Recently, Rhei Rhizoma extracts (RRE) have begun to receive more attention as potential biological response modifiers. In the present study, we studied the antiproliferative effect of RRE on tumor cells and the effect of RRE on macrophage function. A variety of tumor cells and macrophages were treated with RRE at various concentrations. The effect of RRE on cell proliferation was measured by MTT assay and the effect of RRE on the production of nitric oxide (NO) was determined in the macrophage-like cell lines Raw264.7, C6 and peritoneal macrophages (pMQ). RRE inhibited the growth of tumor cells (e.g., B16, HOS). However, the effects of RRE on the production of NO varied with macrophage types. RRE had no effect on C6 cell growth and slightly increased the growth of Raw264.7 cells. In addition, treatment of normal pMQ with RRE enhanced NO production in a concentration-dependent manner, whereas RRE suppressed NO production at $50\;{\mu}g/mL$ in both Raw264.7 and C6 cells. However, RRE suppressed NO production in LPS/IFN-$\gamma$-stimulated C6 cells. Overall, these results suggest that RRE elicits an antiproliferative property and differentially modulates NO production in various macrophages, and have a potential for therapeutic application.
The Rev binding to Rev Responsive Element (RRE) of HIV-1 mRNA plays an important role in the HIV-I viral replication cycle. The disruption of the Rev-RRE interaction has been studied extensively in order to develop a potential antiviral drug. In order to provide the basis for a more promising approach to develop a Rev-RRE binding inhibitor against HIV-I infection, it is necessary to understand the binding modes of the aminoglycoside antibiotics to RRE. In the present study, the binding mode of a modified antibiotic, a neamine conjugated with pyrene and arginine (NCPA), to RRE has been studied by the methods of $T_m$ measurement and spectroscopic analysis of RRE with or without antibiotics. The results confirmed that NCPA competes with Rev in binding to RRE.
제1형 인간 면역결핍 바이러스 (human immunodeficiency virus type 1, HIV-1)의 Rev 단백질에 대하여 야생형보다 10배 더 잘 결합하도록 시험관에서 선택된 RRE40라 명명된 RNA aptamer가 과연 임상적으로 유용한지 알기 위하여 인체의 CD4^+ peripheral blood lymphocytes 세포에서 레트로바이러스 벡터를 이용하여 RRE40 RNA를 발현한 후에 그 세포에서의 HIV-1 증식 현상을 관찰하였다. 그 결과 대조군인 tRNA를 발현하는 유전자가 전달된 세포에 비해 RRE40 RNA를 발현하는 세포에서 보다 더 효과적으로 HIV-1의 증식이 억제되었다. 그러나 바이러스의 증식이 완전히 억제되지는 못 하였고 일시적 또는 감소된 형태로 바이러스 증식이 억제되었다. 이러한 결과는 RRE40 RNA가 decoy로서 세포에서의 HIV-1 증식 억제에 유용함을 시사하지만 RNA decoy를 HIV-1 감염 환자의 치료에 이용하기 위해선 보다 효과적인 유전자 전달방법 및 보다 개선된 RNA decoy의 개발 등이 필요할 것이다.
Allergic contact dermatitis is a common skin disease resulting from specific immunologic to topically applied various allergen. After Dinitrochlorobenzene (DNCB) secondary sensitization, the ICR mice administered Rhemanniae radix extract (RRE) was observed to ascerstain the effect of RRE on allergic contact dermatitis. Purpose of this study was to investigate contact hypersensitivity assay, abdominal skin morphologic changes. Including mast cells and cell-surface glycoconjugates. The change of interleukin 2 (IL-2) receptor (CD25R). ICAM in abdominal skin, lymph node of inguinal region, and electro microscope-morphologic changes of abdominal skin. The results of this study were as follows: 1. The contact hypersensitivity assay, the ear swelling in the RRE had lesser probability than in the ACD Group. 2. In the general morphologic change of skin, hyperplasia of keratinocytes, distribution of vasculogenesis and epidermal lymphocytes infiltration were decreased in the RRE group compared with the ACD group. In epidermal basal layer and prickle layer, cell damage was decreased in the RRE group compared with the group painted with ACD. 3. MasT-cell in dermis was decreased in the RRE group compared with the group painted with DNCB. 4. Distribution of interlukin-2 Receptior positive cell and ICAM positive cell in dermis was decreased in the RRE Group compared with the ACD group. 5. Distribution of helper T-lymphocyte and cytotoxic T-lymphocite in inguinal nodes was decreased in the RRE group, and was observed well in paracortical area and cortical cord. 6. Distribution of apoptotic cell was appeared in the RRE group compared with the ACD group, in skin, dermis. in inguinal nodes, paracortical area observed well. With above results, the restarint of immunosuppression occuring in Allergic contact dermatitis is resulted in the slow progress the effect of Allergic contact dermatitis, and it is thought that ithis fact has a series of relation with apoptosis.
