Parameters on Physiological Responses of Soybean (Glycine max Merr.) to Salinity (염분에 대한 콩의 생리학적 반응지표 연구)

  • The mechanism imparting salt tolerance to crop plants remains still unsolved, although soybean has been classified as a susceptible plant to NaCl. To determine optimum parameters on physiological responses for improving sensitivity of salinity in breeding program, soybean (Glycine max Merr., cv. "Gwan-gan") plants were grown in a greenhouse, treated 20 days after emergence for 7 days with NaCl at 0, 30, 60, and 90mM, corresponding to electric conductivity of 1.2, 4.4, 7.3, and 10.4 dS/m, respectively, and assessed 30 days after treatment. Chlorophyll contents were significantly decreased by NaCl ($0.4{\sim}1.0\;mg/g$) compared to control (1.2 mg/g). Photosynthesis rate by NaCl treatment at $0{\sim}90\;mM$ at flowering stage was ranged from 5.0 (control) to $9.6\;{\mu}mol/m^2/s$. Oxygen for respiration was consumed from 5.4 to $9.7\;{\mu}mol/m^2/s$ so that the ratio of $O_2$ (evolution:consumption) was increased with the increase of NaCl, indicating that $O_2$ consumption seems to go beyond $O_2$ evolution. Water potential of leaf at vegetative stage II was ranged from -0.6 to -1.8 MPa and the highest level was observed at mid-day. Water potential by salt stress was decreased with range of $-2.1{\sim}-2.7MPa$ compared to control. Transpiration was decreased from 17% to 20% by NaCl stress. Water vapor diffusing resistance of intercellular air space was affected significantly, increasing up to $16{\sim}24%$ compared to control by NaCl treatment. Salt-treated soybean tended to accumulate $Na^+$, specially in root, with reduced absorption of N, P, $K^+$, $Ca^{2+}$, and $Mg^{2+}$ contents. Free proline content of soybean leaf as affected by different NaCl concentrations was increased 4.2 times ($184{\sim}434\;{\mu}g/g$) more than control. NaCl also increased activities of nitrate reductase and peroxidase by $28{\sim}161%$ and $3{\sim}22%$, respectively. The results show that physiological characteristics of soybean plants during assay were useful as the best parameters of salt stress or salt tolerance test to improve sensitivity in screening and breeding program among cultivars or germplasms.

The Limnological Survey of Major Lakes in Korea (4): Lake Juam (국내 주요 호수의 육수학적 조사(4) : 주암호)

  • In this study limnological characteristics of Lake Juam was surveyed from June 1993 to May 1994 in order to provides important information regarding water resources. Secchi disc transparency, epilimnetic chlorophyll a (chi-a), total nitrogen (TN), total phosphorus (TP) concentration and primary productivity were in the range of $2.0{\sim}4.5\;m$, $0.9{\sim}13.6\;mgChl/m^3$, 0.78$\{sim}$2.32 mgN/l, $11{\sim}56\;mgP/m^3$, $270{\sim}2.160\;mgCm^{-2}\;day^{-1}$, respectively. On the basis of TP, Chl-a and Secchi disc depth, the trophic state of Lake Juam can be classied as mesotrophic lake. The phosphorus inputs from non-point sources are concentrated in heavy rain episodes during the monsoon season. As a result, phosphorus concentration are higher in summer than in winter. TP loading from the watershed were estimated to be $0.9\;gPm^{-2}yr^{-1}$, which correspond to a boundary of the critical loading ($1.0\;gPm^{-2}yr^{-1}$) for eutrophication. From the results of the algal assay, both phosphous and nitrogen act as limiting nutrients in algal growth. The seasonal succession of phytoplankton community structure in Lake Juam was similar to that observed in other temperate lakes. Diatoms (Asterionella formosa and Aulacoseira granulate var. angustissima)fujacofeira BraHuJafa uar. aHgusHrsiaia) weredominant in spring and winter, cyanobacteria) were dominant in warm season. The organic carbon, nitrogen and phosphorus content of lake sediment were $9.5{\sim}14.0\;mgC/g$, $1.01{\sim}1.82\;mgN/g$ and $0.51{\sim}0.65\;mgP/g$, respectively. The allochthonous organic carbon loading from the watershed and autochthonous organic carbon loading by primary production of phytoplankton were determined to be 1,122 tC/yr and 6,718 tC/yr, respectively. To prevent eutrophication of Lake Juam, nutrient management of watershed should be focus on reduction of fertilizer application, proper treatment of manure, and conservation of topsoil as well as point source.

