Infection Status with the Metacercariae of Heterophyid Trematode in Mullet and Goby Collected from Western Coastal Areas of Cholla-do, Korea

  • The present study was performed to investigate the infection status of heterophyid fluke metacercariae in mullets and gobies collected from some coastal areas of cholla-do. The collected fishes were transferred to the laboratory, artificially digested with pepsin-HCI solution and examined under a stereomicroscope. In 70 mullets examined, the metacercariae of Heterophyes nocens (in 28.6% of fish), Heterophyopsis continua (in 2.9%), Pygidiopsis summa (in 32.9%) and Stictodora spp. (in 14.3%) were detected. The metacercariae of H nocens were detected in mullets from Shinan-gun (in 60.0% of fish), Muan-gun (in 54.4%), Hampyong-gun (in 25.0%) and Yonggwang-gun (in 16.7%). The average metacercarial density per infected fish were 7.6,34, 10 and 5 in each areas. A total of 7 metacercariae of H. continua was detected in only 2 mullets from Hampyong-gun. The metacercariae of p. summa were detected in mullets from Haenam-gun (in 80.0% offish), Muan-gun (in 27.3%), Hampyong-gun (in 16.7%) and Puan-gun (in 100%). The average metacercarial density per infected fish were 350, 14, 5 and 97 in each areas. The metacercariae of Stictodora spp. were detected in mullets from Shinan-gun (in 26.7% offish), Hampyong-gun (in 33.3%) and Puan-gun (in 20.0%). The average metacercarial density per infected fish were 2.8, 18.8 and 2.5 in each areas. In 70 gobies examined, the Metacercariae of H nocens (in 15.7% of fish), H continua (in 47.1%) and Stictodora spp. (in 48.6%) were detected. The metacercariae of H. nocens were detected in gobies from Shinan-gun (in 50.0% of fish) and Yonggwang-gun (in 10.0%). The average metacercarial density per infected fish were 71 and 2 in each areas. The metacercariae of H. continua were detected in gobies from Shinan-gun (in 65.0% of fish), Hampyong-gun (in 70.0%) and Yonggwang-gun (in 60.0%). The average metacercarial density per infected fish were 5.7,6.6 and 3.2 in each areas. The metacercariae of Stictodora spp. were detected in gobies from Shinan-gun (in 80.0% offish), Hampyong-gun (in 70.0%) and Puan-gun (in 40.0%). The average metacercarial density per infected fish were 149,203 and 5.5 in each areas. From the above results, it was confirmed that the mullet and goby from some coastal areas of cholla-do are infected with numerous metacercariae of heterophyid trematodes.

Morphological Characteristic of the Rust Fungi, Uromyces truncicola, and Histological Changes in the Infected Host Tree, Sophora japonica (회화나무 녹병균(Uromyces truncicola)의 형태적 특징과 감염 기주의 조직해부학적 변화)

  • rust fungus, Uromyces truncicola, infected stems, branches and leaves of Sophora japonica trees, and 78% of planted host depending on investigated sites. Teliospores of this fungus were reddish brown, global to oval and $29-37{\times}24-27\;{\mu}m$. The urediniospores were yellowish brown, elliptical to oval with spiny surface and $28-39{\times}19-22\;{\mu}m$. In the infected branches cork layers were thickened, ray parenchymatous cells were distorted, branched or combined and increased in numbers. While ray parenchymatous cells in healthy branches were one or two layers, the cells in the infected branches were three or more layers to shape fusiform. In the infected branches xylem tissues also distorted and yellowish pigments formed in the intercellular space. In the less infected trees the hairs on the branches were dense and long, and the thickness of upper epidermal cell walls were ca 23.3 ${\mu}m$, while in the highly infected trees the hairs were less dense and short, and the thickness of upper epidermal cell walls was ca 17.4 ${\mu}m$. We think individual host trees resistant to this fungus are present.

