옻나무 유래 Flavonoid 처리가 흰쥐 Leydig 세포의 체외배양에서 Testosterone 분비에 미치는 영향 (Effects of Flavonoid from Rhus verniciflua on Testosterone Secretion by Rat Leydig Cells In Vitro)

  • 본 연구의 목적은 옻나무 유래 F가 성숙 수컷 흰쥐의 생식 기능에 직접 미치는 영향을 알아보고자, 횐쥐 Leydig 세포를 분리, 회수하여 체외배양에서 F 단독 혹은 LH와의 공배양하여 배양액의 testosterone 농도를 조사하였다. 1. Leydig 세포를 36시간까지 체외 배양하여 testosterone의 농도를 조사한 결과, 6시간 배양구에 비해 24시간 배양구가 가장 높은 농도(P<0.05)를 나타내었으며 36시간 배양에는 감소되었다. 2. F를 단독 첨가하여 12시간 배양한 실험에서 F 80ng 첨가구에서 유의적으로(P<0.05) 높은 testosterone 분비를 보였다. 3. LH 10ng 및 100ng 첨가구의 시간별 testosterone 분비의 변화로는, LH 10ng 첨가구에서는 6∼12시간 이후, LH 100ng 첨가구에서는 3∼6시간 사이에서 증가를 보였으며, 6시간에는 두 처리구 모두 유의차를 보였다. 4. LH 10ng 첨가 실험에서는 LH 10ng+F 40ng에서 12시간 배양 시, 유의적으로 높은 testosterone 분비를 확인하였다. 5. LH 100ng과 F의 공배양실험에서 3시간 배양한 결과, F20ng 및 F40ng처리구가 대조구에 비해 유의적으로(P<0.05) 높은 testosterone의 분비를 보였다. 6. Leydig 세포에 LH+IGF-I 첨가효과를 비교 검토한 결과, IGF-I 50ng과 100ng 첨가구에서는 대조구에 비해 높게 나타났으나, LH 100ng 단독처리와 유의차는 없었다. 이상의 결과로, 옻나무 유래 Flavonoid는 횐쥐 정소 Leydig 세포의 testosterone 분비를 촉진하는 작용을 하는 것으로 사료되며, 특히 LH+F구에서 testosterone 분비를 더욱 향상시킴으로써 옻나무 유래 F는 포유동물의 생식기능에 중요하게 작용하는 것으로 사료된다.> 이상의 난소당 난자회수율 (22.6개/63.8%)은 비발정기의 것보다 유의적으로 높았다 (P < 0.05). 발정기의 난소에서 110 $\mu\textrm{m}$ 이상의 난자의 MI까지의 핵발달율 (24.3%)은 110 Um이하의 것 또는 비발정기의 110 $\mu\textrm{m}$ 이상 및 이하의 난자의 것보다 유의적으로 높았다 (2.5, 6.8 및 0.0%; P <0.05). 발정기의 110 $\mu\textrm{m}$ 이상 난자의 AT 또는 MII까지의 핵 발달율은 다른 처리군보다 높게 발달하였다. TCM199에서 MI까지의 핵발달율 (21.8%)은 $\alpha$-MEM (10.0%)보다 유의적으로 높았다 (P< 0.05). 그러나 AT까지의 핵 발달율은 TCM199 (7.3%)과 (-MEM (1.1%) 간에는 유의적 차이가 있었으나 (P<0.05), MII까지는 TCM199 (0.9%)과 (-MEM (1.1%)에는 유의차가 없었다. 본 연구결과는 110 $\mu\textrm{m}$ 이상의 난자는 발정기의 난소로부터 더 많은 난자를 회수할 수 있었고, 110 $\mu\textrm{m}$ 이상의 난자들이 MI, AT까지의 핵발달 능력이 높았다. 또한 체외성숙배양액 시 TCM199이 $\alpha$-MEM보다 Mi과AI까지 높은 발달율을 보였다.U/$m\ell$ FSH+10 $m\ell$U/$m\ell$ LH 의 첨가가 생쥐 preantral follicles의 체외배양을 위한 적정조건임을 제시하고 있다., 30, 40 $\mu\textrm{g}$/kg을 각각 2일간 투여했을 때 신장, 비장 및 간의 중량은 정상대조군과

