The anti-inflammatory effect of Colocasia esculenta water extract on mouse ear edema models induced by TPA

  • Background and objective: Dermatitis is a chronic disease accompanied by such symptoms as itching and dry skin. The environment and diet can aggravate dermatitis, so attention to skin care is essential. Colocasia esculenta is used in various manners and for different purposes, including with regard to inflammation, aging, and the digestive system. The anti-inflammatory effect of Colocasia esculenta water extract was confirmed using RAW 264.7 macrophages with regard to male ICR mice. Methods: In the case of the ICR mice, 5% 12-O-Tetradecanoylphorbol-13-acetate (TPA) was used to cause inflammation for 7 days, and 100 μL of Colocasia esculenta water extract and panthenol were administered orally for 10 days. In addition, RT-PCR, NO, ELISA was conducted. Results: As a result of reverse transcription polymerase chain reaction (RT-PCR) using RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS), it was found that Colocasia esculenta water extract reduced the expression of inflammatory cytokines. As a result of hematoxylin and eosin (H&E) staining using mouse ear tissue, Colocasia esculenta water extract reduced ear thickness and showed an effect of suppressing ear edema. In addition, compared to the TPA-treated group, the Colocasia esculenta extract-treated group had reduced nitric oxide (NO) production by 18.23 μM and IL-13 production decreased by 136.55 pg/ml. Conclusion: Colocasia esculenta water extract has been shown to be effective in lowering inflammatory cytokine production. These results suggest that Colocasia esculenta water extracts can be used as natural products to treat dermatitis.

Effects of the ethanol extract of Codonopsis Pilosulae Radix on ovalbumin-induced allergic responses in mice (난알부민 유도 알레르기 면역반응에 대한 당삼(黨參)에탄올추출물의 효능 연구)

  • Objectives : The root of Codonopsis pilosula (Fr.) Nannf. (Codonopsis Pilosulae Radix) has been traditionally used as a oriental medicine with an anti-thrombotic, antidiabetic, anticancer, and anti-gastric ulcer effects and immunological adjuvant. In this study, we investigated the effect of 70% ethanol extract of Codonopsis Pilosulae Radix (CPR-E) on ovalbumin (OVA)-induced allergic responses in mice. Methods : Mice were sensitized (1, 8, and 15 days) with OVA and airway challenged(22, 24, 26, 28, and 30 days) to induced allergic responses. CPR-E extract at doses of 50 and 100 mg/kg/body weight was orally administered from days 21 to 30 consecutively. The levels of allergic mediators such as histamine, OVA-specific immunoglobulin (Ig) E, and Th1/Th2 cytokines such as IFN-${\gamma}$ and IL-4 were measured in the sera of mice by ELISA. The histological change of lung tissue was observed by hematoxylin and eosin (H&E) staining. Results : CPR-E extract significantly decreased the serum levels of histamine, OVA-specific IgE, and IL-4 compared with those of OVA control group, but significantly increased the serum level of IFN-${\gamma}$. Based on H&E staining, CPR-E extract inhibited the infiltration of inflammatory cells into lung tissues with histological changes. Conclusions : These results indicate that CPR-E extract has anti-inflammatory and anti-allergic responses through regulating the cytokine balance, suggesting that the extract may be useful for the treatment of allergic inflammatory diseases such as bronchial asthma and allergic rhinitis.

