We cultured a tubular marine green alga, originally identified as Capsosiphon groenlandicus (J. Agardh) K.L. Vinogradova, from Amaknak Island, Alaska. The alga had an alternation of heteromorphic generations in which tubular monoecious fronds produced quadriflagellate zoospores and/or biflagellate isogametes. The gametes fused to produce cysts or Codiolum-like zygotes with long, tortuous stalks. Cysts and codiola produced 8-16 aplanospores, which germinated in situ to yield upright fronds. Fronds arising from both aplanospores and zoospores displayed a distinctive development in which non-septate colorless rhizoids from the base of the initially uniseriate, Ulothrix-like filament were transformed into septate uniseriate Ulothrix-like photosynthetic filaments. These transformed filaments then developed new basal non-septate rhizoids. This pattern of rhizoids becoming filaments, which then produced new rhizoids, was repeated to yield a tuft of up to 50 fronds. Periclinal and longitudinal divisions occurred in each filament, starting basally, until the mature tubular thallus was achieved. Pyrenoid ultrastructure revealed several short inward extensions of chloroplast lamellae, each of which was surrounded by pyrenoglobuli. Analysis of ribosomal SSU and ITS sequences placed this alga in the family Ulotrichaceae, order Ulotrichales, together with but as a distinct species from North Atlantic Capsosiphon groenlandicus. Analysis of a partial ITS sequence from authentic Capsosiphon fulvescens, the current name of the type of the genus Capsosiphon, indicated that neither our material nor C. groenlandicus belongs in that genus, and we propose a new genus, Pseudothrix, to accommodate both species. We propose P. borealis for the North Pacific entity formerly called C. groenlandicus and make the new combination P. groenlandica for the Atlantic species.

To examine the effects of two endophytic algae, Mikrosyphar zosterae (brown alga) and Ulvella ramosa (green alga), on the host Chondrus ocellatus (red alga), culture experiments were conducted. Four treatments were made: endophyte-free (Chondrus only), endophyte-M (Chondrus + Mikrosyphar), endophyte-U (Chondrus + Ulvella), and endophytes-M U (Chondrus + Mikrosyphar + Ulvella). After 3 weeks, the relative growth rates (RGRs) of frond lengths and the number of newly formed bladelets were examined. M. zosterae formed wart-like dots on C. ocellatus fronds, whereas U. ramosa made dark spots. The RGRs of frond lengths of C. ocellatus were significantly greater in the endophyte-free and endophyte-M treatment groups than in the endophyte-U and endophytes-M U treatment groups, indicating that the growth of host C. ocellatus was inhibited more by the green endophyte U. ramosa than the brown endophyte M. zosterae. The number of newly produced bladelets was greater in the endophyte-U and endophytes-M U groups than in the endophyte-free and endophyte-M treatment groups. These results indicate that the two endophytes inhibit growth of the host C. ocellatus. The negative effects of U. ramosa on C. ocellatus growth were more severe than those caused by M. zosterae. Furthermore, U. ramosa destroyed the apical meristems of C. ocellatus, whereas M. zosterae did not. On the other hand, C. ocellatus showed compensatory growth in the form of lateral branch production as U. ramosa attacked its apical meristems.
The inflammatory bowel diseases (IBD) including ulcerative colitis and Crohn disease are characterized by chronic inflammation throughout the gastrointestinal tract. The prevalence of IBD has been increasing worldwide, and has sometimes led to irreversible impairment of gastrointestinal structure and functions. In the present study, we identified a new sulfoquinovosylmonoacylglycerols (SQMG) (1) together with two known SQMGs (2 and 3) regulating intestinal inflammation from the brown alga Turbinaria ornata. The anti-inflammatory properties of two bioactive SQMGs, 1 and 2 were evaluated using an in vitro co-culture system consisting of human epithelial Caco-2 cells and PMA (phorbol 12-myristate 12-acetate)-differentiated THP-1 macrophages. Treatment with 1 or 2 inhibited the production nitric oxide and prostaglandin E2 induced by lipopolysaccharide and interferon γ challenge. The expressions of inducible nitric oxide synthase and cyclooxygenase 2 were markedly down-regulated in response to inhibition of nuclear factor κB translocation to nucleus. These findings suggest the potential use of the brown alga T. ornata and its biologically active metabolites SQMGs as pharmaceutical adjuvants in the treatment of inflammation-related diseases, including IBD.
