A Study on the Development of Functional Cosmetics Using Silk-gland Powder of Silkworm (누에생실샘 미세분말을 이용한 기능성 화장품 소재 개발에 대한 연구)

  • In this study, silk-gland powder of silkworm was investigated for the usefulness as a potential agent for functional cosmetics, total content of amino acids and DPPH free radical scavenging assay, and clinical trial were done. The analysis of the amino acids of silk-gland powder showed that serine (26.77 %) content was the highest and aspartic acid, glycine, and glutamic acid followed that. DPPH free radical scavenging activity of silk-gland powder was lower than vitamin C (82.3 % v.s. 97 %), The moisture and elasticity were increased in silk-gland powder compared to control cosmetics by 257.2 % and 181.15 %, relatively. These results suggest that silk-gland powder of silkworm may have beneficial properties as a material for cosmeceuticals.

Antimicrobial Activities of Sophorolipids and Its Application for Cosmetics (Sophorolipid의 항균효과와 화장품에의 응용)

  • Some surfactants known as biosurfactants, are produced biologically by yeast and bacteria from various substances. They are more effective and environmentally friendly than many other synthetic surfactants. Antimicrobial activities of sophorolipids produced by Candida bombiocola were investigated against various microorganisms. Minimal Inhibitory concentration (MIC) of sophorolipid against S. mutans, C. xerosis, and P. acnes were 0.005, 0.05, and 0.005%, respectively. The antimicrobiological activities were more effective than those of SLS and APG. Skin exfoliating and moisturizing effects of vehicles with the sophorolipids were tested. The skin turnover time of aqueous solution with 0.25% sophorolipids was similar to that of aqueous solution with 5.0% lactic acid. Higher moisturizing effects on skin showed as the concentration of sophorolipids increased. We suggest that the sophorolipids can be used for cosmetics as an antimicrobiological agent and an active materials of skin moisturizing and exfoliating.

Development of Gel Formulation Containing Alkaloid of Cacao Extract (카카오 추출물의 알칼로이드 성분이 함유된 겔제형의 개발)

  • In this study, methanol (CAMe) and ethanol (CAEt) extract of Theobroma cacao L. were tested to see possibility as natural functional cosmetic agent. Cacao extract and cacao extract loaded gel were evaluated for various properties such as antioxidant, antimicrobial, cell viability, anti-inflammatory. Inhibition of nitric oxide production in lipopolysaccaride (LPS) stimulated raw 264.7cells, at concentration of $0.125{\sim}1{\mu}g/ml$ a dose-dependent manner. Gel formulation (3% extract) were stable for 60day in both extract. In conclusion, gel containing extract of Theobroma cacao L. indicated strong possibility for functional cosmetic ingredient.

  • To minimize the use of animals in toxicity testing, and to reduce the cost in vivo test, more rational test method was described which determines, in the same animal, photoxic and photoallergic potential of a substance, and is daptable to routine testing. The other purpose of this study was to investigate the usefulness of in vivo alternatives ; photostability and spectrophotometric carbonyl assay. In this modified photosensitization model, animal numbers and resting periods, the number and method of topical application were simplified. Two positive photoreactive agents, Benzocaine and 6-methyl coumarine, showed a similar photoallergic potential to that of Ichikawa's method. Two sunscreens, Octyl methoxy cinnamate, Butyl methoxyl dibenzoyl methane, hardly showed photoallergic potentials. The photostability test could be used in the step of prescreening of photosensitization potential because most of the photoreactive agents represented the reduction of more than 20% in the absorbance. And photoreactive agents have a high potential of photosensitization in the sddessment of spectrophotometric carbonyl level although two sunscreens have a low possibility of photosensitization. Therefore this method was assumed as a valuable in vivo alternatives in the respect even in the very low concentrations which phototoxicity test using almonella showed no phototoxic potential.

Anti-inflammatory Effects of Prescription Extracts Containing Forsythia viridissima L. (연교를 함유한 처방단 추출물들의 항염증 효과)

  • Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antiviral, antitumor, antibacterial, antipyretic and anti-aging activities. This work was carried out to investigate the anti-inflammatory effects of the Korean traditional medicinal prescriptions containing Forsythia viridissima extract. The prescriptions containing Forsythia fructus were evaluated for antioxidative effects, inhibitory effects on lipoxygenase activity and on LPS-induced NO and $PGE_2$ production. In human irritation test, they did not show any adverse effect. Based on these results, we suggest that the se prescriptions hold great promise for application as an anti-inflammatory agent for trouble skins such as atopic dermatitis and acne.

