Residue Dissipation Patterns of Indoxacarb and Pymetrozine in Broccoli under Greenhouse Conditions (시설재배 브로콜리 중 Indoxacarb 및 Pymetrozine의 잔류 소실특성)

  • BACKGROUND: This study was carried out to establish pre-harvest residue limits (PHRLs) of indoxacarb and pymetrozine in broccoli under greenhouse conditions, based on dissipation patterns and biological half-lives of pesticides during 10 days after application. METHODS AND RESULTS: The field studies were conducted in two different greenhouse, located in Chungju-si (Field 1) and Gunsan-si (Field 2). Samples were collected at 0, 1, 2, 3, 5, 7 and 10 days after spraying pesticide suspension. The analytical methods for indoxacarb and pymetrozine using HPLC-DAD were validated by recoveries ranging of 94.3-105.4% and 81.8-96.0%, respectively, and MLOQ (Method Limit of Quantification) of 0.05 mg/kg. Biological half-lives of indoxacarb and pymetrozine were 2.9 and 3.2-3.8 days in broccoli, respectively. The lower 95% confidence intervals of dissipation rate constant of indoxacarb were determined as 0.1508 (Field 1) and 0.2017 (Field 2), whereas those of pymetrozine were calculated as 0.1489 (Field 1) and 0.1577 (Field 2). CONCLUSION: The significant differences were not observed between the dissipation rates of indoxacarb and pymetrozine in broccoli. The major factor affecting residue dissipation was the dilution effect by fast growth. The PHRLs for 10 days prior to harvest were recommended as 30.06 (Field 1) and 18.07 (Field 2) mg/kg for indoxacarb, and 4.84 (Field 1) and 4.43 (Field 2) mg/kg for pymetrozine, respectively.

  • Acute methemoglobinemia is induced by various causes, especially ingestion of oxidizing agents such as phenazopyridine, dapsone, and nitrite. Indoxacarb is an oxadiazine insecticide with high insecticidal activity and low mammalian toxicity. It is known to block voltage-gated Na+ channels in insects and mammals, but the mechanism is not yet well understood. We describe a case of a 41-year-old woman with methemoglobinemia that developed following Indoxacarb ingestion, which improved after intravenous injection of methylene blue. This is the first known such case. If signs and symptoms of methemoglobinemia occur after Indoxacarb ingestion, antidotal therapy with methylene blue should be considered as a necessary treatment.

Efficacy of Maxforce and Avion gel baits containing fipronil, clothianidin, and indoxacarb against the German cockroach (Blattella germanica)

  • The present study evaluated the efficacy of Maxforce gel baits containing 0.01% fipronil (Maxforce), 0.05% fipronil (Maxforce Magnum), 1.0% clothianidin (Maxforce Impact), and 0.6% indoxacarb (Avion gel bait; all from Bayer Environmental Science, Clayton, CA, USA) against reference and wild strains (Mobasher, Andisheh, Tamin-e Ejtemaei, and Maskoni) of German cockroach collected in 2016. Test design and bait placement were according to World Health Organization (WHO) procedures for efficacy achieving a 95% mortality rate. Although cockroaches fed on all gel baits, gel baits containing clothianidin and indoxacarb were eaten at higher rates than gel baits containing 0.01% and 0.05% fipronil. The cumulative mortality rates reached 100% for all cockroach strains at 32, 28, 28 and 40 h after eating baits containing 0.01% fipronil, 0.05% fipronil, clothianidin, and indoxacarb, respectively. Efficacy (a 95% mortality rate) after exposure (consumption) of gel baits containing 0.05% fipronil and clothianidin was seen after 28 h, compared with 32 h for 0.01% fipronil and 40 h for indoxacarb. In conclusion, commercial gel baits have improved and may be more effective than spray formulations against German cockroaches.

  • In order to know the residual pattern of some pesticides and predict to the degradation period until below MRL, we experimented chlorothalonil, indoxacarb, lufenuron, metalaxyl and methomyl for Chinese cabbage. They were frequently detected pesticides in Chinese cabbage by NAQS (National Agricultural product Quality management Service) monitoring survey. In this experiment, we sprayed those pesticides 10days before harvest and analyzed 0, 1, 2, 3, 5, 7, 10 day samples to establish logical equation and to calculate $DT_{50}0$. During the cultivating period, the residue amount of chlorothalonil was changed from $55.58\;mg\;kg^{-1}$ (0 day) to $20.08\;mg\;kg^{-1}$ (10 day), $DT_{50}$ was 7.45 days, indoxacarb was $7.85\;mg\;kg^{-1}$ (0 day) to $1.46\;mg\;kg^{-1}$ (10 day), and 4.2 days, lufenuron was $1.57\;mg\;kg^{-1}$ (0 day) to $0.49\;mg\;kg^{-1}$ (10 day), and 5.85 days, metalaxyl was $8.12\;mg\;kg^{-1}$ (0 day) to $0.10\;mg\;kg^{-1}$ (10 day), and 175 days, and methomyl was $11.51\;mg\;kg^{-1}$ (0 day) to $0.80\;mg\;kg^{-1}$ (10 day), and 2.42 days at single dose application, respectively. The $DT_{50}$ of double amount in those pesticides were 9.05 days in chlorothatonil, 7.09 days in indoxacarb, 8.82 days in lufenuron, 3.32 days in metalaxyl, and 2.72 days in methomyl, respectively.

  • Insecticidal activity, systemic and residual effects, and effects on enzyme activities (esterase, acetylcholinesterase, glutathione S-transferase) of indoxcarb were evaluated against Plutella xylostella. The insecticide was very effective against larvae of P. xylostella. Also, indoxacarb showed only residual effect to P. xylostella when applied to vegetable leaves. It inhibited acetylcholinesterase activity, but didn't do esterase and glutathione S-transferase activities.

