Endothelial Cell Products as a Key Player in Hypoxia-Induced Nerve Cell Injury after Stroke

  • Objective : Activated endothelial cells mediate the cascade of reactions in response to hypoxia for adaptation to the stress. It has been suggested that hypoxia, by itself, without reperfusion, can activate the endothelial cells and initiate complex responses. In this study, we investigated whether hypoxia-induced endothelial products alter the endothelial permeability and have a direct cytotoxic effect on nerve cells. Methods : Hypoxic condition of primary human umbilical vein endothelial cells[HUVEC] was induced by $CoCl_2$ treatment in culture medium. Cell growth was evaluated by 3,4,5-dimethyl thiazole-3,5-diphenyl tetrazolium bromide [MTT] assay Hypoxia-induced products [$IL-1{\beta},\;TGF-{\beta}1,\;IFN-{\gamma},\;TNF-{\alpha}$, IL-10, IL-6, IL-8, MCP-l and VEGF] were assessed by enzyme-linked immunosorbent assay. Endothelial permeability was evaluated by Western blotting. Results : Prolonged hypoxia caused endothelial cells to secrete IL -6, IL -8, MCP-1 and VEGF. However, the levels of IL -1, IL -10, $TNF-{\alpha},\;TGF-{\beta},\;IFN-{\gamma}$ and nitric oxide remained unchanged over 48 h hypoxia. Hypoxic exposure to endothelial cells induced the time-dependent down regulation of the expression of cadherin and catenin protein. The conditioned medium taken from hypoxic HUVECs had the cytotoxic effect selectively on neuroblastoma cells, but not on astroglioma cells. Conclusion : These results suggest the possibility that endothelial cell derived cytokines or other secreted products with the increased endothelial permeability might directly contribute to nerve cell injury followed by hypoxia.

Vulnerability Assessment of Water Quality and Aquatic Ecosystem to Climate Change in Korea using Proxy Variables (대리변수를 이용한 한반도 수질 및 수생태계 부문의 기후변화 취약성 평가)

  • This study aims at assessing vulnerability of water quality and aquatic ecosystem to climate change by using proxy variables. Vulnerability to climate change is defined as a function of exposure to climate, sensitivity, and adaptive capacity. Detailed proxy variables were selected considering availability and then standardized by re-scaling concept. After adequate weights were assigned to standardized proxy variables by Delphi technique, vulnerability index was calculated. As results, vulnerability of adjacent regions to coastal areas include water quality and aquatic ecosystem is relatively higher than that of inland areas, and especially adjacent region to the western and southeast seas, and Jeju show high vulnerabilities. Vulnerability in the future was performed based on A1B scenario (IPCC, 2000). Temporally, the increase of vulnerability from 2050s to 2100s may be larger than the increase from 2000s to 2050s. Because vulnerability index was estimated through the relationship among various proxy variables, it is important to consider characteristics of local region with measurements and policies for reduction of sensitivity and enhancement of adaptive capacity on climate change. This study is expected to be useful in planning adaptation measures and selecting priority to the policy on climate change.

  • Large pressure on the growing population has increased rapid change in the LULC (land use/land cover) patterns in the watershed area. Spatial distribution of LULC information and its changes are desirable for any effective planning, managing and monitoring activities. The aim of the study is to produce the 1,50,000 scaled LULC change map for the sub-watershed, Western Moyar, India using the multi-temporal satellite image dataset of IRS LISS III images for the year 1989, 1999, and 2002. About 9 classes are extracted using onscreen visual interpretation techniques for all the three years. The change detection analysis was performed using matrix method for period I (1989-1999) and period II (1999-2002). The study reveals that the changes noticed in period II (1999-2002) is comparatively more than period I (1989-1999), which is dynamic information to protect the sub-watershed area from the deterioration and paves the way to for the sustainable development.

