This study was conducted to evaluate the susceptibility of rice(Oryza sativa L.) cultivars to herbicide thiobencarb through the determination of protein patterns using SDS-PAGE. In both greenhouse and laboratory screening tests, IR 10198-66-2 and IR 9660-50-3-1 showed relatively tolerant response to thiobencarb, while IR 22, IR 31802-48-2-2-2 and IR 20656-R-R-R-6-1 were susceptible to it. The total protein content of susceptible cultivars markedly decreased as the thiobencarb concentrations increased, but tolerant cultivars such as IR 10198-66-2 and IR 9660-50-3-1 showed very slight changes suggesting that protein synthesis of susceptible cultivars may be inhibited by thiobencarb. The protein profiles of the tolerant cultivars were not much affected by thiobencarb treatment. However, the protein spots with molecular weights of 14.4 kD and 55 kD in the susceptible cultivar of IR 22 disappeared with the treatment of 3 ppm thiobencarb, indicating that differential susceptibilities of rice cultivars against thiobencarb can be attributed to their difference in protein metabolism affected by thiobencarb.

The dehydration responsive element binding protein 2C (DREB2C) is a dehydration responsive element/C-repeat (DRE/CRT)-motif binding transcription factor that induced by mild heat stress. Previous experiments established that overexpression of DREB2C cDNA driven by the cauliflower mosaic virus 35S promoter (35S:DREB2C) resulted in increased heat tolerance in Arabidopsis. We first analyzed the proteomic profiles in wild-type and 35S:DREB2C plants at a normal temperature ($22^{\circ}C$), but could not detect any differences between the proteomes of wild-type and 35S: DREB2C plants. The transcript level of DREB2C in 35S: DREB2C plants after treatment with mild heat stress was increased more than two times compared with expression in 35S:DREB2C plants under unstressed condition. A proteomic approach was used to decipher the molecular mechanisms underlying thermotolerance in 35S:DREB2C Arabidopsis plants. Eleven protein spots were identified as being differentially regulated in 35S:DREB2C plants. Moreover, in silico motif analysis showed that peptidyl-prolyl isomerase ROC4, glutathione transferase 8, pyridoxal biosynthesis protein PDX1, and elongation factor Tu contained one or more DRE/CRT motifs. To our knowledge, this study is the first to identify possible targets of DREB2C transcription factors at the protein level. The proteomic results were in agreement with transcriptional data.
We examined the enhancement of thermotolerance for storage conferred on Lactobacillus acidophilus 30SC by adaptation to different stresses. The viable cells of Lactobacillus acidophilus 30SC were compared with their viability prior to heating at $45,\;55^{\circ}C\;and\;60^{\circ}C$. Heat-adapted ($45^{\circ}C$ for 15 min) L. acidophilus 30SC in MRS broth exhibited higher survivability at lethal temperature of $55^{\circ}C$ than control. Cellular protein profiles of L. acidophilus 30SC during heat adaptation were examined with SDS-PAGE, and scanning electron microscopy. When L. acidophilus 30SC was heat-adapted at $55^{\circ}C$ for 15min, 5 new protein spots of ca $8\~45\;kDa$ size were observed on 2D SDS-PAGE. It was presumed that new proteins of L. acidophilus 30SC were produced to adapt to the environment of higher growth temperature.

A new bacterial disease of sesame(Sesamum indicum) was observed on field-grown plants in Suwon, Hongchun and Yeonchun in 2000. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Isolations made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Two bacterial strains were purified and used for farther tests. Pathogenicity of strains was confirmed on 3-week-old sesame plants sprayed with bacterial suspensions containing $10^{8}cfu/ml$ of phosphate buffered saline. The Biolog and fatty acid analyses of the two strains(SL3451 and SL3476) 1mm sesame leaf blight showed that they could be identified as ft campestris pv. sesami because of their high similarity to the tester strain(X. campestris pv. sesami LMG865) with a match probability of $100\%$. The bacterium grew well between 18 and 36$^{\circ}C$, but optimum temperature was $27^{\circ}C$ on LB broth. This is the first report of bacterial blight of sesame in Korea. Symptoms of bacterial blight of sesame are difficult to differentiated with those of bacterial leaf spot caused by Pseudomonas syringae pv. sesami.

Tomato spotted wilt virus (TSWV) occurred abruptly with a high incidence rate in 14 vegetable crops in Anyang area, Gyunggido in 2004. TSWV was identified by the symptoms on the indicator plants, immunocaptured reverse transcription-polymerase chain reaction (IC/RT-PCR), virion captured (VC)RT-PCR and RT-PCR using total RNA from the infected plants. Vegetable crops infected with TSWV included table tomato, cherry tomato, red pepper, lettuce, chicory, red leaf chicory, red mustard, dragon tongue, treviso, potato, perilla, sesame, pumpkin, and ssamchoo (hybrid of oriental cabbage and cabbage). The incidence of TSWV in fields ranged from 30 to $100\%$. TSWV usually produced necrosis, wilt and/or severe mosaic with typical single or double ring spots on the leaves. TSWV could be detected in Frankliniella occidentalis collected from the crops in the fields with $90\%$ rate by IC/RT-PCR.