Seedling Quality and Early Yield after Transplanting of Paprika Nursed under Light-emitting Diodes, Fluorescent Lamps and Natural Light (발광다이오드, 형광등 및 자연광 하에서 육묘된 파프리카의 묘소질 및 정식 후 초기 수량)

  • This study was conducted to analyze the seeding quality of paprika and the growth and early yield after transplanting of paprika nursed under artificial light and natural light. In this study, blue LED, red LED, and white fluorescent lamps (FL) were used as artificial lighting sources. Photoperiod, average photosynthetic photon flux, air temperature, and relative humidity in a closed transplants production system (CTPS) were maintained at 16/8 h, $204{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 26/$20^{\circ}C$, and 70%, respectively. Leaf length, leaf width, leaf area, top fresh weight and dry weight of paprika seedlings, and chlorophyll content in paprika leaves nursed under LED and fluorescent lamps for 21 days after experiment were significantly affected by light treatments. As compared with the control (white FL), leaf area of paprika grown under blue LED, red LED, and natural light was decreased by 63%, 63%, and 28%, respectively. Top dry weight of paprika grown under blue LED, red LED, and natural light was 64%, 50%, and 22%, respectively, compared with the control. Number of leaves on 18 days after transplanting showed with red LED, blue LED, and natural light by 86%, 84%, and 48%, respectively, compared with the control. On 114 days after transplanting, paprika nursed under blue LED and red LED had relatively short plant height. This result might be caused that the elongation of its internodes was suppressed by the illumination of sole blue or red light. Average number of fruits per plant harvested during 4 weeks after first harvest was 3.5 with red LED, 3.3 with blue LED, 1.0 with natural light, and 2.2 with control, respectively. Early yield of paprika nursed under red LED, blue LED, natural light, and control were 453 g/plant, 403 g/plant, 101 g/plant, and 273 g/plant, respectively. Larger fruit of 136 g was harvested with red LED treatment. Even though the early yield of paprika was greatly increased with artificial lighting, but total yield was almost similar as the harvest period after transplanting in greenhouses was lengthened. From the above results, we could understand that paprika nursed under white FL, blue LED, and red LED showed good growth after transplanting and was early harvested by a week as compared to the natural light. Therefore, the white FL, blue LED, and red LED as the artificial lighting sources in CTPS could be strategically used to enhance the seedling quality, to shorten the harvest time, and to increase the yield of paprika.

Effect of Timing of Nutrient Starvation during Transplant Production on the Growth of Runner Plants and Yield of Strawberry 'Seolhyang' (딸기 '설향' 육묘기 양분 공급 중단 시기가 자묘 생육 및 수량에 미치는 영향)

  • This study was conducted to investigate the effects of timing of nutrient starvation during transplant production on growth of runner plants and yield of strawberry 'Seolhyang' (Fragaria ${\times}$ ananassa). Nutrient solution supply at the level of EC (electrical conductivity) 0.8 $dS{\cdot}m^{-1}$ was terminated at interval about 10 days between July 25 and September 5. As a result, the growth of above-ground part was inhibited while root growth increased when the nutrient starvation treatment had been brought forward to July 25. It also reduced the T/R ratio significantly and chlorophyll content was tended to be lower than the other treatment. In addition, it significantly promoted the budding, flowering and harvest of first flower cluster. On the other hand, the period of harvest was delayed more than two weeks when the nutrients were continuously supplied after the middle of August. An accumulated marketable fruit yield per plant until the end of January and February was 169 and 266g, respectively in the treatment of nutrient starvation on July 25, which was 71 and 12% increase, respectively, as compared with those in the treatment of September 5. Therefore, the appropriate nutrient starvation in the late season of strawberry nursery period could be expected the increase in yield and income during the winter season by promoting the flower bud differentiation as reducing the endogenous nitrate level of the plantlet.