Effect of the water extract of Peucedani Japonici Radix on ovalbumin-induced allergic asthma in mice (난알부민으로 유도된 천식 마우스에서 식방풍(植防風)의 알레르기 면역반응 조절효과 연구)

  • Objectives : The root of Peucedanum japonicum Thunberg (Peucedani Japonici Radix; PJR) has been traditionally used as an herbal medicine for the treatment of anti-headache, anti-paralysis, anti-cancer, vascular protection, and blood pressure regulation. In this study, we investigated the anti-allergic effect of PJR water extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at days 1, 8 and 15 with OVA and airway challenged at days 22, 24, 26, 28, and 30 to induced allergic asthma. PJR-W extract at doses of 100 and 300 mg/kg/body weight (bw) was orally administered during OVA challenge once per a day. The levels of allergic mediators such as immunoglobulin (Ig) E, and Th1/Th2 cytokines (IFN-${\gamma}$ and IL-4) were measured in the sera of mice by ELISA. The histological change of lung tissue was observed with hematoxylin and eosin (H&E) staining. Results : The administration of PJR-W extract significantly decreased the serum levels of IgE, IL-4, and IFN-${\gamma}$ compared with those of OVA control group. In H&E staining, PJR-E extract inhibited OVA-induced airway inflammation and the inflammatory cells infiltration in the peribronchial regions of the lung. Conclusions : These results indicate that PJR-W extract has an anti-inflammatory and anti-allergic effect on allergic response through the down-regulation of allergic mediators, suggesting that this herb may be used as a useful source for the treatment of allergic inflammatory diseases such as asthma.

The Protective Effects of Acupressure at Feng Shi against Chronic Alcohol-induced Muscle Atrophy in Rats (흰쥐의 풍시(風市)혈(GB31) 자극이 알콜성 근위축에 미치는 효과)

  • Purpose : Excessive alcohol causes damage to skeletal muscles, leading to the development of a specific disease entity called alcoholic myopathy. Chronic inflammation is related as an underlying mechanism for the loss of muscle mass induced by alcohol. Pro-inflammatory cytokines such as TNF-α (tumor necrosis factor-α) and IL-6 (interleukin-6) play a role in this process. The acupuncture point Feng Shi (GB 31) is located on the midline of the lateral aspect of the thigh, above the transverse popliteal crease. This acupoint is used for the treatment of weakness, atrophy, numbness, and post-stroke symptoms of lower limbs. The purpose of this study was to investigate the effect of Feng Shi stimulation on muscle atrophy caused by chronic alcohol administration. Method : Young male Sprague-Dawley rats were randomly divided into three groups of eight each: Normal, Control, and GB31. The rats in the Control and GB31 groups were orally given 25 % ethanol (5 ㎖/kg, body weight) daily for 4 weeks. The Normal group was similarly administered saline. The acupressure at Feng Shi was treated to rats in the GB31 group. After 4 weeks, the body weight, muscle weight and cross-sectional area of gastrocnemius were assessed and the histological changes in gastrocnemius muscle fiber were observed by hematoxylin and eosin staining. Moreover, TNF-α and IL-6 expressions were immunohistochemistrically evaluated. Results : Acupressure stimulation at Feng Shi had a protective effect on the weight reduction of the gastrocnemius muscle caused by alcohol intake, and had an effect of suppressing anatomical change in muscle fiber and decreasing the average cross-sectional area. Also, the immunoreactivities of TNF-α and IL-6 in the GB31 group were decreased. Conclusion : These results suggest that acupressure at Feng Shi has protective effects on chronic alcohol-induced muscle atrophy by inhibiting pre-inflammatory proteins such as TNF-α and IL-6.

The Effectiveness of 448-kHz Capacitive Resistive Monopolar Radiofrequency for Subcutaneous Fat Reduction in a Porcine Model

