Cytotoxic Activities of Brominated Sesquiterpenes from the Red Alga Laurencia okamurae

  • Four known sesquiterpenes, laurinterol (1), isolaurinterol (2), aplysinal (3) and aplysin (4) were isolated from the Korean red alga Laurencia okamurae off Cheju Island, Korea. Their structures were identified by comparison with the literature data. Compounds 1-4 showed potent cytotoxicity against A549, SK-OV-3, SKMEL-2, XF498, and HTl5 cell lines with $EC_{50}$ values ranging from 1.2 to 17.6 ${\mu}g/ml$.

  • Four prostate-type triterpenes were isolated from a methanol extract of Alismatis Rhizoma by bioassay-guided isolation using in vitro cytotoxic assay. The compounds were identified as alisol B 23-acetate (1), alisol C 23-acetate (2), alisol B (3), alisol A 24-acetate (4) by spectroscopic methods. Amongst the compounds, alisol B (3) showed significant cytotoxicity against SK-OV3, B16-F10, and HT1080 cancer cell lines with $ED_50$ values of 7.5, 7.5, $4.9\mu\textrm{g}/ml$, respectively.

Study on Antitumor Activity and Antimetastatic effect of Kamigumguesingihwan(KGSH) (가미금궤신기환(加味金櫃腎氣丸)의 항암(抗癌) 및 항전이(抗轉移) 효과(效果)에 관(關)한 연구(硏究))

  • To evaluate the antitumor activity and antimetastatic effects of Kamigumgusingihwan(KGSH) studies have ken done. The results were obtained as follows: 1. KGSH extracts exhibited a weak cytotoxicity against A549, SK-OV-3, B16-F10, and SK-MEL-2 cell lines. But exhibited potent cytotoxicity against P388 cell line in a dose-dependent manner. 2. The concentration inhibiting adhesion of A549, to complex extracellular matrix up to below 30% of control was recognized at $10^{-3}g/ml$ of KGSH 3. KGSH extracts showed a weak inhibitoty effect on DNA topo-isomerase I from calf thymus. 4. The T/C% was 137% in KGSH treated group in S-180 bearing ICR mice. 5. In pulmonary colonization assay, a number of colonies in the lungs were decreased significantly in KGSH treated group as compared with control group. 6. In hematological changes in B16-BL6 injected C57BL/6, numbers of WBC were decreased insignificantly in KGSH treated groups, and also those of platelet were increased insignificantly in KGSH treated groups as compared with control. 7. In CAM assay, KGSH extracts inhibited angiogenesis at $15{\mu}g/egg $concentration significantly as compared with control. Taken together these results, it is strongly demonstrated that KGSH significantly suppressed tumor metastasis by blocking cell adhesion to extracellular matrix. Therefore, KGSH is expected to be clinically a potent antimetastatic drug for the prevention and treatment of cancer.

Radical Scavenging Activity and Cytotoxicity of Maysin(C-glycosylflavone) isolated from Silks of Zea mays L.

  • Maysin, a C-glycosylflavone, was isolated from the silks of maize, Zea mays L. The ESI mass spectrum indicates that molecular weight of maysin is $577\textrm{M}^+$m/z, and the ether-linked sugar is rhamnose, $431\textrm{M}^+$m/z (MW$^{+}$-146). The DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical scavenging activity of maysin was higher than that of rutin. However, as compared with its aglycon luteolin, maysin showed the relatively moderate DPPH scavenging activity mainly due to the glycosylation of two sugars moieties, keto-fucose and rhamnose. In the in vitro cytotoxicity test against the five human tumor cell lines such as lung (A549), ovarian (SK-OV-3), melanoma (SK-MEL-2), central nerve system (XF-489), and colon (HCT-15), maysin exhibited the relatively weaker activities than cisplatin. The $\textrm{ED}_{50}$ values of maysin were 62.24, 43.18, 16.83, 37.22, and 32.09/$m\ell$, respectively. Result suggests that maysin is a potential cytotoxicity compound, particularly for human colon, central nerve system, and melanoma tumors.s.

