Blood Pressure and Serum Lipids in Relation to Serum Gamma-glutamyltransferase Activity in Men of Rural Area (농촌 지역 남성들에서 혈청 Gamma-glutamyltransferase 활성도와 혈압 및 혈청 지질의 관련성)

  • Objectives: To examine the association between serum gamma-glutamyltransferase(GGT) activity level and blood pressure, serum lipids in the male residents of rural area, we analyzed cross-sectionally the data of 379 male health examinees aged 40 years and older from rural areas in Gyeongsangnam-do prefecture in Korea. Methods: Blood pressure and concentration of serum lipids were compared between high and low level of serum GGT activity by t-test or Wilcoxon rank sum test. Possible confounding effects of age, body mass index and coffee drinking were adjusted by analysis of covariance. Results: Adjusted values of systolic and diastolic blood pressure were higher in the group of high level of serum GGT activity in non-drinkers(P=0.055 and P=0.074 respectively) and drinkers(P=0.284 and P=0.398) of alcohol. Adjusted serum total cholesterol level was also higher in the group of high level of serum GGT activity in non-drinkers(P=0.052) but not in drinkers(P=0.981) of alcohol. In serum triglyceride, adjusted level was significantly higher in the group of high level of serum GGT activity in both non-drinkers(P=0.035) and drinkers(P=0.002) of alcohol. Conclusions: These results suggest the association of serum GGT activity and serum triglyceride, total cholesterol, or blood pressure in non-drinkres of alcohol.

  • Poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] microspheres were prepared using solvent evaporation technique. P(3HB-co-4HB) with 3.9 mol% 4HB was synthesized by fed-batch culture of Ralstonia eutropha. The effects of concentration and type of surfactant (Tween 80, sodium dodecylsulfate, and polyvinyl alcohol), addition of dispersion stabilizer (Acacia), concentration of polymer and model drug (bovine serum albumin) on particle size of the microspheres and their in vitro drug release characteristics were investigated. The average particle size of the microspheres decreased with the addition of dispersion stabilizer and increased with the concentration of surfactant, drug and polymer. Amount of drug release increased with decreasing particle size of the microspheres.

  • Effects of organic solvents fraction from Ainsliaea acerifolia ethanol extract on alcohol metabolism in rats were examined and the results were as follows: Ethanol souble fraction, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic alcohol dehydrogenase(ADH) activity, on the other hand, the fraction insouble in ethanol was found to cause an increase ethanol concentration in the blood and inhibit ADH activity.

Effect of an Oriental Herbal Composition, Jang Baek Union (JBU), on Alcohol-Induced Hangover and CCl4-Induced Liver Injury in Rats (한약재 조성물이 흰쥐의 알코올성 숙취 및 사염화탄소 유발 간 손상에 미치는 영향)

  • Effects of the extract of oriental herbal composition, Jang Baek Union (JBU), on hangover and liver function were studied in rats. To investigate anti-hangover effect, alcohol concentration and acetaldehyde concentration of blood were measured after oral ethanol treatment. Blood alcohol concentration was significantly reduced by pre-treatment of JBU extract. The effect of JBU extract on the blood acetaldehyde concentration was more significant than commercial product used as a positive control. To investigate hepato-protective effect, serum GOT and GPT levels and histological changes of liver tissue were analyzed in $CCl_4$-treated rats. JBU extract adminstration significantly inhibited the increase of serum GPT and GOT levels induced by $CCl_4$-treatment. Moreover, histological injuries of liver tissue such as fatty changes and sinusoidal leukocytosis were inhibited by JBU extract.

  • In order to investigate the influences of Panax ginseng (white ginseng and red ginseng) on the anesthetic effect and toxic effect of alcohol, experimental studies .had been carried out with albino rabbits, mongolian dogs and mice. The anesthetic effect of alcohol was observed by measuring the induction time, .anesthetic time, recovery time and duration from the beginning of induction to , the recovery of anesthesia (total time), respectively. and toxic effect ($LD_{50}$) of alcohol was measured. In addition to these experiments, al cohol concentration in .blood, blood sugar level, serum transaminase (GOT and GPT) activities and serum alkaline phosphatase activity were measured. Also in order to study the clinical effect of alcohol in healthy students, code .substitution, response time and muscle coordination were tested. The results were obtained as follows. 1. In the rabbits and mongolian Jags, the induction time of anesthesia by the administration of alcohol was delayed by the pretreatment of ginseng but recovery time and total time of anestksia were markedly shortend. 2. The bleed alcohol concentration was decreased by the pretreatment of ginseng , but not affected in mongolian dogs. 3. The blood sugar level, serum transaminase (GOT and GPT) activities and alkaline phoshatase activity in rabbits and dogs induced by the administration of alcohol were affected by the Pretreatment of ginseng. Because those were included within normal ranges, the differnces have no remarkable significance. 4. Liver alcohol dehydrogenase activity of rabbit was increased by the treatment of ginseng, especially it was markedly increased by the treatment of red ginseng 5. The average lethal dose of alcohol to mice was increased by the pretreatment. of ginseng, especially it was markedly increased by the pretreatment of red .ginseng. 6. In the clinical experiments, the blood alcohol concentration induced by alcohol administration was not affected by the pretreatment of ginseng whereas the bleed sugar level was increased. Blood alcohol concentration and bleed sugar level were measured after three hours alcohol administration. 7. The response time of healthy students administered with alcohol was markedly shortened by the pretreatment of ginseng but the experiments on the code substitution and muscle coordination were not affected.

