Interactions between secreted GRA proteins and host cell proteins across the parasitophorous vacuolar membrane in the parasitism of Toxoplasma gondii

  • Interactions between GRA proteins of dense granules in Toxoplasma gondii and host cell proteins were analyzed by yeast two-hybrid technique. The cMyc-GRA fusion proteins expressed from pGBKT7 plasmid in Y187 yeast were bound to host cell proteins from pGADT7-Rec-HeLa cDNA library transformed to AH109 yeast by mating method. By the selection procedures, a total of 939 colonies of the SD/-AHLT culture, 348 colonies of the $X-\alpha-gal$ positive and PCR, 157 colonies of the $X-\beta-gal$ assay were chosen for sequencing the cDNA and finally 90 colonies containing ORF were selected to analyze the interactions. GRA proteins interacted with a variety of host cell proteins such as enzymes, structural and functional proteins of organellar proteins of broad spectrum. Several specific bindings of each GRA protein to host proteins were discussed presumptively the role of GRA proteins after secreting into the parasitophorous vacuoles (PV) and the PV membrane in the parasitism of this parasite.

Experimental Study on Bond Behavior of Retrofit Materials by Bond-Shear Test (부착전단 실험에 의한 보강재료의 부착거동 실험 연구)

  • A variety of retrofit material such as CFRP, GFRP, and PolyUrea have been developed for strengthening RC structures and infrastructures. From previously reported research results, the capacity of strengthened concrete structures was dictated by the behavior of the interface between retrofit material and concrete. In this study, bond-shear test was carried out to estimate the bond behavior between retrofit material and concrete using a newly developed test grip. The test results of load and slip relation and energy absorption capacity of each retrofit material were obtained. These test results will provide basic information for retrofit material selection to achieve target retrofit performance.

Transient Adverse Neurologic Effects of Spinal Pain Blocks

  • Objective : Chronic neck or back pain can be managed with various procedures. Although these procedures are usually well-tolerated, a variety of side effects have been reported. In this study we reviewed cases of unexpected temporary adverse events after blocks and suggest possible causes. Methods : We reviewed the records of patients treated with spinal pain blocks between December 2009 and January 2011. The types of blocks performed were medial branch blocks, interlaminar epidural blocks and transforaminal epidural blocks. During the first eight months of the study period (Group A), 2% mepivacaine HCL and triamcinolone was used, and during the last six months of the study period (Group B), mepivacaine was diluted to 1% with normal saline. Results : There were 704 procedures in 613 patients. Ten patients had 12 transient neurologic events. Nine patients were in Group A and one was in Group B. Transient complications occurred in four patients after cervical block and in eight patients after lumbar block. Side effects of lumbar spine blocks were associated with the concentration of mepivacaine (p<0.05). The likely causes were a high concentration of mepivacaine in five patients, inadvertent vascular injection in three patients, intrathecal leak of local anesthetics in one, and underlying conversion disorder in one. Conclusion : Spinal pain blocks are a good option for relieving pain, but clinicians should always keep in mind the potential for development of inevitable complications. Careful history-taking, appropriate selection of the anesthetics, and using real-time fluoroscopy could help reduce the occurrence of adverse events.

Analysis on Characteristics of Agricultural Heritage in GIAHS sites (세계중요농업유산(GIAHS) 등재지역 농업유산의 특성 분석)

  • Since FAO introduced the Globally Important Agricultural Heritage Systems(GIAHS) in 2002, 36 sites of 15 countries so far have been listed on GIAHS. This study aims to find the important agricultural heritages of Korea and to prepare the methods for them to be selected as GIAHS. We have analyzed the proposals of the 36 GIAHS listed in order to study the characteristics of their components of the agricultural heritage which worked for being selected. To analyze the components of the agricultural heritage first, agricultural heritage was classified into 13 types and 42 components in light of GIAHS criteria. Then central themes were set to analyze the relevant contents in the proposals. They were, the type of GIAHS Site, significant agricultural landscape, the agriculture-forestry-fisheries-livestock linkage systems, the multi-layered, inter-cropping, circulation cultivation systems, the soil and water management systems, the conservation of agrobiological diversity and genetic resources, the history of the agricultural heritage, the succession of traditional farming techniques, the cultural diversity and so on. Most GIAHS are located in mountains, grasslands, rivers and coasts, desert than in plains, through which GIAHS assure us that it is the heritage of human challenge to overcome the harsh geographic environment and maintain a livelihood. In these sites the traditional farming techniques are carried on, such as mountain clearings, terraced rice paddies, and burn fields, and the unique irrigation systems and agricultural landscapes are well maintained, and the eco-friendly traditional farming techniques utilizing abundant forest resources and agriculture are well handed down. The origin or home of crop growing, a variety of genetic crop storage, the world's largest crop producer and preserving cultural diversity are also important factors for the selection.