Objective: To investigate the clinical effect of riboflavin sodium phosphate on prevention of radiotherapy related esophagitis (RRE). Methods: This retrospective study involved 55 patients with middle and advanced esophageal cancer who were divided into an experimental group of 28 and a control group of 27 patients. Those in the experimental group were treated with riboflavin sodium phosphate combined with conventional symptomatic treatment during radiotherapy; while patients in control group received the latter alone. The incidence and degree of RRE were compared after radiotherapy. Results: The incidences of RRE in experimental and control group were 53.5% and 81.4%, respectively (p<0.05); the incidence of stages III and IV RRE in the experimental group was 17.8%, while in the control group it was 44.4% (p<0.05). Conclusion: Riboflavin sodium phosphate could significantly prevent RRE and reduce the incidence of stage III and IV disease. These results were worthy of further confirmation by randomized controlled trials.
Intracellular expression of RNA decoys, such as TAR or RRE decoy, has been previously shown to protect immune cells from human immunodeficiency virus type 1 (HIV-1) replication by inhibiting the binding of the HIV-1 regulatory protein to the authentic HIV RNA sequence. However, HIV-1 challenge experiments of primary human T cells, which express the RNA decoy, demonstrated that the cells were only transiently protected, and hence, more improved protocols for HIV-1 inhibition with the RNA decoys need to be developed. In this report, in order to develop a more effective RNA decoy, we analyzed and compared the ability of a series of RNA decoy derivatives in inhibiting HIV-1 replication in CEM cells. Using an improved tRNA cassette to express high levels of RNA decoy transcripts in cells, we found that co-expression of both TAR and RRE decoys, in the form of an aligned sequence in a single transcription cassette, much more potently blocked cells from HIV-1 than the expression of only one kind of RNA decoy. This observation will have an important implication for experiments involving optimization of clinical applications in RNA decoy-based gene therapy against HIV-1.
HIV-1 Rev단백질은 바이러스의 구조단백질의 발현조절에 매우 중요한 역할을 하며RNA가 1차발현된 원형대로 또는 일부만이 절단/재조합된 상태로 세포질에 집적되는 것을 가능하게 한다. env gene에 존재하는 Rev-responsive element RNA는 복잡한 RNA구조를 지니면서 Rev의 기능을 위하여 필수적으로 필요시 된다. 그러나 이러한 절차의 완전한 진행을 위해서는 핵단백질이 요구되는 것으로 추정된다. 본 연구에서는 electrophoretic mobility shift, UV-crosslinking 또는 SDS/PAGE등의 실험을 통하여 36/37, 56, 41, 76, 150 kD등의 핵단백질등이 RRE RNA와 결합반응하는 것을 발견하였으며 특히 36/37 과 56 kD 단백질을 5분간의 자외선조사에 의해 특히 RRE RNA에 결합하는 핵단백질들은 gel mobility shift assay에서 Rev RNA인식결합에 경쟁적인 특징을 나타내고 있다.
Deletion of smpd3 induces osteogenesis and dentinogenesis imperfecta in mice. smpd3 is highly elevated in the parietal bones of developing mouse calvaria, but not in sutural mesenchymes. Here, we examine the mechanism of smpd3 regulation, which involves BMP2 stimulation of Runx2. smpd3 mRNA expression increased in response to BMP2 treatment and Runx2 transfection in C2C12 cells. The Runx2-responsive element (RRE) encoded within the -562 to -557 region is important for activation of the smpd3 promoter by Runx2. Electrophoretic mobility shift assays revealed that Runx2 binds strongly to the -355 to -350 RRE and less strongly to the -562 to -557 site. Thus, the smpd3 promoter is activated by BMP2 and is directly regulated by the Runx2 transcription factor. This novel description of smpd3 regulation will aid further studies of bone development and osteogenesis.
This paper proposes a gradation image processing method for the development of a Road Sign Recognition Platform (RReP), which aims to facilitate the rapid and accurate management and surveying of approximately 160,000 road signs installed along the highways, national roadways, and local roads in the cities, districts (gun), and provinces (do) of Korea. RReP is based on GPS(Global Positioning System), IMU(Inertial Measurement Unit), INS(Inertial Navigation System), DMI(Distance Measurement Instrument), and lasers, and uses an imagery information collection/classification module to allow the automatic recognition of signs, the collection of shapes, pole locations, and sign-type data, and the creation of road sign registers, by extracting basic data related to the shape and sign content, and automated database design. Image division and merging, which were applied in this study, produce superior results compared with local binarization method in terms of speed. At the results, larger texts area were found in images, the accuracy of text recognition was improved when images had been gradated. Multi-threshold values of natural scene images are used to improve the extraction rate of texts and figures based on pattern recognition.

RBFII의 cDNA에 대한 염기서열을 결정하였으며 기존에 연구된 바 있는 RBFII(RBF 또는 TRBP로 보고되었으며 본 연구에서는 RBFII와 구분하기 위해 RBFI으로 명명)과 대부분의 경우 공통적인 ORF를 지니는 것으로 나타났다. 그러나 5’말단에서는 공통적인 ORF 가 RBFI의 경우 21개의 아미노산을 의미하는 63 nt가 Lac-Z의 N-말단에 연결된데 비해서 특이한 46개 아미노기를 의미하는 138nt가 존재함이 밝혀졌다. 5’-말단에 처음 나타나는 ATG 및 부근의 염기서열을 분석해 볼 때 양 cDNA는 5’말단이 완전하지 않은 것으로 사료된다.