Pop Art-Inspired Fashion (패션에 나타난 팝 아트의 영향)

  • Throughout the history of fashion and fashion collections, fashion design has been influenced by fine arts. Philosophy and concept of fine arts has been inspiration on the development of fashion design which brings on the close interrelation between fine arts and modern fashion. In order to analyze the affect of fine arts such as Pop art on fashion this study inquires into new perspective that considers different social contexts on the premise that acknowledges the essential difference between the genre of fine arts and design. This study researches the influence of Pop art which has been inspiration on fashion designers since the birth in the 1960s and often appears in recent fashion trends. In view of the results achieved in this study, Pop art-inspired fashion does not concern the aesthetic contemplation of everyday life in western society anonymously as in Pop art but deals with pop art as new ideas in a way that adopts images randomly from designer's convenience which is equivalent to the conception of pastiche. In addition, it was inferred that Peter Pan syndrome exert influence as a mental process and Kidult trend operate on Pop art-inspired fashion as a social phenomenon. On the basis of the theoretical background, the formative features in Pop art-inspired fashion from Spring/Summer 2000 to Spring/Summer 2004 collection has been analyzed. The results fall on the following four categories; those are the use of Pop color which resembles the Hard-edge technique in Pop art, direct appropriation of Pop art such as Andy Warhol and Roy Lichitenstein's works on clothes and accessories, adaptation of Pop art's subject using brand names of mass products or icons in mass culture as design motives, and application of representation method in Pop art such as Andy Warhol's silk screen techniques or Tom Wesselman's composition of pictures.

Functional Implication of the tRNA Genes Encoded in the Chlorella Virus PBCV-l Genome

  • The prototype Chlorella virus PBCV-l encodes 11 tRNA genes and over 350 protein-encoding genes in its 330 kbp genome. Initial attempts to overexpress the recombinant A189/192R protein, a putative virus attachment protein, in E. coli strain BL21(DE3) SI were unsuccessful, and multiple protein bands were detected on Western blots. However, the full-length A189/192R recombinant protein or fragments derived from it were detected when they were expressed in E. coli BL21 CodonPlus (DE3) RIL, which contains extra tRNAs. Codon usage analysis of the a189/192r gene showed highly biased usage of the AGA and AVA codons compared to genes encoded by E. coli and Chlorella. In addition, there were biases of XXA/U($56\%$) and XXG/ C($44\%$) in the codons recognized by the viral tRNAs, which correspond to the codon usage bias in the PBCV-1 genome of XXA/U ($63\%$) over those ending in XXC/G ($37\%$). Analysis of the codon usage in the major capsid protein and DNA polymerase showed preferential usage of codons that can be recognized by the viral tRNAs. The Asn (AAC) and Lys (AAG) codons whose corresponding tRNA genes are duplicated in the tRNA gene cluster were the most abundant (i.e., preferred) codons in these two proteins. The tRNA genes encoded in the PBCV-l genome seem to play a very important role during the synthesis of viral proteins through supplementing the tRNAs that are frequently used in viral proteins, but are rare in the host cells. In addition, these tRNAs would help the virus to adapt to a wide range of hosts by providing tRNAs that are rare in the host cells.

Life in Old Age and Intergenerational Dependency: An Exploration of Aging Humanities (노년의 삶과 세대의존: 노년인문학의 탐색)

  • Old age is often compared as climbing mountains which requires walking out of breath, yet with wider vision. And it is also likened to the estuary where the river flows slowly and broadly into the open sea. Socially, old age has been regarded as a symbol of wisdom and reflection, and elderly people often take the role of sage who leads the community. On the other hand, the dementia, gray hair and wrinkles of old age were sometimes perceived as the decline of intellect and vitality. Especially, in the digital age in which technology makes people more sensitive to physical artificiality, the evaluation of the old age becomes more complex and obscure. In other words, some elderly people can not escape from Confucious convention of the elders first, which causes the denouncement by younger generations. On the other hand, some elderly people are becoming more adaptable to the trend of young people, emerging as the new elderly people. The anti-aging movement, early adaptation of IT, bioengineering regimen also strong for the advanced age. However, as the new elderly people are active in many fields of society, they also face intergenerational conflicts in some areas where remains the overlap between young people and them due to the limited openings in economy and culture. This study is a transdisciplinary research which can be called old age humanities. First of all, this paper looks at the aspects of lifestyles and intergeneration conflicts in old age in four Korean and Western literary works about the old people, and also searches how to improve the quality of the later life of old people, Overall, this paper aims to explore the way the old people can achieve the full life with the help of intergenerational dependency through building aging humanities and new communities for old people.

