Establishment of Callus Induction and Plant Regeneration System from Mature Seeds of Miscanthus sinensis (억새(Miscanthus sinensis) 성숙 종자로부터의 캘러스 유도 및 식물체 재분화 체계 확립)

  • This study was conducted to establish the tissue culture system for Korean domestic Miscanthus sinensis, which is used in various purposes such as forage, and bio-energy resources. With the mature seed of Miscanthus, optimum concentrations of plant growth regulators were identified for an efficient callus induction and regeneration. Among the treatments of 1~10 $mg{\cdot}L^{-1}$ 2,4-D, IBA, or NAA, callus induction rate was highest (85.3%) on MS medium containing 5 $mg{\cdot}L^{-1}$ 2,4-D. Under the condition, the callus were efficiently induced and proliferated with comparably lower frequencies of callus browning. In shoot regeneration, the treatment of NAA combined with BAP seemed to contribute more efficient conditions to shoot regeneration than those of NAA with Kinetin or 2-iP. Especially, regeneration efficiency and number of regenerated plants were 83.7% and 5.5 in 3 $mg{\cdot}L^{-1}$ NAA with 5 $mg{\cdot}L^{-1}$ BAP, respectively, which were higher frequencies than those in NAA with Kinetin or 2-iP. In results, 5 $mg{\cdot}L^{-1}$ 2,4-D and 3 $mg{\cdot}L^{-1}$ NAA combined with 5 $mg{\cdot}L^{-1}$ BAP were efficient for embryogenic callus induction and regeneration of Miscanthus. This system would be useful for mass-propagation and developing new cultivars via tissue culture of Miscanthus sinensis.

Combined Expression of Metastasis Related Markers Naa10p, SNCG and PRL-3 and its Prognostic Value in Breast Cancer Patients

  • Combinations of multiple biomarkers representing distinct aspects of metastasis may have better prognostic value for breast cancer patients, especially those in late stages. In this study, we evaluated the protein levels of N-${\alpha}$-acetyltransferase 10 protein (Naa10p), synuclein-${\gamma}$ (SNCG), and phosphatase of regenerating liver-3 (PRL-3) in 365 patients with breast cancer by immunohistochemistry. Distinct prognostic subgroups of breast cancer were identified by combination of the three biomarkers. The Naa10p+SNCG-PRL-3-subgroup showed best prognosis with a median distant metastasis-free survival (DMFS) of 140 months, while the Naa10p-SNCG+PRL-3+subgroup had the worst prognosis with a median DMFS of 60.5 months. Multivariate analysis indicated Naa10p, SNCG, PRL-3, and the TNM classification were all independent prognostic factors for both DMFS and overall survival (OS). The three biomarker combination of Naa10p, SNCG and PRL-3 performed better in patients with lymph node metastasis, especially those with more advanced tumors than other subgroups. In conclusion, the combined expression profile of Naa10p, SNCG and PRL-3, alone or in combination with the TNM classification system, may provide a precise estimate of prognosis of breast cancer patients.

Effect of Plant Growth Regulators on in vitro Cultured Atractylodes Hybrid 'Dachul' (A. macrocephala x A. japonica) (기내배양 백출 교잡종 '다출'(Dachul, Atractylodes macrocephala x A. japonica)에 미치는 생장조절제처리효과)

  • This study was conducted to establish the tissue culture system for Atractylodes plant which is most frequently used in oriental medicine. Root and auxiliary bud of Dachul cv., which is Atractylodes hybrid (A. macrocephala x A. japonica), were used as target tissues for in vitro culture. In root culture, callus induction rate was higher in the treatment of BAP combined with NAA than others, however, 2-iP was more effective for callus proliferation and root induction. Although calli were effectively induced from the root and proliferated in lower concentration of cytokinin combined with higher auxin, root tissue was inappropriate for shoot regeneration. For plant regeneration with axillary bud, BAP combined with NAA was more effective than 2-iP with NAA or IBA. Number of regenerated plant per bud was 3.8, which was highest, and stem diameters was shown as 5.0mm under the conditions of 1 mg/L BAP combined with 1 mg/L NAA. Although, plant height was tend to be higher in 2-iP than BAP, number of the regenerated plant was lower via versus. Furthermore, root proliferation of regenerated plant was more effective in higher concentration of sucrose (7%) than in lower concentration (3%). In results, auxiliary bud was an efficient target tissue for producing regenerated plant of Atractylodes under the conditions of 1 mg/L BAP combined with 1 mg/L NAA and higher concentration of sucrose was effective for root proliferation of regenerated plants.

