Ginsenoside Rd inhibits the expressions of iNOS and COX-2 by suppressing NF-κB in LPS-stimulated RAW264.7 cells and mouse liver

  • Ginsenoside Rd is a primary constituent of the ginseng rhizome and has been shown to participate in the regulation of diabetes and in tumor formation. Reports also show that ginsenoside Rd exerts anti-oxidative effects by activating anti-oxidant enzymes. Treatment with ginsenoside Rd decreased nitric oxide and prostaglandin $E_2$ ($PGE_2$) in lipopolysaccharides (LPS)-challenged RAW264.7 cells and in ICR mouse livers (5 mg/kg LPS; LPS + ginsenoside Rd [2, 10, and 50 mg/kg]). Furthermore, these decreases were associated with the down-regulations of inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 and of nuclear factor (NF)-${\kappa}B$ activity in vitro and in vivo. Our results indicate that ginsenoside Rd treatment decreases; 1) nitric oxide production (40% inhibition); 2) $PGE_2$ synthesis (69% to 93% inhibition); 3) NF-${\kappa}B$ activity; and 4) the NF-${\kappa}B$-regulated expressions of iNOS and COX-2. Taken together, our results suggest that the anti-inflammatory effects of ginsenoside Rd are due to the down-regulation of NF-${\kappa}B$ and the consequent expressional suppressions of iNOS and COX-2.

A Qualitative Study of the Aging Experience among Korean Older Adults Residing in Urban, Rural, and the US Immigrant Context (도시, 농촌, 재미 이민사회에 거주하는 한국노인의 노화 경험에 관한 질적 연구)

  • Korea has undergone rapid modernization and globalization process in the last few decades. The purpose of this study is to explore how shifting traditional values are impacting Korean elderlies and their perceptions of aging experience. In order to understand how changing social and cultural practices are affecting older adults, the similarities and differences among Korean elderlies living in three different geographical locations are examined in a multidimensional comparative framework. Life story interviews were conducted with 30 Korean elderlies (10 urban residents in Korea, 10 rural residents in Korea, and 10 residents in the US). Paying attention to within-group variation, this study particularly focused on analyzing the psychological, social, emotional, and spiritual experiences that made lives meaningful and resilient in old age in spite of physical decline. Based on analysis of narrative, the following major areas were identified as having positive or negative influence on aging experiences: physical health, emotional health, family relationship, work, leisure/social activity, and spirituality. Common themes of aging well across all settings included: "life not indebted to their offspring," "physical and mental health," and "dying well." Spirituality played a critical role in cultivating perspectives on life and accepting aging process, which were rooted in specific religious traditions participants identified (e.g., Christianity or Buddhism). Interesting differences in the meaning of work and leisure were also found between elderlies residing in Korea and the US immigrant context. One unique factor continuing to negatively affect Korean elderly in rural community was related to shame of not obtaining higher education. Implications for future research are also addressed.

Anti-skinaging effects of Gryllus bimaculatus on ERM-CZ100-exposed human diploid fibroblasts (미세먼지 유발 피부노화에 대한 쌍별귀뚜라미의 예방 효과)

  • Purpose: Increasing levels of domestic fine dust (DFD) have emerged as a serious problem that threatens public health by causing chronic respiratory diseases and skin aging. The present study was performed to investigate the inhibitory effects of Gryllus bimaculatus (the two-spotted cricket), which has recently attracted attention as an edible insect in South Korea, on DFD-induced aging and inflammation. Methods: To verify that DFD causes skin aging and investigate the anti-aging effect of an aqueous ethanolic-Gryllus bimaculatus extract (AE-GBE), human diploid fibroblasts (HDF) were treated with 100 ㎍/mL of European reference material (ERM)-CZ100 dust for 24 hrs in the presence or absence of 100 ㎍/ml AE-GBE. Aging and cellular toxicities were assessed by measuring reactive oxygen species (ROS) levels, DNA fragmentation, and β-galactosidase activity. The protein levels of cyclooxygenase (COX) 2, matrix metalloproteinase (MMP)-1, and collagen were measured by western blot, and the mRNA expressions of inflammation-related genes were assayed by quantitative reverse transcriptase polymerase chain reaction. Results: Treatment with ERM-CZ100 induced an aged phenotype in HDF cells, as evidenced by increased ROS levels, DNA fragmentation, and senescence-associated β-galactosidase activity, but cotreatment with AE-GBE significantly reduced these inductions. The mRNA expressions of pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α, induced by ERM-CZ100 were also reduced by AE-GBE cotreatment, which also reduced COX2 expression. Moreover, ERM-CZ100-induced MMP-1 expression and reduced collagen type I expression were recovered by AE-GBE treatment. Conclusion: These results suggest that AE-GBE is a potential treatment for domestic fine dust-induced skin inflammation and inflammaging.