국내에서 규모가 가장 큰 우포늪과 목포늪의 식물플랑크톤 군집의 특성을 파악하기 위해 1998년 1월부터 12월까지 식물플랑크톤의 종조성, 현존량, 우점종, 종 다양성 및 우점도 지수 등을 조사하였다. 우포늪의 식물플랑크톤 군집의 종조성은 녹조류와 규조류에 의해 우점되었으며, 현존량의 최고 밀도는 Oscillatoria sp.와 Micractinium psuillum이 bloom을 형성한 5월과 11월에 나타났다. 현존량의 강별 구성은 겨울을 제외한 전조사기간 동안 녹조류와 남조류가 우점하였고, 남조류는 4월부터 9월까지 그리고 11월에 높은 현존량을 나타내었으며 특히 5월과 7월에 매우 높은 우점도를 나타내었다. 규조류와 유글레나류는 1월과 2월에 현존량의 구성비가 높게 나타났으나 총 현존량이 적어 큰 의미는 없는 것으로 판단되었다. 우점종은 녹조류의 Micractinium pusillum은 가을과 겨울에 우점종으로 출현하였고, 남조류 Oscillatoria sp.가 두 정점에서 모두 4월부터 9월까지 그리고 11월에 우점종으로 출현하였으며, Actinastrum hantzschii var. fluviatile과 Monoraphidium contortum, 그리고 Cryptomonas sp. 등이 일시적인 우점종으로 출현하였으나 규조류는 우점종으로 출현하지 않았다. 종 다양성은 우점도 지수와 역의 상관도를 나타내었으며, 현존량이 최고치인 Micractinium pusillum과 Oscillatoria sp.가 bloom을 형성하여 절대적인 우점도를 나타낸 5, 6월 및 11월에 종 다양성은 가장 낮았다.
다당류 및 플로로탄닌 등을 주성분으로 하는 갈조추출물 NX42가 인지능력 향상에 미치는 영향을 평가하기 위한 in vitro 및 동물실험을 수행하였다. 그 결과 NX42는 acetylcholinesterase에 대하여 온화하지만 용량의존적인 억제효과($IC_{50}=600-700\;{\mu}g/mL$)를 나타내었다. NX42로부터 추출된 플로로탄닌 분획은 현저히 높은 용량 의존적 억제 효과($IC_{50}=54\;{\mu}g/mL$)를 나타내었다. 반면, 플로로탄닌이 제거된 분획과 푸코이단은 억제효과가 없었다. NX42 및 플로로탄닌 분획은 과산화수소에 의해 유발된 산화스트레스 조건 하에서의 SK-N-SH 세포의 파괴를 유의성 있게 억제한 반면, 플로로탄닌이 제거된 분획과 푸코이단은 보호효과를 나타내지 않았다. 스트레스 조건 하에 있는 마우스의 학습능력에 미치는 효과를 평가한 결과, NX42를 섭취한 마우스의 경우에는 섭취하지 않은 경우에 비하여 유의성 있게 개선된 학습능력을 나타내었으며, 이는 in vitro 실험 결과를 바탕으로 볼 때, NX42에 함유된 플로로탄닌의 acetylcholinesterase 억제 활성 및 신경보호활성에 의한 것으로 해석된다.
The design of appropriate chemically modified electrodes should allow development of new voltammetric measurement schemes with enhanced selectivity and sensitivity. Microorganism like algae has high ability to trap toxic and heavy metal ions and different affinities for metal ions. A copper(II) ion-selective carbon paste electrode was constructed by incorporating alga Anabaena into a conventional carbon paste mixture, and then the film of 10% Nafion was coated to avoid the swelling of the electrode surface. Copper ion could be deposited at the 25% algamodified electrode for 15 min without the applied potential while stirring the solution by only immersing the electrode in a buffer (pH 4.0) cot1taining copper(II). Temperature was controlled at $35^{\circ}C$. After preconcentration was carried out the electrode was transferred to a 0.1 M potassium chloride solution and was reduced at -0.6 volt at $25^{\circ}C$. The differential pulse anodic stripping voltammetry was employed. A well-defined oxidation peak could be obtained at -0.1 volt (vs SCE). In five deposition / measurement / regeneration cycles, the responses were reproducible and relative standard deviations were 3.3% for $8.0{\times}10^{-4}M$ copper(II). Calibration curve for copper was linear over the range from $2.0{\times}10^{-4}M$ to $1.0{\times}10^{-3}M$. The detection limit was $7.5{\times}10^{-5}M$. Studies of the effect of diverse ions showed that the coexisting metal ions had little or no effect for the determination of copper. But anions such as cyanide. oxalate and EDTA seriously interfered.
The unicellular green alga Dunaliella salina is a halotolerant eukaryotic organism. Its halophytic properties provide an important advantage for open pond mass cultivation, since D. salina can be grown selectively. D. salina was originally described by E. C. Teodoresco in 1905. Since that time, numerous isolates of D. salina have been identified from hypersaline environments on different continents. The new Dunaliella strain used for this study was isolated from the salt farm area of the west coastal side of South Korea. Cells of the new strain were approximately oval- or pear-shaped (approximately $16-24\;{\mu}m$ long and $10-15\;{\mu}m$ wide), and contained one pyrenoid, cytoplasmatic granules, and no visible eyespot. Although levels of $\beta$-carotene per cell were relatively low in cells grown at salinities between 0.5 to 2.5 M NaCl, cells grown at 4.5 M NaCl contained about a ten-fold increase in cellular levels of $\beta$-carotene, which demonstrated that cells of the new Korean strain of Dunaliella can overaccumulate $\beta$-carotene in response to salt stress. Analysis of the ITS1 and ITS2 regions of the new Korean isolate showed that it is in the same clade as D. salina. Consequently, based on comparative cell morphology, biochemistry, and molecular phylogeny, the new Dunaliella isolate from South Korea was classified as D. salina KCTC10654BP.