  • We examined the effect of modulation of PKA isozymes on the growth of human ovarian cancer cells. Three ovarian cancer cell lines, 2774, SK-OV-3, and OVCAR-3, were examined in this study. The treatment of 5 uM 8-CI-cAMP, which has been known to down-regulate RI (or type 1 PKA) and up-regulate RII (or type II PKA), markedly inhibited the growth of all cell lines (50-80% at day 6). To test whether alteration in PKA regulatory subunits level can change the growth characteristics of ovarian cancer cells, we introduced RIIB- expression construct and Rla antisense-expression construct into 2774 cells. The overexpression of RIIB down-regulated Rla protein, and the antisense-expression of Rla up-regulated RIIB protein, showing that the intracellular levels of RI and RII are reciprocally regulated. In both cases, cell growth was reduced by 30% at day 2. These results indicate that the growth of ovarian cancer cells is controlled by the signals from PKA isozymes, and the modulation of PKA isozymes can be employed for the human ovarian cancer therapy.

Glioblastoma-Specific Anticancer Activity of Pheophorbide a from the Edible Red Seaweed Grateloupia elliptica

  • The chlorophyll-related compound pheophorbide a (Pa) was successively purified from an edible red seaweed, Grateloupia elliptica, using silica, octadecyl silica column chromatography and reversed phase-high-performance liquid chromatography, as well as the cell cycle inhibitory and apoptotic effects of Pa being investigated in U87MG glioblastoma cells. The Pa exhibited strong anticancer effects in the absence of direct photo-irradiation against various cancer cell lines, including U87MG, SK-OV-3, and HeLa cells. Among the cancer cells, the strongest anticancer activity of Pa exhibited on U87MG cells with $IC_{50}$ values of 2.8 ${\mu}g/ml$. In addition, Pa specifically had cytostatic activity on glioblastoma cells rather than human umbilical vein endothelial cells. Analysis of the cell cycle distribution showed that Pa induced G0/G1 arrest of U87 MG cells. In addition, arrested cells induced late apoptosis and DNA degradation under dark condition. These results suggest that Pa isolated from G. elliptica is a potential glioblastoma-specific anticancer agent without side effects on normal cells.

Importance of Sulfonylimidazolidinone Motif of 4-Phenyl-1-arylsulfonylimidazolidinones for Their Cytotoxicity: Synthesis of 2-Benzoyl-4-phenyl[1,2,5]thiazolidine-1,1-dioxides and Their Cytotoxcity

  • For probing the importance of planarity of imidazolidinone motif of 4-phenyl-1-(benzenesulfonyl)imidazolidinones 1 for their cytotoxicity, 4-phenyl-2-(benzoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2a), 4-phenyl-2-(p-toluoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2b), 4-phenyl-2-(phenylcarbamoyl)[1,2,5]thiadiazolidine-1,1-dioxide (3a), and 4-phenyl-2-(p-tolylcarbamoyl)[1,2,5]thiadiazolidine-1,1-dioxide (3b) were prepared along with their regioisomers (5a, 5b, 9a, 9b) and their cytotoxicity were measured against human lung carcinoma (A549), human colon carcinoma (COLO205), human ovarian cancer (SK-OV-3), human leukemic cancer (K562), and murine colon adenocarcinoma (Colon26) cell lines in vitro. All compounds prepared do not show any activity against all five cancer cell lines unlike 1. Compounds 1 possess planarity of imidazolidinone, especially in sulfonylurea moiety ($-SO_2$NHCONH-). However compounds 2 and 3 have nonplanar 5-membered ring, [1,2,5]thiadiazolidine-1,1-dioxides. Such structural differentiation might result in the loss of activity. Therefore the inactivity of 2 and 3 could also be an indication for the necessity of planarity of imidazolidinone ring of 1 for their cytotoxic activity.

  • A series of 8-alkyl-and 8-aryl-8,9-dihydro-7H-isoindolo[5,6-g]quinoxaline-7,9-diones were synthesized using sultine chemistry as a key step in good yield. These compounds were evaluated for their in vitro cytotoxicity against six human cancer cell lines (HCT15, SK-OV-3, A549, SNB19, MCF7 and MCF7/ADR).

  • The methanol extract of the seed of Myristica fragrans (myristicaceae) demonstrated a potent inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2(melanoma), XF498 (central nerve system) and HCT-15(colon). The MeOH extract was fractionated into three portions by serial solvent partition i,e., EtOAc soluble part, BuOH soluble part and remaining water layer. Among them, the EtOAc soluble part of the extract demonstrated a potent inhibition on the proliferation of cultured human tumor cells, Bioassay-guided fractionation of the EtOAc soluble part led to the isolation of six lignan constituents, i.e., safrole(1), machilin A (2), licarin B (3), macelignan (4), mesodihydroguaiaretic acid (5) and myristargenol A (6) as well as a large amount of myristic acid as active ingredients. Structures of the isolated active components (1-6) were established by chemical and spectroscopic means.