Thermal Effluent Effects of Domestic Sewage and Industrial Wastewater on the Water Quality of Three Small Streams (Eung, Chiljang and Buso) during the Winter Season, Korea (동계 저온기의 소하천 수질에 미치는 하·폐수의 온배수 영향)

  • The sewage and wastewater (SAW) are a well-known major source of eutrophication and greentide in freshwaters and also a potential source of thermal pollution; however, there were few approaches to thermal effluent of SAW in Korea. This study was performed to understand the behavioral dynamics of the thermal effluents and their effects on the water quality of the connected streams during winter season, considering domestic sewage, industrial wastewater and hot spring wastewater from December 2015 to February 2016. Sampling stations were selected the upstream, the outlet of SAW, and the downstream in each connected stream, and the water temperature change was monitored toward the downstream from the discharging point of SAW. The temperature effect and its range of SAW on the stream were dependent not only on the effluent temperature and quantity but also on the local air temperature, water temperature and stream discharge. The SAW effects on the stream water temperature were observed with temperature increase by $2.1{\sim}5.8^{\circ}C$ in the range of 1.0 to 5.5 km downstream. Temperature effect was the greatest in the hot spring wastewater despite of small amount of effluent. The SAW was not only related to temperature but also to the increase of organic matter and nutrients in the connected stream. The industrial wastewater effluent was discharged with high concentration of nitrogen, while the hot spring wastewater was high in both phosphorus and nitrogen. The difference between these cases was due to with and without chemical T-P treatment in the industrial and the hot spring wastewater, respectively. The chlorophyll-a content of the attached algae was high at the outlet of SAW and the downstream reach, mostly in eutrophic level. These ecological results were presumably due to the high water temperature and phosphorus concentration in the stream brought by the thermal effluents of SAW. These results suggest that high temperature of the SAW needs to be emphasized when evaluating its effects on the stream water quality (water temperature, fertility) through a systematized spatial and temporal investigation.

Effects of OTR Film Type on the Quality of Lepidium sativum L. Baby Leaf Vegetable during MA Storage (큰다닥냉이 어린잎채소의 MA저장 중 OTR 필름 종류가 품질에 미치는 영향)

  • This study was conducted to examine the effect OTR film type on the quality of garden cress (Lepidium sativum L.) baby leaf during MA storage. Garden cress harvested at baby leaf size of 10cm plant height packed with 1,300 cc, 10,000 cc, 20,000 cc, 40,000 cc, and $80,000cc{\cdot}m^{-2}{\cdot}day^{-1}{\cdot}atm^{-1}$ OTR (oxygen transmission rate) films and MP (micro-perforated) film, and then stored at $8^{\circ}C$ for 10 days. All of the OTR film treatments showed a decrease of 0.5% fresh weight until the storage end date and a 1.3% decrease in the MP film treatment. The oxygen concentration in the packaging during storage was maintained at 18% or more in 20,000 cc, 40,000 cc, and 80,000 cc OTR film treatments, while the 1,300 cc OTR film treatment decreased to 11% at the storage end date. And the concentration of carbon dioxide was steadily increased in the 1,300 cc and 10,000 cc OTR film treatments to show the levels of 4.5% and 3.4%, respectively, and the other OTR film treatments showed a concentration of less than 1%. Ethylene concentration in the package was maintained at the highest level of $3-5{\mu}L{\cdot}L^{-1}$ in the 1,300 cc treatment during the storage period. The lowest odor and the highest quality of appearance were observed in the 1,300 cc treatment, but the MP film treatment and the other OTR treatments lost marketable quality due to yellowing. The color of garden cress baby leaf was changed the lowest in 1,300 cc treatment that showed the highest chlorophyll content and Hue angle value, lowest $b^*$ value, present of yellowing at end of storage date. Therefore, 1,300 cc treatment which was most effective for yellowing and odor suppression during storage is considered to be suitable for packaging of garden cress baby leaf.