Diagnostic Paper Chip for Reliable Quantitative Detection of Albumin using Retention Factor (체류 인자를 이용한, 알부민의 정량 분석용 종이 칩)

  • Herein we present a diagnostic paper chip that can quantitatively detect albumin without external electronic reader and dispensing apparatus. We fabricated a diagnostic paper chip device by printing wax barrier on the paper and wicking it with citrate buffer and tetrabromophenol blue to detect albumin in sample solution. The paper chip is so simple that we dropped a sample solution at sample pad and measure the ratio of two travel distances of the sample solvent and albumin under the name of retention factor. Our result confirmed that the retention factor was constant in the samples with same concentration of albumin and useful determinant for the measurement of albumin concentration. The paper chip is affordable and equipment-free, and close to ideal point-of-care test in accordance with the assured criteria, outlined by the World Health Organization. We assume that this diagnostic paper chip will expand the concept of colorimetric determination and provide a inexpensive diagnostic method to aging society and developing country.

Arabidopsis Transcription Factor ANAC032 Enhances Salinity and Drought Tolerance

  • The plant-specific NAC transcription factors control various biological processes, including plant development and stress responses. We have isolated an ANAC032 gene, one of the NAC transcription factor family, which was highly activated by multi-abiotic stresses, including high salt and drought in Arabidopsis. Here, we generated transgenic plants constitutively expressing ANAC032 and its knockout to identify the functional roles of ANAC032 in Arabidopsis under abiotic stress responses. The ANAC032-overexpressing plants showed enhanced tolerance to salinity and drought stresses. The anac032 knockout mutants were observed no significant changes under the high salt and drought conditions. We also monitored the expression of high salt and drought stress-responsive genes in the ANAC032 transgenic plants and anac032 mutant. The ANAC032 overexpression upregulated the expression of stress-responsive genes, RD29A and ERD10, under the stresses. Thus, our data identify that transcription factor ANAC032 plays as an enhancer for salinity and drought tolerance through the upregulation of stress-responsive genes and provides useful genetic traits for generating multi-abiotic stress-tolerant forage crops.

Effect of variation of water retention characteristics due to leachate circulation in municipal solid waste on landfill stability

  • This study investigated the effect of water retention characteristics between aged and fresh Municipal Solid Waste (MSW) on the stability of the landfill. A series of transient numerical modeling for the slope of an MSW landfill was performed considering the variation of water retention characteristics due to leachate circulation. Four different scenarios were considered in this analysis depending on how to obtain hydraulic conductivity and the aging degree of materials. Unsaturated hydraulic properties of the MSW used for the modeling were evaluated through modified hanging column tests. Different water retention properties and various landfill conditions, such as subgrade stiffness, leachate injection frequency, and gas and leachate collection system, were considered to investigate the pore water distribution and slope stability. The stability analyses related to the factor of safety showed that unsaturated properties under those varied conditions significantly impacted the slope stability, where the factor of safety decreased, ranging between 9.4 and 22%. The aged materials resulted in a higher factor of safety than fresh materials; however, after 1000 days, the factor of safety decreased by around 10.6% due to pore pressure buildup. The analysis results indicated that using fresh materials yielded higher factor of safety values. The landfill subgrade was found to have a significant impact on the factor of safety, which resulted in an average of 34% lower factor of safety in soft subgrades. The results also revealed that a failed leachate collection system (e.g., clogging) could result in landfill failure (factor of safety < 1) after around 298 days, while the leachate recirculation frequency has no critical impact on stability. In addition, the accumulation of gas pressure within the waste body resulted in factor of safety reductions as high as 24%. It is essential to consider factors related to the unsaturated hydraulic properties in designing a landfill to prevent landfill instability.

