Effect of Hexane Extract of Acori graminei Rhizoma on the Growth of Chloramphenicol Resistant Bacteria

  • The combination of hexane extract (E4) of rhizome of Acorus gramineus with chloramphenicol (Cm) was applied to Gram negative Cm resistant microbials to find the possibility of clinical use and to clarify the relationship of the activity of chloramphenicol acetyltransferase (CAT). The combination of $1,000\;{\mu}g/ml$ of E4 and $8\;{\mu}g/ml$ of Cm entirely ceased the growth of S. aureus SA2, a gram positive resistant strain to 10 antibiotics. But in Gram negative strains which possess CAT activity, some showed considerably strong resistances to Cm and some did weakly.

  • We studied the folilar wiping effects of antioxidants (ascorbate, glutathione and sodium azide), which effectively inhibited the chlorophyll bleaching or completely recorved the early stage of photosynthesis of Panax ginseng C.A. Meyer, on photosynthesis, stomatal resistance, free sugar, starch, and total saponin contents of ginseng under the excess light intensity (45 kLux) during 6 days. Ascorbate and glutathione, endogenous antioxidant, recovered photosynehtsis and stomatal resistance, and reduced the photoinhibition by the excess light intensity (45 kLux) on free sugar, starch and total saponin contents. But sodium azide, exogenous $^{1}O_2$ quencher, showed negative effect. Therefore, we assumed that carbohydrates and saponin metabolisms of ginseng by antioxidants (ascorbate, glutathione) were normal. For the reduction of inhibition by excess light in ginseng a program for the higher activation of antioxidants and antioxidative enzymes in ginseng leaf will be desirable.   • Finishing materials used during the construction of reinforced concrete structures aid in providing resistance to carbonation and help ensure the durability of a structure. However, detailed examinations of this phenomena using data gained from long-term outdoor exposure are not only lacking, but also are not taken into account as factors affected by the local environment. In this research, the velocity coefficient in terms of carbonation is compared as a difference according to the local region and the averaged annual temperature, and the carbonation-preventive effects of finishing materials are analyzed. As an outcome of this study, the results of long-term carbonation can be evaluated from carbonation resistance R induced by an acceleration carbonation test.

  • Bacterial growth inhibition by lead, zinc and cadmium was measured by using modified Tris minimal medium. The toxicity against Escherichia coli strain was in the order of zinc> cadmium> lead, and the Escherichia coli strain overexpressing iron-containing superoxide dismutase 2 of Streptomyces coelicolor A3(2) was found to have resistance to heavy metals.

  • Lactic acid producing bacteria were isolated from baby feces and characterized to be used as a probiotic with anti Helicobacter pylori functions. The selected bacteria had inhibition activity on the adherance and growth of H. pylori. These bacteria had additional beneficial characteristics for the probiotic such as antibacterial activity, antitumor activity, immunostimulation activity, resistance to antibiotic and bile salt, ability to bind to the intestinal cells, and safe for the human use.

Effects of Fermented Lotus Extracts on the Differentiation in 3T3-L1 Preadipocytes (3T3-L1 전지방세포에서 연잎-연근 혼합 발효물의 지방세포 분화 억제 효과)

  • Objectives: This study was performed to evaluate the effects of fermented lotus extracts on the inhibition of differentiation in 3T3-L1 preadipocytes. Methods: Extracts of lotus leaf and lotus root were fermented using 4 different probiotics separately, including Lactobacillus plantarum, Lactobacillus rhamnosus, Bifidobacterium breve, and Bifidobacterium longum. Inhibition of preadipocyte differentiation was examined by Oil red O dye staining. Expressions of adipogenic transcription factors including CCAAT/enhancer binding proteins (C/$EBP{\alpha}$) and peroxisome proliferators-activated receptor ${\gamma}$ ($PPAR{\gamma}$) were analyzed by real time polymerase chain reaction and Western blotting analysis. Results: Fermented lotus extracts inhibited adipogenic transcription factors by inhibiting preadipocytes differentiation. All of the groups fermented by 4 kinds of probiotics showed reduction in Oil Red O dye staining. Bifidobacterium breve showed the most effective inhibition of C/$EBP{\alpha}$. Bifidobacterium breve and Bifidobacterium longum showed the best downregulation of $PPAR{\gamma}$ expressions compared with the control and the unfermented lotus group. Conclusions: Fermented lotus extracts showed significant effects on inhibition of preadipocyte differentiation in 3T3-L1 preadipocytes showing correlation with insulin sensitivity and lipid metabolism related with obesity.

  • Acne vulgaris, the most common skin disease. can be formed as only a few comedons or severe inflammatory lesions. The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.

Cosmetic Effects of Dietary Fiber from Mozuku, Cladosiphon novae-caledoniae Kylin (큰실말(Cladosiphon novae-caledoniae Kylin) 부산물의 화장품소재 특성)

  • To investigate the effect of dietary fiber from mozuku, Cladosiphon novae-caledoniae kylin (C. novae-caledoniae kylin) on the skin care, we measured anti-oxidant activity, anti-microbial activities, tyrosinase activity inhibition and elastic activity. B16F10 melanoma cell (MTT assay) were used to measure cell viability. MC and MI exhibited in vitro antibacterial activity against Staphyloccus aureus (S. aureus) and MRSA without antifungal activity. Mozuku extract (MS) showed excellent tyrosinase inhibition effect compared to arbutin as a positive control (to 49% for tyrosine). The wrinkle-improving effect was relatively low. However, wrinkle-improving effect was relatively low. DPPH free radical scavenging activity was 89% in a concentrations at $500{\mu}g/mL$. These results indicate that the mozuku extracts may be an effective cosmetic ingredient for skin whitening.

Inhibition of mitoNEET induces Pink1-Parkin-mediated mitophagy

  • MitoNEET, a mitochondrial outer membrane protein containing the Asn-Glu-Glu-Thr (NEET) sequence, controls the formation of intermitochondrial junctions and confers autophagy resistance. Moreover, mitoNEET as a mitochondrial substrate undergoes ubiquitination by activated Parkin during the initiation of mitophagy. Therefore, mitoNEET is linked to the regulation of autophagy and mitophagy. Mitophagy is the selective removal of the damaged or unnecessary mitochondria, which is crucial to sustaining mitochondrial quality control. In numerous human diseases, the accumulation of damaged mitochondria by impaired mitophagy has been observed. However, the therapeutic strategy targeting of mitoNEET as a mitophagy-enhancing mediator requires further research. Herein, we confirmed that mitophagy is indeed activated by mitoNEET inhibition. CCCP (carbonyl cyanide m-chlorophenyl hydrazone), which leads to mitochondrial depolarization, induces mitochondrial dysfunction and superoxide production. This, in turn, contributes to the induction of mitophagy; mitoNEET protein levels were initially increased before an increase in LC3-II protein following CCCP treatment. Pharmacological inhibition of mitoNEET using mitoNEET Ligand-1 (NL-1) promoted accumulation of Pink1 and Parkin, which are mitophagy-associated proteins, and activation of mitochondria-lysosome crosstalk, in comparison to CCCP alone. Inhibition of mitoNEET using NL-1, or mitoNEET shRNA transfected into RAW264.7 cells, abrogated CCCP-induced ROS and mitochondrial cell death; additionally, it activated the expression of PGC-1α and SOD2, regulators of oxidative metabolism. In particular, the increase in PGC-1α, which is a major regulator of mitochondrial biogenesis, promotes mitochondrial quality control. These results indicated that mitoNEET is a potential therapeutic target in numerous human diseases to enhance mitophagy and protect cells by maintaining a network of healthy mitochondria.