Effects of Testosterone, 17β-estradiol, and Progesterone on the Differentiation of Bovine Intramuscular Adipocytes

  • The aim of this study was to investigate the effects of testosterone, 17$\beta$-estradiol, and progesterone on the differentiation of bovine intramuscular adipocytes (BIA). Stromal-vascular (SV) cells were obtained from M. longissimus dorsi of 20 months old Korean (Hanwoo) steers, and were cultured in DMEM containing 5% FBS. The proliferated BIA were induced to differentiate with 0.25 $\mu$M dexamethasone, 0.5 mM 1-methyl-3-isobutyl-xanthine and 10 $\mu$g/ml insulin. During differentiation, the cells were treated with testosterone, 17$\beta$-estradiol, and progesterone at concentrations of $10^{-10}$, $10^{-9}$, and $10^{-8}$ M, respectively, for 12 days. Regardless of its concentration, testosterone remarkably reduced lipid droplets in the cytosol of BIA. On the other hand, 17$\beta$-estradiol and progesterone increased the accumulation of lipid droplets in BIA. Testosterone significantly (p<0.05) decreased GPDH activities with a dose-dependent pattern. 17$\beta$-Estradiol treatment onto BIA during differentiation, however, increased GPDH activity showing the highest activity (11.3 nmol/mg protein/min) at $10^{-10}$ M. Treatment of BIA with progesterone also increased (p<0.05) GPDH activity with the highest activity (13.8 nmol/mg protein/min) at $10^{-9}$ M. In conclusion, the results in the current study suggest that testosterone inhibits differentiation of BIA by suppressing GPDH activity while 17$\beta$-estradiol and progesterone have adverse effects.

Effect of Phellius linteus water extract on benign prostatic hyperplasia

  • Benign prostatic hyperplasia (BPH) is one of the most common diseases among elderly men. As the old-age population is increasing recently, it is to our interest to observe the growing BPH within them. In BPH, the dihydrotestosterone (DHT) acts as promotes prostate growth. It inhibits enzyme $5{\alpha}$-reductase that is involved in the conversion of testosterone to the DHT activity which reduces the excessive prostate growth. Through experiments, the effects of Phellius linteus water extract performed on the BPH rats were induced by testosterone treatments. For 12 weeks, Sprague-Dawley rats were treated with testosterone for the induction of BPH. Rats were divided into four experimental groups: the not treated group (N), the testosterone injection and D.W treatment group (TN), the testosterone injection and Phellinus linteus treatment group (TP) and testosterone injection and finasteride treatment group (TF). Prostate weight, volume and weight ratio in the TP group and the TF group were significantly lower than the TN group. Testosterone and DHT levels in the TN group were significantly higher than that of the N group. And the TP group was significantly decreased than that of the TN group. While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation; the TP and TF groups showed trophic symptoms and were lined by flattened epithelial cells, thus, the stromal proliferation is relatively low as compared to the TN group. These suggest that Phellinus linteus water extracts may be an useful remedy for treating the benign prostatic hyperplasia.

hCG가 TeBG에 미치는 영향 (Effect of hCG on TeBG)

