There are many sources of systematic variations in cDNA microarray experiments which affect the measured gene expression levels like differences in labeling efficiency between the two fluorescent dyes. Print-tip lowess normalization is used in situations where dye biases can depend on spot overall intensity and/or spatial location within the array. However, print-tip lowess normalization performs poorly in situation where error variability for each gene is heterogeneous over intensity ranges. We proposed the new print-tip normalization methods based on support vector machine regression(SVMR) and support vector machine quantile regression(SVMQR). SVMQR was derived by employing the basic principle of support vector machine (SVM) for the estimation of the linear and nonlinear quantile regressions. We applied our proposed methods to previous cDNA micro array data of apolipoprotein-AI-knockout (apoAI-KO) mice, diet-induced obese mice, and genistein-fed obese mice. From our statistical analysis, we found that the proposed methods perform better than the existing print-tip lowess normalization method.
This study aims to analyze the efficiency of the government policy of Nanotechnology which is expected to minimize nanotechnology's potential risk, using the methodology of conjoint analysis and market share analysis. The attributes of conjoint analysis were divided into potential risk factor and the policy factor. It was found that the policy factor could alleviate the potential risk, subsequently increasing consumers' utility. Additionally, the government certification was more powerful than the mandatory labelling. The market share also increased in result of the nanotechnology-applied product with the certification or labeling either. The result of this study can be used as a reference to related policy makers in the fields of Nanotechnology.
The Escherichia coli K-12 and B strains are among the most frequently used bacterial hosts for scientific research and biotechnological applications. However, omics analyses have revealed that E. coli K-12 and B exhibit notably different genotypic and phenotypic attributes, even though they were derived from the same ancestor. In a previous study, we identified a limited number of proteins from the two strains using two-dimensional gel electrophoresis and tandem mass spectrometry (MS/MS). In this study, an in-depth analysis of the physiological behavior of the E. coli K-12 and B strains at the proteomic level was performed using six-plex isobaric tandem mass tag-based quantitative MS. Additionally, the best lysis buffer for increasing the efficiency of protein extraction was selected from three tested buffers prior to the quantitative proteomic analysis. This study identifies the largest number of proteins in the two E. coli strains reported to date and is the first to show the dynamics of these proteins. Notable differences in proteins associated with key cellular properties, including some metabolic pathways, the biosynthesis and degradation of amino acids, membrane integrity, cellular tolerance, and motility, were found between the two representative strains. Compared with previous studies, these proteomic results provide a more holistic view of the overall state of E. coli cells based on a single proteomic study and reveal significant insights into why the two strains show distinct phenotypes. Additionally, the resulting data provide in-depth information that will help fine-tune processes in the future.
Ovarian cancer is the most lethal gynecological malignancy, and specific biomarkers are important needed to improve diagnosis, prognosis, and to forecast and monitor treatment efficiency. There are a lot of pathological factors, including reactive oxygen species (ROS), involved in the process of cancer initiation and progression. The oxidative modification of proteins by ROS is implicated in the etiology or progression of disorders and diseases. In this study, a labeling experiment with the thiol-modifying reagent biotinylated iodoacetamide (BIAM) revealed that a variety of proteins were differentially oxidized between normal and tumor tissues of ovarian cancer patients. To identify cysteine oxidation-sensitive proteins in ovarian cancer patients, we performed comparative analysis by nano-UPLC-$MS^E$ shotgun proteomics. We found oxidation-sensitive 22 proteins from 41 peptides containing cysteine oxidation. Using Ingenuity program, these proteins identified were established with canonical network related to cytoskeletal network, cellular organization and maintenance, and metabolism. Among oxidation-sensitive proteins, the modification pattern of Collagen alpha-1(VI) chain (COL6A1) was firstly confirmed between normal and tumor tissues of patients by 2-DE western blotting. This result suggested that COL6A1 might have cysteine oxidative modification in tumor tissue of ovarian cancer patients.
Viral and non-viral vectors have been used in the delivery of genetic materials into animal cells and tissues, with each approach having pros and cons. Non-viral vectors have many useful merits such as easy preparation, low immunity and size tolerance of a transgene when compared to those of viral vectors. Delivery specificity may be achieved by complex formation between receptor ligands and a non-viral vector. In the present study, non-viral vector systems are investigated in an effort to find a practical delivery means for gene therapy, Receptor-ligand interaction between transferrin-receptor and transferrin was utilized for efficient gene transfer into cancer cells. A plasmid vector, pcDNA3 (LacZ) was ligated with a small duplexed oligo fragment in which a Biotin- VN$^{TM}$ phosphoramidite was placed in the middle of the oligo. The plasmid vector labeled by biotin was then conjugated with biotin-labeled transferrin via streptavidin. This trimeric conjugates were delivered to a hepatoma cell line, HepG2. The delivery efficiency of the trimeric conjugate was 2-fold higher than that of cationic liposomes used for transfection of a plasmid vector. These results demonstrate that a plasmid vector can be efficiently transferred into cells by forming a trimeric complex of plasmid vector-linker-ligand.

