Inhibitory Effect of Hizikia fusiformis Solvent-Partitioned Fractions on Invasion and MMP Activity of HT1080 Human Fibrosarcoma Cells

  • Matrix metalloproteinases (MMPs) are endopeptidases that take significant roles in extracellular matrix degradation and therefore linked to several complications such as metastasis of cancer progression, oxidative stress, and hepatic fibrosis. Hizikia fusiformis, a brown algae, was reported to possess bioactivities, including but not limited to, antiviral, antimicrobial, and anti-inflammatory partly due to bioactive polysaccharide contents. In this study, the potential of H. fusiformis against cancer cell invasion was evaluated through the MMP inhibitory effect in HT1080 fibrosarcoma cells in vitro. H. fusiformis crude extract was fractionated with organic solvents, $H_2O$, n-BuOH, 85% aqueous MeOH, and n-hexane (n-Hex). The non-toxicity of the fractions was confirmed by MTT assay. All fractions inhibited the enzymatic activities of MMP-2 and MMP-9 according to the gelatin zymography assay. Cell migration was also significantly inhibited by the n-Hex fraction. In addition, both gene and protein expressions of MMP-2 and -9, and tissue inhibitor of MMPs (TIMPs) were evaluated by reverse transcription-polymerase chain reaction and Western blotting, respectively. The fractions suppressed the mRNA and protein levels of MMP-2, MMP-9 while elevating the TIMP-1 and TIMP-2, with the $H_2O$ fraction being the least effective while n-Hex fraction the most. Collectively, the n-Hex fraction from brown algae H. fusiformis could be a potential inhibitor of MMPs, suggesting the presence of various derivatives of polysaccharides in high amounts.

  • High-pressure (HP) injection injury to the upper extremity often causes a very serious clinical problem, leading to poor outcomes, including amputation, so that a true surgical emergency is required. The outcomes can be improved with emergent wide surgical debridement. However the diagnosis of these injuries is often delayed due to underestimated evaluation at first appearance and lack of common knowledge of the seriousness of this injury. The type and pressure of the infecting material is an important factor in prognosis and organic solvents infected pressure injury can cause poor outcome and increased amputation rate. In this case, we report on reconstruction of HP oil-based paint injection injuries of the finger using T-shaped pedicles and multiple venous anastomoses. In this concept, arterial flow can be maintained by the reverse flow of distal anastomosis when there is difficulty with the proximal anastomosis. And venous flow can be preserved by deep and superficial vein anastomosis. This concept has various advantages including preserving patency of the pedicle in chronic vasculopathy or trauma cases and maintaining the arterial flow by the reverse flow of distal anastomosis and can improve the free flap survival by a two vascular anastomosis system.

  • 2,2-Bis(3-amino-4-hydroxyphenyl)hexafluoropropane(BAPAF)과 pyromelltic dianhydride(PMDA), 3,3',4,4'-benzophenontetracarboxylic dianhydride(BTDA), 4,4'-(hexafluoroisopropylidene)-bis(phthalic anhydride)(6FDA)을 사용하여 용해성 폴리이드를 합성하였다. 합성된 폴리이미드를 여러가지 유기용제(acetone, NMP, DMAc, DMSO, THF, DMF)를 이용하여 용해성 실험을 행한 결과 열적 이미드화한 경우에는 녹지 않고 팽윤 되었으나 화학적 이미드화한 경우에는 모든 구조가 여러 유기용제에 대해 좋은 용해성을 나타냈다. 이러한 용해도 차이는 화학적 이미드화할 경우 탈수제로 들어간 acetic anhydride에서 나온 $CH_3COO^-$가 하이드록실기와 에스테르 반응을 하여 분자사슬에 결합한데 기인한다. 합성된 폴리이미드의 유리전이온도는 열적 이미드화 경우가 더 높은 값을 나타냈으며 TGA 측정 결과 모든 구조가 $300^{\circ}C$ 이상까지 열적 안정성을 나타내었다. 합성된 폴리이미드의 결정성은 XRD를 이용하여 조사한 결과 모두 무정형으로 나타났다.