  • In the previous experiment, authors have shown that during the latter half of estrous cycle there was an increase in plasma testosterone level in the rats stimulated with hCG. To determine the physiologic significance of elevated plasma testosterone, changes of the plasma concentrations of TeBG and testosterone following hCG stimulation were analyzed in the rats having a regular 5 day cycle. The rats were divided into three groups; the control, the rats stimulated with single hCG on the day of proestrus and stimulated with hCG throughout the entire cycle. Blood samples were obtained once a day for an estrous cycle and analyzed for the binding capacity of TeBG using ammonium sulphate precipitation method and testosterone concentration by means of radioimmunoassay. Followings were the results; 1) There was no significant variation in the binding capacity of TeBG in peripheral blood during the estrous cycle of the control rats. 2) No cyclic variation in the binding capacity of TeBG was observed in the rats stimulated with single hCG on proestrus. although the levels tended to be higher in the rats with stimulation than in the control rats. 3) Continual stimulation of hCG produced a marked increase in the binding capacity of TeBG especially on the day of metaestrus. 4) The changes in the plasma level of testosterone followed the same basic pattern seen in the TeBG binding capacity. 5) From above results, the followings were suggested. a. hCG related increase of the binding capacity of TeBG is probably secondary to a modest increase in estrogen as well. b. hCG related increase of plasma testosterone in female rats is not entirely due to excess production rather in part due to decreased metabolism induced by the rise in TeBG. c. It seems likely that most of elevated testosterone shown in the rat stimulated with hCG is bound to TeBG and only small portion is unbound form which influence cellular activity. It is rather possible that an increase in TeBG could augment estrogen activity.

백굴채(白屈菜) 추출물의 피지생성 억제효과 (Inhibitory Effect of Extract of Chelidonii Harba on Sebum Synthesis)

  • Sebum is secreted due to the effect of androgen, which start to be secreted at puberty. Androgens have profound effects on the physiology of the sebaceous gland, Using the human sebocyte cell line SZ95, we investigated the inhibitory effect of Chelidonii Harba (CH) on the subum production. Our results showed that numerous cytoplasmic lipid droplets were examined by Oil red staining and lipid droplets were increased markedly by testosterone. Cell viability was dose-dependently decreased by CH as compared with untreated cells, while total lipid content and cholesterol slightly were increased by CH. Testosterone significantly stimulated the synthesis of total lipid and the synthesis of specific sebaceous lipids such as cholesterol and triglyceride. Combined treatment with CH and testosterone resulted in a lower lipid synthesis than with testosterone alone. Especially cholesteol content was reduced by combined treatment with CH and testosterone. These results indicate that CH inhibits the testosterone-induced lipid synthesis in SZ95 cells and acts antagonistically to androgen at the cellular level.

Alleviating effects of the mixture of Elaeagnus multiflora and Cynanchum wilfordii extracts on testosterone deficiency syndrome

  • Testosterone deficiency syndrome (TDS), also known as late-onset hypogonadism, is a clinical and biochemical syndrome associated with advanced age and characterized by deficient serum testosterone levels. The Elaeagnus multiflora fruit (EMF) and Cynanchum wilfordii (CW) have been used in traditional herbal medicine. This study aimed to investigate the therapeutic effects of EMF and CW mixtures (at the ratios of 3:7, 5:5, and 7:3) on TDS using TM3 cells and aging male rats. EMF, and mixtures of EMF and CW (at the ratios of 3:7, 5:5, and 7:3) significantly increased testosterone levels in TM3 cells (p <0.05). The rats were orally administered EMCW (EMF and CW mixed at the ratio of 3:7 50, 100 and 200 mg/kg/day) for 4 weeks consecutively. After 4 weeks of EMCW administration, latency time on the rotarod test, and serum testosterone and dehydroepiandrosterone sulfate levels were significantly increased (p <0.05 and p <0.01). Moreover, the levels of globulin-bound sex hormones were decreased in the EMCW-fed groups. However, prostate-specific antigen levels did not differ among the groups. These results suggest that EMCW can be effectively used to alleviate TDS.

