Characteristics of nonalcoholic fatty liver disease induced in wistar rats following four different diets

  • BACKGROUND/OBJECTIVES: The prevalence of nonalcoholic fatty liver disease (NAFLD) has increased worldwide in parallel with overnutrition characterized by high-fat and high-carbohydrate intake. Our objective was to establish, in 16 weeks, a model of NAFLD in Wistar pathogen-free rats following four dietary types. MATERIALS/METHODS: Forty (6 weeks old) healthy Wistar male rats, weighing an average of 150 g were randomly divided into four groups of ten and assigned a diet with the same quantity (15 g/rat/day), but with different composition. The moderate-fat (MF) group was fed a moderate-fat diet (31.5% fat and 50% carbohydrates), the high-fat (HF) group was fed a fat-rich diet (51% fat), the high-sucrose (HS) group and the high-fructose (HFr) group were fed a carbohydrate-rich diet (61%). The carbohydrate contents of the HS group was composed of 60.3% sucrose while that of the HFr group was composed of 59.3% fructose. RESULTS: At week 16, the HF group had the highest percentage of cells enriched in fat (40%) and the highest weight and liver weight (P < 0.05). The HFr group showed significantly higher levels of serum triglycerides, alanine aminotransferase and adiponectin at week 16 as compared to week 1 (P < 0.05). CONCLUSIONS: The 15 g/rat/day diet composed of 51% fat or 61% carbohydrates enriched mainly in fructose may induce characteristics of NAFLD in rats.

한국인의 식이 섭취 상태에 따르는 흰쥐의 성장 발달에 미치는 영향 (A Study of Growth a Development of Rats Fed by Korean Diet Patterns)

  • This study was designed to compare the metabolic effects of varicous types of Korean diet. 40 males and same number of females of Albino rats, divided into eight groups, 5 rats each. 1 Group; Sugar casein standard Group 2 Group; Rice casein standard Group 3 Group; Rice group 4 Group; 65% Rice +35% Anchovy 5 Group; 65% Rice +35% Chinese cabbage 6 Group; 65% Rice+10% Anchovy +25% Chinese cabbage 7 Group; 65%. Rice+26% Chinese cabbage+3.5% Anchovy+4% Bean+1% Potatoes+0.2% Sea Weeds+0.3% Seasonings 8 Group; 47% Rice+24% Chinese cabbage+20.5% Anchovy+5% Bean+1% Potatoes+1.8% Sea Weeds+0.7% Seasonings The rats were kept in individual cage and given 8 different diet for 10 weeks. The result of this study were elucidated as fallow. Food intake of sugar casein standard group and rice casein standard group and Seoul diet pattern group were high, Rice diet group showed low food intake. F.E.R, P.E.R, body weight, organ weight were the similar results. The nitrogen content in various organs were no great difference, but nitrogen metabolism and total nitrogen retention were significant differences. The lipid content in the liver showed no significant differences, but fecal lipid and serum cholesterol showed significant differences. This study showed the glucose content in urine and feces were due to the dietary carbohydrate content. In other word, the results of this study showed no significant differences between sugar casein standard group and rice casein standard group, but significiant differences between standard group and experimental group.

한국산 잣기름이 콜레스테롤 첨가식이로 사육한 토끼의 혈장 총지방산 조정에 미치는 영향 (Effect of Supplement of Korean Pinenut Oil on Plasma Total Fatty Acid Composition in Cholesterol-fed Rabbits)

