Effects of Yukgunja-Tang on Secretion of Gastric Juice and Movements of Isolated Stomach (육군자탕(六君子湯)이 위액(胃液) 분비(分泌) 및 적출(摘出) 위(胃) 운동(運動)에 미치는 영향(影響)에 관한 연구(硏究))

  • The extracts of Yukgunja-tang which has been used in some of gastroin testinal disorders were examined for secretion of gastric juice and movement of isolated stomach of rabbit. Experiments were performed with the dried extract of Yukgunja-tang (sample I) and the dried extract compound made of each drug of Yukgunja-tang (sample II). The secretion of gastric juice at the doses of 25.0 and 50.0mg/kg p.o. of sample I showed the decreases of 34.3 and 43.3%, respectively, as compared with control group, whereas the secretion at a dose of 76.8g/kg of sample II showed 43.1%. In gastric acid secretion, sample I showed 50.0% decrease at a dose of 50.0mg/kg and sample II showed 46.2% decrease at a dose of 76.8mg/kg. In pepsin output sample I showed 17.8 and 23.4% decreases at a dose of 25.0 and 50.0mg/kg, respectively, whereas sample II showed 22.5% decrease at a dose of 76.8mg/kg. The isolated stomach of rabbit showed dose-responsive contraction by addition of sample I, however the contraction was not observed by sample II.

Effect of Medial Amygdala on Gastric Acid Secretion and Plasma Gastrin Concentration in Conscious Rats (흰쥐에서 내측 편도체가 위산 분비와 혈장 Gastrin 농도에 미치는 영향)

  • This study was undertaken to investigate the effect of medial amygdala on the gastric acid secretion and plasma gastrin concentration in the rats with chronic gastric fistula. After the medial nucleus of amygdala was damaged bilaterally by radiofrequency a. c. through stereotaxically inserted electrodes, the gastric juice was collected in the basal and histamine-stimulated states for 1 hour. The gastric juice was also collected while the medial nucleus of amygdala was stimulated with biphasic square wave in the both states. After the collection of the gastric juice, blood samples were drawn from the abdominal aorta for the radioimmunoassay of plasma gastrin. The results were as follows: 1) The damage of the medial amygdala significantly decreased the gastric juice volume and the acid output in the histamine-stimulated state. 2) The electrical stimulation of the medial amygdala significantly increased the gastric juice volume and the acid output in the histamine-stimulated state, and the acid output in the basal state. 3) The damage of the medial amygdala significantly decreased the plasma gastrin concentration but the electrical stimulation of the medial amygdala did not affect the plasma gastrin concentration. It is therefore suggested that the medial amygdala has a facilitatory influence on the histamine-stimulated gastric acid secretion in rats, and the influence may not be attributed to gastrin release.

Effects of Hyangsayukgunjatang and Hyangsayukgunjatang-ga-kyenegum, moryobun on experimental gastric ulcers in rats (향사육군자탕(香砂六君子湯) 및 향사육군자탕가계내김모려분(香砂六君子湯加鷄內金牡蠣粉)이 백서(白鼠)의 위궤양(胃潰瘍)에 미치는 영향(影響))

  • The following study looked at the effects of Hyangsayukgunjatang and Hyangsayukgunjatang-ga-kyenegum, moryobun on gastric ulcers in rats. In the 1st experiment, ulcers were induced by method of shay's pylorus-ligated in rats. Subjects were administered 2 kinds of medicine, Hyangsayukgunjatang and Hyangsayukgunjatang-ga-kyenegum, moryobun, which were taken orally. The quantity of gastric juice secretion, total acidity of gastric juice, HCI secretion and serumgastrin content were measured and tissues of gastric mucosa epithelium were observed. 1. The quantity of gastric juice secretion in Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treated groups showed significant decrease incomparision with the control group. 2. In relation to total acidity of gastric juice, only Hyangsayukgunjatang -treated groupshowed significant decrease in comparison with the control group. 3. HCI secretion of Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treated groups showed significant decrease in comparision with the control group. 4. Content of serum gastrin, which were present in Hyangsayukgunjatang -treated and Hyangsayukgunjatang-ga-kyenegum, moryobun- treated groups showed significant decrease in comparision with the control group. 5. The degree of ulcer occurrence & parietal cell numbers and chief cell numbers of Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treatedgroups, which were observed by macrographic & histologic method showed signicant decreasein comparison with the control group. Hyangsayukguniatang-treated group prooved to be increasingly effective. From above the experiment results it can be concluded that Hyangsayukgunjatang will be more clinical effective on curing gastric ulcers induced by method of Shay's pylorus-ligated rather than that of Hyangsayukgunjatang-ga-kyenegum, moryobun.