  • In this study, ultrastructural change of the bile duct fibroblast at infected rat with Clonorchis sinensis, and the distribution of lectin receptors and actin protein in cultured bile duct infected with Clonorchis sinensis. It explored using colloidal gold label complex with lectin WGA purified from wheat germ (Triticum vulgaris) and anti actin antibody purified actin (43 kDa) isolated from chicken back muscle. The lectin WGA with protein A gold complex labeled sections of the cultured fibroblast revealed gold particles specifically distributed on the multi vesicular form Golgi complex and cell surface of the fibroblast. The actin antibody with protein A gold complex labeled sections of the cultured fibroblast revealed gold particles specifically distributed on the cytoplasm of the fibroblast. Labeling of cultured fibroblast in rat bile duct infected with Clonorchis sinensis was then quantified and compared to that of cultured Fibroblast in Rat Bile duct. These results indicate that lectin WGA receptors are located in the multi vesicular form Golgi complex in the cytoplasm to the cytoplasmic process of the Rat bile duct fibroblast infected with Clonorchis sinensis. Therefore, the GlcNAc and NeuNac regions on the cell surface and cytoplasmic process appear to be functionally associated with cell-recognition and protection from other cell of the tissue, and linked with secretion and exocytosis of the fibroblst cytoplasm. GlcNAc and NeuNAc product in the multi vesicular form Golgi complex then it is transported to cell surface. Actin protein is many appears that infected fibroblast rather than normal fibroblast. The fibroblast of infected with Clonorchis sinensis are against of the physical and chemical stimulation. Then development of cytoplasmic process is relative some stimulation.

Pregnancy Rates and Outcomes of HIV-Infected Women in Korea

  • Background: Antepartum, intrapartum, and postpartum preventive measures with antiretroviral drugs, appropriate delivery methods, and discouraging breastfeeding significantly decrease the risk of mother-to-child transmission of human immunodeficiency virus (HIV) infection. Herein, we investigated the pregnancy outcomes in HIV-infected Korean women. Methods: We retrospectively reviewed medical records of childbearing-age HIV-infected women between January 2005 and June 2017 at four tertiary care hospitals in Korea. Results: Among a total of 95 HIV infected women of child-bearing age with 587.61 years of follow-up duration, 15 HIV-infected women experienced 21 pregnancies and delivered 16 infants. The pregnancy rate was 3.57 per 100 patient-years. Among the 21 pregnancies, five ended with an induced abortion, and 16 with childbirth including two preterm deliveries at 24 and 35 weeks of gestation, respectively. The two preterm infants had low birth weight and one of them died 10 days after delivery due to respiratory failure. Among the 14 full-term infants, one infant was small for gestational age. There were no HIV-infected infants. Conclusion: The pregnancy rate of HIV-infected women in Korea is lower than that of the general population. Although several adverse pregnancy outcomes were observed, mother-to-child transmission of HIV infection was successfully prevented with effective preventive measures.

Pathogenesis of Human Norovirus Genogroup II Genotype 4 in Post-Weaning Gnotobiotic Pigs

  • Norovirus is the most common cause of acute gastroenteritis. Its pathogenesis is poorly understood owing to the difficulty of establishing viral infection in animal models. Here, post-weaning gnotobiotic pigs were infected with human norovirus genogroup II genotype 4 (HuNoV GII.4) to investigate the pathogenesis and replication of the virus. Three groups of four pigs were infected with $1{\times}10^5$, $1{\times}10^6$, or $1{\times}10^7$ genomic equivalent (GE) copies of HuNoV GII.4. Four pigs were used as negative controls. Blood and rectal swab samples were collected after viral infection, and gross legions were examined after necropsy. Diarrhea was induced in 25% and 75% of pigs infected with $1{\times}10^6$ and $1{\times}10^7$ GE copies, respectively. Viral shedding was detected in 50%, 75%, and 50% of pigs infected with $1{\times}10^5$, $1{\times}10^6$, and $1{\times}10^7$ GE copies, respectively. Viremia was detected in 25% of pigs infected with either $1{\times}10^6$ or $1{\times}10^7$ GE copies. When gross lesions of gastroenteritis were investigated, the ileum walls of the infected pigs were thinner than those of the controls. Villi atrophy and inflammatory cell infiltration were identified in the ileum of each infected pig. Viral capsid was identified in the jejunum, ileum, colon, spleen, and mesenteric lymph node. Virus replication was newly verified in the spleen and mesenteric lymph nodes by detection of negative-sense viral RNA. In conclusion, HuNoV GII.4 could induce acute gastroenteritis and replicate in the extra-intestinal lymphoid tissues in post-weaning gnotobiotic pigs. Therefore, such pigs would be a suitable animal model for studying the pathogenesis and replication of HuNoV.