Bacterial hemoglobin from Vitreoscilla (VHb) is recognized as a good fusion protein for the soluble expression of foreign protein. In this study, we generated a monoclonal antibody (MAb) against VHb for its detection. For the rapid screening of MAb, a protein chip technology based on the Alexa-488 (A488) dye labeling method was introduced. In order to fabricate the chip, the VHb protein was chemically coupled to the chip surface and then the culture supernatants of 84 hybridoma cell lines were spotted onto the VHb chip. The bound MAbs were measured by A488-modified anti-mouse IgG. A single spot (MAb A10) exhibited significantly high signal intensity. The immunoblot analysis evidenced that the MAb A10 can detect VHb-fused proteins with high specificity.
Since the import ban of plastic waste in China has been enforced, plastic wastes were not properly collected and recycled in Korea. Hence, the management strategies for plastic waste in Korea should be improved by examining the regulations and policy in developed countries such as United States, Japan, EU and United Kingdom. The management strategy for the recycling cycle should be implemented to expand the labeling system of separation and discharge, reduce the consumption of plastic products, automate the separation and sorting method in recycling facilities, and improve the economical efficiency of the recycling cycle. The concept of residual waste (secondary waste) in the material flow analysis should be implemented to identify the shortage point in the plastic waste stream. Finally, the cooperation with international communities is required for a transboundary movement of plastic waste, which includes participation at the working group of international standards to recycle plastic waste.

The role of the library is as a public institution that provides academic information to a variety of people, including students, the general public, and researchers. These days, as the importance of lifelong education is emphasized, libraries are evolving beyond simply storing and lending materials to complex cultural spaces that share knowledge and information through various educational programs and cultural events. One of the problems library user's faces is locating books to borrow. This problem occurs because of errors in the location of borrowed books due to delays in updating library databases related to borrowed books, incorrect labeling, and books temporarily located in different locations. The biggest problem is that it takes a long time for users to search for the books they want to borrow. In this paper, we propose a system that visually displays the location of books in real time using an AI vision sensor and LED. The AI vision sensor-based book location guidance system generates a QR code containing the call number of the borrowed book. When the AI vision sensor recognizes this QR code, the exact location of the book is visually displayed through LED to guide users to find it easily. We believe that the AI vision sensor-based book location guidance system dramatically improves book search and management efficiency, and this technology is expected to have great potential for use not only in libraries and bookstores but also in a variety of other fields.
The protein nutritional quality of foods has become an important factor to food processors with the advent of nutritional labeling regulations for foods. Then, as is true today, the officially approved assay for protein nutritional quality was the rat based protein efficiency ratio(PER) bioassay. The PER bioassay requires a minimum of 28 days to performe, and is therefore not applicable to routine quality assurance use by the food industry. Within the past ten years there has been a research emphasis placed on the development of rapid, inexpensive, biological and/or chemical based assays for protein nutritional quality. It was hoped that if a rapid assay could be developed and thoroughly tested, it could be used in lieu of the PER bioassay in the day-to-day quality assurance screening of food ingredients and products. The rapid assays developed in the hope of attaining this goal have been based on microorganisms, proteolytic enzymes, and amino acid profiles, as well as combinations of the above. In this review, it will be described and briefly discussed many of procedures which had contributed conceptually as well as practically to the development of in vitro methods for the evaluation of protein quality. Special emphasis will be placed on the C-PER(computed protein efficiency ratio) assay which combines data from in vitro protease digestion and amino acid composition to predict protein nutritional quality designed by Satterlee et al. (1980), and the DC-PER(discriminant computed PER) which is a method of estimating protein quality based on rat assay and in vitro digestibility obtained using solely essential amino acid data will be also introduced.
본 연구에서는 UAV (Unmanned Aerial Vehicle)로 촬영한 이미지를 활용하여 수치지도 지형지물 표준 코드에서 정의하고 있는 건물 8종에 대하여 딥러닝 기반의 객체 탐지 분석을 수행하였다. UAV로 촬영한 이미지 509매에 대하여 이미지 라벨링을 하였고 YOLO (You Only Look Once) v5 모델을 적용하여 학습 및 추론을 진행하였다. 실험 및 분석은 오픈소스 기반의 분석 플랫폼과 알고리즘을 적용하여 데이터를 분석하였으며 분석결과 88%~98%의 예측 확률로 건물 객체를 탐지하였다. 또한 학습데이터의 구축 및 반복 학습의 과정에서 건물 객체 탐지의 높은 정확도를 위해 필요한 학습 방식 및 모델 구축방식을 분석하였고, 학습한 모델을 다른 영상자료에 적용하는 방안을 모색하였다. 본 연구를 통해 고효율 심층 신경망과 공간정보데이터가 융합하는 모델을 제안하며 공간정보데이터와 딥러닝 기술의 융합은 향후 공간정보데이터 구축의 효율성, 분석 및 예측의 정확도 향상에 많은 도움을 제공할 것이다.
