Antioxidant and Antihemolytic Activities of Ethanol Extracts of Carpesii Fructus and Farfarae Flos (학슬 및 관동화 에탄올 추출물의 항산화 및 항용혈 효과)

  • Objectives : To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Carpesii Fructus and Farfarae Flos. Methods : Aerial parts of Carpesii Fructus and Farfarae Flos were extracted with 80% ethanol. Antioxidant activity of Carpesii Fructus or Farfarae Flos extract was evaluated by employing three different assays, i.e., 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzthiazoline-6-suphonic acid) diammonium (ABTS) scavenging and reducing power activities. Also, anti-hemolytic activity of Carpesii Fructus or Farfarae Flos extract was determined using [2,2'-azobis (2-methylpropionamidine) dihydrochloride (AAPH)]-induced hemolysis, glutathione (GSH) depletion and malondialdehyde (MDA) formation in normal rat red blood cells (RBC) or plasma. Results : The extracts obtained from Carpesii Fructus and Farfarae Flos dose-dependently increased the scavenging activity on DPPH- or ABTS-induced radicals and the reducing power activities. Carpesii Fructus and Farfarae Flos were similar to the scavenging activity and the reducing power of butylated hydroxy anisole effect at high concentration ($1,000{\mu}g/mL$). RBC oxidative hemolysis and plasma MDA formation induced by AAPH were significantly suppressed by the extracts of Carpesii Fructus and Farfarae Flos in a dose-dependent manner. Also, Carpesii Fructus and Farfarae Flos extracts prevented the depletion of cystosolic antioxidant GSH in RBCs. Carpesii Fructus generally had better than the free radical scavenging activity, the reducing power and anti-hemolytic effects of Farfarae Flos. Conclusions : These results suggest that Carpesii Fructus and Farfarae Flos may have value as the potential antioxidant and anti-hemolytic medicinal plant.

Comparison of Anti-oxidant Activity and Marker Compounds in Eucommiae Cortex Samples from Regional Groups (지역별 두중(杜仲)의 지표성분 및 항산화 활성 비교)

  • Objectives : It is necessary to manage herbal medicines based on effectiveness by comparing the efficacy of herbal medicines by region. In this study, we compared anti-oxidative activity and marker compounds of Eucommiae Cortex by regional groups. Methods : Eucommiae Cortex grown and harvested in Gangjin, Sancheong, Yeongwol, Jangsu, and Jecheon were used. Eucommiae Cortex was extracted in distilled water at $100^{\circ}C$ for 3 hours, and filtered. filtered. Extract samples were freeze-dried at $-80^{\circ}C$. Comparison of anti-oxidant activity in Eucommiae Cortex samples from regional groups was measured in 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between regional groups of Eucommiae Cortex. In addition, high-performance liquid chromatography (HPLC) analysis was conducted to compare pinoresinol diglucoside concentration by regional groups. Results : HPLC analysis found that pinoresinol diglucoside concentration, widely known as the marker compound of Eucommiae Cortex, was the highest in Gangwon Yeongwol. There was a significant difference in anti-oxidative activity of Eucommiae Cortex between regional groups as discovered in DPPH, ABTS and FRAP assays. DPPH free radical scavenging was the highest in Jeonbuk Jangsu. ABTS free radical scavenging was the highest in Jeonbuk Jangsu and Chungbuck Jecheon. FRAP was the highest in Jeonbuk Jangsu. Conclusions : Although pinoresinol diglucoside concentration was high, anti-oxidative activity was not proportionately high. Pinoresinol diglucoside concentration was the highest in Gangwon Yeongwol. Anti-oxidative activity was the highest in Jeonbuk Jangsu.

Antioxidant Activities of Hot Water and Ethanol Extracts from Portulacae Herba (마치현 열수 및 에탄올 추출물의 항산화 활성)

  • Objectives : This study aims to provide basic data about Portulacae Herba (PH) extracts as natural antioxidants by considering diverse antioxidant activities of PH depending on solvents. Methods : The samples of PH were pulverized, and A hot water and a 70% EtOH were stir-extracted for two hours three times repeatedly in a water bath with a temperature of 95 degrees and at room temperature respectively to measure 7 kinds of antioxidant activities. Results : There were significant differences in total phenol content, because the total phenol content of the 70% EtOH extract was higher than the hot water extract's, and the total flavonoid content of the 70% EtOH extract($4.40{\mu}g/mg$) was nearly 3.8 times higher than the hot water extract's($1.16{\mu}g/mg$). DPPH and ABTS radical scavenging activities were 70% EtOH extract showed a little higher activity than the hot water extract, and at a concentration of $500{\mu}g/mg$, the highest scavenging activity was found in the 70% EtOH extract, not in the control group. Hydroxyl radical and $Fe^{2+}$ chelatingactivities were slightly higher in hot water extract than in 70% EtOH extract, and increased in a dose-dependent manner. Nitrite scavenging activities increased dose-dependently in the hot water and the 70% EtOH extract, regardless of the pH level, and scavenging activity of the 70% EtOH extract was higher at pH 1.2 than at pH 3.0. Conclusions : In conclusion, it is thought that PH hot water and 70% EtOH extract have antioxidant activities, and can be used as natural antioxidants in future.