  • The role of NAA(1-Naphthaleneacetic acid) and BA(6-Benzyladenine) in the adventitious shoot regeneratlon from Populus leaf segments and changes in the pattern of RNA and protein synthesis were investigated. The adventitious shoot regeneration octured at the basal cut end of Populus leaf segments. This process was effected by many factors, including wounding culture conditions, light and plant growth regulators etc. The highest adventitious shoot regeneration frequency was obtained at $0.01mg/\ell$ NAA with $0.2mg/\ell$ BA. In this condition adventitious shoot starved to regenerate on the 13th day of oullure. The most optimal hormone composition for RNA and protein synthesis was $0.01mg/\ell$ NAA with $0.2mg/\ell$ BA. The content of RNA and protein was greater at the proximal part. In the course of adventitious shoot regeneration, the proteins associated with polar-regeneration appeared at the proximal part of populus leaf segment.

  • This experiment was carried out to clarify the effect of pyroligneous liquor and coconut water on plantlet multiplication and in vitro flowering of Dendrobium moniliforme. Plantlet growth and multiplication was good in 1.0 ml/L pyroligneous liquor treatment which was added to the basal media of 3 g/L hyponex and 4 g/L peptone ($H_{3} P_{4}$) containing 0.1 mg/L NAA and 1.0 mg/L kinetin. In the treatment of 30 ml/L coconut water showed good results on plantlet growth and multiplication. In vitro flowering showed highest rate in the treatment of 1.0 ml/L pyroligneous liquor and 30 ml/L coconut water which were added to the basal media of $H_{3} P_{4}$ containing 0.1 mg/L NAA and 1.0 mg/L kinetin.

Development of Wound-treatment Formulation Using Plant Growth Regulators for Wound Healing of Some Tree Species (수피상처치료를 위한 식물생장조절물질 기반 상처유합제 개발)

  • New tree wound-treatment formulations (WTF) were developed in this study. Stimulating effects of plant growth regulating substance on callus formation were evaluated in vivo twig disc culture of Zelkova serrata, Ziziphus jujuba, Pinus densiflora, Ginkgo biloba. and Aesculus turbinata. Based on the evaluation, WTF were prepared with 2,4-D, NAA, and IBA using xanthan gum as an extending agent. WTF were pasted on $2{\times}10cm$ artificial wound on the stem and the effects were evaluated 12 weeks later with the thickness of new callus-tissues. The effects varied with tree species and the growth regulators. In Z. serrata and A. turbinata, 2 mg/L of 2,4-D was the best WTF. In P. koraiensis and G. biloba, 8 mg/L of NAA and 1 mg/L of IBA were the best, respectively. Callus formation was quite lower in G. biloba compared to other species. Stimulating effect of thiophanate-methyl paste was excellent only in Z. serrata and lower than certain WTF in other species. Vaseline, which was used generally as an WTF, remained longer on the wound and causing decay. From the results, it could be recommended that 2,4-D, NAA and IBA were the best WTF for Z. serrata/A. turbinata, P. koraiensis and G. biloba, respectively.

  • The embryos of Pinus taeda, P. rigida, and P. taeda ${\times}$ rigida were cultured for adventitious shoot regeneration in vitro. Culture media were modified from Gresshoff and Doy (MGD), Murashige and Skoog (MMS), Lloyd and McCown (MLM), and Schenk and Hildebrandt (MSH). NAA was added to initiation media at a concentration of 0.1 or 0.01 mg/l. BAP was used at the concentrations of 0.1. 0.5, 1, 2, or 5mg/l. Each explant was induced for 3-4 weeks on solid medium. All explants were cultured up to 16 weeks. Illumination was about $1506{\pm}540lux$ at the level of the tissues in the growth room with a temperature of $25{\pm}2^{\circ}C$. A 16-hour photoperiod per 24 hours was used. Half-strength medium was used for all the subcultures. For shoot production by loblolly pine, MMS, MLM, or MSH is preferred with 5 mg/l BAP with either 0.1 or 0.01 mg/l NAA. For shoot production by pitch pine, MMS, MLM, or MSH is recommended with 2 or 5 mg/l BAP with 0.1 mg/l NAA. For shoot production by the hybrid pine, MMS or MLM is more effective with 1, 2 or 5 mg/l BAP with 0.1 mg/l NAA. There were no differences recognized among the species tried in the patterns of bud formation and shoot development. Different composition of media, in major and minor salts or possibly in vitamins, should be tested for the two developmental stages of adventitious shoots ; the induction of shoot buds and the elongation of them into shoots.