The Effects of Taheebo Extracts on Gastric Secretion and Gastric Injury in Rats (타히보 추출물의 흰쥐 위액 분비 및 위 손상에 미치는 영향)

  • The effects of methanol and water extracts of Taheebo were investigated on gastric secretion, gastric lesion and ulcer in rate. Experimental gastric lesion and ulcer was produced in rats using the following methods : HCl.aspirin-induced lesion, HCl.ethanol-induced lesion, indomethacin-induced ulcer and water-immersion stress ulcer model. In addition, the amount of gastric secretion in pylorus-ligated rats for 4 hours was determined. Water extracts of Taheebo significantly inhibited HCl.aspirin-induced gastric lesion at 1,000mg/kg, po in rats. Likewise, Water extracts of Taheebo caused significant inhibition of indomethacin-induced ulcer at oral dose of 1,000mg/kg. The lesion induced by HCl.ethanol was significantly reduced by both water and methanol extracts of Taheebo. It also showed significant antiulcer activity in water-immersion stress ulcer, respectively. In gastric secretion experiments, methanol extracts of Taheebo also showed significant inhibition of gastric juice secretion, acidity and acid output at doses 500 and 1,000mg/kg. These results may suggest that Taheebo shows antigastritic and antiulcerative action in rats in part by the inhibition of gastric juice secretion and acidity.

Effect of Cingulate Cortical Ablation on Gastric Acid Secretion and Plasma Gastrin Concentration in Rats (흰쥐의 위산분비 및 혈장 Gastrin농도에 미치는 대상회전 제거의 영향)

  • It has been recently reported that cingulate cortex mar facilitate gastric acid secretion, but its facilitatory mechanism on the gastric acid secretion is still unclear. This study was undertaken to investigate the facilitatory mechanism of the cingulate cortex upon gastric acid secretion in rats. Twenty·three male albino rats were divided into the cingulate(N= 13) and the operated control(N= 10) groups. The cingulate group in which cingulate cortex was removed by suction through a slit-shaped opening on each side of, and parallel to, the sagittal suture. In the operated control group, the surgical procedure was ended with the skull opening and the incision of dura mater. The gastric juice was collected via a chronic gastric cannula after 24 hours of fast, with water ad libitum. The juice was collected continuously for 6 hours, starting 3 hours prior to the injection of gastric secretagogue, pentagastrin$(12\;{\mu}g/kg)$ or histamine dihydrochloride $(320\;{\mu}g/kg)$. Three one·hour samples were obtained before ana after the administration of each secretagogue. The two agents were injected separately and subcutaneously at intervals of 1 week, the blood samples were drawn from the abdominal aorta for the radioimmunoassay of postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil. 1) After pentagastrin administration, the volume of gastric juice tended to decrease, but its acidity tended to increase in the cingulate group compared with those of the operated control group. However, there was no any difference in the acid output between the two groups. 2) Histamine-stimulated acid output and volume of the gastric juice of the cingulate group decreased significantly compared with those of the operated control group, while there was not significantly different in the acidity between the two groups. 3) Before pentagastrin or histamine administration, any change was not observed in the gastric acid secretion following the cingulate cortical ablation. 4) Postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil was insignificantly lower in the cingulate group than in the operated control group. It is inferred from the above results that the cingulate cortex exerts a facilitatory influence upon the histamine-stimulated gastric acid secretion in rats, and its influence may not be mediated by the stimulation of gastrin secretion.

  • This study was undertaken to investigate the effect of cingulate cortical ablation upon gastric secretion and its components in rats. 23 male rats were divided into the cingulate(N=9) and the operated control(N= 14) groups. Cingulate cortex was ablated through a slit-shaped opening(1 mm in width, 5 mm in length) which was made symmetrically on both sides of, and parallel to, the sagittal suture by removing a bone flap from frontal bone on each side. In the operated control group, the surgical procedure was ended by the removal of the bone flap. Under light ether anesthesia, experimental animals were placed in a restraining jacket of fine mesh wire and gastric juice was collected for 5 hours via a canula which had been inserted through the anterior abdominal wall into the antral portion of the lumen of the stomach. Volume of the gastric juice was measured, and total acid output and free acid output were titrated with 0.04 N NaOH solution by using phenolphthalein and Topfer's reagent at indicator, and chloride ion output was estimated by means of chloridometer. Results obtained were that volume, total acid output, free acid output and chloride ion output of the gastric juice were higher significantly in the cingulate group than in the operated control group. It is inferred from the above results that the cingulate cortex exerts a fascilitatory influence upon gastric secretion and acid output in rats.