Comparison of Nerve Growth Factor Induction and Anti-aging Activity Using Dried Gastrodia and Fermented Gastrodia Extracts (건천마와 발효천마 추출물의 항노화 활성과 신경성장인자 유도 효과 비교)

  • This study was performed to examine the increasing biological activities of both nerve growth factor induction and anti-aging activity of gastrodia (Gastrodia elata) with fermentation process. Antioxidant activities, taken in the fermenting, were investigated to verify utility value of gastrodia for functional food and cosmetics. Fermented gastrodia extracts showed higher antioxidant activities than dried gastrodia extracts. During the routine check of all the practical use potential as functional food, inhibition effect of angiotensin converting enzyme (ACE) and xanthine oxidase (XOase) was tested among water extracted dried gastrodia (WEDG), water extracted fermented gastrodia (WEFG), 70% ethanol extracted dried gastrodia (EEDG) and 70% ethanol extracted fermented gastrodia (EEFG). DPPH was shown as WEDG = $64.14{\pm}0.89%$, WEFG = $66.21{\pm}1.03%$, EEDG = $82.25{\pm}0.52%$, and EEFG = $82.36{\pm}2.37%$. ABTS was shown as WEDG = $54.15{\pm}1.37%$, WEFG = $60.24{\pm}2.25%$, EEDG = $59.18{\pm}1.86%$, and EEFG = $77.17{\pm}4.23%$. Therefore, ACE activity was dramatically inhibited by EERG while there was no difference of XOase inhibition between EEFG and EERG. Nerve growth factor (NGF) activity was measured and indicated about 40% increased neurite growth effect. To conclude, biologically active compounds of gastrodia were increased by fermentation process. It seems to be that ferment gastrodia enhance the use ranges from functional food to fuctional cosmetics, and to all processing industry.

  • The discovery of insulin receptor substrate (IRS) proteins and their role to link cell surface receptors to the intracellular signaling cascades is a key step to understanding insulin and insulin-like growth factor (IGF) action. Moreover, IRS-proteins coordinate signals from the insulin and IGF receptor tyrosine kinases with those generated by proinflammatory cytokines and nutrients. The IRS2-branch of the insulin/IGF signaling cascade has an important role in both peripheral insulin response and pancreatic $\beta$-cell growth and function. Dysregulation of IRS2 signaling in mice causes the failure of compensatory hyperinsulinemia during peripheral insulin resistance. IRS protein signaling is down regulated by serine phosphorylation or protea-some-mediated degradation, which might be an important mechanism of insulin resistance during acute injury and infection, or chronic stress associated with aging or obesity. Under-standing the regulation and signaling by IRS1 and IRS2 in cell growth, metabolism and survival will reveal new strategies to prevent or cure diabetes and other metabolic diseases.

  • The anthropometric measurement and the photometric measurement for have been conducted 358 children from 4 years old to 6 years old to design children's clothing. The characteristics of body type were grasped among total 44 measurement items according to the aging, being extracted seven factors from factor analysis. From cluster analysis among the seven factors the three body types were classified. Type 1 shows low fat body with a small figure and short upper body. The body has a round belly and waist. Type 2 shows the fattest body with an average height. The body has been backed with lower shoulders. Type 3 shows common fat body with a high height and a big figure. The body shows crooked round back with a big upper body and rising shoulders.

  • Insulation diagnostic tests for high voltage motor stator winding were conducted to analyze the insulation characteristics. Test motors were manufactured same factory and year(1996). Insulation characteristics of moisture winding are different from those of deteriorative winding. Insulation resistance and disscipation factor test results are sensitive to moisture winding. AC current, disscipation factor tip-up and PD test results are sensitive to deteriorative winding. Also, Capacitance value for stator winding insulation material has characteristic of increasing in moisture winding.