Induction of IFN-β through TLR-3- and RIG-I-Mediated Signaling Pathways in Canine Respiratory Epithelial Cells Infected with H3N2 Canine Influenza Virus

  • Canine influenza virus (CIV) induces acute respiratory disease in dogs. In this study, we aimed to determine the signaling pathways leading to the induction of IFN-β in a canine respiratory epithelial cell line (KU-CBE) infected with the H3N2 subtype of CIV. Small interfering RNAs (siRNAs) specific to pattern recognition receptors (PRRs) and transcription factors were used to block the IFN-β induction signals in H3N2 CIV-infected KU-CBE cells. Among the PRRs, only the TLR3 and RIG-I expression levels significantly (p < 0.001) increased in CIV-infected cells. Following transfection with siRNA specific to TLR3 (siTLR3) or RIG-I (siRIG-I), the mRNA expression levels of IFN-β significantly (p < 0.001) decreased, and the protein expression of IFN-β also decreased in infected cells. In addition, co-transfection with both siTLR3 and siRIG-I significantly reduced IRF3 (p < 0.001) and IFN-β (p < 0.001) mRNA levels. Moreover, the protein concentration of IFN-β was significantly (p < 0.01) lower in cells co-transfected with both siTLR3 and siRIG-I than in cells transfected with either siTLR3 or siRIG-I alone. Also, the antiviral protein MX1 was only expressed in KU-CBE cells infected with CIV or treated with IFN-β or IFN-α. Thus, we speculate that IFN-β further induces MX1 expression, which might suppress CIV replication. Taken together, these data indicate that TLR3 and RIG-I synergistically induce IFN-β expression via the activation of IRF3, and the produced IFN-β further induces the production of MX1, which would suppress CIV replication in CIV-infected cells.

Effect of severe acute respiratory syndrome coronavirus 2 infection during pregnancy in K18-hACE2 transgenic mice

  • Objective: This study aimed to examine the influence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection on pregnancy in cytokeratin-18 (K18)-hACE2 transgenic mice. Methods: To determine the expression of hACE2 mRNA in the female reproductive tract of K18-hACE2 mice, real-time polymerase chain reaction (RT-PCR) was performed using the ovary, oviduct, uterus, umbilical cord, and placenta. SARS-CoV-2 was inoculated intranasally (30 μL/mouse, 1×104 TCID50/mL) to plug-checked K18-hACE2 homozygous female mice at the pre-and post-implantation stages at 2.5 days post-coitum (dpc) and 15.5 dpc, respectively. The number of implantation sites was checked at 7.5 dpc, and the number of normally born pups was investigated at 20.5 dpc. Pregnancy outcomes, including implantation and childbirth, were confirmed by comparison with the non-infected group. Tissues of infected mice were collected at 7.5 dpc and 19.5 dpc to confirm the SARS-CoV-2 infection. The infection was identified by performing RT-PCR on the infected tissues and comparing them to the non-infected tissues. Results: hACE2 mRNA expression was confirmed in the female reproductive tract of the K18-hACE2 mice. Compared to the non-infected group, no significant difference in the number of implantation sites or normally born pups was found in the infected group. SARS-CoV-2 infection was detected in the lungs but not in the female reproductive system of infected K18-hACE2 mice. Conclusion: In K18-hACE2 mice, intranasal infection with SARS-CoV-2 did not induce implantation failure, preterm labor, or miscarriage. Although the viral infection was not detected in the uterus, placenta, or fetus, the infection of the lungs could induce problems in the reproductive system. However, lung infections were not related to pregnancy outcomes.