  • The present study was carried out in atheroscleorotic New Zealend white(NZW) rabbits. to evaluate the effect of dietary supplementation with Korean pinenut oil, on plasma total fatty acid composition. In study I, NZW rabbits were fed 10 weeks on a commercial chow diet supplemented with 5% of energy as fats(soybean oil or pinenut oil) or 10% of energy as fats(soybean oil or pinenut oil) with the addition of 1% cholesterol to the diet. Nineteen fatty acids ranged from myristic acid (14:0) to cervonic acid (22:6 ${\omega}3$) were identified in all the samples. The c5, c9, $c12{\sim}18$ : 3 acid was not reported in the fatty acid methyl ester profiles of each group because it was included in the linoleic acid peak. The major constitutent fatty acids in the chow diet group were linoleic acid, oleic acid, palmitic acid and ${\alpha}$-linolenic acid. In the cholesterol group, oleic acid, linoleic acid and palmitic acid were the major fatty acids. In plasma of cholesterol-fed animals, the levels of 16:1 ${\omega}$ 7 and 18:1 1 ${\omega}$ 9 were increased. Linoleic acid was the major fatty acid in soybean oil/cholesterol and pinenut oil/cholesterol groups. Plasma linoleic acid levels were significantly incresed from 4 to 6% by the supplementation of 5% soybean or 5% pinenut oil in the cholesterol diet for 5 weeks, compared to cholesterol group. Plasma 16 : 1 ${\omega}$ 7 levels in animals fed with 5 or 10% pinenut oils were significantly lower than in those fed cholesterol for 5 weeks. After 10 weeks on the soybean oil and pinenut oil diet there were no significant differences in the fatty acid composition. In study II, the fatty acid composition was not affected by the types or levels of oils supplemented for 5 weeks. After 10 weeks on the oil diets 16:1 ${\omega}$ 7 and 18:1 ${\omega}$ 9 were decreased in 10% soybean in oil/cholesterol and 10% pinenut oil/cholesterol groups, compared to cholesterol group.

조선 인조(仁祖)의 질병관리 중 약죽(藥粥)의 적용과 의미에 관한 고찰 - 승정원일기 기록을 중심으로 - (Review of Application of Medicinal Porridges by King-Injo of the Joseon Dynasty - Based on the Records from The Daily Records of the Royal Secretariat of Joseon Dynasty -)

  • During the Joseon Dynasty, medicinal foods derived from herbs were often more effective than traditional medicines. In addition, the royal family of the Joseon Dynasty believed that foods could be used as various disease treatments. Grain-based foods, especially medicinal porridges (藥粥), were most frequently used for diet therapy. We investigated various types of diet-related diseases suffered by King Injo (仁祖) as well as how the diseases were treated using medicinal porridges based on information in the SeungjeongwonIlgi (承政院日記), which is the daily record of the Royal Secretariat of the Joseon Dynasty. This study examined the SeungjeongwonIlgi of King Injo from his1st year (1623) to 27th year (1649) on a website database maintained by the National Institute of Korean History. According to the records, King Injo suffered from severe diarrhea several times due mainly to febrile disease (煩熱症) as well as abdominal dropsy (脹滿) throughout his entire life. Major diseases affecting King Injo were due to his unhealthy eating habits and psychological factors. For treatment, royal doctors prescribed around 15 medicinal porridges, including nelumbo (seed) porridge (Yeonja-juk), milk porridge (Tarak-juk), Chinese dioscorea porridge (Sanyak-juk), mungbean porridge (Nokdu-juk), perilla seed porridge (Imja-juk), adzuki-bean porridge (Pat-juk), soybean porridge (Kong-juk), Korean-leek porridge (Buchu-juk), and so on, in addition to other medical treatments. Diet therapy using medicinal porridges has been used throughout history since the Joseon Dynasty period. However, knowledge of traditional diet therapy and medicinal porridges used by monarchs in the Joseon Dynasty is insufficient. Therefore, in-depth study is needed to understand the theory of traditional medicinal foods as well as explore their application to patients in the context of modern medicine.

부상, 반부상, 침강 및 습사료가 조피볼락의 성장 및 어체 조성에 미치는 영향 (Effects of Extruded-floating, Slow-sinking, Fast-sinking or Moist Pellet Diets on the Growth and Body Composition in Korean Rockfish(Sebastes schlogeli))