Effects of the Hovenia dulcis THUMBER var. Koreana Nakai Extracts on Hepatoprotective Activities and Improvement of Hepatofunction

  • The Hovenia extracts showed a significant hepato-protective activity against alcohol and CC4-induced hepatotoxicity. Injection of CC4 and alcohol induced liver injury and increased serum GOT (glutamic oxaloacetic transaminase) and GPT (glutamic pyuvic transaminase) levels in injured rats. The ADH (alcohol dehydrogenase)-like and ALDH (aldehyde dehydrogenase)-like activities in the Hovenia extracts were studied. Their activities in the extracts of Hovenia wood were higher than the extracts of other parts of Hovenia. And then Hovenia wood extracts have high effects on the alcohol and acetealdehyde degradation. The Hovenia extracts were decreased concentration of fusel oil. The ADH-like and ALDH-like activities in the Hovenia wood extract were 1.14 $unit/{\mu}l$, 0.33$unit/{\mu}l$ respectively.

Comparative Effects of Dietary Quercetin and Rutin in Rats Fed with the Lieber-DeCarli Ethanol Diet

  • Flavonoids including quercetin and rutin are a group of naturally occurring compounds widely distributed in plants, especially in buckwheat. Thus, cereal and the leaf of the plant have increasingly used as a source of nutritional and functional foods such as noodle, cake or soup in Korea, Japan and other countries. This study investigated comparative effects of dietary rutin rich in buckwheat and its aglycone, quercetin, on serum biomarkers and antioxidant parameters in rats treated with chronic ethanol. Rats were fed with the liquid diets prepared by the method of Lieber Decarli. Serum alanine transaminase (ALT) and aspartate transaminase (AST) activities increased significantly by alcohol feeding. Dietary flavonoids including rutin, quercetin and their mixtures (1/1, v/v) decreased significantly the activities of serum ALT whereas the feeding of quercetin decreased only the activity of serum AST. The concentration of serum malondialdehydes elevated by chronic alcohol feeding decreased markedly in all the experimental groups that were fed with the flavonoids; however, the combined administration of quercetin or rutin, but not that of rutin or quercetin alone decreased significantly the concentration of liver malondialdehydes to the normal range in rats fed without ethanol. Our results suggested that dietary combined mixture of rutin and quercetin might be effective in ameliorating adverse responses seen in rats exposed to ethanol chronically.

Studies on Voice Changes Associated with Alcohol Intake (음주에 따른 음성 변화에 대한 고찰)

  • Background and Objectives : It has been pointed out that alcohol intake in human beings induces changes in voice register and maximum phonation time. These changes supposedly result from injection of the vibratory vocal folds. The purpose of this study was to clarify the voice changes associated with alcohol intake and the changes of laryngeal mucosa. Materials and Methods : The subjects included 29 volunteers, including 20 men and 9 women ranging in age from 22 to 31 years. Alcohol intake was accomplished by oral administration of 23% soju 1 bottle (255cc). Serum alcohol concentration levels were evaluated hourly for 3 h after ingestion of alcohol. Seven measurements were performed at pre-alcohol intake and post-alcohol intake hourly : fundamental frequency, jitter, shimmer, noise to harmonic ratio as the acoustic analysis, maximal phonation time, mean flow rate, and subglottal pressure as the aerodynamic analysis. The changes of laryngeal mucosa were evaluated by flexible laryngoscope at each measurement. Results : By comparing the acoustic and aerodynamic data and laryngeal mucosa before and after alcohol intake, there were not remarkable changes (p>0.05). Conclusion : The voice and laryngeal mucosa have not remarkably changed according to alcohol concentration in this study. Furthermore studies on the voice change induced by multiple alcohol concentrations are required.

Effects of Dietary Protein Levels on the Serum Lipids and Enzyme Activities of Ethanol-Administered Rats (에탄올을 투여한 흰쥐에서 식이 단백질 섭취 수준이 혈청중 지방 및 효소활성에 미치는 영향)

  • This study was undertaken to investigate effects of alcohol and dietary protein levels on serum lipids and enzyme activities in 15 week-old male rats given a normal diet. Rats were divided into 8 groups : control group (16% protein 16PC) and 8%(8PE) 16%(16PE) and 24% protein groups(24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight organ weight and various blood components were determined at 4 and 10 weeks. Body weight gain organ weight hemoglobin concentration and hematocrit value were not influenced by ethanol and dietary protein levels. The levels of total cholesterol HDL-cholesterol and phospholipid in serum were not affected by ethanol consumption. Serum triglyceride concen-trations after 10 weeks were significnatly increased ethanol-treated group compared with that of control group and the effect was greater in low protein group than control group. Serum ALP activity was significantly higher in 8PE group than other group but there was no influence by ethanol consumption.

  • As an initial step for evaluating hepatoprotective components against alcohol-induced toxicity, the effect of various fractions from Aloe vera on alcohol metabolism in rats were examined and the results were as follows: Water soluble fraction, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic ADH activity. On the other hand, the fractions soluble in organic solvent was found to cause an increase in the blood ethanol concentration and inhibit ADH activity. Further fractionation of the water soluble fraction by ultrafiltration system gave four subfractions corresponding to molecular weight and treatment of them in rats demonstrated that subfraction of M.W. > 30,000 exhibited the most potent enhancing activity of ethanol methabolism.
