Construction of Candida antarctica Lipase B Expression System in E. coli Coexpressing Chaperones (대장균에서의 Chaperone 동시 발현을 통한 Candida antarctica Lipase B 발현 시스템 구축)

  • Recently, Candida antarctica lipase B (CalB) draws attention from industries for various applications for food, detergent, fine chemical, and biodiesel, because of its characteristics as an efficient biocatalyst. Since many industrial processes carry out in organic solvent and at high temperature, CalB, which is stable under harsh condition, is in demand from many industries. In order to reform CalB promptly, the expression system which has advantages of ease to use and low cost for gene libraries screening was developed using E. coli. The E. coli strains, Rosettagami with competence for enhanced disulfide bond formation, Novablue, and $DH5{\alpha}$, were exploited in this study. To obtain the soluble CalB, the pCold I vector expressing the cloned gene at $15^{\circ}C$ and the chaperone plasmids containing groES/groEL, groES/groEL/tig, tig, dnaK/dnaJ/grpE, and dnaK/dnaJ/grpE/groES/groEL were used for coexpression of CalB and chaperones. The colonies expressing functional lipase were selected by employing the halo plate containing 1% tributyrin, and the CalB expression was confirmed by SDS-PAGE. E. coli Rosettagami and $DH5{\alpha}$ harbouring groES/groEL chaperones were able to express soluble CalB effectively. From a facilitative point of view, E. coli $DH5{\alpha}$ is more suitable for further mutation study.

  • Asbestos or its applications have been used for long times and for various purposes in our life because of their merits, namely fire resistance, electric insulation and chemical resistance capacity etc. Despite of theses many merits, one of the problems of asbestos is shown toxicity according to its fiber type. So we need data to solve about this problem. In this paper, we study on the technical method of asbestos waste treatment and on the trends of asbestos researches and developments by the analysis of its patents and DWPI database materials. As a result, the asbestos-waste treatment data in the its related patents is used 267 cases to analyze. These data are divided into 86(32.5%) cases of solid waste disposal(B09B). 41(16.6%) cases of separation(B01D) and 27(10.2%) of lime, magnesia, slag, cement and their composites(C04B).

Preparation of Nanostructures Using Layer-by-Layer Assembly and Applications (층상자기조립법을 이용한 나노구조체의 제조와 응용)

  • We introduce a novel and versatile approach for preparing self-assembled nanoporous multilayered films with antireflective properties. Protonated polystyrene-block-poly (4-vinylpyrine) (PS-b-P4VP) and anionic polystyrene-block-poly (acrylic acid) (PS-b-PAA) block copolymer micelles (BCM) were used as building blocks for the layer-by-layer assembly of BCM multilayer films. BCM film growth is governed by electrostatic and hydrogen-bonding interactions between the oppositely BCMs. Both film porosity and film thickness are dependent upon the charge density of the micelles, with the porosity of the film controlled by the solution pH and the molecular weight (Mw) of the constituents. PS7K-b-P4VP28K/PS2K-b-PAA8K films prepared at pH 4 (for PS7K-b-P4VP28K) and pH 6 (for PS2K-b-PAA8K) are highly nanoporous and antireflective. In contrast, PS7K-b-P4VP28K/PS2K-b-PAA8K films assembled at pH 4/4 show a relatively dense surface morphology due to the decreased charge density of PS2K-b-PAA8K. Films formed from BCMs with increased PS block and decreased hydrophilic block (P4VP or PAA) size (e.g., PS36K-b-P4VP12K/PS16K-b-PAA4K at pH 4/4) were also nanoporous. Furthermore, we demonstrate that the nanostructured electrochemical sensors based on patterning methods show the electrochemical activities. Anionic poly(styrene sulfonate) (PSS) layers were selectively and uniformly deposited onto the catalase (CAT)-coated surface using the micro-contact printing method. The pH-induced charge reversal of catalase can provide the selective deposition of consecutive PE multilayers onto patterned PSS layers by causing the electrostatic repulsion between next PE layer and catalase. Based on this patterning method, the hybrid patterned multilayers composed of platinum nanoparticles (PtNP) and catalase were prepared and then their electrochemical properties were investigated from sensing $H_2O_2$ and NO gas. This study was based on the papers reported by our group. (J. Am. Chem. Soc. 128, 9935 (2006); Adv. Mater. 19, 4364 (2007); Electro. Mater. Lett. 3, 163 (2007)).

A 3~5 GHz UWB Up-Mixer Block Using 0.18-μm CMOS Technology

  • This paper presents a direct-conversion I/Q up-mixer block, which supports $3{\sim}5$ GHz ultra-wideband(UWB) applications. It consists of a VI converter, a double-balanced mixer, a RF amplifier, and a differential-to-single signal converter. To achieve wideband characteristics over $3{\sim}5$ GHz frequency range, the double-balanced mixer adopts a shunt-peaking load. The proposed RF amplifier can suppress unwanted common-mode input signals with high linearity. The proposed direct-conversion I/Q up-mixer block is implemented using $0.18-{\mu}m$ CMOS technology. The measured results for three channels show a power gain of $-2{\sim}-9$ dB with a gain flatness of 1dB, a maximum output power level of $-7{\sim}-14.5$ dBm, and a output return loss of more than - 8.8 dB. The current consumption of the fabricated chip is 25.2 mA from a 1.8 V power supply.