  • Background and Objectives The effectiveness of many physiotherapy modalities in reducing subcutaneous fat has been investigated in numerous previous studies. However, to the best of our knowledge, there have been no attempts to determine the effectiveness of physiotherapy modalities in body contouring. The present report determined the effect of 448-kHz capacitive resistive monopolar radiofrequency (CRMRF) in a porcine model. Materials and Methods This study investigated the effect of selective destruction of the subcutaneous fat layer in abdominal fat tissue using CRMRF. The effects of two types of CRMRF (capacitive electric transfer (CET) and resistive electric transfer (RET)) treatment were evaluated using regular digital photography in addition to thermal imaging evaluation, ultrasound measurement, hematological evaluation, and histologic analyses (H&E (hematoxylin and eosin), Oil red O, and immunohistochemistry staining). Results Preclinical evaluation was performed to obtain the data for comparison of the safety and efficacy of the subcutaneous fat reduction after applying CRMRF using CET and RET. After treatment, the thermal transmission was effective, and a 42-47℃ temperature change was observed in the fat layer while an approximately temperature of 42℃ was confirmed on the skin surface. Moreover, after the application of both types of CRMRF treatment, fibrotic septa were observed in the adipose tissue induced by heat at the treatment sites. TUNEL staining was also performed to confirm the process of apoptosis in the adipocytes. Conclusion These results suggest that both CET and RET for CRMRF treatment are safe and effective for subcutaneous fat reduction in a porcine model.

  • Systemic sclerosis is an autoimmune disease characterized by inflammatory reactions and fibrosis. Myofibroblasts are considered therapeutic targets for preventing and reversing the pathogenesis of fibrosis in systemic sclerosis. Although the mechanisms that differentiate into myofibroblasts are diverse, transforming growth factor β (TGF-β) is known to be a key mediator of fibrosis in systemic sclerosis. This study investigated the effects of extracellular vesicles derived from human adipose stem cells (ASC-EVs) in an in vivo systemic sclerosis model and in vitro TGF-β1-induced dermal fibroblasts. The therapeutic effects of ASC-EVs on the in vivo systemic sclerosis model were evaluated based on dermal thickness and the number of α-smooth muscle actin (α-SMA)-expressing cells using hematoxylin and eosin staining and immunohistochemistry. Administration of ASC-EVs decreased both the dermal thickness and α-SMA expressing cell number as well as the mRNA levels of fibrotic genes, such as Acta2, Ccn2, Col1a1 and Comp. Additionally, we discovered that ASC-EVs can decrease the expression of α-SMA and CTGF and suppress the TGF-β pathway by inhibiting the activation of SMAD2 in dermal fibroblasts induced by TGF-β1. Finally, TGF-β1-induced dermal fibroblasts underwent selective death through ASC-EVs treatment. These results indicate that ASC-EVs could provide a therapeutic approach for preventing and reversing systemic sclerosis.

HISTOPATHOLOGICAL STUDY ON CARDIOMYOPATHY IN EXPERIMENTALLY INDUCED DIABETIC RATS (실험적으로 유도된 당뇨백서의 심근병증에 관한 조직병리학적 연구)

  • Diabetes mellitus revealed a chronic disorder of lipid, carbohydrate and protein metabolism characterized by insulin deficiency, and a striking tendency toward development of atherosclerosis, microangiopathy, nephropathy, neuropathy and recently cardiomyopathy etc. The mechanism of heart failure in patients with diabetic cardiomyopathy is not clear but diabetic cardiomyopathy usually occurs in persons with long standing diabetes. After diabetes induced in made Sprague- Dawley strain rats by injection of streptozotocin(60mg/kg), cardiac tissue with hematoxylin-eosin and Masson's trichrome stain was examined at 3 days, 1, 2, 4, 6 weeks later under light microscope. The results were obtained as follows : 1. In H&E stain of control group, myocardiac cells were shorter than skeletal muscle cell, which was branched out and connected each other at terminal with striation, intercalated disk and nucleus at center of cell. 2. In MT stain of control group, a few of collagen fibrile were seen at periva scular interstium, but wasn't seen between skeletal muscle fiber, and cardiac muscle was seen in various size. 3. In MT stain of experimental group, increased collagen fiber deposition at perivascular interstiums were seen periodically. 4. In MT stain of experimental group, increased collagen fiber deposition at interstitial matrix between perimyocardiac cells were seen at 3 day, 4 weeks and 6 weeks after DM induction. 5. In H&E stain of experimental group, partial degeneration of myocardiac cells was seen after 4 weeks of DM induction. From above results, streptozotocin induced diabetes mellitus increased collagen around perivascular and between intercellular matrix in heart.