Assessment of Malignancy in Brain Tumors by 3T MR Spectroscopy

  • Purpose: To assess clinical proton MR spectroscopy (MRS) as a noninvasive method for evaluating tumor malignancy at 3T high field system. Methods: Using 3T MRI/MRS system, localized water-suppressed single-voxel technique in patients with brain tumors was employed to evaluate spectra with peaks of N-acetyl aspartate (NAA), choline-containing compounds (Cho), creatine/phosphocreatine (Cr) and lactate. On the basis of Cr, these peak areas were quantificated as a relative ratio. Results: The variation of metabolites measurements of the designated region in 10 normal volunteers was less than 10%. Normal ranges of NAA/Cr and Cho/Cr ratios were 1.67$\pm$018 and 1.16:1:0.15, respectively. NAA/Cr ratio of all tumor tissues was significantly lower than that of the normal tissues (P=0.005). Cho/Cr ratio of high-grade gliomas was significantly higher than that of low-grade gliomas (P= 0.001), Except 4 menigiomas, lactate signal was observed in all tumor cases. Conclusions: The present study demonstrated that the neuronal degradation or loss was observed in all tumor tissues. Higher grade of brain tumors was correlated with higher Cho/Cr ratio, indicating a significant dependence of Cho levels on malignancy of gliomas. This results suggest that clinical proton MR spectroscopy could be useful to predict tumor malignancy. Effect of plant growth regulators and carbon source on the shoot regeneration and rooting of 'Wonhwang' pear (Pyrus pyrifolia L.) (배 '원황'의 재분화와 발근에 미치는 식물생장조절물질과 탄소원의 영향)

  • The aim of this research is to establish shoot regeneration system for 'Wonhwang' pear (Pyrus pyrifolia L.) using various concentrations of 1-Naphthaleneacetic acid (NAA) 0.01, 0.05, 0.1, 0.5 mg/L in combination with benzylaminopurine (BA) 3, 5, 10 mg/L. Medium containing 4.4 g/L of Murashige and Skoog (MS) medium with vitamins containing 8 g/L of plant agar and 30 g/L of sucrose with NAA 0.05 mg/L and BA 3 mg/L showed 13.3% of shoot regeneration rate. 'Wonhwang' showed no root growth on existing rooting media of P. pyrifolia cv. Niitaka, 'Whang-keumbae' and 'Bae Yun No. 3'. We evaluated the effect of concentration and kinds of plant growth regulators and carbon source to establish efficient rooting condition for 'Wonhwang' pear. In the result of using various concentrations of NAA 0.5 mg/L and 1.0 mg/L in combination with indolebutyric acid (IBA) 3, 5, 10 mg/L, rooting rate of 24% was observed using 1/4 Linsmaier and Skoog (LS) medium supplemented with 7.5 g/L glucose as carbon source and IBA 1.0 mg/L with NAA 1.0 mg/L.

Intersubgeneric Protoplast Fusion of Pelargonium aridum (Ligularia) and P.zonale(Ciconium) (Pelargonium aridum과 P. zonale 아속간의 원형질체융합)

  • In an attempt to obtain intersubgeneric somatic hybrids of Pelargonium aridum and P.zonale, protoplast isolated from the two species were fused by using polyethylene glycol(PEG) and electorfusion methods. Protoplast were isolated from cotyledon and leaf tissues using MS medium containing 550 mM sucrose, 0.7% cellulase (Onozuka R-10) and 0.4% Macerozyme. The optimum number of protoplasts per mL of culture medium was 6 x 10$^4$. Protoplast fused by the electrofusion method were more active than by PEG method. Heterokaryotically fused protoplasts formed calli when cultured in MS medium containing 550 mM glucose, 1 to 2 mg/L NAA and 0.5 to 1 mg/L BA.
