AILU type preconditioners for a two-dimensional combined P2P1 finite element formulation of the interaction of rigid cylinder with incompressible fluid flow have been devised and tested by solving fluid-structure interaction (FSI) problems. The FSI code simulating the interaction of a rigid cylinder with an unsteady flow is based on P2P1 mixed finite element formulation coupled with combined formulation. Four different preconditioners were devised for the two-dimensional combined P2P1 finite element formulation extending the idea of Nam et al., which was proposed for the preconditioning of a P2P1 mixed finite element formulation of the incompressible Navier-Stokes equations. It was found that PC-III or PC-IV among them perform well with respect to computational memory and convergence rate for some bench-mark problems.
Preconditioners for a two-dimensional combined finite element formulation have been devised and tested for fluid-structure interaction (FSI) problems. The FSI code simulating the interaction of a elastic body with an unsteady flow is based on P2P1 finite element combined formulation. It has been shown that two preconditioners among them perform well with respect to computational memory and convergence for a bench-mark problem. Based on the verification of the preconditioners for the two-dimensional combined formulation, four preconditioners are proposed for the problem of an elastic body interacting with a flow.
In order to design a 24 hr sustained release preparation of sulindac for oral administration, fast release pellet (FR), slow release pellet (SR) and two combined formulation (1 : 1 and 1 : 2) were prepared. The pharmacokinetic effect of such preparations has been evaluated using rabbits as a suitable in vivo model, and tested in man. Dose determination was carried out using curve fitting according to RSTPJP II program. In bioavailability test using rabbit, AUCs of sulindac in a few designed formulations were similar to each other. $C_{max}$- of RF and SR were 1.8 times and 1.2 times higher, respectively, compared to that of combined formulation (FR:SR=1:1). While plasma concentration of FR and SR decreased rapidly, that of combined formulation (FR:SR 1:1) lasted at the level close to $C_{max}$ for 24 hrs. Plasma concentration of sulfide form from the combined pellet(FR:SR=1:1) lasted for 24 hrs, and its AUC value was 1.4-fold, 2.7-fold. and 1.2-fold greater than FR pellet, SR pellet and combined pellet (FR:SR 1 : 2). Thus, the combined pellet of 1:1 ratio was found to be the most effective for oral sustained release formulation. Bioavailability test in human showed that AUC of sulfide from TSRP (1 : 1) was approximately 1.5 times greater than total AUC of Immbaron$^{\circledR}$ administered twice in a day. While $T_{max}$ of sulfide from lmmbaron$^{\circledR}$ was 4.33 +/- 1.37 hr (lst administration) and 3.33 ${pm}$ 0.82 hr (2nd administration), respectively, that of sulfide from TSRP increased to 7.17 ${pm}$ 2.86 hr. Plasma concentration of sulfide from TSRP was sustained at more, than 1.0 ${\mu}g{\cdot}$hr/ml until 24 hrs after one dose administration. In addition, TSRP may decrease local adverse reaction in the stomach, since plasma concentration of sulfide from the combined pellet was low within 2hrs in the stomach. In conclusion, it is suggested that TSRP formulation may be effective for oral 24 hr sustained release formulation of sulindac dosing 300 ~ 350mg once a day.
Combined formulation developed for the fluid-particle mixture is introduced to simulate the biocube-fluid mixture flow, which is utilized for sewage disposal. Some tricky boundary conditions are introduced in order to simulate the effect of screen wall and air bubble, which is injected from the bottom of sewage reservoir. It has been shown that a circulated flow pattern, which was observed in experiment, is reproduced from the present numerical simulation. Furthermore, the effect of biocube density on the distribution pattern of biocube is also studied. It has been shown that a biocube whose density is slightly smaller than that of surrounding fluid or neutrally buoyant one are optimal for the uniform distribution of biocube.
A splitting method for the direct numerical simulation of solid-liquid mixtures is presented, where a symmetric pressure equation is newly proposed. Through numerical experiment, it is found that the newly proposed splitting method works well with a matrix-free formulation fer some bench mark problems avoiding an erroneous pressure field which appears when using the conventional pressure equation of a splitting method. When deriving a typical pressure equation of a splitting method, the motion of a solid particle has to be approximated by the 'intermediate velocity' instead of treating it as unknowns since it is necessary as a boundary condition. Therefore, the motion of a solid particle is treated in such an explicit way that a particle moves by the known form drag (pressure drag) that is calculated from the pressure equation in the previous step. From the numerical experiment, it was shown that this method gives an erroneous pressure field even for the very small time step size as a particle velocity increases. In this paper, coupling the unknowns of particle velocities in the pressure equation is proposed, where the resulting matrix is reduced to the symmetric one by applying the projector of the combined formulation. It has been tested over some bench mark problems and gives reasonable pressure fields.