Effect of Systemic Fungicide on Total Hemocyte Count and Hemolymph Biochemical Changes in Silkworm, Bombyx mori L., infected with Beau-veria bassiana

  • Silkworm diseases are major constraint in silk cocoon production. Among silkworm diseases, white muscar-dine is highly contagious and most common in winter and rainy seasons. It is suggested that hemocytes involve in defense against invasion of Beauveria bassi-ana and systemic fungicide/chemicals prevent the proliferation of fungi in the hemolymph or preventing the growth of the fungi in the body cavity through enhancing the hemocyte mediated defense response. In the present study the influence of systemic fungicide on hematological changes in silkworms infected with Beauveria bassiana was reported. It is observed that the total hemocyte counts increased in the hemolymph up to 5 days post inoculation in systemic fungicide treated batches while in the inoculated control the increase was up to 3 days indicating the positive hemocyte mediated response in silkworm treated with systemic fungicide. After 2 days in the inoculated control as the multiplication and growth of mycelia increased, defense capacity of the silkworm was decreased. The biochemical changes were also observed in the hemolymph of silkworm infected with B. bassiana. In silkworm infected with Beauveria bassi-ana, the total protein content increased whereas total carbohydrate and total lipids decreased as the infection progresses. In the case of systemic fungicide treated batches the increase in total protein content was comparatively higher and decrease in total carbohydrate and lipids were comparatively lower than the inoculated control.

Machine Vision Based Detection of Disease Damaged Leave of Tomato Plants in a Greenhouse (기계시각장치에 의한 토마토 작물의 병해엽 검출)

  • Machine vision system was used for analyzing leaf color disorders of tomato plants in a greenhouse. From the day when a few leave of tomato plants had started to wither, a series of images were captured by 4 times during 14 days. Among several color image spaces, Saturation frame in HSI color space was adequate to eliminate a background and Hue frame was good to detect infected disease area and tomato fruits. The processed image ($G{\sqcup}b^*$ image) by OR operation between G frame in RGB color space and $b^*$ frame in $La^*b^*$ color space was useful for image segmentation of a plant canopy area. This study calculated a ratio of the infected area to the plant canopy and manually analyzed leaf color disorders through an image segmentation for Hue frame of a tomato plant image. For automatically analyzing plant leave disease, this study selected twenty-seven color patches on the calibration bars as the corresponding to leaf color disorders. These selected color patches could represent 97% of the infected area analyzed by the manual method. Using only ten color patches among twenty-seven ones could represent over 85% of the infected area. This paper showed a proposed machine vision system may be effective for evaluating various leaf color disorders of plants growing in a greenhouse.

Factors Influencing Clinical Nurses' Nursing Intention for High Risk Pathogen Infected Patient (임상간호사의 고위험 병원체 감염 환자 간호의도 영향요인)

  • Purpose: The purpose of this study was to determine factors related to clinical nurses' nursing intention for high risk pathogen infected patient. Methods: The participants were 230 clinical nurses from four university hospitals which located in B and D metropolitan cities. Data were collected from September 14 to 25 in 2015 and were analyzed using a hierarchical regression. Results: The mean of nursing intention score for high risk pathogen infected patient for the participants was $110.62{\pm}12.68$ (range 36~180). The most influential factor for nursing intention was professional self concept of nurses (${\beta}=.321$), and organizational group culture (${\beta}=.166$) was the next influential factor. The level of education and working department were also statistically significant contributors. Conclusion: It is necessary to increase clinical nurses' nursing intention for high risk pathogen infected patient to improve professional self concept of nurses and organizational group culture.
