Germicide 8-HQS was more effective than {TEX}$Al_{2}(SO_{4})_{3}${/TEX} in the aspect of solution uptake and fresh weight. Treatment of 250 mg/L 8-HQS +3% sucrose showed the longest vase life (24.67±2.52 days) and the lowest rate of leaf chlorosis (or senescence). Addition of {TEX}$Al_{2}(SO_{4})_{3}${/TEX} and sucrose accelerated leaf chlorosis and increased diameter of flower, respectively. Sucrose treatment prolonged the vase life of flowers. Except control and 250 mg/L 8-HQS, content of chlorophyll was rapidly decreased after 6 days at upper, middle and lower leaves in order.

Arabidopsis AHL gene contains 4 exons encoding a putative protein highly homologous to the yeast salt-sensitive enzyme HAL2, a 3'(2'),5'-bisphosphate nucleotidase involving in reductive sulfate assimilation. AHL cDNA complemented yeast met22 (hal2) mutant. AHL fusion protein expressed in E. coli exhibited $Mg^{2+}$-dependent, 3'-phosphoadenosine 5'-phosphate (PAP)-specific phosphatase activity. $Li^+,\;Na^+,\;K^+$ and $Ca^{2+}$ ions inhibit the enzyme activity by competing with $Mg^{2+}$ for the active site of the enzyme. The enzyme activity was also sensitive to ${\mu}M$ concentrations of toxic heavy metal ions such as $Cd^{2+},\;Cu^{2+}$ and $Zn^{2+}$, but was not recovered by addition of more $Mg^{2+}$ ions, suggesting that these ions inactivate the enzyme with a mechanism other than competition with $Mg^{2+}$ ions. Inhibition of the AHL enzyme activity may result in accumulation of PAP, which is highly toxic to the cell. Thus, the AHL enzyme could be one of the intial targets of heavy metal toxicity in plants.

The antioxidant activity of Bacillus polyfermenticus SCD was studied by partially purified culture extracts using various methods: ammonium sulfate precipitation, adsorption to Diaion HP-20 columns using polar solvents, and extraction using non-polar solvents. The 1,1-diphenyl-2-picyrylhydrazyl (DPPH) radical scavenging activity of these partially purified fractions was then investigated. The precipitate isolated using 75%-saturated ammonium sulfate was shown to contain about 77.2% DPPH radical scavenging activity. Using the Diaion HP-20 resin adsorption method, the fraction obtained using 60% ethanol and 60% methanol possessed 76.7 and 89.5% DPPH radical scavenging activity, respectively. Fractions obtained by extracting with the non-polar solvents 80 mg/mL chloroform, 80 mg/mL n-hexane, 80 mg/mL ethyl acetate, and 80 mg/mL butanol contained 68.4, 75.0, 70.7, and 87.5% DPPH radical scavenging activity, respectively. Further study is needed to characterize the antioxidant substance(s) released by B. polyfermenticus SCD cultures.

The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and combination with herbal medicine on dextran sodium sulfate (DSS)-induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n = 9): group I-normal group, group II-DSS control group, group III-DSS + sulfasalazine (30 mg/kg), group IV-DSS + sulfasalazine (60 mg/kg), group V-DSS + sulfasalazine (30 mg/kg) + Radish Extract mixture (30 mg /kg) (SRE). DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. SRE supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SRE treatment significantly reduced the expression of pro-inflammatory signaling moleculesthrough suppression both mitogen-activated protein kinases(MAPK) and nuclear factor-kappa B ($NF-{\kappa}B$) signaling pathways, and prevented the apoptosis of colon. Moreover, SRE administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase. This is the first report that Radish extract mixture combined with sulfasalazine protects against experimental UC via the inhibition of both inflammation and apoptosis, very similar to the standard-of-care sulfasalazine.
Chromium(III) recovery from tanning wastewater by means of adsorption on activated carbon and elution with sulfuric acid was studied. Tests were carried out at laboratory scale on an effluent of industrial origin. Initially, proteinaceous materials and fats were separated via sieving followed by ultrafiltration. The chemical composition of the sample thus precleansed was (in g/L): 1.09 chromium(III); 10.36 sulfate; 11.10 sodium; 9.57 chloride; 0.40 proteinaceous materials; and 0.20 fats. Adsorptions were made at 20, 30, and $40^{\circ}C$, establishing what temperature favored chromium(III) uptake. At $40^{\circ}C$, the maximum cation fixation was 40.2 mg/g, and the lowest content in an equilibrium solution was 3.9 mg/L. As regards sodium, chloride, and sulfate, the concentrations before and after the treatment were similar. Likewise, it was found that protons were also retained, modifying the pH of the liquid medium. Adsorption isotherms were analyzed using the Langmuir, Temkin, and Freundlich models. Finally, the extraction of the adsorbed tanning agent with sulfuric acid was evaluated. A recovery of 96.5% was achieved with 0.9 N at $70^{\circ}C$ (13.23 g/L $Cr^{3+}$; 42.98 g/L sulfate; and 0.40 g/L NaCl).
During the period from April to September 2002, the size distributions of ambient aerosol were measured at the coastal site at Hamduk in Jeju Island. Na$\^$+/, K$\^$+/, Mg$\^$2+/, Ca$\^$2+/and Cl$\^$-/ exhibited mostly a bimodal coarse mode size distribution, while ammonium and sulfate were mainly in the fine size range, with maximum at around 0.54$\mu\textrm{m}$. The average molar concentration ratio of ammonium to sulfate for fine particles was equal to 2.0${\pm}$0.9. Nitrate was evenly found in both the coarse and fine modes. Elements like Al, Fe, Cu, Mg, Na, Ti, Sr and Mn were dominant in coarse particles, with the maximum at around 5.25$\mu\textrm{m}$. S and Pb were mainly in the submicrometer size range. Other elements with a fine and coarse modes were V, Ni, Cu, Ba and Mo. The patterns of the size distributions of trace elements measured at the downtown in Jeju City were very similar to those at the coastal site in Hamduk. However, the amplitude of size fractional concentrations at Jeju City was narrower than that at Hamduk. While the mass median diameters for the chemical species originated from the natural origin such as marine and crust were relatively large, those for ammonium, sulfate, S and Pb were very small.
Acid mine drainage (AMD) resulting from pyrite oxidation in mining areas, subsequently leads to soil acidification accompanied by lowering pH and high concentration of metals and metalloids in its surrounding environment. Regarding to this, the microbial amelioration has been considered as a promising option for a more cost-effective and eco-friendlier countermeasure, compared to the use of alkaline chemicals. This study was aimed to evaluate influencing factors in microbially-mediated amelioration of acidic soil spiked by simulated AMD. For this, microcosm experiments were conducted by acid-neutralizing bacterial consortium (dominated by Klebsiella sp. and Raoultella sp.) under the various conditions of AMD spikes (0-2,500 mg SO42-/L), together with acidic mine soil (0-100 g) or sphagnum peat (0-5 g) in the 200 mL of nutrient medium. The employed bacterial consortium, capable of resisting to high level of sulfate concentration (up to 1,500 mg SO42-/L) in low pH, generated the ammonium while concomitantly reduced the sulfate, subsequently contributing to the effective soil stabilization with an evolution of soil pH up to neutral. Furthermore, it demonstrates that suitable condition has to be tuned for successful microbial metabolism to facilitate with neutralization during practical application.