Characterization of Heterogeneous Interaction Behaviour in Ternary Mixtures by Dielectric Analysis: The H-Bonded Binary Polar Mixture in Non-Polar Solvent

  • The heterogeneous association behaviour of various concentration binary mixtures of mono alkyl ethers of ethylene glycol with ethyl alcohol were investigated by dielectric measurement in benzene solutions over the entire concentration range at 25 ${^{\circ}C}$. The values of static dielectric constant $\epsilon_0$ of the mixtures were measured at 1 MHz using a four terminal dielectric liquid test fixture and precision LCR meter. The high frequency limiting dielectric constant $\epsilon_\infty$ values were determined by measurement of refractive index $n_D$ ($\epsilon_\infty\;=\;n_D\;^2$). The measured values of $\epsilon_0$ and $\epsilon_\infty$ were used to evaluate the values of excess dielectric constant $\epsilon^E$, effective Kirkwood correlation factor $g^{eff}$ and corrective correlation factor $g_f$ of the binary polar mixtures to obtain qualitative and quantitative information about the H-bond complex formation. The non-linear behaviour of the observed $\epsilon_0$ values of the polar molecules and their mixtures in benzene solvent confirms the variation in the associated structures with change in polar mixture constituents concentration and also by dilution in non-polar solvents. Appearance of the maximum in $\epsilon^E$ values at different concentration of the polar mixtures suggest the formation of stable adduct complex, which depends on the molecular size of the mono alkyl ethers of ethylene glycol. Further, the observed $\epsilon^E$ < 0 also confirms the heterogeneous H-bond complex formation reduces the effective number of dipoles in these polar binary mixtures. In benzene solutions these polar molecules shows the maximum reduce in effective number of dipoles at 50 percent dilutions. But ethyl alcohol rich binary polar mixtures in benzene solvent show the maximum reduce in effective number of dipoles in benzene rich solutions.

  • 여러 종류의 전해질이 존재할 때, 수계 및 비수계 분산매체에서 저온균일침전법으로 제조된 침상형태의 일차입자를 갖는 나노 크기의 루틸상 TiO$_2$분말에 대한 분산 안정성을 조사하였다. 제타전위 측정은 수계 및 비수계 분산매체에 전해질 첨가가 TiO$_2$입자 표면의 전위 역전을 유발하는 것을 보여주었다. 비수계 분산매체에 분산되어 있는 TiO$_2$입자 사이에 작용하는 정전기적 반발력은 수계 분산매체에서보다 크게 관찰되었고, 이것은 점도, 유전 상수와 같은 유기 용매의 물리적 특성과 밀접한 연관이 있음을 알 수 있었다. pH, 전해질의 농도와 이온의 원자가는 TiO$_2$입자의 표면전위를 크게 변화시켰고, TiO$_2$입자의 분산 거동을 사실상 주도하였다.

  • 버섯에서 추출되는 생리활성물질얀 ${\beta}$-glucan은 탁월한 항암 효과를 갖는 물질이다. 특히 Agaricus blazei에 $\beta$-glucan이 많이 포함되어 있는 것으로 보고되어져 있다. 본 연구의 목적은 마이 크로웨이브를 사용하여 아가리쿠스버섯으로부터 ${\beta}$-glucan을 추 출하는 공정의 추출효율을 극대화하는 것으로, 다양한 추출시간 과 마이크로웨이브 출력을 이용하여 최적 운전조건을 결정하였 다. 최적 마이크로웨이브의 최적 출력은 200, 500, 650 watt의 경우에서 실험하여 결정하였다. 추출물질의 구조와 농도 분석은 nuclear magnetic resonance spectroscopy와 infrared spectroscopy 를 이용하여 수행하였다 본 실험 결과로 마이크로웨이브를 이 용한 추출의 다당류 수율이 전통적 인 열수추출방법보다 향상됨을 확인하였고, 소량의 용매와 짧은 추출 시간으로 향상된 수율을 얻을 수 있음을 알 수 있었다.

코아 가교 양친성 고분자 나노입자를 이용한 고함량 유용 약물 담지 고분자 나노입자 제조 (Preparation of Valuable Compounds Encapsulated Polymer Nanoparticles with High Payload Using Core-crosslinked Amphiphilic Polymer Nanoparticles)