호로파와 야관문 복합추출물의 남성갱년기 개선에 관한 연구 (Effect of a Mixed Extract of Fenugreek Seeds and Lespedeza cuneata on Testosterone Deficiency Syndrome)

  • 본 연구에서는 야관문, 호로파 복합추출인 YHM이 남성갱년기증상의 완화에 영향을 미치는 지를 알아보기 위해 흰쥐에게 YHM을 4주간 투여한 후, testosterone, SHBG, PSA, 체지방, 혈중지방, 근육량, 운동수행능력, 정자수, 간독성 등에 미치는 영향을 조사하였다. YHM 투여 후 남성갱년기의 지표로 사용되는 남성호르몬인 testosterone이 유의적으로 증가하였고, testosterone과 결합하여 기능을 저하시키는 SHBG의 농도는 감소하였다. 또한 남성호르몬 보충요법에서 나타날 수 있는 전립선 관련 부작용을 알아보기 위해 PSA를 측정하였으나, 대조군과 별다른 차이를 나타내지는 않았다. 지방조직과 근육량을 측정한 결과 지방 조직은 줄이고 근육량은 늘리는 것으로 나타나 남성갱년기에서 흔히 있는 근육량 감소 및 체지방 증가를 완화시킬 수 있을 것으로 생각된다. 아울러 혈중지질농도에 있어서도 YHM 투여군이 대조군에 비해 총 콜레스테롤과 중성지방은 감소한 반면 고밀도 콜레스테롤은 증가하였음을 관찰하였다. 강제수영을 통한 운동수행능력 측정에서도 YHM 투여군이 대조군에 비하여 수영시간이 길었고 이는 YHM의 투여가 운동수행능력 향상에 도움을 준 것으로 볼 수 있다. 총 정자수와 활동성 정자수를 측정한 결과에서도 YHM 투여군이 유의적으로 증가한 것으로 나타났고 이는 정자형성과 활동성에 영향을 주는 testosterone 농도의 증가에 기인한 것으로 생각된다. 마지막으로 YHM의 투여가 간독성을 가지는지를 알아보기 위해 AST와 ALT를 측정한 결과 대조군과 별다른 차이를 나타내지 않음으로써 간독성을 가지지는 않는 것으로 판단하였다. 또한, 실험결과 전반적으로 저농도 투여군인 YHM-40에서 고농도 투여군인 YHM-80에서 보다 긍정적인 효과를 나타냈다. 이는 YHM의 효과가 농도 의존적이지 않다는 것을 의미하며, 최적의 생체 이용률을 나타내는 농도가 YHM 40mg/kg인 것으로 판단된다. 결론적으로 YHM의 투여는 남성갱년기에 나타나는 여러 가지 지표들을 개선시키는 것으로 나타났고, 이를 활용하여 남성갱년기 개선에 도움을 줄 수 있는 건강기능성식품 소재로 개발할 수 있을 것으로 기대된다.

수화기제탕(水火旣濟湯)이 주령별 흰쥐의 혈청과 테스토스테론에 미치는 영향 (Effects of Suhwagije-tang Distillate on Serum and Testosterone in Aging Rats)

  • 1. Objectives The purpose of this study is to investigate the effects of oral administration of distilled Suhwagije-tang in aging rats. 2. Methods The SD rats used in this experiment were 6, 48 and 68 weeks old. These groups were treated with distilled water(DW), 1% Vitamin C(Vit. C) and 50% Suhwagije-tang(SGT) distillate respectively, once a day for 4 weeks. Blood samples were taken from heart and the levels of various parameters in serum were analysed. 3. Results and Conclusions (1) At all concentration of SGT, survival rates of liver cells were higher than the control group. (2) The rate of which the 10W-SGT group gained weight was significantly higher than that of the control groups. (3) The levels of albumin in serum of 10W-SGT group were significantly increased in comparison to those of the DW group. The levels of ALP, total cholesterol and HDL-cholesterol in serum of 52W-SGT group were significantly decreased in comparison to those of the DW group. (4) The levels of testosterone in serum of SGT groups showed no significantly changes in comparison to those of the control groups. Testosterone levels were naturally decreased with advancing years, so 72W-DW group's testosterone level was significantly decreased in comparison to those of 10W-DW group's. In contrast, 72W-SGT group's level was the highest in comparison to those of the control groups and those of deviation from 10W, 52W and 72W in SGT groups was also smaller than control groups'. (5) The levels of glucose, total-bilirubin, ALT, AST, BUN, CRP, hs-CRP, homocystein, Na, K, Na/K ratio, Cl and Ca in serum of SGT groups showed no significantly changes in comparison to those of the control groups. These results suggest that oral administration of Suhwagije-tang distillate should be effective in promoting growth and preventing rapid decline of testosterone in aging rats does not cause any imbalance in liver, kidney and electrolyte.

