Effect of Jaeumgeonbigagamtang (JGT) on Restraint-induced Oxidative Stress in Mouse Brain

  • Objectives: This study was performed to investigate the effect of Jaeumgeonbigagamtang (JGT) onrestraint-induced oxidative stress in the mouse brain. Methods: After treatment with JGT, CBC, ROS, MDA, TAC, SOD, activity of catalase, and total GSH content were analyzed. Results: JGT had a strong antioxidant activity by in vitro assay as presented GEAC. JGT treatment significantly ameliorated decrease of blood WBC and increase of platelet count. JGT (50mg/kg) treatment significantly ameliorated increase of MDA and GSH content level in brain tissue. JGT (100mg/kg) treatment significantly ameliorated increase of MDA and activity of TAC level in brain tissue. JGT (200mg/kg) treatment significantly ameliorated increase of ROS, MDA, activity of TAC level and depletion of catalase level in brain tissue. Conclusion: The present study demonstrated antioxidant activity in brain tissue. This result would be consistent with the long clinical efficacy of JGT, and this finding may provide a strong possibility of JGT as a drug candidate for brain-specific multiple disorders and symptoms.

Effects of Kaempferol on Lippolysaccharide-induced Inflammation in Mouse Brain (Kaempferol이 LPS로 유도된 생쥐 중추신경계 염증에 미치는 영향)

  • Objectives : Brain inflammation early activates the microglia and activated microglia secrete a variety of pro-inflammatory cytokines. Kaempferol, which is a flavonoid in Cuscutae Semen, shows a wide range of physiological activities, including neurons protection and anti-inflammatory actions through inhibition of pro-inflammatory mediators. The present study examined the modulatory effect of kaempferol on cytokines [tumor necrosis factor- alpha ($TNF-{\alpha}$), interleukin-1beta ($IL-1{\beta}$) and interleukin-6 (IL-6)] and cyclooxygenase-2 (COX-2) mRNA expression and microglia activation in the brain tissue of the mouse. Methods : Kaempferol was administered orally three doses of 10, 20 and 30 mg/kg respectively, once 1 hour before the lippolysaccharide(LPS) (3 mg/kg, i.p.) injection. Brain tissue was removed at 4 hours after LPS injection. Cytokines and COX-2 mRNA expression in the brain tissue was measured by the quantitative real-time polymerase chain reaction (PCR) method. Iba1 expression was calculated by western blotting method. Microglia was observed with immunohistochemistry. Immunohistochemistry stained microglia was analyzed by using ImageJ software. Results : Kaempferol 20 and 30 mg/kg was significantly attenuated the expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 mRNA. Kaempfrol 10, 20 and 30 mg/kg significantly attenuated COX-2 mRNA expression in the brain tissue. Kaempferol 30 mg/kg significantly suppressed the increase of Iba1 protein expression by LPS. Kaempferol 30 mg/kg significantly decreased the number of microglia in the cerebral cortex and the number and cell size of microglia in the hypothalamic region and the area percentage of ionized calcium binding adaptor molecule 1(Iba1)-expressed microglia in the hippocampus. Conclusions : This results indicate that kaempferol plays an anti-inflammatory role in the brain.

Characterization of tissue-specific mbu-3 gene expression in the mouse central nervous system

  • Mbu-3 is a novel mouse brain unigene that was identified by digital differential display. In this study, expression of the gene was chased through developmental stages and the protein product was identified in the brain. The cDNA sequence was 3,995-bp long and contained an ORF of 745 AA. Database searches revealed that the chicken SST273 gene containing LRR- and Ig-domain was an mbu-3 orthologue. Tissue specificity for the gene was examined in embryos and in brains at post-natal and adult stages. During the embryonic stages, mbu-3 was localized to the central nervous system in the brain and spinal cord. In the early post-natal stages, the gene was evenly expressed in the brain. However, with aging, expression was confined to specific regions, particularly the hippocampus. The protein was approximately 95 kDa as determined by Western blot analysis of brain extracts.

Tumor Segmentation in Multimodal Brain MRI Using Deep Learning Approaches

  • A brain tumor forms when some tissue becomes old or damaged but does not die when it must, preventing new tissue from being born. Manually finding such masses in the brain by analyzing MRI images is challenging and time-consuming for experts. In this study, our main objective is to detect the brain's tumorous part, allowing rapid diagnosis to treat the primary disease instantly. With image processing techniques and deep learning prediction algorithms, our research makes a system capable of finding a tumor in MRI images of a brain automatically and accurately. Our tumor segmentation adopts the U-Net deep learning segmentation on the standard MICCAI BRATS 2018 dataset, which has MRI images with different modalities. The proposed approach was evaluated and achieved Dice Coefficients of 0.9795, 0.9855, 0.9793, and 0.9950 across several test datasets. These results show that the proposed system achieves excellent segmentation of tumors in MRIs using deep learning techniques such as the U-Net algorithm.