Effect of TJGB on the liver of high-fat diet-fed mice and the viability of HepG2 cells (고지방식이를 급여한 마우스의 간과 HepG2 세포에서 TJGB의 효과에 대한 연구)

  • Objectives: This study was performed to investigate the effect of TJGB on the liver of high-fat diet (HFD)-fed mice and the cell viability of HepG2 cells. Methods: After a week adaptation, 8-week-old C57BL/6N mice were fed with a 45% HFD or normal diet for 3 weeks. For the next 9 weeks, the mice were divided into 6 groups: normal diet group; HFD group; HFD plus orlistat group; HFD plus Ephedra sinica Stapf (ES) group; HFD plus low dose of TJGB group; HFD plus high dose of TJGB group. To estimate the effect of TJGB in the liver of HFD-fed mice, the protein expressions of phospho-acetyl-CoA carboxylase (p-ACC) and liver X Receptor (LXR) were determined by Western blot assay. The cell viability of ES and TJG was also evaluated in HepG2 cells. Results: The administration of TJGB had little effect on the protein expressions of p-ACC and LXR in the liver of HFD-fed mice. And the cytotoxicity was showed above 7.8 ㎍/mL in HepG2 cells. Conclusion: Further research is needed to evaluate the mechanism of TJGB on hepatic steatosis and cytotoxicity in HepG2 cells.

Variation of Lactate Dehydrogenase Isozymes in Angelfish (Pterophyllum scalare) according to Acute Environmental Change (급격한 환경변화에 대한 angelfish (Pterophyllum scalare) 젖산탈수소효소 동위효소의 변화)

  • In this study, the properties and gene expression of the lactate dehydrogenase (EC 1.1.1.27, LDH) isozyme were studied in angelfish (Pterophyllum scalare) - known for their adaptation to the low oxygen environment of the tropics - which were acclimated to acute temperature change ($27{\pm}0.5{\rightarrow}18{\pm}0.5^{\circ}C$) and dissolved oxygen (DO) change ($6{\pm}1{\rightarrow}18\;ppm$) for 2 hours. The properties of the LDH isozymes were confirmed in the native-polyacrylamide gel electrophoresis, Western blot analysis and enzyme activity measurement. Liver- and eye-specific Ldh-C gene were expressed in liver, eye and brain tissues. Through Western blot analysis, the LDH $A_4$ isozyme was shown to have a more cathodal mobility relative to the $B_4$ isozyme. In the liver tissue, the LDH $A_4$ isozyme increased with temperature drop while the $B_4$ isozyme decreased. The LDH $A_4$ and $C_4$ isozymes increased with DO increment, while the $B_4$ isozyme decreased. In the eye tissue, the LDH $A_4$ and B4 isozymse increased with temperature drop while the $B_4$ isozyme decreased. The LDH $A_4$ and $B_4$ isozymes increased with DO increment, but the $C_4$ isozyme and isozymes including the subunit C decreased. In the heart tissue, LDH activity increased with DO increment, as well as the LDH $B_4$ isozyme. In the brain tissue, the LDH $A_4$ and $B_4$ isozymes increased with temperature drop. The LDH $B_4$ isozyme increased with DO increment. Accordingly, since the liver- and eye-specific Ldh-C are influenced by changes in DO and the LDH $B_4$ and $C_4$ isozymes are relatively controlled in the liver and eye tissues, the $C_4$ isozyme can be considered to have a lactate oxidase function.