This study was conducted to develop a easy seedling method to test large amounts of barley and wheat cultivars for resistance of powdery mildew. In addition, we also examined the resistance of genetic resources that have been used in barley and wheat breeding programs in Korea. This seedling test used paper bag with seeds can be completed within three weeks. to-day seedlings were available to inoculation and 8 days was needed for result reading. This method can test at least 180 collections at one time. we can identify the two resistant types by leave symptoms showed non infection and necrotic spots. Among the 79 Korean barley cultivars, only two cultivar, 'Sangrokbori' and 'Dajinbori' were resistant and 'Jejubori' showed moderate resistant. There was no resistant in hulless barley and wheat cultivars. It was same results in comparison of earlier resistant reports in field test. We confirm that this method could using in test of powdery mildew resistance in barley and wheat. Among the 1,401 genetic resources using in Korean breeding program, malting barley has more resistant collections comparing to 796 hulled and hulless barley and 273 wheat germplasms.

A pathogen that causes a new disease on green pumpkin in the nursery and the field was characterized and identified. Symptoms of the disease on green pumpkin were water soaking lesions and spots with strong yellow halo on leaf, brown lesion on flower, and yellow spot on fruit. The bacterial isolates from the leaf spot were pathogenic on the 8 curcubitaceae crop plants, green pumpkin, figleaf gourd, wax gourd, young pumpkin, zucchini, cucumber, melon, and oriental melon, whereas they did not cause the disease on sweet pumpkin and watermelon. They were Gram-negative, rod shape with polar flagella, fluorescent on King's B agar and LOPAT group 1a by LOPAT test. Their Biolog substrate utilization patterns were similar to Pseudomonas syringae pv. syringae's in Biolog database. Phylogenetic trees with 16S rRNA gene sequences and multilocus sequence typing (MLST) with nucleotide sequences of 4 housekeeping genes, gapA, gltA, gyrB, rpoD and those of P. syringae complex strains in the Plant Associated and Environmental Microbes Database (PAMDB) showed that the green pumpkin isolates formed in the same clade with P. syringae pv. syringae strains. The clade in MLST tree was in the genomospecies 1 group. The phenotypic and genotypic characteristics suggested that the isolates from green pumpkin lesion were P. syringae pv. syringae.

This study develops the tire/pavement noise measuring trailer that can minimize the effect of various environmental noise and evaluate road noise characteristics without regard to time and spots. As literature reviews, it is examined into road noise characteristics and case studies of various CPX Trailer, and made a development plan of device. Based on these, HEART CPX Trailer is developed and conduct experiments for verification of the semi-anechoic chamber suitability, precision and field application of device. In the result of the experiment for verification of semi-anechoic chamber suitability, we can make sure that it is suitable and reliable among frequency bands. In the test of repetition measurement, we can verify excellent measurement accuracy among speeds and microphone's locations. In case of field tests using HEART CPX Trailer, it is founded that the result is similar to CPB test result carried out by Chung-Ang university in 2003. It can be concluded caused that the effect of environmental noise in and around the road including mechanical noise of a tow car was excluded. This paper confirmed and suggested that HEART CPX Trailer is suitable for evaluation of real road noise characteristics.

Rabbit hemorrhagic disease is a highly acute and fatal viral disease caused by rabbit hemorrhagic disease virus (RHDV). Since first outbreak in Korea 1987, RHDV has been continually affected in the country, but the pattern of outbreak seem to be changed. In this study, to understand the pathogenesis of the new RHDVa serotype, we therefore carried out to inoculate RHDVa to rabbits, and to examine the sequential histopathologic changes and viral distribution. Macroscopically, various sized dark red or white spots or appearance were observed in the liver, lung, kidney uterus and ureter. In euhanized rabbits, significant pathologic findings such as infiltration of heterophils and mononuclear cells were observed at 24 hours after inoculation (HAI), and these were sequentially extended periportal to centrilobular area. However, in dead rabbits, severe hepatic degeneration and/or necrosis with relatively weak inflammatory responses were observed. RHDV antigens began to detect in liver, spleen, and lung from 12 HAI by PCR. Immunohistochemically, RHDV positive cells were seen in only liver from 24 HAI, and the degree of immunogen reactivity was stronger in dead rabbits than in euthanized ones. In conclusion, RHDVa caused the subacute or chronic infection accompanying low mortality and moderate to severe inflammatory reaction in rabbits, suggesting the possibility that RHD could become endemic.
Recently, as a part of the production of spatial information by smart cities, three-dimensional reproduction of structures for reverse engineering has been attracting attention. In particular, terrestrial LiDAR is mainly used for 3D reproduction of structures, and 3D reproduction research by UAS has been actively conducted. However, both technologies produce blind spots due to the shooting angle. This study deals with vertical structures. 3D model implemented through SfM-based image analysis technology using UAS and reproducibility and effectiveness of 3D models by terrestrial LiDAR-based laser scanning are examined. In addition, two 3D models are merged and reviewed to complement the blind spot. For this purpose, UAS based image is acquired for artificial rock wall, VCP and check point are set through GNSS equipment and total station, and 3D model of structure is reproduced by using SfM based image analysis technology. In addition, Through 3D LiDAR scanning, the 3D point cloud of the structure was acquired, and the accuracy of reproduction and completeness of the 3D model based on the checkpoint were compared and reviewed with the UAS-based image analysis results. In particular, accuracy and realistic reproducibility were verified through a combination of point cloud constructed from UAS and terrestrial LiDAR. The results show that UAS - based image analysis is superior in accuracy and 3D model completeness and It is confirmed that accuracy improves with the combination of two methods. As a result of this study, it is expected that UAS and terrestrial LiDAR laser scanning combination can complement and reproduce precise three-dimensional model of vertical structure, so it can be effectively used for spatial information construction, safety diagnosis and maintenance management.
