Method Validation for Monitoring of Agricultural Worker Exposure to Insecticide Fenthion (살충제 Fenthion에 대한 농작업자 노출 측정을 위한 분석/시험방법 검증)

  • Exposure measurement of agricultural worker to pesticide is one of important part of health risk assessment of pesticide. Therefore exposure matrices, apparatus, instruments and methods must be validated in advance to field experiment. In this study, method validation with an organophosphorus insecticide fenthion was carried out for exposure monitoring of agricultural worker. LOD and LOQ were 0.01 and 0.05 ng, respectively. Calibration curve linearity ($R^2$ > 0.999) and reproducibility (C.V. < 3%) were also excellent. Recovery at LOQ, 10LOQ and 100LOQ levels from gloves, socks, mask, patch, solid sorbent, glass fiber filter was 76~113% (C.V. < 3%). Trapping efficiency was 95~105% while no breakthrough was observed. Method validation for the exposure monitoring was established successfully through several experiments. Such method validation can be usually performed in laboratory and not much different for each pesticide so that, this techniques will be applied widely in research for pesticide exposure monitoring by combination with body surface area and respiration rates.

Analysis on the Microbial Community Structure of Red Granule in the Anaerobic Ammonium Oxidation Reactor (혐기성 암모늄 산화 반응기 내 붉은색 입상슬러지의 미생물 군집구조 분석)

  • Extremely slow growing anammox(anaerobic ammonium oxidation) bacteria were cultivated using a combination of UASB(Upflow Anaerobic Sludge Blanket) reactor seeded with anaerobic granular sludge and carbon-fiber cultivating reactor. After 180 days of continuous cultivation, average nitrogen removal rate showed 0.54 kg $N/m^3-day$ when 0.6 kg $N/m^3-day$ of nitrogen loading was applied. The black granule was changed to brown and red granule as continuous operation, and the red granule was highly dependant on the high anammox activity. Microbial community structure of red granule in the UASB reactor was analyzed by molecular methods such as gene cloning, phylogenetic tree analysis, and FISH(Fluorescence In Situ Hybridization) method. As a result of gene cloning and phylogenetic tree analysis, 5 kinds of phylum were found to be Planctomycetes, Proteobacteria, Acidobacteria, Chlorobi and Chloroflexi. 13 clones were matched to anammox bacteria among 51 clones in the red anammox granule. In-silico test which used cloning information and FISH probe of the AMX368 was conducted to detect the presence of anammox bacteria in the red anammox granule. As a result of in-silico test only one clone was exactly matched to AMX368 but 11 clones was mutated one base among 18 bases representing all 12 clones are anammox bacteria. A filamentous Chloroflexi might be related to the granulation of anammox bacteria. As a result of FISH analysis, anammox bacteria was abundant in the red anammox granule.

Identification and Physiological Characters of Intestinal Bacteria of the Black Soldier Fly, Hermetia illucens (아메리카동애등에 장내세균 동정과 생리적 특징)

  • The black soldier fly, Hermetia illucens, larvae may depend on indigenous bacteria in the intestine to feed and digest diverse food sources. To prove this hypothesis, we isolated and identified the intestinal bacteria of the black soldier fly for their digestive and antimicrobial abilities. The last instar larvae had long digestive tracts, which were about seven times longer than its body length. An individual of H. illucens larvae possessed a total of $5.0{\pm}10^6$ bacteria in the whole intestine, of which more than 98% bacteria were located in the hindgut. Three different bacterial isolates cultured on nutrient agar (NA) medium were detected in the intestine and identified as Morganella morganii, Providencia rettgeri and Bacillus halodurans by Biolog microbial identification system. Analysis of 16S rDNA sequences of the intestinal bacteria detected the additional bacteria of Proteus mirabilis, Providencia alcalifaciens, and Providencia sp. These intestinal bacteria cultured on NA medium exhibited high resistance to 4 antibiotics and inhibited growth of other microbes which are mainly plant pathogens. Also, these bacteria exhibited catalytic activities to degrade cellulose, lipid, proteins, and carbohydrates. These results suggest that H. illucens larvae possess intestinal bacteria that may play crucial roles in their digestive physiology.