The Degradation Patterns of Three Pesticides in Perilla Leaf by Cultivation, Storage and Washing (깻잎의 재배, 저장 및 세척에 따른 잔류농약 분해특성)

  • Three pesticides commonly used in perilla leaf were subjected to a field residue trial to ensure safety of terminal residues in the harvest. The residual patterns of three pesticides, which were dimethomorph, indoxacarb and procymidone were examined after applying with the recommended and double dose and their DT50 were calculated. Also degradation patterns of pesticides at storage $4^{\circ}C$ were compared to those at $20^{\circ}C$, and removal rates of pesticides by washing perilla leaf with water were measured. Biological half-lives of dimethomorph, indoxacarb and procymidone were $2.91{\sim}3.11,\;2.53{\sim}3.14\;and\;2.62{\sim}2.92$ days, respectively. During the storage period, the degradation patterns were appeared more obviously at $20^{\circ}C$ than $4^{\circ}C$. Removal rates of dimethomorph, indoxacarb and procymidone were $51.3{\sim}73.3%,\;74.1{\sim}91.3%\;and\;57.9{\sim}81.6%$by various washing methods.

  • This experiment was carried out to investigate the residue patterns of two insecticides, indoxcarb and thiamethoxam, commonly used for Chinese cabbage, under greenhouse conditions. The pesticides were sprayed onto Chinese cabbage at the recommended dose and double of the recommended dose 10 days before the prearranged harvest and then sampling was done at 0.17, 1, 2, 3, 4, 5, 6, 8, 10, and 12 days after spraying. The amounts of their residues in the crop were analyzed with an HPLC. The limit of detection(LOD) of both indoxacarb and thiamethoxam was 0.01 mg $kg^{-1}$ and mean recoveries of indoxacarb and thiamethoxam were from 97.91 to 104.36% and from 97.07 to 105.49%, respectively. Half-lives of indoxacarb and thiamethoxam were 3.4 and 2.3 days at the recommended dose and 3.3 and 3.5 days at the doubled dose, respectively. The ratios of the EDI to ADI by intake the crop harvested 10 days after spraying were less than 4% of their ADIs.

Operator Exposure to Indoxacarb Wettable Powder and Water Dispersible Granule during Mixing/loading and Risk Assessment (Indoxacarb의 수화제 및 입상수화제 살포액 조제 시 농작업자의 노출량 측정 및 위해성 평가)

  • Exposure and risk assessments were conducted to evaluate the relative safety of mixing/loading work of indoxacarb between wettable powder (WP) and water dispersible granule (WG). Hand exposure was monitored using cotton gloves while inhalation exposure was measured using personal air monitor. Method validation for the exposure monitoring was established successfully through several experiments. Limit of determination and limit of quantitation were 0.25 and 1 ng, respectively. $R^2$ of calibration curve linearity was more than 0.9999 and reproducibility was 0.7-6. Recovery of indoxacarb from gloves, solid sorbent and glass fiber filter at three different levels was 81.5-108.8%. Trapping efficiency and breakthrough tests gave 981.5-108.8% of recovery. During mixing/loading procedure, hand exposure amount (75 percentile of 30 repetitions) for indoxacarb WP was 6 folds (459.8 mg/kg a.i) than that of WG (81.4 mg/kg a.i). This result indicates that WG has less drift than WP thanks to its granular type of formulation. Inhalation amount was $10^{-8}-10^{-7}%$ of spray mixture prepared and $10^{-4}-10^{-3}%$ of hand exposure. In inhalation case, no significant differences were observed between two formulations. Margin of safety was calculated for risk assessment using male Korean average body weight and acceptable operator exposure level as the important exposure factors. Mixing/loading procedures for both of the formulations were considered to be of least risk because calculated MOS values were more than 1.

  • From the results achieved by the evaluation of toxicities to an aphid predator, Aphidoletes aphidimyza Rondani eggs with 47 pesticides used for cucumber farming, 22 pesticides including emamectin benzoate showed low toxicity. 12 pesticides with flonicamid among 22 pesticides tested on eggs showed low toxicity (mortality, 0~2.7%) to A. aphidimyza lavae and indoxacarb + teflubenzuron and chlorantraniliprole showed toxicity of 35.4% and 64.6%, respectively. 14 pesticides including indoxacarb + teflubenzuron and chlorantraniliprole demontrating low toxicity against A. aphidimyza lavae showed low toxicity to A. aphidimyza pupae and adults as well.

Selection of Crop Protectant for Friendly Environmental Control of Spodopfera exigua (Lepidoptera: Noctuidae) (파밤나방(Spodoptera exigua)의 환경친화적 방제를 위한 작물보호제의 선발)

  • For the development of friendly environmental control of the beet armyworm, Spodoptera exigua that is too hard to control in the field, 25 insecticides were chosen from 58 registered to the beet armyworm, and bioassayed. There are 12 insecticides with neurotoxical activities, 10 with insect growth regulators and 3 Bacillus thuringiensis products. Among 12 insecticides with neurotoxical activities, mortality of S. exigua was 100% with emamectin benzoate (EC) and indoxacarb (WP) within 3 and 5 days after application, respectively. Otherwise, WG and SC of indoxacarb, Indoxacarb + etofenprox (WP) and pyridalyl (EW) were showed up to 91 %. Methoxyfenozide + spinosad (SC) was better than any other insect growth regulator as 100% mortality within 3 days after application. And methoxyfenozide (WP), tebufenozide (WP) and methoxyfenozide (SC) were 92% by 5 days. However, 3 kinds of B. thuringiensis products were showed under 35% mortality within 5 days from first spray.