Gastric Ulcer Healing Effects of Dioscorea japonica, Halloysite and Ostrea gigas Mixtures

  • A novel gastric ulcer healing formulation, a mixture of Dioscoreae Rhizoma, Halloysitum Rubrum and Ostreae Testa (Dihaos), was examined for gastric ulcer healing effects. The effect of Dihaos was assessed in various gastric ulcer models in rats. Oral administration of Dihaos significantly reduced HCl-ethanol-induced gastric ulcers. Dihaos also significantly reduced gastric and duodenal ulcers induced by cysteamine. Ostreae Testa decreased secretion of gastric juice and increased the pH of gastric juice. Furthermore, the extracts of Dioscoreae Rhizoma affected the cell proliferation of MKN 74 cells. These results suggest that the healing effect of Dihaos on gastrohemorrhagic lesions results from its protective effect against acid secretion and proliferation of mucosal cells in induced gastric ulcers in rats.

Effects of Intravenous Catecholamine on Gastric Acid, Gastrin and Secretin Secretion in Basal State of the Rat (정맥 주입한 Catecholamine이 흰쥐의 기초상태시 위산, Gastrin 및 Secretin 분비에 미치는 영향)

  • This study was conducted to investigate the effects of epinephrine and norepinephrine on basal gastric acid secretion and plasma gastrin and secretin concentration in the conscious rat. One hundred and eighty-four albino rats with gastric cannula were used after 18 hours or more of fast, with water ad libitum. In a restraint cage for collection of gastric juice, physiological saline (0.9% NaCl) was continuously infused into the jugular vein through a catheter for one hour at a rate of 1 ml/hr (control period). Immediately after the control period, epinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$, norepinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$ or physiological saline (1 ml/hr) was infused for another one hour. Gastric juice was collected at one hour interval for two hours infusion period. Adrenergic antagonists, phentolamine and propranolol were injected into the jugular vein 5 min prior to the infusion of epinephrine or norepinephrine at a dose of 0.2 mg/0.1 ml. Blood was sampled from the jugular vein for the radioimmunoassay of plasma gastrin and secretin after the collection of gastric juice. The results were as follows: 1) Both epinephrine and norephinephrine significantly increased gastric acid output in a dosedependent manner. 2) The effects of epinephrine and norepinephrine on the gastric acid secretion were antagonized by the pretreatment with phentolamine and propranolol. 3) Plasma gastrin and secretin concentrations were not significantly affected by the intravenous infusion of epinephrine and norepinephrine. It can be inferred from the above results that epinephrine and norepinephrine facilitate gastric acid secretion in conscious rats and the mechanism of which is attributed to ${\alpha}\;and\;{\beta}$ adrenergic receptors rather than gastrin and secretin.

  • In a preliminary screening of the plant extracts for the antigastritic action in rats, the extract of Aralia elata(Araliaceae) showed positive activity in HCI plus ethanol induced gastric lesion. Systematic fractions with hexane, chloroform, ethyl acetate and butanol resulted in the most patent activity with the butanol fraction: This butanol fraction at the oral dose of 200 mg/kg exhibited significant inhibition of absolute alcohol induced gastric lesion which was more potent than 100 mg/kg of cimetidine and had significant stimulation of mucus secretion. The butanol fraction showed significant decreases in the ulcer indices of Shay ulcers and inhibition of gastric juice secretion with acid output in pylorus-ligated stomachs of rats. It also suppressed the acetic acid induced gastric ulcer. These results might suggest that the butanol fraction had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and stimulation of mucin secretion in the stomachs of rats.

  • As a result of administration of Coptidis Rhizoma extract and Zingiberis Rhizoma extract to Rat and Rabbit, its effect to the Gastric Juice Volume, Acidity, Pepsin out put of Rat and the movement of Intestine, Pulse Rate of Rabbit was as follows; 1. In the secretion of Gastric Juice, the Juice Volume in Coptidis Rhizoma administered group in contrast to Control Group, has increased in 30mg the increasing rate 21.4%, and has decreased in 300mg the decreasing rate 113.9%. But the Juice Volume in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60, 120mg, and has remarkably decreased in 60mg the decreasing rate 62.9%. 2. The Gastric Acidity in Coptidis Rhizome administered group, in contrast to Control Group, has increased in 30, and 75mg, but no significant value, and has decreased in 300mg the decreasing rate 104.7%. But the Gastric Acidity in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60 120mg, and has remarkably decreased in 60mg the decreasing rate 105.6%. 3. The Pepsin out-put in Coptids Rhizoma administered group, in contrast to Control group, has increased in 30mg the increasing rate 30.3%, and has decreased in 150, 300mg, no significant value. But the Pepsin out-put in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60, 120mg, and has remarkably decreased in 60mg the decreasing rate 86.4%. 4. In the movement of Intestine, the contraction Coptidis Rhizoma administered group, in contrast before the administration, has increased in 12mg, but has begun to decrease in 30mg, and has remarkably decreased in 70mg. The contraction in Zingiberis Rhizoma administered group, in contrast before the administration, has all increased in 30, 60, 120mg. 5. In Pulse Rate, the pulsation, in contrast to Control group after 60, 120 180, 240min. has decreased in Coptidis Rhizoma administered group, and has increased in Zingiberis Rhizoma administered group, but no significant value.