Glycyrrhizin Attenuates MPTP Neurotoxicity in Mouse and $MPP^+$-Induced Cell Death in PC12 Cells

  • The present study examined the inhibitory effect of licorice compounds glycyrrhizin and a metabolite $18{\beta}$-glycyrrhetinic acid on the neurotoxicity of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in the mouse and on the 1-methyl-4-phenylpyridinium ($MPP^+$)-induced cell death in differentiated PC12 cells. MPTP treatment increased the activities of total superoxide dismutase, catalase and glutathione peroxidase and the levels of malondialdehyde and carbonyls in the brain compared to control mouse brain. Co-administration of glycyrrhizin (16.8 mg/kg) attenuated the MPTP effect on the enzyme activities and formation of tissue peroxidation products. In vitro assay, licorice compounds attenuated the $MPP^+$-induced cell death and caspase-3 activation in PC12 cells. Glycyrrhizin up to $100{\mu}M$ significantly attenuated the toxicity of $MPP^+$. Meanwhile, $18{\beta}$-glycyrrhetinic acid showed a maximum inhibitory effect at $10{\mu}M$; beyond this concentration the inhibitory effect declined. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid attenuated the hydrogen peroxide- or nitrogen species-induced cell death. Results from this study indicate that glycyrrhizin may attenuate brain tissue damage in mice treated with MPTP through inhibitory effect on oxidative tissue damage. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid may reduce the $MPP^+$ toxicity in PC12 cells by suppressing caspase-3 activation. The effect seems to be ascribed to the antioxidant effect.

Effects of Ginseng Radix plus Crataegi Fructus on the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}$ amyloid peptide(${\beta}A$). (인삼산사복합방(人蔘山査複合方)이 Alzheimer성 치매 병태(病態) 생쥐의 뇌조직 손상에 미치는 효과)

  • Objectives : This research was investigated the effect of the Ginseng Radix plus Crataegi Fructus on the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. Methods : Observed a change of the injury of brain tissue and reduced the infarction area of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Results : 1. The Gin-CF extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. 2. The Gin-CF extract reduced the Tau protein, GFAP protein, and presenilin1/presenilin2 protein (immunohistochemistry) of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Conclusion : These results suggest that the Ginseng Radix plus Crataegi Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Ginseng Radix plus Crataegi Fructus extract for Alzheimer's disease is suggested for future research.

Effect of Maternal Dietary $\omega$3 and $\omega$6 Polyunsaturated Fatty Acids on the Fatty Acid Composition of the Second Generation Rat Brain (어미 쥐의 $\omega$3계 및 $\omega$6계 지방산 식이가 제2세대 쥐의 뇌조직 지방산 성분에 미치는 영향)

  • The change in fatty acid composition in brain tissue of the second generation rats(Sprague-Dawley strain) was studied using four different fat diets(Corn oil=CO, Soybean oil=SO, Perilla oil=PO, Fish oil=FO, 10% by Wt). The experimental diets were started from pregnancy in four different groups, each consisting of 9 rats. The seound generation rats were fed the same diet as their mothers. Animals were anesthetized with ether at 0, 3, 9 & 16 weeks of age. Whole brains were dissected out, brain tissues were, then, homogenized and lipids were extracted from brain tissues. The fatty acid compositions were measured after methylation by gas-liquid chromatography at 0, 3, 9 and 16 weeks of age of offspring. The changes in the relative concentrations of polyunsaturated fatty acids(PUFA) or more specifically docosahexaenoic acid(22 : 6, $\omega$3, DHA), the major $\omega$3 fatty acid component in rat brain at different age were similar to changes in the amount of DNA in brain tissue showing the maximum value during the lactation. The changes in saturated fatty acid(SFA) content showed a contrasting patten to those of PUFA, while monounsaturated fatty acid(MUFA) increased steadily throughout the experimental period. At birth, the relative concentrations of $\omega$3 series fatty acids the relative concentrations of PUFA, MUFA and SFA converged to very similar values respectively regardless of the dietary fatty acid compositions. In brain tissue, it is of value to note that while changes in relative concentrations of linoleic acid (18 : 2, $\omega$6, LA) and arachidonic acid(20 : 4, $\omega$6, AA) showed a precursor-product-like relationship, $\alpha$-linolenic acid(18 : 3, $\omega$3, $\alpha$-LnA) and DHA showed a different pattern. Even when the $\omega$3 fatty acid content in very low in maternal diet(CO), the second generation rat brain tissues appeared to secure DHA content, suggesting an essential role of this fatty acid in the brain. The fact that a large amount of $\alpha$-LnA in the maternal diet did not have a significant effect on the second generation rat brain $\alpha$-LnA content, indicated that DHA seemed essential component for the brain development in our experimental condition. In all groups, the relative content of $\alpha$-LnA in the brain tissues remained relatively constant throughout the experimental period at the very low level. The study of the specific concentrations and essential role(s) of DHA in each parts of brain tissue is needed in more details.