  • 사료물성과 단백질원 배합비에 따른 조피볼락의 성장차이를 구명하기 위하여, 실험사료의 단백질원을 북양어분과 갈색어분으로 한 사료와 경제적인 측면을 고려하여 어분의 배합 비율을 낮추는 대신, 대두박, 콘글루텐 밀, 육분 및 우모분의 비율을 늘린 2종의 사료를 설계하여, 각각 EP(extruded-floating pellet, 부상사료), SP(slow-sinking pellet, 반부상사료), FP(fast-sinking pellet, 침강사료)와 MP (moist pellet, 습사료) 형태의 배합사료를 제조하였다. 평균체중 75 g의 조피볼락을 각 수조에 50마리씩 수용하여 $2(diet\times)4(diet type)\times3(replication)$ factorial design으로 15주간 사육하였다. 사육 실험한 결과, 실험사료와 사료형태에 따른 최종평균체중, 증체율, 사료효율, 사료 섭취율, 단백질 효율, 간중량비 및 내장중량비는 전혀 차이가 없는 것으로 나타났다(P>0.05). 실험 종료시의 실험 구별, 어체 부위별 일반성분 중에 사료별, 사료 형태별로 모두 간과 근육의 단백질 및 근육의 수분과 지질 함량은 실험구간별로 유의적인 차이가 없었다(P>0.05). 위와 같이 사료의 물성이나 침강 속도는 조피볼락의 성장, 영양소 이용효율, 사료 섭취율 및 체성분에 영향을 미치지 않은 것으로 나타나, 양식 현장에서 사료 형태에 관계없이 적절히 선택하여 사용할 수 있을 것으로 전망된다.

Effect of Diet and Water Intake on Aquaporin 2 Function

  • Appropriate control of diet and water intake is important for maintaining normal blood pressure, fluid and electrolyte homeostasis in the body. It is relatively understood that the amount of sodium and potassium intake directly affects blood pressure and regulates ion transporters; Na and K channel functions in the kidney. However, little is known about whether diet and water intake regulates Aquaporin (AQP) function. AQPs, a family of aquaporin proteins with different types being expressed in different tissues, are important for water absorption by the cell. Water reabsorption is a passive process driven by osmotic gradient and water permeability is critical for this process. In most of the nephron, however, water reabsorption is unregulated and coupled to solute reabsorption, such as AQP1 mediated water absorption in the proximal tubule. AQP2 is the only water channel founded so far that can be regulated by hormones in the kidney. AQP2 expressed in the apical membrane of the principal cells in the collecting tubule can be regulated by vasopressin (antidiuretic hormone) controlling the final volume of urine excretion. When vasopressin binds to its receptor on the collecting duct cells, it stimulates the translocation of AQP2 to the membrane, leading to increased water absorption via this AQP2 water channel. However, some studies also indicated that the AQP2 is also been regulated by vasopressin independent mechanism. This review is focused on the regulation of AQP2 by diet and the amount of water intake on salt and water homeostasis.

Anti-inflammatory and anti-diabetic effects of brown seaweeds in high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: Seaweeds have been reported to have various health beneficial effects. In this study, we investigated the potential anti-obesity and anti-inflammatory effects of four types of domestic brown seaweeds in a high-fat diet-induced obese mouse model and bone marrow-derived macrophages (BMDM). MATERIALS/METHODS: Male C57BL/6N mice were fed low-fat diet (LFD), high-fat diet (HFD) or HFD containing Undaria Pinnatifida, HFD containing Laminaria Japonica (LJ), HFD containing Sargassum Fulvellum, or HFD containing Hizikia Fusiforme (HF) for 16 weeks. RESULTS: Brown seaweed supplementation did not affect long-term HFD-associated changes in body weight or adiposity, although mice fed HFD + LJ or HFD + HF gained slightly less body weight compared with those fed HFD at the beginning of feeding. Despite being obese, mice fed HFD + LJ appeared to show improved insulin sensitivity compared to mice fed HFD. Consistently, we observed significantly reduced blood glucose concentrations in mice fed HFD + LJ compared with those of mice fed HFD. Although no significant differences in adipocyte size were detected among the HFD-fed groups, consumption of seaweeds decreased formation of HFD-induced crown-like structures in gonadal adipose tissue as well as plasma inflammatory cytokines. BMDM from mice fed HFDs with seaweeds showed differential regulation of pro-inflammatory cytokines such as IL-$1{\beta}$ and IL-6 compared with BMDM from mice fed HFD by LPS stimulation. CONCLUSION: Although seaweed consumption did not prevent long-term HFD-induced obesity in C57BL/6N mice, it reduced insulin resistance (IR) and circulation of pro-inflammatory cytokines. Therefore, seaweeds may ameliorate systemic inflammation and IR in obesity partially due to inhibition of inflammatory signaling in adipose tissue cells as well as bone marrow-derived immune cells.

