Phellinus linteus의 인공재배 (Artificial Cultivation of Phellinus linteus)

  • Phellinus linteus(상황버섯 또는 목질진흙버섯)의 인공재배에 대하여 연구하였다. 통나무, 상수리나무, 오리나무, 밤나무를 기질로 사용하였을 때, 자실체 수확량(Biological efficiency)은 상수리나무가 $9.4{\pm}1.6$으로 가장 높았다. 자실체 형성온도와 배양 기간은 각각 $25^{\circ}C$와 약 10개월로 나타났다.

상황버섯 (Phellinus linteus)으로부터 항산화 및 항균 효과 (Antioxidant and Antimicrobial Activities of Cambodian Mushroom, Phellinus linteus)

  • This study was peformed to determine the antioxidant, antimicrobial activity and antifungal of the cambodian mushroom, Phellinus linteus, in various concentrations from the mushroom extracts. It's extracts were found to cause significant free radical scavenging effects on DPPH (1,1-diphenyl-2-picrylhydrazyl). This extracts exhibited in vitro broad-spectrum antimicrobial activity of Gram-negative, Gram-positive bacteria and without antifungal activity. On the other hand, the extracts had not significant hemolytic activity against human red blood cells. These results indicate cambodian mushroom, Phellinus inteus having antioxidant and antimicrobial activity without hemolytic activity maybe useful as therapeutic agents.

배양상황버섯의 저밀도지질단백질 (LDL)산화에 미치는 영향 (Effect of the Cultivated Fruit body of Phellinus linteus on Low Density Lipoprotein Oxidation)

  • Phellinus linteus (Hymenocaetaceae) has been used for the treatment of gastric cancer, noninsulin dependant diabetes, diarrhea, and menstrual irregularity. The antioxidative effect of cultivated fruit body of Phellinus linteus on low density lipoprotein oxidation was investigated. The MeOH and water extracts were examined by TEARS assay on human plasma LDL in vitro system. The MeOH ex. showed antioxidative activity, and then was fractionated into Precipitates, Water Insoluble fr., Water fro and ether fro The results showed that the lipophilic fractions, Water Insoluble fr. and Ether fr. of cu 1-tivated fruit body of Phellinus linteus, inhibited the oxidative modification of LDL.

  • The effects of dissolved oxy-gen(DO) concentration and DO control modes on cell growth in a 4L computer-controlled bioreactor system were investigated using Phellinus linteus WI-001, a producer of protein-polysaccharides having potent anticancer activi-ties. When DO was controlled at about 20%, maximum cell concentration and specific growth rate of the strain were observed to increase 36% and 64%, respectively, as compared to the experiment performed without DO control. By adopting cascade automatic control of DO ar 20% in a mixed automatic control mode using computer-con-trolled program, 19.5g/L of maximum cell concentration was obtained. These results showed that the mixed auto-matic control mode was the effective method for enhancing cell growth of the shear sensitive Phellinus linteus.

캄보디안 상황버섯의 항암 및 면역조절작용에 대한 연구 (Study on Antitumor and Immunomodulatory Effects of Cambodian Phellinus linteus)

  • Phellinus Iinteus from Cambodia was confirmed to have a homologous DNA sequencec to Phellinus Iinteus. Antitumor and immunomodulatory activities were evaluated with aquous extract of Cambodian Phellinus Iinteus(CPL). CPL didn't show any significant cytotoxicity on HT1080, Sarcoma 180 and B16BL6, whereas it inhibited the relaxation of DNA topoisomerase I from the concentration of 250ug/ml. In the pulmonary colonization assay it inhibited pulmonary metastasis by B16BL6 in C57BL6 mice to 36%, 36.9% and 55.5% at various doses of 2 mg, 20 mg and 50 mg. From FACS analysis with splenocytes pretreated with CPL, it significantly increased lymphoblast and induced production of IL-2. These results indicate Cambodian Phellinus Iinteus has antitumor and immunomodulatory activities still suggesting more study on its mechanism and effective compound in detail.