Blue-light Induces the Selective Cell Death of Photoreceptors in Mouse Retina (청색광에 의한 마우스 망막손상에서 선택적 광수용세포의 사멸)

  • Purpose: The study was conducted to determine that photoreceptors of mouse having pigment in RPE(retinal pigment epithelium) can be damaged by blue-light and apoptosis of specific cells among photoreceptors are induced by blue-light, and to assist the investigation of AMD(Age-related macular degeneration) mechanisms and development of AMD drugs. Methods: C57Black mice were injured by irradiating $2800{\pm}10lux$ of 463 nm LED for 6 hours after 24 hours dark adaptation and eyes were enucleated 1, 3, 7 days. Damage of retina induced by blue-light was determined by western blotting GFAP(Glial fibrillary acidic protein) expression. In the light-injured retina, cell death of photoreceptors was determined by TUNEL(Terminal deoxynucleotidyl transferase dUTP nick end labeling) assay. ERK(Extracellular signal-regulated kinases), JNK, and SRC(sarcoma) expression were assessed by western blotting to determine regulated pathway. Blue light-injured retina were immunostained with antibodies against Opsin and Rhodopsin as markers of photoreceptors to compared the damage cone cells with rod cells. Results: After 1, 3 and 7 days from exposure to blue-light, thickness of retina was more decreased than control, and more decreased at nuclear layer than at outer plexiform layer and GFAP expression was increased day 1 after blue-light injured. While phosphorylated ERK and SRC protein expressions at day 1 were increased after blue-light injured, phosphorylated c-JUN was decreased. Fluorescence intensity analysis showed that markers of cone and rod cells were decreased after blue-light injured and Opsin was more decreased than Rhodopsin. Conclusions: The study suggests possibilities that the blue-light promotes retinal damage and causes apoptotic cell death via ERK and SRC pathway in mouse retina, and blue-light retinal damage is more induced cone cells apoptosis than rod cells directly.

Sugarcane bagasse as exclusive roughage for dairy cows in smallholder livestock system

  • Objective: The study aimed to evaluate sugarcane bagasse as roughage in lactating cow on feed intake, digestibility, ingestive behavior, milk production and composition, and microbial protein synthesis. Methods: Ten Girolando cows at initial body weight of $450{\pm}25.6kg$ and at $143.7{\pm}30.7days$ in milk were assigned in two $5{\times}5$ Latin square designs. Five 21-day experimental periods were adopted ($1^{\circ}$ to 14-day: diets adaptation period; $15^{\circ}$ to 21-day: data collection and sampling period). The diets consisted of four different levels of sugarcane bagasse (45%, 50%, 55%, and 60%) and a control diet, commonly adopted in the region, based on spineless cactus (25% sugarcane bagasse), formulated to meet 12 kg/d milk yield. Results: The dry matter (DM), organic matter (OM), and total digestible nutrients intakes and DM and OM digestibilities observed for 45% and 50% bagasse inclusion were similar to control diet, while that 55% and 60% bagasse inclusion were lower. Cows fed control diet, and bagasse diets of 45%, and 50% levels had the nutritional requirements attended, that guaranteed 12 kg/d of milk yield. The crude protein intake and digestibility of cows fed 45%, 50%, and 55% of bagasse inclusion were similar to control diet. The neutral detergent fiber (NDF) intake and digestibility differ for all bagasse diets related to control diet, while the non-fiber carbohydrates intake and digestibility for cows fed 45% of bagasse were similar for control diet. The intakes and digestibilities of nutrients decreased linearly in function of bagasse inclusion; NDF and indigestible NDF intakes did not vary. The ruminating time, feeding and rumination efficiency, microbial protein synthesis and milk yield decreased linearly with sugarcane bagasse inclusion. Conclusion: Sugarcane bagasse decreases milk production; however, its inclusion level in between 45% to 50% associated to concentrate could replace diets based on spineless cactus for crossbred dairy cow's producing 12 kg/d of milk.