Morphological, molecular and chromosomal studies were carried out on Tauya basicrassa, an endemic kelp species distributed on the northern continental coast of the Sea of Okhotsk in Russia. The sporophytes of T. basicrassa grow up to 3-6 m long, 1.8-2.2 m wide, and 6.5-7 kg wet weight. The thallus has a blade with very thick narrow basal portion and thinner and much broader upper portion, which usually splits into 3 bullated lobes. A dwarf laminariacean alga, which did not show any morphological similarity to the other species of the order Laminariales, was found from the same locality. The blade of this alga is thin and soft, reached 26-34 cm long and 6-6.5 cm wide and had 4 longitudinal rows of bullations that covered the entire blade. Molecular analysis showed that the dwarf alga has 100% sequence identity in plastid-encoded RuBisCo spacer, mitochondrial cytochrome c oxidase subunit 1 and nuclear-encoded rDNA genes with normal sporophytes of T. basicrassa, indicating that they are different life forms of the same species. Fluorescent DAPI staining showed that the nucleus in the normal sporophyte was 50-65% larger than those of the dwarf ones. Chromosome count using acetocarmine staining showed n = ca. 20 for the normal sporophytes of T. basicrassa and n = ca. 10 for the dwarf one. These results suggest that the dwarf thallus is a haploid parthenosporophyte of T. basicrassa, which developed in nature. This is the first evidence of parthenosporophytes of the laminariacean algae occurring naturally in the field.
An increase in seawater temperature owing to global warming is expected to substantially limit the growth of marine algae, including Pyropia yezoensis, a commercially valuable red alga. To improve our knowledge of the genes involved in the acquisition of heat tolerance in P. yezoensis, transcriptomes sequences were obtained from both the wild-type SG104 P. yezoensis and heat-tolerant mutant Gy500. We selected 1,251 differentially expressed genes that were up- or downregulated in response to the heat stress condition and in the heat-tolerant mutant Gy500, based on fragment per million reads expression values. Among them, PyHRG1 was downregulated under heat stress in SG104 and expressed at a low level in Gy500. PyHRG1 encodes a secretory protein of 26.5 kDa. PyHRG1 shows no significant sequence homology with any known genes deposited in public databases to date. However, PyHRG1 homologs were found in other red algae, including other Pyropia species. When PyHRG1 was introduced into the single-cell green alga Chlamydomonas reinhardtii, transformed cells overexpressing PyHRG1 showed severely retarded growth. These results demonstrate that PyHRG1 encodes a novel red algae-specific protein and plays a role in heat tolerance in algae. The transcriptome sequences obtained in this study, which include PyHRG1, will facilitate future studies to understand the molecular mechanisms involved in heat tolerance in red algae.
A bacterial strain, labeled $UR11^T$, was isolated from green alga Ulva pertusa collected from Jeju Island, Korea. $UR11^T$ was identified as a gram-negative, rod-shaped, motile by gliding and aerobic bacterial strain with yellow colonies on R2A plates. The strain $UR11^T$ grew over at a temperature range of $10^{\circ}C$ to $30^{\circ}C$ (optimally at $25^{\circ}C$), a pH range of 6.0-11 (optimally at pH 7.0) and a Nacl range of 0.5-5% Nacl (w/v). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain $UR11^T$ was a member of the genus Flavobacterium. Strain $UR11^T$ shared close similarity with F. jejuensis $EC11^T$ (98.0%) F. jumunjinense $HME7102^T$ (96.1%), F. haoranii $LQY-7^T$ (95.3%), F. dongtanense $LW30^T$ (95.1%), and F. ahnfeltiae 10Alg $130^T$(94.9%). The major fatty acids (>5%) were $iso-C_{15:0}$ (33.9%), $iso-C_{15:1}$ G (12.4%), $iso-C_{17:0}$ 3-OH (9.0%), $isoC_{16:0}$ (7.0%) and $iso-C_{15:0}$ 3-OH (6.3%). The major polar lipids were phosphatidylethanolamine, seven unknown aminolipids, two unknown aminopolarlipids and two unknown lipids. DNA-DNA hybridization value was 58% at strain $UR11^T$ with F. jejuensis $EC11^T$. Based on phenotypic, chemotaxonomic and phylogenetic evidence, strain $UR11^T$ represents a novel species of the genus Flavobacterium, for which the name Flavobacterium jocheonensis sp. nov. is proposed. The type strain is Flavobacterium jocheonensis is $UR11^T$ (=KCTC $52377^T$ =JCM $31512^T$).