A combined computational fluid dynamics(CFD)-Kirchhoff method is presented for predicting high-speed impulsive noise generated by a hovering blade. Two types of Kirchhoff integral formula are used; one for the classical linear Kirchhoff formulation and the other for the nonlinear Kirchhoff formulation. An Euler finite difference solver is solved first to obtain the flow field close to the blade, and then this flow field is used as an input to a Kirchhoff formulation to predict the acoustic far-field. These formulas are used at Mach numbers of 0.90 and 0.95 to investigate the effectiveness of the linear and nonlinear Kirchhoff formulas for delocalized flow. During these calculiations, the retarded time equation is also carefully examined, in particular, for the cases of the control surface located outside of the sonic cylinder, where multiple roots are obtained. Predicted results of acoustic far-field pressure with the linear Kirchhoff formulation agree well with experimental data when the control surface is at the certain location(R=1.46), but the correlation is getting worse before or after this specific location of the control surface due to the delocalized nonlinear aerodynamic flow field. Calculations based on the nonlinear Kirchhoff equation using a linear sonic cylinder as a control surface show a reasonable agreement with experimental data in negative amplitudes for both tip Mach numbers of 0.90 and 0.95, except some computational integration problems over a shock. This concliudes that a nonlinear formulation is necessary if the control surface is close to the blade and the flow is delocalized.
The objective of this study was to develop effective oral formulations of rhEGF for gastric ulcer healing using polycarbophil. hydroxypropylcellulose(HPC) and sucralfate as its bioadhesive bases. Cytoprotective effects of rhEGF, cell proliferation and differentiation. on the ulcers induced by ethanol or acetic acid in rats were studied. rhEGF release from HPC formulation was much faster than that from polycarbophil formulation. HPC formulation combined with small amount of sucralfate showed much slower release of rhEGF than only HPC base only. rhEGF preparations with bioadhesive polymers showed better effects on the healing of gastric ulcers than EGF solution when administered orally. When rhEGF preparations were administered at once and the animals were under starvation, polycarbophil formulation showed better effect on gastric ulcers than HPC formulation. Otherwise, when rhEGF preparations were given more than three times and the rats were fed normally, HPC formulation showed good healing efficacy of ulcers compared to polycarbophil formulation. rhEGF showed dose-dependent effect on the healing of both chronic and acute ulcers.
The conventional segregated finite element formulation produces a small and simple matrix at each step than in an integrated formulation. And the memory and cost requirements of computations are significantly reduced because the pressure equation for the mass conservation of the Navier-Stokes equations is constructed only once if the mesh is fixed. However, segregated finite element formulation solves Poisson equation of elliptic type so that it always needs a pressure boundary condition along a boundary even when physical information on pressure is not provided. On the other hand, the conventional integrated finite element formulation in which the governing equations are simultaneously treated has an advantage over a segregated formulation in the sense that it can give a more robust convergence behavior because all variables are implicitly combined. Further it needs a very small number of iterations to achieve convergence. However, the saddle-paint-type matrix (SPTM) in the integrated formulation is assembled and preconditioned every time step, so that it needs a large memory and computing time. Therefore, we newly proposed the P2PI semi-segregation formulation. In order to utilize the fact that the pressure equation is assembled and preconditioned only once in the segregated finite element formulation, a fixed symmetric SPTM has been obtained for the continuity constraint of the present semi-segregation finite element formulation. The momentum equation in the semi-segregation finite element formulation will be separated from the continuity equation so that the saddle-point-type matrix is assembled and preconditioned only once during the whole computation as long as the mesh does not change. For a comparison of the CPU time, accuracy and condition number between the two methods, they have been applied to the well-known benchmark problem. It is shown that the newly proposed semi-segregation finite element formulation performs better than the conventional integrated finite element formulation in terms of the computation time.
In this paper, we present a time domain combined field integral equation formulation (TD-CFIE) to analyze the transient electromagnetic response from dielectric objects. The solution method is based on the method of moments which involves separate spatial and temporal testing procedures. A set of the RWG functions is used for spatial expansion of the equivalent electric and magnetic current densities, and a combination of RWG and its orthogonal component is used for spatial testing. The time domain unknowns are approximated by a set of orthonormal basis functions derived from the Laguerre polynomials. These basis functions are also used for temporal testing. Use of this temporal expansion function characterizing the time variable makes it possible to handle the time derivative terms in the integral equation and decouples the space-time continuum in an analytic fashion. Numerical results computed by the proposed formulation are compared with the solutions of the frequency domain combined field integral equation.
For a large deformation nonlinear dynamic analysis of two-dimensional frictional contact, the linear complementarity formulation combined with a linearization is used. The solution procedure is based on the total Lagrangian formulation with a predictor and corrector scheme. For contact searching, a hierarchical scheme with a circular territory is used. A second-order approximation of displacements is used to detect impact time and position. The formulation is illustrated by means of numerical examples.