  • 본 연구에서는 반응성 비닐기를 가지고 있는 반응성 양친성 고분자 전구체(Reactive Amphiphilic Reactive Polymer Precursor) (RARP)를 이용하여 제조된 소수성 세그먼트들이 가교된 코아 가교 양친성 고분자(Core-crosslinked Amphiphilic Polymer) (CCAP) 나노입자와 나노침전법을 사용하여서 소수성 유용물질을 고함량으로 담지할 수 있는 새로운 공정을 제안하였다. 극성이 각기 다른 유기용매(에탄올, 아세톤, 테트라하이드로퓨란(THF))들과 소수성 세그먼트 분자량이 다른 CCAP를 사용하여서, 모델 유용 약물인 ${\alpha}$-tocopherol의 담지 효율, 담지량 및 약물 담지 나노입자의 크기와 안정성 변화를 조사하였다. 소수성 세그먼트 분자량이 큰 CCAP와 소수성 용매인 THF를 용매로 사용한 경우에 가장 높은 유용 약물 담지량, 담지 효율을 나타내는 안정한 나노입자가 형성이 되었다. 즉 CCAP 나노입자들의 물리적 화학적으로 견고한 나노 구조로 인해서 33 wt%의 높은 담지량과 97% 이상의 담지 효율을 가지면서 물속에서 70 nm의 크기의 안정한 유용 약물 담지 고분자 나노입자를 제조할 수 있었다.

크롬(VI)-피라진 착물을 이용한 알코올류의 산화반응과 메카니즘 (Mechanism for the Oxidation Reaction of Alcohols Using Cr(VI)-Pyrazine Complex)

  • 6M HCl 용매 하에서 피라진과 chromium (VI) trioxide의 반응을 통하여 PZCC (크롬 (VI)-피라진 착물)을 합성하였다. 적외선분광광도법(IR), 유도결합 플라즈마(ICP) 등으로 구조를 확인하였다. 여러 가지 용매 하에서 PZCC을 이용하여 벤질알코올의 산화반응을 측정한 결과, 용매의 유전상수 값이 증가함에 따라 반응수율이 증가했다. 그 순서는 N,N'-디메틸포름아미드 > 아세톤 > 클로로포름 > 시클로헥센이었다. 산($H_2SO_4$) 촉매를 이용한 N,N'-디메틸포름아미드 용매 하에서, PZCC은 벤질알코올(H)과 그의 유도체들($p-OCH_3$, $m-CH_3$, $m-OCH_3$, m-Cl, $m-NO_2$)을 효과적으로 산화시켰다. 전자받개 그룹들은 반응속도가 감소한 반면에 전자주개 치환체들은 반응속도를 증가시켰고, Hammett 반응상수(${\rho}$)값은 -0.70 (308 K)이었다. 본 실험에서 알코올의 산화반응 과정은 속도결정단계에서 수소화 전이가 일어났다.

소나무 부위별 추출물의 항산화 활성 (Antioxidant Activities of Extracts from Different Parts of the Pine Tree)

  • This study was conducted to investigate the antioxidant activities of extracts from various parts of the pine tree, which is known as a good source of functional food material. While ethanol extraction yields of pine bud and cone were higher than water extraction yields of pine bud and cone, water extraction yield of pine needle was higher than ethanol extraction yield of the pine needle. The content of polyphenols in the pine cone ethanol extract was 5 times higher than that in the pine bud and needle. Further, the content of flavonoids in the pine cone ethanol extract was 8 times higher than that in the pine bud and needle. DPPH radical scavenging effect of the pine cone ethanol extract was 3~5 times higher that of the pine bud and needle extract. Regardless of the extraction solvents, trolox equivalent antioxidant capacity (TEAC) and ferric reducing antioxidant power (FRAP) of the pine cone were stronger than those of the other parts of the pine tree. Taken together, it can be expected that the pine cone can be practically used as an antioxidant substance in food and beauty industries.

자생 산사(Crataegus pinnatifida BUNGE)씨의 추출 용매에 따른 항산화 활성 (Antioxidative Activity of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts Using Various Solvents)

  • The purpose of this study was to examine the antioxidative activity of feral haw (Crataegus pinnatifida BUNGE) seed extracts using 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v), n-butanol and ethyl acetate (EA). The total phenol content of the five extracts ranged from 37.29 mg/g to 55.53 mg/g. Moreover, the content was high in the 70% methanol and, 70% ethanol extracts, but low in the n-butanol extract. On the contrary, the total flavonoid content decreased in the order of n-butanol (2.93 mg/g), EA (2.67 mg/g), 70% methanol (1.00 mg/g), 70% ethanol (0.88 mg/g) and CM (0.67 mg/g) extracts. The $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay and, superoxide dismutase (SOD) like ability decreased in the order of 70% methanol, 70% ethanol, CM, EA and, n-butanol extracts; further, a similar tendency was also observed in the total phenol contents. Overall, these results indicated that the antioxidative activity of feral haw seeds was closely related to the total phenol and flavonoid contents. Therefore, haw seeds might be usefully applied to natural antioxidants as well as functional foods.