The Stimulatory Effects of Epimedium koreanum Nakai Extract on Melanogenesis (음양곽 추출물의 멜라닌 생성 촉진 효과)

  • To develop a natural stimulating agent for hair melanogenesis, we investigated the stimulatory effects of Epimedium koreanum Nakai extract (EKNE) on melanogenesis. EKNE both increased melanin content in B16 melanoma cells up to about 104 % and intracellular tyrosinase activity up to about 95 % at a concentration of $50\;{\mu}g/mL$ without cell cytotoxicity (below $100\;{\mu}g/mL$). From the result of western blot, we could find that EKNE dose-dependently induce the expression of TRP-2. Also, EKNE increased melanogenesis up to about 25 % at a concentration of 5 % (w/v) in back hair of C3H/Hej mouse. These results suggest that EKNE has an effect to stimulate melanin formation through the induction of tyrosinase activity and TRP-2 expression which are involved in melanin synthesis in melanocyte. Therefore, we suggest that EKNE could be used as a useful melanogenesis stimulator for development of natural hair cosmetics.

  • We have studied the effects of red ginseng root extract on the derease of MMS-induced gemotoxicity in cultured NIH3T3 cells. The increase in survival and the recovery from DNA synthesis inhibition in MMS-treated cells as a function of normal medium incubation time was potentiated, at a rate higher than those in UV-irradiated cells, by the presence of the ginseng extract. The extract also increased the MMS-induced excision repair as determined by unscheduled DNA synthesis. The amount of MMS-induced DNA single strand breads that are accumulated by polymerase inhibitors was increased, but as a rate lower rate than in UV-induced strand break, by the presence of the extract. These results suggest that the red ginseng extract increase MMS-induced repair and could be used as a reagent for protectiong alkylating agent-induced genotoxicity and cytotoxicity.

Effect of the Processed Selaginella tamariscina on Antioxidation and Inhibition of Matrix Metalloproteinase (수치에 의한 권백의 항산화 효과와 MMP 발현 저해 효과)

  • Selaginella tamariscina with the popular Korean name Keoun Back, is a traditional medicinal plant for therapy of advanced cancer patients in the Orient. In this study, we evaluated anti-aging activity of S. tamariscina using processed technology and investigated diverse biological activities of processed S. tamariscina (PST) as an anti-aging ingredient of cosmetics. PST, heated with sand, used to different purpose compared with origin in medicine. PST raises total phenol concentration and enhances the DPPH radical scavenging activity. For testing intracellular ROS scavenging activity, the cultured human dermal fibroblasts were analyzed by increase in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20 mJ/cm^2$ after treatment of PST. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced in a dose-dependent manner by PST. Taken together, 4hese results suggest that PST may act as an anti-aging agent by preventing the skin cell from damage induced by UV irradiation, and imply that PST may be useful as a new ingredient for anti-aging cosmetics.

  • To develop a new anti-inflammation agent for cosmetics, we investigated the anti-inflammatory activity of Fallopia sachalinensis RonseDecr. fruit extracts (FSR). FSR inhibited the pre-inflammatory cytokines ($1L-1{\beta}$, IL-6, TNF-${\alpha}$) and reduced NO production of RAW 264.7 macrophage cells in a dose-dependent manner and decreased the activity to about 51% at a concentration of $100{\mu}g/mL$. FSR suppressed not only the mRNA expression of iNOS and COX-2, but also the protein expression of iNOS and COX-2 in macrophage. The results suggested that FSR has considerable potential as a cosmetics ingredient with an anti-inflammatory effect.

Anti-Inflammatory Effect of Germinated Mung Bean and Hairdye Applications (발아 녹두 추출물의 항염증 효능 및 염모제 응용)

  • In this study, a research of anti-inflammatory effect and application for cosmetics after extracting of germinated mung bean. Germinated mung bean extract was showed high NO constraining effect compared with $250{\mu}g/m{\ell}$ concentrated mung bean extract and germinated mung bean extract. Also shown is the highest nitrogen oxide inhibiting effect that using 1,3-butylene glycol when comparing the results of extraction conditions of a water-soluble, and extracted mung bean germinated for 5 days. The result of applying the germinated mung bean extract hairdye inflammatory reaction happens many showed nitrogen oxides higher inhibitory effect than that hair dye containing germinated mung bean extract is not included. Therefore, germinated mung bean extract is expected to the abirritant and anti-inflammatory agent as the material for cosmetic.
