Dual-purpose Reagent for Separating Paper with Tape and Developing Fingerprints on the Paper Simultaneously (종이에 부착된 테이프를 박리하는 동시에 지문을 현출하는 이중목적시약(dual-purpose)에 관한 연구)

  • Tapes are usually used in violent crimes and may contain important evidence. Separating adhesive side should be preceded to collect these evidence. In this study, we researched a novel dual-purpose reagent that can separate the tape without damaging and develop the fingerprints on porous surface when the tape is attached to A4 paper. As a result, the reagent of 1:2 ratio of 1,2-IND stock solution and HFE-7100 can separate without damaging the adhesive surface and develop fingerprints on separated A4 paper.

The Effects of Jihwangyeumja and GamiJihwangyeumja water extract on The Cultured Primary Hippocampal Cell Damaged by XO/HX (지황음자와 가미지황음자 유출액이 XO/HX로 손상된 배양 해마신경세포에 미치는 효과)

  • The purpose of this study is to examine the toxic effects caused by xanthine oxidase/hypoxanthine(XO/HX) and the effects of herbal extracts such as JHYJ and GJHYJ on the treatment of the toxic effects. For this purpose, experiments with the cultured hippocampal cells from new born mice were done. The results of these experiments were as follows. 1. XO/HX, a oxygen radical-generating system, decreased the survival rates of the cultured cells on XTT assay and INT assay, the amount of DNA syntheses, and the amount of neurofilaments, and increased the lipid peroxidation. 2. JHYJ and GJHYJ have the efficacy of increasing the survival rates of the cultured cells. 3. JHYJ and GJHYJ have the efficacy of increasing the amount of neurofilaments and of decreasing the lipid peroxidation. 4. JHYJ and GJHYJ have the efficacy of increasing the amount of DNA syntheses. From the above results, it is suggested that Jihwangyeumja and Gamijihwangyeumja have marked efficacy as a treatment for the damages caused by the XO/HX-mediated oxidative stress. And Jihwangyeumja and Gamijihwangyeumja are thought to have certain pharmacological effects. Further dinical study of this pharmacological effects of Jihwangyeumja and Gamijihwangyeumja should be complemented.

In vitro Effects of Epigallocatechin Gallate on Sister Chromatid Exchange in the Lymphocytes Exposed to Glyphosate (글라이포세이트 노출로 인한 DNA손상에 대한 녹차의 예방적 효과)

  • Purpose: Green tea is known as a potent anti-oxidant, anti-carcinogen, and genetic protector. Glyphosate (N-phosphonomethyl glycine) is a widely used non-selective herbicide that causes DNA damage. The present study was conducted to investigate the protective effects of green tea in human blood lymphocytes exposed to glyphosate using the Sister Chromatid Exchange (SCE) frequency method. Methods: Peripheral blood was obtained from 10 volunteers and cultured through four different conditions. Four groups were divided into control, glyphosate only (300 ng/mL), glyphosate and low ($20{\mu}m$) concentrations of epigallocatechin gallate (EGCG) and glyphosate and high ($100{\mu}m$) concentrations of EGCG. Results: The glyphosate exposed groups had a higher mean SCE frequency ($10.33{\pm}2.50$) than the control group ($6.38{\pm}2.28$, p<0.001). The low concentrations of EGCG groups had a lower mean SCE frequency ($9.91{\pm}1.93$) than the glyphosate-only group, although this difference was not significant (p=0.219). However, the high concentration group ($9.49{\pm}1.85$) had a significantly lower SCE frequency than the glyphosate-only group (p=0.001). Conclusion: EGCG has a gene protective effect in human lymphocytes exposed to the genotoxicity of glyphosate in the case of high concentrations.

Antioxidant and Alpha-Glucosidase Inhibitory Activity of Strychnos nux-vomica Extracts (마전자 추출물의 항산화 및 Alpha-Glucosidase 저해 활성)

  • The physiological activities of Strychnos nux-vomica extracts were investigated through the total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), comet assay, and $\alpha$-glucosidase inhibitory activity. S. nux-vomica extracts were prepared with methanol, ethanol, and acetone. The methanol extract showed the highest phenolic content (71 mg/100 g gallic acid equivalents). Pretreatment with S. nux-vomica extracts resulted in significant reductions in oxidative DNA damage at all of the concentrations tested ($1{\sim}50\;{\mu}g/mL$). The $\alpha$-glucosidase inhibitory activity of a methanol extract was 12.8% at the concentration of 1 mg/mL. Therefore, these results indicate that S. nux-vomica might be a noble potential candidate exhibiting antioxidant and $\alpha$-glucosidase inhibitory activity.

Biological Hazard of Electromagnetic Field Exposure: A Review (전자기파의 생체 위해성에 관한 소고)

  • The safety of human exposure to an ever-increasing number and diversity of electromagnetic field (EMF) sources both at work and at home has clearly become a public health issue. To date many $in$ $vivo$ and $in$ $vitro$ studies revealed that EMF exposure can alter cellular metabolism, endocrine function, immune activity, reproductive function, and fetal development in animal system. The major parameters found to be altered in cells or individuals following EMF exposure include an increase of free radicals, DNA damage, cancer risk, developmental defect, and reproductive dysfunctions. Epidemiological studies reported EMF can increase life-threatening illnesses such as leukemia, brain cancer, amyotrophic lateral sclerosis, clinical depression, suicide, and Alzheimer's disease has been identified. These effects of EMF exposure differ according to duration of exposure, frequency of waves, and strength (energy) of EMF. In the present review, we briefly introduced the physical properties of EMF and summarized the effect of EMF on human and wildlife animals according to types of EMF, duration of exposure at cellular and organism levels.