Experimental Analysis on Filtration-Permeation: Influence of the Type and Sheets of Filter Media, and Filtration Pressure (여과-투과에 대한 실험적 분석: 여과매체의 종류, 여과매체의 매수, 여과압력의 영향)

  • The average specific cake resistance, the most important indicator for cake filtration and solid-liquid separation, is measured by filtration experiment. But the exact value is difficult to measure because of the other influences such as sedimentation during filtration. This study, a little more stable method named filtration-permeation is proposed for measuring average specific cake resistance. The filtration-permeation is composed of permeation of particle eliminated water through pre-formed cake by filtration. Using 1 wt% calcium carbonate suspension, the filtration-permeation experiments were performed for 8 kinds of filter media at the conditions of 0.5 atm and 0.2 atm, 1 and 3 sheets of filter media. At each specific condition, three to five times filtration-permeation were accomplished. As a result, stable permeation speed is measured. According to this experimental result, the characteristics of permeation and the effect of sedimentation are analyzed with Ruth's equation. The one way analysis of variance (one way ANOVA) is applied to the average specific cake resistances of filtration and permeation obtained with the selected three kinds of filter media. The average specific cake resistances between 0.5 atm and 0.2 atm by filtration do not distinguished, but those by permeation is perfectly distinguished. The experimental results during permeation have a very narrow distribution than that measured during filtration. The analysis of filtration experiments, it was verified that the resistance of filter medium by traditional method is of no significance. Finally, the migration of small particles through the medium composed of fiber glass at low pressure was studied.

Antiproliferative Effect of Extracts from Corydalis heterocarpa on Human Cancer Cells (염주괴불주머니 (Corydalis heterocarpa) 추출물의 암세포 성장 억제 효과)

  • Whole plants of Corydalis heterocarpa were extracted twice with $CH_2Cl_2$ and MeOH in turn. The combined crude extracts were concentrated in vacuo and then partitioned between $CH_2Cl_2$ and $H_2O$. The organic layer was fractionated with n-hexane and 85% aq. MeOH, and the aqueous fraction was also further fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions were evaluated in AGS, HT1080, U-937, MCF-7 and HT-29 human cancer cells using MTT assay. The inhibitory effects of solvent fractions were increased in a dose-dependent manner. Among these tested samples, 85% aq. MeOH fraction showed the most potent inhibitory effect on the growth of human cancer cells. These results suggest that active compounds having much stronger anticancer effect can be isolated from Corydalis heterocarpa.

EXPERIMENTAL RECONSTRUCTION OF LARYNX WITH STERNOMASTOID MYOPERIOSTEAL FLAP (흉유돌근골막 피판을 이용한 후두 재건)