Medical Image Compression using Adaptive Subband Threshold

  • Medical imaging techniques such as Magnetic Resonance Imaging (MRI), Computed Tomography (CT) and Ultrasound (US) produce a large amount of digital medical images. Hence, compression of digital images becomes essential and is very much desired in medical applications to solve both storage and transmission problems. But at the same time, an efficient image compression scheme that reduces the size of medical images without sacrificing diagnostic information is required. This paper proposes a novel threshold-based medical image compression algorithm to reduce the size of the medical image without degradation in the diagnostic information. This algorithm discusses a novel type of thresholding to maximize Compression Ratio (CR) without sacrificing diagnostic information. The compression algorithm is designed to get image with high optimum compression efficiency and also with high fidelity, especially for Peak Signal to Noise Ratio (PSNR) greater than or equal to 36 dB. This value of PSNR is chosen because it has been suggested by previous researchers that medical images, if have PSNR from 30 dB to 50 dB, will retain diagnostic information. The compression algorithm utilizes one-level wavelet decomposition with threshold-based coefficient selection.

  • In this paper, we give some results on ${\frac{zf^{\prime}(z)}{f(z)}}$ for the certain classes of holomorphic functions in the unit disc $E=\{z:{\mid}z{\mid}<1\}$ and on ${\partial}E=\{z:{\mid}z{\mid}=1\}$. For the function $f(z)=z^2+c_3z^3+c_4z^4+{\cdots}$ defined in the unit disc E such that $f(z){\in}{\mathcal{A}}_{\alpha}$, we estimate a modulus of the angular derivative of ${\frac{zf^{\prime}(z)}{f(z)}}$ function at the boundary point b with ${\frac{bf^{\prime}(b)}{f(b)}}=1+{\alpha}$. Moreover, Schwarz lemma for class ${\mathcal{A}}_{\alpha}$ is given. The sharpness of these inequalities is also proved.

  • This paper, we presents a RF protection technique of antenna switch by improving the power handling capability in worst case environment mode for mobile phone applications without critical payment of circuit performances such as insertion loss, isolation and ACBV (AC breakdown voltage). By applying a additional capacitive path located in front of the antenna in cell-phone, it performs the effective reduction of input power in high voltage standing wave ratio (VSWR) condition. Under the all-path off condition which causes a high VSWR, it achieved 37.7dBm power handling level as high as 5.7dB compared to that of conventional one at 2GHz. In addition, insertion loss and isolation performances were 0.31dB and 42.72dB at 2 GHz, respectively which were almost similar to that of the conventional circuit. The proposed antenna switch was fabricated in 130nm CMOS SOI technology.

  • In this text, we investigate some fixed point results in double-controlled cone metric spaces using several contraction mappings such as the B-contraction, the Hardy-Rogers contraction, and so on. Additionally, we prove the same fixed point results by using rational type contraction mappings, which were discussed by the authors Dass. B. K and Gupta. S. Also, a few examples are included to illustrate the results. Finally, we discuss some applications that support our main results in the field of applied mathematics.

  • Existing B2B, B2C computer systems and applications that executed business trans-actions were the client- server based architecture which consists of heterogeneous hardware and software including personal computers and mainframes. Due to the active boom of electronic business, integration and compatibility of exchanged data, applications and hardwares have emerged as hot issue. This paper designs and implements a message transport system and a document transformation system in order to solve the interoperability problem of integrated logistics system in e-Business when doing electronic business. Message transport system integrated ebMS 2.0 which is standard business message exchange format of ebXML, the international standard electronic commerce framework, and JMS of J2EE enable to ensure reliable messaging. The document transformation system could convert non-standard XML documents into standard XML documents and provide the web services after integrating message system. Using suggested business scenario and various test data, our message oriented system preyed to be interoperable and stable. We participated ebXML messaging interoperability test organized by ebXML Asia Committee ITG in oder to evaluate and certify the suitability for message system.

A chord error conforming tool path B-spline fitting method for NC machining based on energy minimization and LSPIA

  • Piecewise linear (G01-based) tool paths generated by CAM systems lack $G_1$ and $G_2$ continuity. The discontinuity causes vibration and unnecessary hesitation during machining. To ensure efficient high-speed machining, a method to improve the continuity of the tool paths is required, such as B-spline fitting that approximates G01 paths with B-spline curves. Conventional B-spline fitting approaches cannot be directly used for tool path B-spline fitting, because they have shortages such as numerical instability, lack of chord error constraint, and lack of assurance of a usable result. Progressive and Iterative Approximation for Least Squares (LSPIA) is an efficient method for data fitting that solves the numerical instability problem. However, it does not consider chord errors and needs more work to ensure ironclad results for commercial applications. In this paper, we use LSPIA method incorporating Energy term (ELSPIA) to avoid the numerical instability, and lower chord errors by using stretching energy term. We implement several algorithm improvements, including (1) an improved technique for initial control point determination over Dominant Point Method, (2) an algorithm that updates foot point parameters as needed, (3) analysis of the degrees of freedom of control points to insert new control points only when needed, (4) chord error refinement using a similar ELSPIA method with the above enhancements. The proposed approach can generate a shape-preserving B-spline curve. Experiments with data analysis and machining tests are presented for verification of quality and efficiency. Comparisons with other known solutions are included to evaluate the worthiness of the proposed solution.