  • 본 연구는 Phellinus linteus와 관련 담자균류 속간의 IGS I 영역의 분석에 기초한 분류체계를 정립하기 위해서 실시하였다. 실험에 사용된 Phellinus linteus 균주의 IGS I 영역은 730 bp였고 다른 담자균류의 IGS I 영역과 비교시 5' 말단부위의 $1{\sim}280\;base$까지는 각 균주들간의 보존성이 높게 나타났으며 3' 말단부위로 갈수록 보존성이 감소하다가 다시 증가하는 경향을 보였다. 담자균류의 IGS 영역에 대한 연구가 앞으로 진행된다면 ITS 영역의 비교와 더불어 새로운 계통분류지표로 이용될 수 있을 것이다.

장수상황버섯(Phellinus baumii)의 병재배 특성 (Cultural Characteristics of Phellinus baumii Grown in Bottle)

  • 장수상황버섯(Phellinus baumii)은 현재 원목재배가 주종을 이루는 있는데 자실체 생육기간이 길고 원목수급문제가 발생할 가능성이 있고 톱밥의 경우 목재부산물로 다량 발생하고 있어 이를 활용한 톱밥병재배을 실시하였다. 장수상황버섯(Phellinus baumii)의 톱밥병재배를 실시한 결과 참나무톱밥 + 미강(4 : 1)배지가 배양속도와 균사밀도는 가장 높았지만 자실체를 형성하지 못하였고 반면에 참나무톱밥 100% 배지는 균사밀도와 배양속도는 낮았지만 병배지에서 자실체를 형성할 수 있었고 850 ml병에 비해서 1,100 ml 병 처리가 수량성과 생체효율이 약간 높은 결과가 나타났다.

The Culture Conditions for the Mycelial Growth of Phellinus spp.

  • Phellinus genus belonged to Hymenochaetaceae of Basidiomycetes and has been well known as one of the most popular medicinal mushrooms due to high antitumor activity. This study was carried out to obtain the basic information for mycelial culture conditions of Phellinus linteus, P. baumii, and P. gilvus. According to colony diameter and mycelial density, the media for suitable mycelial growth of them were shown in MEA, glucose peptone, and MCM. The optimum temperature for mycelial growth was $30^{\circ}C$. Carbon and nitrogen sources were man nose and malt extract, respectively. The optimum C/N ratio was 10:1 to 5:1 with 2% glucose concentration, vitamin was thiamine-HCl, organic acid was succinic acid, and mineral salt was $MgSO_{4}{\cdot}7H_{2}O$.

  • Objectives : This study was carried out to investigate the effects of Phellinus igniarius Quel extract on the Anti-inflammatory, Anti allergy, Anti-oxidant and Anti-wrinkle. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe TNF-$\alpha$, IL-8 by ELISA Results : Phellinus igniarius Quel extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Phellinus igniarius Quel extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

ITS 염기서열에 기초한 차가버섯과 근연속간 유연관계분석 (Phylogenetic Relationships between the Genus Inonotus and its Related Genera Based on the Nucleotide Sequences of Internal Transcribed Spacers)

  • 국내에 유통 중인 약용버섯의 분류체계 확립과 차가버섯 근연속간의 유연관계를 밝히기 위해 이들 버섯의 ITS 부위 염기서열을 비교 분석하였다. 조사결과, 약용버섯으로 주로 시판되고 있는 국내유통균주는 총 6개의 속(Phellinus, Inonotus, Sparassis, Fomes, Ganoderma, Hericium)으로 나누어짐을 알 수 있었고 그 중에서 기존에 잘 알려진 상황버섯(Phellinus linteus)과 최근 들어 수입양이 급증하고 있는 차가버섯이 대부분을 차지하고 있는 것으로 확인되었다 일명 차가버섯(Inonotus obliquus)으로 유통 중인 15개 제품 중 중국에서 수입되어진 한 균주(59번)가 P. pini로 확인되었으며 일본에서 수입되어진 균주(51, 52번)가 P. baumii와 P. linteus와 유사종 혹은 동일종으로 확인되었다.