Thoracoscopic Lung Biopsy for Diffuse Interstitial Lung Disease (미만성 간질성 폐질환에서 비디오 흉강경을 이용한 폐생검)

  • Diffuse interstitial lung disease due to a wide variety of conditions are amenable to diagnosis by means of clinical evaluation, bronchoalveolar lavage, transbronchial biopsy and lung biopsy. The recently introduced technique of thoracoscopic wedge biopsy provides the potential advantages of greater selection of biopsy site and reduced postoperative chest pain compared with the standard open lung biopsy Video assisted thoracoscopic lung biopsy was performed in twenty patients for diagnosis of diffuse in- terstitial lung disease during the period from January 1993 to June 1994. The ages of the patients ranged from 18 to 69 years(mean 48.5 years), and the patients consisted of 10 males and 10 females. In all twenty, thoracoscpic lung biopsy samples were obtained; 9 from left lower lobe, 7 from right lower lobe, 2 from right upper lobe, 1 from left upper lobe, and 1 from right middle lobe. All patients were correctly diagnosed. The two common diseases were usual in erstitial pneumonia(UIP)(8120) and mil- liary tuberculo si s(4120) . Mean duration of the chest drainage was 3.2 days. There was no complication.

Selection of Grading Deviations to Develop Improved Baseball Leg Guards for Size Suitability and Efficiency (사이즈 적합성과 효율성이 향상된 야구 다리보호대 개발을 위한 그레이딩 편차 선정)

  • Baseball is a life sport that gives vitality to people. However, currently available leg guards do not have a variety of sizes and it is very difficult to purchase a leg guard that is suitable for the size of a specific consumer. Therefore, this study investigated the size system of a leg guard sold in the market and suggested a grading deviation suitable for the legs of Korean males. The results of the study were as follows. First, most brands were sold only in three size systems as children's, teenagers, and adults. Adult size systems were not subdivided. Second, Korean male legs were classified into Group 1 (large girth and height and lower body muscle) and Group 2 (small girth and height and low underbody muscle). Third, the size system is based on vertical items such as height and mid-thigh circumference. Each group produced 10 sizes. The smallest height or the largest height was also found to be about 4.5 to 5.0cm smaller or larger than the sample size for each group. The total length of the leg guard was about 2.5cm when the height was increased or decreased by one size. It was confirmed that the deviation of the circumference of the mid-thigh should be about 2.0cm larger or smaller than the sample size even if the height is the same. The deviation of the knee circumference and ankle circumference was smaller than the other circumference. In conclusion, the dimension combinations of the leg guard must be set differently to provide a leg guard with high size suitability according to leg type.

Optimization of Human Embryonic Stem Cells into Differentiation of Dopaminergic Neurons in Vitro: I. Additive Effect of Neurotrophic Factor on Human Embryonic Stem Cells

  • Embryonic stem cells are capable of differentiating into a variety of cell lineages. However, the ultimate results of differentiation in vitro greatly depend on the duration of treatment and kinds of differentiating inducers added. In order to investigate the efficiencies of various differentiation inducers and the methods of treatment, we examined differentiation patterns of human embryonic stem cell (hESC, MB03) according to several different protocols. Exp. I) Upon differentiation using retinoic acid and ascorbic acid (RA/AA), embryoid bodies (EB, for 4days) derived from hESC was exposed to Rh (10$^{-6}$ M) and AA (50 mM) for 4 days, and were allowed to differentiate in N2 medium for 7, 14, 21, or 28 days. Exp. II) When bFGF was used, neuronal precursor cells were selected for 8 days in N2 medium after EB formation. After selection, cells were expanded at the presence of bFGF (20 ng/ml) for another 6 days followed by a final differentiation in N2 medium for 7, 14, 21 or 28 days. Exp. III) In addition, to examine the effects of neurotrophic factors in the production of mature neurons, groups of cells were exposed to either BDNF (5 ng/ml) or TGF-$\alpha$(10 ng/ml) during the 28 days of final differentiation. Differentiation patterns of RA/AA or bFGF treated groups were very similar; approximately 82% and 83% of the cells, respectively, were positive for anti-NF200 antibody, while it was about 10% and 11%, respectively, for anti-NF160 antibody in 28 days in N2 medium. Alsor, cells expressing TH were as low as 5%, while the cells doubled when matured at the presence of either BDNF or TGF-$\alpha$. Cells immunoreactive to anti-GAD antibody were approximately 20%. These results suggest that a maturation step rather than differentiation induction step, which is formation of EB, effects more decisively to the ultimate differentiation pattern.