Anti-oxidant Activity and Skin Barrier Function Measured in Different Solvent Fractions Obtained from Agrimoniae Herba (용아초(龍芽草) 유기용매 분획물의 항산화 및 피부 장벽 개선 효과)

  • Objectives : Agrimoniae Herba is a herbal medicine widely distributed in Asia and contains flavonoids including catechin, quercitrin, rutin, hyperoside, and quercetin. This study aimed to investigate the anti-oxidant activity and skin barrier function of different solvent fractions (Hexane; methylene chloride, MC; ethyl acetate, EA; n-butanol, Bu; Water) obtained from Agrimoniae Herba. Methods : Anti-oxidant activity of different solvent fractions obtained from Agrimoniae Herba was investigated through total polyphenol contents, total flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity measurements. Then, filament aggregating protein (Filaggrin), Type I collagen, ceramide synthase (CERS) 3, and CERS4 were analyzed to evaluate the skin barrier strengthening effect of different solvent fractions obtained from Agrimoniae Herba on UVB-stimulated HaCaT cells. Results : As a result of measuring total polyphenol contents, total flavonoid contents, DPPH free radical scavenging activity, and ABTS radical scavenging activity, antioxidant activity was found to be excellent in the order of EA > Bu > MC > Hexane > Water. As a result of measuring mRNA gene expression of Type I collagen, Filaggrin, CERS3, and CERS4 after UVB-stimulated was applied to HaCaT cells treated with different solvent fractions obtained from Agrimoniae Herba, it was found to increase significantly in the Bu-treated group. Conclusion : Our findings show that the Bu sample obtained from Agrimoniae Herba has excellent anti-oxidant ability, which increases Type I collagen, Filaggrin, and ceramide synthetase in UVB-stimulated HaCaT cells to control the skin barrier improvement effect.

An Experimental Study on the Antioxidant and Anti-inflammatory Effects of Goryeon-hwa (고련환(苦練丸)의 항산화 및 항염증 효과에 관한 실험 연구)

  • Objectives: The purpose of this study is to evaluate the antioxidant and anti-inflammatory effects of Goryeon-hwan (GRH), which is mentioned in ≪Donguibogam≫ that treats leukorrhea. Methods: In this study, the antioxidant efficacy of GRH was evaluated by measuring the total polyphenol and flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, and ROS production through RAW264.7 cells. The concentration of GRH cytotoxicity was confirmed through the cell viability of RAW264.7 cells, and the production of NO, the production of Cytokine through ELISA assay, and the expression of genes through Real-time PCR were measured to evaluate anti-inflammatory efficacy. Protein phosphorylation and protein expression were measured through Western blot analysis. Results: As a result of the experiment, GRH contained polyphenol and flavonoid, and concentration-dependent increased DPPH radical scavenging activity and ABTS radical scavenging activity and decreased ROS production. The anti-inflammatory efficacy measurement results showed a significant decrease in NO and Cytokine production in the GRH administration group compared to the control group. In terms of gene expression and protein expression, there was a significant decrease in iNOS, COX-2, IL-1β, IL-6, and TNF-α depending on the concentration, and a significant increase in HO-1 and NQO1. Protein phosphorylation measurements showed a concentration-dependent significant decrease in the GRH group at ERK and p38. Conclusions: As a result, the study experimentally confirmed the antioxidant and anti-inflammatory effects of GRH, suggesting that it may be used as a treatment for various gynecological inflammatory diseases including vaginitis.