  • 후두암의 수직절제 수술후의 후두 재건 목적은 기관절개를 통하지 않고 후두를 통한 호흡이 가능하도록 하면서 연하시 기도흡인을 피하기 위한 sphincter 능력의 보존, 그리고 발성이 가능하도록 성대진동 mechanism을 재건하는데 있다. 오늘날 수많은 후두학자들에 의해서 고안 개발된 다양한 재건 방법이 있다. 연자는 광범위한 후두절제에 따른 큰 결손을 메우기 위해 bulky하면서도 점막 상피의 재생이 용이한 재건 피판으로 흉유돌근골막 피판을 이용하여 후두 결손부를 재건하여 보았다. 방법은 성견 3마리를 대상으로하여 thiopental sodium 정맥주사로 전신마취를 시행한 후 설골에서부터 흉골상까지 경부 정중앙의 피부를 절개하고 후두를 노출시켜 후두 수직절제술을 시행하였다. 흉유돌근과 흉골에 부착된 골막을 박리하여 흥유돌근골막 피판을 제작한 후 골막이 후두강 안쪽으로 되도록하여 골막연과 후두점막을 봉합하였다. 그리고 3, 5, 9개월에 후두적출술을 시행하여 병리조직학적 및 수술후 경과를 관찰하였다. 3실험견 모두 기관절개술 없이 후두를 통한 호흡이 가능하였고 흡입성 폐렴이나 피하기종등의 합병증없이 창상이 치유되었으며 발성도 양호하였다. 이식된 골막위로 신생혈관의 출현과 함께 점막재생이 잘되 있었다. 단지 봉합사 주위에 소량의 육아종이 관찰되었다. 조직학적으로는 섬유조직위로 편평상피가 재생되었으며 성문하부에서는 일부 섬모가 있는 호흡기 점막도 관찰되고 골막하부에 신생골 형성은 관찰할 수 없었다. 골막 피판은 그 유연성 때문에 결손부위의 점막연에 맞춰도 tension이 없고 공기 누출이 되지 않게 봉합이 가능할 뿐 아니라 점막이 재생할 수 있는 frame-work의 역할을 하는 것으로 사료되었다. 이상과 같은 사실로 미루어 흉유돌근 자체가 견실하고 골막에 혈류공급이 잘되어 창상치유에 좋을 뿐 아니라 큰 후두결손부의 재건이 가능하리라고 사료되었다.로서 몇가지 앞으로의 치료지침에 도움이 되는 결과를 얻었기에 보고하는 바이다. 1) 성별 분포는 남자 16(39 %), 여자 25 (61%)이었으며 1 : 1.5의 빈도를 보였다. 2) 연령 분포는 20대와 30대에서 남녀 모두 25명으로 대부분을 차지하였다. 3) 부식제의 종류는 빙초산이 26명 (63.4 %)으로 대부분을 차지하였고 염산 7 (17.1 %) Lye 3 (0.7 %) 의 순이었다. 4) 음독후 12시간내에 식도경술을 받은 환자가 3명(0.7 %) 12-24시간에 받은 환자가 17명(41.5 %), 24 - 48시간에 받은 환자가 11명(26. 8%)으로 48시간 내에 시행받은 환자가 전체의 75.6%를 차지하였다. 5) 식도경 검사상 나타난 식도화상은 Grade I 11명 (26.8%) G.ade II 18(43.9%) Grade III 7명(17.1%) 이었으며 Grade II 인 경우가 18명(43.9%)로 가장 많았으며 Grade I 11명(26.8 %), Grade III 7명 (17.1 %) Normal 5명 (12.2 %) 순이었다. 6) 조기 식도경 검사에서 41명중 oral cavity burn이 없었던 경우가 15명(36. 1 %) 이었으며, oral cavity burn이 있었던 26명중 5명(19 .2 %)에서 Esophageal burn이 없었다 특히 Esophageal burn의 Grade II, III 25명 중 9명(29.6 %)에서 oral cavity burn이 없었다. 7) 식도 부식중 환자의 치료 원칙으로 Grade I, II, III에서 항생제 및 보존적 치료를 하였으며 Grade I에선 oral feeding을 시켰고 Steroid는 경우에 따라 투여하였으며 Grade III에선 원칙적으로 사용치 않았다. 식도조영술은 Grade I II III에서 3주 후에 모두 시행하였다. 8) 3주 후 식도조영술을 실

Primary bone tumors of fibula (비골에 발생한 원발성 골 종양)

  • Purpose: The purpose of this study was to investigate on primary bone tumors of fibula. Materials and Methods: Patients who received operative treatment from January 1993 to December 2005 for primary bone tumors of fibula were analyzed for clinical outcomes. Results: There were 17 patients(10 males, 7 females) who were diagnosed for primary bone tumors of fibula. Osteochondroma was the most common diagnosis and others were osteosarcoma, fibrous dysplasia. The malignant bone tumors were 10.5% of all fibula tumors. There were 12 cases on proximal, 4 cases on mid shaft, and 1 cases on distal part of fibula. At the final follow up, 14 patients reported patient satisfaction of either excellent or good. Conclusion: Bone tumor of fibula doesn't need reconstruction after the resection unless it invades tibia or its site distal, which enables easy surgical resection. However, generally it is rare and accompany with vague symptoms, so careful approach to diagnosis is necessary because late diagnosis can mean the needs for amputation.