Corn silk extract improves benign prostatic hyperplasia in experimental rat model

  • BACKGROUND/OBJECTIVES: This study was conducted to investigate the effect of a corn silk extract on improving benign prostatic hyperplasia (BPH). MATERIALS/METHODS: The experimental animals, 6-week-old male Wistar rats, were divided into sham-operated control (Sham) and experimental groups. The experimental group, which underwent orchiectomy and received subcutaneous injection of 10 mg/kg of testosterone propionate to induce BPH, was divided into a Testo Only group that received only testosterone, a Testo+Fina group that received testosterone and 5 mg/kg finasteride, a Testo+CSE10 group that received testosterone and 10 mg/kg of corn silk extract, and a Testo+CSE100 group that received testosterone and 100 mg/kg of corn silk extract. Prostate weight and concentrations of dihydrotestosterone (DHT), $5{\alpha}$- reductase $2(5{\alpha}-R2)$, and prostate specific antigen (PSA) in serum or prostate tissue were determined. The mRNA expressions of $(5{\alpha}-R2)$ and proliferating cell nuclear antigen (PCNA) in prostate tissue were also measured. RESULTS: Compared to the Sham group, prostate weight was significantly higher in the Testo Only group and decreased significantly in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups (P < 0.05), results that were consistent with those for serum DHT concentrations. The concentrations of $(5{\alpha}-R2)$ in serum and prostate as well as the mRNA expression of $(5{\alpha}-R2)$ in prostate were significantly lower in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups than that in the Testo Only group (P < 0.05). Similarly, the concentrations of PSA in serum and prostate were significantly lower in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups (P < 0.05) than in the Testo Only group. The mRNA expression of PCNA in prostate dose-independently decreased in the Testo+CSE-treated groups (P < 0.05). CONCLUSIONS: BPH was induced through injection of testosterone, and corn silk extract treatment improved BPH symptoms by inhibiting the mRNA expression of $(5{\alpha}-R2)$ and decreasing the amount of $(5{\alpha}-R2)$, DHT, and PSA in serum and prostate tissue.

Effects of testosterone on the orchidectomized rats

  • The aim of this study was to determine the changes of body weight, organ weight, hematological values and biochemical parameters by testosterone (Testos) on the orchidectomized (Orch) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. Orch group received operation and no treatment. Orch+Testos received operation and testosterone. The body weights of each group increased, but that of Orch+Testos group was significantly lower in Orch+Testos group than in all the other groups. There were significant differences (P<0.05, P<0.001) of body weights between Orch+Testos group and all the other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart weights were significantly lower (P<0.001) in the Orch+Testos group than in all the other groups. The liver weights in the Orch+Testos group were significantly differences in comparison with those in the Sham (P<0.001) and Orch group (P<0.05). On the other hand, there were no significantly differences in the organ weights of spleen and kidney between the Orch+Testos group and the any other groups. The hematological values of white blood cell (WBC), red blood cell (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were no significant differences in any other groups. The concentrations of serum total protein and albumin increased significantly (P<0.05) in the Orch+Testos group as compared to that in the Orch group. However, there were no significant differences in Ca, IP and Mg in any other groups. We conclude that testosterone was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that testosterone may influence the process of lipid packaging and absorption in the orchidectomized rats.