  • Structural differences in various divisions of the rabbit colon were investigated using light and scanning electron microscopy. For light microscopic study, various Portions of the colon from seven rabbits (2.5 kg body weight) were fixed in 10% neutral buffered formalin, and paraffin sections were stained with hematoxylin-eosin. Tissues for scanning electron microscopy were fixed in 1% glutaraldehyde-1.5% paraformaldehyde and postfixed in 1% $OsO_4$, dehydrated to 100% alcohol, transfered to isoamilacetate and dried by the critical point method. Subsequently, specimens were coated with gold and viewed with a JSM-35C scanning electron microscope. The colon displays a morphological diversity along its proximo-distal axis. Five regions can be discerned based on the macroscopic and microscopic characteristics. 1) The first segment immediately distal to the cecocolical junction possessing three teniae is approximately 5 cm ($4{\sim}6cm$) in length, and displays irregular folds of the mucosa oriented transversely similar to those of the cecum. 2) The second segment possessing three teniae is about 7 cm ($5{\sim}8cm$) in length, and is characterized by the papilla-like protrusions on the mucosal surface. 3) The third segment, possessing a single tenia is about 16 cm ($12{\sim}20cm$) in length, and also displays the papilla-like protrusions similar to the aforegoing segment. 4) Fusus coli, approximately 4 cm ($3{\sim}5cm$) in length, is free of teniae and exhibits longitudinal folds on the mucosal surface. These four portions together constitute the proximal colon. 5) The distal colon reaches a length of about 58 cm ($53{\sim}55cm$) and shows a pattern of surface irregularities with minor ridges on the mucosal folds.

  • Summary of background data: At the wound from life of the human being very regarding at and a healing process the wound in the portion which is important meantime the many research is accomplished and healing process at the wound the research is small very from the physical therapy territory. Purpose: This study was performed to examine the effects of cold therapy and pulsed ultrasound treatment in the process of acute wound healing. To this end, we measured changes in the length of the wounds, and observed tissues through an optical microscope in order to evaluate the healing process of the acute wounds. Methods: We divided twenty Sprague-Dawley rats into four experimental groups of five rats each and treated them for three days after wound creation. Then we extracted tissues from the wounds on day 6th after wound creation and then took them out for hematoxylin-eosin(HE) staining. We measured changes in the length of the wounds every other day. Result: We were able to detect significant statistical differences in the cold therapy and pulsed ultrasound-treated wounds compared with control wounds. We observed tissues through an optical microscope and found the tissues in cold therapy and pulsed ultrasound-treated wounds healed well. Conclusions: Overall results indicated that the use of cold therapy and pulsed ultrasound treatment were effective in the process of acute wound healing.

  • Objectives : Primo vascular system is known to new circulatory system and suggested as a anatomical structure of meridian system. Primo-vessels are present throughout the whole body. The purpose of this study is to identify primo tissues taken from falciform ligament and to compare with organ surface primo tissue, blood vessel and lymph vessel. Methods : Male Sprague-Dawley rats (8weeks old, 250~320g) used for this study. The medial line of the abdomen was dissected and searched for primo tissues in falciform ligament and on the internal organs using stereomicroscope. We performed serial cross section and histological investigations. The tissues were stained with hematoxylin-eosin and Masson's trichrome. Results : 1. The primo tissues attached on the falciform ligament had white color and length of 5~8mm. 2. We could observe primo tissues are classified with ligament tissues. 3. Histologically, primo tissue on falciform ligament and organ surface primo tissue could be considered same tissue. Conclusions : In this study, we observed primo tissue discovered on the falciform ligament. And we also histologically compared such discovered primo tissue with organ surface primo tissue. Consequently, we could consider that two tissues have histological similarity and possibility of connection in one network system.