Comparision of Chemical Components of Angelica gigas Nakai and Angelica acutiloba Kitagawa (토당귀와 일당귀의 화학성분 비교)

  • Chemical components of domestic Angelica gigas Nakai and Angelica acutiloba Kitagawa were analyzed. Proximate analysis of each species showed crude protein 18.1% and 13.4%, crude lipid 8.9% and 4.3%, crude fiber 8.6% and 9.4%, crude ash 7.4% and 8.2%, and carbohydrate 57.0% and 64.7%, respectively. Contents of potassium which was found to be the most abundant mineral in both species were 2,740.0 mg% and 2,582.8 mg%, and those of sucrose were 0.4 % and 0.3% respectively while neither fructose nor glucose were detected in each species. Major fatty acids in Angelica gigas Nakai and Angelica acutiloba Kitagawa were linoleic acid (60.8% and 59.5%), palmitic acid (17.4% and 15.3%), oleic acid (8.8% and 7.7%) (respectively) but there was no significant difference between two species. Arginine was revealed as the most abundant amino acid in both species with 2,599.8 mg% in Angelica gigas Nakai and 1,543.4 mg% in Angelica acutiloba Kitagawa. Angelica gigas Nakai and Angelica acutiloba Kitagawa also were shown to contained 10.5 mg% and 12.2 mg% $vitamin\;B_1$, 0.1 mg% and 0.04 mg%, $vitamin\;B_2$, 4.3% and 0.8% decursin, and 988.0 mg% and 900.0 mg% tannin, respectively.

A Study on the Shear Strengthening Characteristic of Reinforced Concrete T-shaped Beams (철근콘크리트 T형보의 전단 보강 특성에 관한 연구)

  • Most of studies on existing strengthening methods were mainly on increase of stiffness and strength of shear strengthening to rectangular beam. As concrete of beam and slab is poured simultaneously on the characteristics of construction in reinforced concrete beam-slab structure, adjacent slab uniformed after hardening has T-shaped beam cross section which makes the flange of beam, enhances the stiffness of the beam and widens the area supporting compressive strength, but available data of flexural behavior of T-shaped beam are lacking. In this research the T-shaped beams would be made, then the reinforced effects and structural properties can be estimated according to the kinds of reinforced materials and reinforced position. The conclusions are shown as below. To sum up the experimental results, The specimen which was reinforce by CB embedded inside of concrete indicated excellent resistive behavior, internal force and stiffness when it was destroyed. The steel plate reinforced specimen of stiffness and internal force were increase but it expressed lower reinforce effects because of lowering anchored force between concrete. Fiber sheet strengthening showed superior effects but the interfacial delamination was found due to the lack of anchored force in destruction. So the measure is needed now.

Recent Progress and Perspectives of Solid Electrolytes for Lithium Rechargeable Batteries (리튬이차전지용 고체 전해질의 최근 진전과 전망)

  • Nonaqueous organic electrolyte solution in commercially available lithium-ion batteries, due to its flammability, corrosiveness, high volatility, and thermal instability, is demanding to be substituted by safer solid electrolyte with higher cycle stability, which will be utilized effectively in large-scale power sources such as electric vehicles and energy storage system. Of various types of solid electrolytes, composite solid electrolytes with polymer matrix and active inorganic fillers are now most promising in achieving higher ionic conductivity and excellent interface contact. In this review, some kinds and brief history of solid electrolyte are at first introduced and consequent explanations of polymer solid electrolytes and inorganic solid electrolytes (including active and inactive fillers) are comprehensively carried out. Composite solid electrolytes including these polymer and inorganic materials are also described with their electrochemical properties in terms of filler shapes, such as particle (0D), fiber (1D), plane (2D), and solid body (3D). In particular, in all-solid-state lithium batteries using lithium metal anode, the interface characteristics are discussed in terms of cathode-electrolyte interface, anode-electrolyte interface, and interparticle interface. Finally, current requisites and future perspectives for the composite solid electrolytes are suggested by help of some decent reviews recently reported.