Development of Host-Vector Systems for Lactic Acid Bacteria (유산균의 Host-Vector System 개발)

  • Lactic acid bacteria (LAB) are widely used for various food fermentation. With the recent advances in modern biotechnology, a variety of bio-products with the high economic values have been produced using microorganisms. For molecular cloning and expression studies on the gene of interest, E. coli has been widely used mainly because vector systems are fully developed. Most plasmid vectors currently used for E, coli carry antibiotic-resistant markers. As it is generally believed that the antibiotic resistance markers are potentially transferred to other bacteria, application of the plasmid vectors carrying antibiotic resistance genes as selection markers should be avoided, especially for human consump-tion. By contrast, as LAB have some desirable traits such that the they are GRAS(generally recognized as safe), able to secrete gene products out of cell, and their low protease activities, they are regarded as an ideal organism for the genetic manipulation, including cloning and expression of homologous and heterologous genes. However, the vec-tor systems established for LAB are stil insufficient to over-produce gene products, stably, limiting the use of these organisms for industrial applications. For a past decade, the two popular plasmid vectors, pAM$\beta$1 of Streptococcus faecalis and pGK12 theB. subtilis-E. coli shuttle vector derived from pWV01 of Lactococcus lactis ssp. cremoris wg 2, were most widely used to construct efficient chimeric vectors to be stably maintained in many industrial strains of LAB. Currently, non-antibiotic markers such as nisin resistance($Nis^{r}$ ) are explored for selecting recombi-nant clone. In addition, a gene encoding S-layer protein, slp/A, on bacterial cell wall was successfully recombined with the proper LAB vectors LAB vectors for excretion of the heterologous gene product from LAB Many food-grade host vec-tor systems were successfully developed, which allowed stable integration of multiple plasmid copies in the vec-mosome of LAB. More recently, an integration vector system based on the site-specific integration apparatus of temperate lactococcal bacteriophage, containing the integrase gene(int) and phage attachment site(attP), was pub-lished. In conclusion, when various vector system, which are maintain stably and expressed strongly in LAB, are developed, lost of such food products as enzymes, pharmaceuticals, bioactive food ingredients for human consump-tion would be produced at a full scale in LAB.

  • The theses is an analytical comparison research between 'Skills Link' and 'Do Dream' : the former is an effective program to establish a variety of intervention strategies and the institutional foundation for employment support of disadvantaged youth, performed by ESDC in the level of the nation, and the latter is the program which is carried out by K-Dream Center of MGEF in South Korea. As for the research method, comparison and analysis have been fulfilled, focusing on the employment support program. The research includes program background, goal, participation subject, selection standard, program contents, support period, support cost, and program effect. It suggests that we should design the systematic program for the youth in the transition to independency and perform constant support. It is necessary to select beneficiary on the consideration of the characteristics and situation of the disadvantaged youth. In addition, it is required to secure consistency of the program and enlarge the support period. For the sake of successful performance of the independent living program, it is a necessity to motivate the youth and introduce the ways to encourage the business sector to take part in.

Evolutionary Theory and Twentieth Century Music -Focused on 4'33" by John Cage- (진화론으로 바라본 20세기 현대 음악 -존 케이지 4분 33초를 중심으로-)

  • 20th century music has been recognized as disconnected. Some criticized that it is not music. This controversy has been continued over decades in music history. This study applied Darwin's evolutionary theory to John Cage's 4'33", the most popular music in 20th century and tried to prove musical continuity embedded in it. 4'33" has always been a subject of controversy because it has no sound. However, the study found out that 4'33" has the common musical factor, the rest. 4'33" can be interpreted as the variation of the rest in evolutionary view. The rest has undergone a variety of variations in form and meaning since the beginning of the musical notation and was suitable for a new musical environment changed in 20th century. In the evolutionary view, 4'33" is evolved music, not disconnected.