사상체질 식이중재연구 현황분석 : Participant, Intervention, Comparison, Outcome (PICO)를 중심으로 (Analysis on Intervention Studies of Sasang Constitutional Diet : Participant, Intervention, Comparison, and Outcome (PICO))

  • Objectives The purpose of this study was to evaluate intervention studies on Sasang Constitutional diet (SCD) through the checks of Participant, Intervention, Comparison, and Outcome (PICO) Methods Randomized controlled trial (RCT) and non-randomized study for intervention (NRSI) about SCD were searched in 4 Korean core databases and other sources, and then PICO was checked. Results 1. Total 10 studies were conducted with 1 RCT and 9 NRSIs. 2. Participants were people with no specific disease, or patients with essential hypertension, hyperlipidemia, obesity, or stroke with diabetes or hyperlipidemia. Most studies were conducted on groups of various Sasang Constitutional types except Taeyangin. 3. Two studies provided participants with meals and exercise. Three studies, instead of providing meals directly, taught participants how to eat SCD on their own. 4. NRSIs have tested the effectiveness of various outcome measures without the presentation of primary outcome, and then concluded that all outcomes were ineffective or some are effective. 5. There was no mention of adverse events. In most studies, a single doctor of Korean medicine diagnosed Sasang Constitution the QSCC II questionnaire. The intervention period ranged from three weeks to three months, and recent studies have conducted interventions for 12 weeks. Conclusions Intervention studies about SCD which were conducted so far have shown problems on the study design of PICO items. The study design and implementation that carefully consider how to maintain similarity between groups, minimize the risk of bias, set primary outcome measure, and control the diet are required.

단식중인 흰쥐에서 지방섭취가 생존 대사에 미치는 영향 (The Effects of Dietary Fat on Survival Metabolism of Fasting Rat)

  • 완전 단식군의 흰쥐(FO)와 단식중에 소기름(FT), 옥수수기름(FC), 들깨기름(FP)만을 각각 투여하고 비교하니 FO군은 대부분 6일까지 생존하고 FT, FC, FP군은 대부분 16일까지 생존하였다. 폐사 직전의 모든 흰쥐 체중은 -24.5~-25%로 감소되었다. 단식중 유지 섭취는 다소 생명을 연장하나 식이 유지 종류에 따른 생존기간과 체중 변화에 미치는 영향에는 괄목할 차이가 없었다. 간장과 신장 무게 변화도 체중변화와 유사한 양상을 보였다. FO군의 혈중 총 cholesterol 농도, 중성지방 농도, 혈당은 대폭 감소되고 GPT 활성값과 BUN 농도는 상당 수준으로 증가되었다. FC군과 FP군에서 유지의 n-6/n-3비율 혹은 불포화도 특성 차이가 총 cholesterol농도, 중성지방농도, 혈당농도, BUN농도, 단식 초기의 GPT활성도에 영향을 주지 못했다. 그러나 기아사 직전의 FP군에서 GPT활성도가 FT, FC군보다 높았다. 본 연구에서 결론은 기아 상태의 흰쥐에 유지 단독섭취는 유지 종류에 관계없이 생존일수를 다소 비슷하게 연장하였고 n-6/n-3비율, 불포화도 특성차이가 전반적으로 단식중의 대사에 미치는 영향에서 별다른 차이가 없었음을 알 수 있었다.

암 환자의 정보요구 분석 (Information Needs on Patients with Cancer in Korea)

  • Purpose: To explore what particular types of information were important to patients diagnosed with cancer. Methods: Seventy three patients with cancer at an outpatient clinic and hospitalized patients in W Christian Hospital Korea, responded. The structured questionnaire developed by the investigator based on previous studies. Results: There was a significant negative relationship between age and the score of informational need (r=-.307, p<.05). Level of education, and level of monthly income were related to level of informational need. The top three informational priorities according to the time since diagnosis were 'Self care during treatment', 'Health food and diet', 'Likelihood of recurrence', 'Follow up care' and 'Side effects'. The top three informational priorities for patients with breast cancer were 'Likelihood of recurrence', 'Metastasis possibility', 'Treatment options', and 'Side effects. For patients with stomach cancer, they were 'Follow up care', 'Healthy food and diet', 'Likelihood of recurrence', and 'Metastasis possibility', and for patients with colon/rectal cancer, they were 'Side effects', 'Healthy food and diet', 'Likelihood of recurrence', and 'Self care during treatment'. Conclusion: The assessment of information needs based on demographic factors and disease-related factors is critical in helping patients with cancer to manage their illness.