Physicochemical Properties and Antioxidative Activities of White Radish Tea by Different Preparation Methods (가공 방법을 달리한 무차의 이화학적 품질 특성과 항산화 활성)

  • The objective of this study was to investigate their physicochemical properties and antioxidant activities of white radish teas such as pH, color value, total polyphenol contents, total flavonoids, DPPH radical scavenging activity, reducing power and sensory evaluation. They were processed by drying(AD), drying and roasting(ADR), steaming and drying(SAD), and steaming, drying, and roasting(SADR). As a result, the pH of ADR, and SADR tended to be significantly lowered and SADR was the lowest at $5.48{\pm}0.14$(p<0.001). Additionally, ADR and SADR were significantly different in color and browning value, compared to AD and SAD. There was no significant difference in reducing sugar content depending on processing methods. The total polyphenol content and total flavonoid content of the teas were significantly increase after roasting. DPPH radical scavenging activity was significantly higher in ADR and SDAR than in AD or SAD by 1.1~1.5 times(p<0.05). Compared with ADR and SADR after roasting, the sensory characteristics of AD and SAD were significantly lower than those of roasted ADAD and SAD, respectively. From the above results, it was concluded that the roasting process is a major process that affects the physicochemical quality characteristics and antioxidant activities.

The Experimental Study on Anti-inflammation and Anti-oxidation of Indigo Naturalis and Rehmanniae Radix (청대(靑黛)와 생지황(生地黃)의 항염 및 항산화 효과에 대한 실험적 연구)

  • Objectives : This experiment study was performed to investigate the anti-inflammation and anti-oxidant effects of Indigo Naturalis(IN) and Rehmanniae Radix(RR) which were herbs for clearing heat. Methods : Anti-inflammation and anti-oxidation effects of IN and RR that were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results : 1. IN and RR groups were not showed cell toxicity. 2. In the inhibitory ability of NO production, IN groups were better than RR groups, but there were no statistical significances among the groups. 3. In the scavenging for DPPH radical, IN groups were better than RR groups, but there were no statistical significances among the groups. Conclusions : These results showed that the IN group was better than the RR group in treating skin eruptions, especially those such as erythema, purple spot, papule, pustules which appear in acute inflammation.

Assessment on In Vitro Antioxidant Properties of Common Thistle (Cirsium pendulum Fisch.) Plant Parts

  • Antioxidant activity of the extract fractions from leaves, stems, roots and flowers of Cirsium pendulum Fisch. was investigated. The results showed the greatest antioxidant activities in leaves by Rancimat, TBA and DPPH methods. Extracts of common thistle plants dose-dependently increased DPPH free radical scavenging activity, The extract from flowers and its hexane fraction showed the strongest antioxidant activity. HPLC analysis showed that BuOH fraction of the leaves had the highest amount of antioxidant chlorogenic and p-coumaric acids at 5.38 and 9.71 mg $100\;g^{-1}$, respectively. It implies that common thistle plants had potent antioxidant activity, and their activities were differently exhibited depending on plant part and solvent fraction.

The Experimental Study on Anti-inflammation and Anti-oxidation of GaeYongHwan (개용환(改容丸)의 항염 및 항산화 효과에 대한 실험적 연구)

  • Objectives : This study was performed to investigate the anti-inflammatory and anti-oxidantic effects of GaeYongHwan(GYH) extract which has been used for patients with acnes. Methods : Anti-inflammatory and anti-oxidantic effects of GYH extract were tested in terms of inhibitory ability of Nitric oxide(NO) production, 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity and anti-bacterial effects against Propionibacterium acnes(P. acnes). Results : 1. All GYH treated groups did not show cytotoxicity. 2. Treatment with $100{\mu}g/m{\ell}$ of GYH extract lowered production levels of NO significantly compared to non-treated control or normal. 3. All of GYH treated groups did not show DPPH free radical scavenging activities. 4. All of GYH treated groups did not show anti-baterial action against P. acnes. Conclusions : These results imply that GYH extract has anti-inflammatory effect to treat acnes.

A New Furofuran Lignan with Antioxidant and Antiseizure Activities from the Leaves of Petasites japonicus

  • A new furofuran lignan (1) was isolated from the n-butanol fraction of the methanolic extract of the leaves of Petasites japonicus (Sieb. et Zucc.) Maxim. (Compositae). The structure of compound 1 was determined to be $2{\alpha}-(4'-hydroxy-3'-methoxyphenyl)-6{\alpha}-(4"-hydroxy-3"-methox­yphenyl)-8{\alpha}-hydroxy-3, 7-dioxabicyclo[3.3.0]octane\;4'-O-({\beta}-D-glucopyranoside)$ by spectroscopic methods including 2D-NMR. In further studies, it was found that the compound 1 expressed an antioxidant activity in DPPH radical scavenging assay, and moreover, ameliorated the seizure in kainic acid-treated mice.