The experimental study on the distribution of radioactive phosphorus-32 to several organs in one-day and seven-days old chicks (초생추(初生雛)의 각(各) 장기(臟器)에 있어서 방사성(放射性) 린(燐)$(^{32}P)$의 흡수분포(吸收分布)에 관한 실험적 연구)

  • Radioactive Phosphorus $(^{32}P)$ was administered intramuscularlly to the newly hatched chicken in the purpose of determination of the uptake and the distribution, as related to sex and hour differences of the several organs of the bodies. $2\;{\mu}\;of\;^{32}P$ was administered to each chick, and the distribution of 32P was observed in 1 hour and 24 hours after administration. In this experiment 80 heads of chicken were used(40 chicken were one day and 40 chicken were 7 days old) and the results obtained as follows: 1. The tissue showed an uptake rate of $^{32}P$ dose per 100 milligram of tissue in one day old chicken, with the following sequence: Males (1 hour): Femur. Liver. G., Muscle. Testis. Brain (24 hour): Femur, Testis, Gastrocnemius Muscle, Liver, Brain. Female(1 hour): Femur, Liver, Gastronemius Muscle, Ovary, Brain. (24 hour): Femur, Liver, Gastrocnemius Muscle, Ovary, Brain. 2. In 1 hour, the uptake rate of $^{32}P$ of the tissues showed significant difference between the male and the female except the gastrocnemius muscle and the brain in one day old group, but they were no significance except the testis and ovary after 24 hours. 3. The distribution of $^{32}P$ of the tissues exhibited higher in 1 hour than in 24 hours except the femur, the brain of the male and female, the brain and gastrocnemius muscle of the female in one day old group. 4. The tissue showed an uptake rate of $^{32}P$ dose per 100 miligram of tissue in 7days old chicken, with the following sequence: Male (1 hour): femur, liver, gastrocmenius muscle, testis, brain. (24 hour): femur, testis, gastrocmenius muscle, liver, brain. Female(1 hour): femur, liver, gastrocmenius muscle, ovary, brain. (24 hour): femur, ovary, liver, gastrocmenius muscle, brain. 5. The distribution of $^{32}P$ of the tissues showed no significant difference between the male and the female except the testis and ovary after 24 hours in 7 days old chicken group. 6. The distribution of $^{32}P$ the tissues exhibited higher in 1 hour in 24 hours except the femur, the brain of the male and the female, the brain and the ovary of the female in 7 days old chicken group.

  • BSE (bovine spongiform encephalopathy)-like symptoms salivation, gait disorder, and the downer syndrome occurred to 8 heads of cattle at six cattle farms in Gyeonju, east eastern Gyeongsang province in 2013. Eight brain tissue samples using a vibration saw (Multimaster$^{(R)}$ FEIN, Germany) were collected referring to "The brain removal technique of transmisible spongiform encephalopathy sampling DVD" by Animal And Plant Quarantine Agency. Brain removal techinque using a vibration saw on the thick, solid frontal bone of cattle is more convenient for incision and removal than an ax for BSE testing.

The Eye/Brain Radioactivity Ratio for Assessment of Graves' Ophthalmopathy (Graves병 안구증에서 $^{99m}Tc-DTPA$ 뇌신티그라피를 이용한 안구/뇌 방사능비에 관한 연구)

  • In Graves' disease, changes in orbital tissue and structure are casued by inflammatory infiltation, which induces increase of capillary permeability and breakdown of blood-tissue barriers. Using the uptake of $^{99m}Tc-DTPA$ in inflammatory lesion, Eye/Brain radioactivity ratios in brain scintigraphy were evaluated in 15 normal controls and 40 Graves' patients. The results were as follows; 1) Eye/Brain radioactivity ratio was significantly higher in Graves' ophthalmopthy group than in control group (p < 0.005). 2) In Graves' ophthalmopathy, Eye/Brain radioactivity ratio was significantly higher in active (progressive) group than in inactive (non-progressive) group (p < 0.05). 3) There was no correlation between class of ATA classification of Graves' ophthalmopathy and Eye/Brain radioactivity ratio. 4) There was no correlation between Eye/Brain radioactivity ratio and serum activity of TBII. In conclusion, Eye/Brain radioactivity ratio using $^{99m}Tc-DTPA$ brain scintigraphy may be useful to determine the activity of Graves' ophthalmopathy and whether treatment of Graves' ophthalmopathy is necessary or not.