Partial genomic sequence of baulovirus associated with white spot syndrome (WSBV) isolated from penaeid shrimp P. chinensis (대하새우로부터 분리한 WSBV의 게놈서열 분석)

  • Baculovirus associated with white spot syndrome (WSBV) is the causative agent of a disease with high mortalities and causes severe damage to shrimp cultures. In this study, we analyzed a recombinant clone (E3) obtained from a viral genomic library to characterize the causative agent in diseased shrimp Penaeus chinensis with white spot syndrome. According to the analysis of nucleotide sequence of E3, this clone did not showed considerable sequence homology with those of other known viruses, including baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV), indicating that WSBV is a novel virus causing a serious disease in P. chinensis. Based on the sequence of E3 clone, a pair of PCR primers was designed. After 30 cycles of amplification, a specific product of the expected size was detected only if the total nucleic acids extracted from the diseased shrimp was used as a template DNA, suggesting that this method can be used to diagnose the virus infection in diseased shrimp.

Detection of a mollicute-like organism in kuruma shrimp, Penaeus japonicus (보리새우(Penaeus japonicus)에 감염된 Mollicute-like Organism)

  • A filamentous form of mollicute-like bacterium was detected during a routine health survey of wild adult kuruma shrimp Penaeus japonicus. The kuruma shrimp were native to Japan and were imported to Korea. The histology showed no pathological changes. Transmission electron microscopy revealed extensive infections of hepatopancreatic epithelial cell by a pleomorphic, filamentous intracellular bacterium. The filamentous bacterium was of about 60 nm in diameter and 300 nm to more than $1{\mu}m$ in length. Teh morphology of bacteria were filamentous and branched with terminal blebs, or knobs, on the branches. They lacked the cell wall, and were bounded by the plasma membrane. They contained typical prokaryotic ribosomes and fibrillar DNA-like strands. No additional internal structure has been observed. They are considered to be mollicutes, based upon the morphological appearance and upon the cellular location.

Lack of p53 Gene Nucleotide Change in Mutation Hot Spots During HeLa Cell Apoptosis by Adriamycin (아드리아마이신에 의한 HeLa 세포의 자살 과정 중 p53 유전자의 돌연변이 빈발 부위에서의 핵산 변화의 부재)

  • Apoptosis is an important event in the anticancer drug therapy. p53 was demonstrated to serve a key component to lead tumor cell death by inducing apoptosis. However, recent study showed the presence of p53 independent apoptotic pathway (Gaftenhaus et al., 1996). We were curious to know it apoptosis induced by adriamycin, a genotoxic anticancer agent, involved p53 gene mutation. Thus this study investigated the p53 gene mutation status among HeLa cell population during apoptosis induced by adriamycin. Under our experimental condition, 12 hour treatment of 1 ${\mu}m$ adriamycin caused apoptosis which was monitored by DNA fragmentation assay. In order to see the p53 gene mutation status, exons of 5, 7 and 8 of p53 gene, where previously reported p53 mutation hot spots reside, were amplified by PCR and nucleotide sequence change was scanned. However, no nucleotide change was observed among apoptotic HeLa cell population. Therefore this study demonstrated that adriamycin induced apoptosis without causing p53 gene damage.

Isolation and Characterization of White Spot Syndrome Baculovirus in Cultured Penaeid Shrimp (Penaeus chinensis) (양식새우(Penaeus chinensis)에서의 White Spot Baculovirus의 분리 및 특성)

  • Beginning in the summer of 1993, a serious mortality among cultured penaeid shrimp occurred in the western sea of Korea. The typical sign of this disease was white spots inside the surface of the carapace. Cytopathic effect (CPE) were not observed by virus in CHSE-214, RTG-2, but not by pH 11. A nonoccluded rod-shaped form virus was observed by electron microscopy in the lymphoid organ. The virion was bacilliform virus and sourrounded by a virion envelope. Its virion protein was found to be similar to hypodermal and hematopoietic necrosis virus (HHNBV) by analysis of virion proteins in SDS-PAGE. The genome of virus is double stranded DNA molecule whose full length was about 114kb. It was similar to penaeus acute viremia (PAV) of Japan.

  • Present study investigated the effect of Nuva-Lite irradiation on the labial oral mucosa of the albino rats. The specimens were embedded in paraffin wax and stained with histological and histochemical procedures such as hematoxylin-eosin stain, Feulgen reaction, methyl green-pyronin stain, alloxan Schiff reaction, -SH group reaction, PAS reaction, colloidal iron reaction and toluidine blue stain. The specimens 24 hours to 3 days after ultraviolet irradiation exhibited mild intracellular edema in the prickle cell layer. On 6th and 10th day after irradiation rete peg hyperplasia was prominent. Ultraviolet irradiation caused diminution of Feulgen reacted DNA. However, sulfhydril and ${\alpha}$-amino acid radicals were increased at the upper layer of stratified squamous epithelium after irradiation.