Difference of Absorption and Anatomical Responses to Protoporphyrinogen Oxidase-Inhibiting Herbicides in Wheat and Barley (Protoporphyrinogen Oxidase 저해형 제초제에 대한 밀과 보리의 흡수 및 해부하적 차이)

  • Laboratory and greenhouse studies were conducted to determine differential sensitivities on absorption of $^{14}$ C-oxyfluorfen and the anatomical responses in wheat and barley to protoporphyrinogen oxidase-inhibiting herbicides [oxyfluorfen (2-chloro-1- (3-ethoxy -nitrophen-oxy)-4-(trifluoromethyl) benzene, acifluorfen(5-[2-chloro-4-(trifluoromethyl) phenoxy]-2-nitro-benzoic acid), bifenox(methyl-5-(2, 4-dichlorophenoxy)-2-nitrobenzoate) and oxadiazon(5-tert-butyl-3-(2, 4-dichloro-5-isopropoxyphenyl)-1, 3, 4-oxadiazol-2-one)]. I$_{50}$ value of the tolerant wheat cultivars to oxyfluorfen was about 10$^{-4}$ , whereas that of the susceptible barley cultivars was about 10$^{-6}$ M, showing significant difference between the two groups. When foliage were applied with acifluorfen, bifenox or oxadiazon, the oxyfluorfen-tolerant wheat showed less decreased in shoot fresh weight and chlorophyll content than the susceptible barley. Also, when soil-applied with these herbicides test plants showed similar tendency in foliar application. Electrolyte leakage from the tissue treated with these compounds was the more influenced in the barley than the wheat. Malondialdehyde(MDA) production as index of lipid peroxidation was greater in the barley than the wheat by treatment of these compounds. Therefore, the differential sensitivities of wheat and barley to protoporphyrinogen oxidaseinhibiting herbicides was showed by our greenhouse and in vitro experiment. The absorption rates of $^{14}$ C-oxyfluorfen were higher in the barley than the wheat. And this tendency was showed appararitly difference by increase of treatment durations. After the oxfluorfen and oxadiazon treatment, the tolerant wheat did not show the structural damage in leaf surface, but the susceptible barley was damaged in the leaf waxy layer. However, the acifluorfen and bifenox treatment showed no difference between wheat and barley. The anatomical changes by these compounds treatment were not observed in the tolerant wheat but epidermal cell and mesophyll cell were highly broken in the susceptible barley.

Effect of different light sources and ventilation on in vitro shoot growth and rooting of a rare and endangered species, Tsuru-rindo(Tripterospermum japonicum) (희귀 및 멸종위기 식물 덩굴용담의 기내생장에 미치는 광질 및 환기효과)

  • Effects of light generated by LEDs on shoot growth and rooting of Tsuru-rindo(Tripterospermum japonicum) were evaluated. Apical shoots(one or two node with 3-4 leaves) were cultured on MS basal medium with 3% sucrose and maintained for four weeks under five different light qualities: fluorescent lamp(F), 100% red LED(R), 70% red LED+30% blue LED(R7B3), 50% red LED+50% blue(R5B5), or 100% blue LED(B). Rooting was promoted by both red light and fluorescent lamp, and the effect was further promoted under the ventilation. Red light enhanced shoot node elongation, whereas blue light appeared to suppress it. Growth of shoots and leaves were enhanced under the ventilation irrespective of the different light qualities. Under the ventilated condition, total fresh weight of plants was highest in R7B3 LED as 257.7 mg per plant. Dry matters, which are used for index of plant growth, were lowest under red light, whereas it was highest under blue light. The dry matter was inclined to getting higher by ascending the ratio of blue light and red light. Total chlorophyll content was highest in both R7B3 LED and R5B5 LED under ventilation as 29.5 and 31.2, respectively. Above results suggest that light quality optimization could be an important factor to foster in vitro growth of the species. Ventilation treatment appeared to be another important factor to induce normal shoot growth and rooting.

Comparison of Quality Characteristics of Sesame leaf Cleaned with Various Electrolyzed Water during Storage (다양한 전기분해수 세정처리에 따른 깻잎의 저장중 품질특성 비교)

  • This study was carried out to investigate the cleaning effect of sesame leaf, the sterilization effect and physicochemical properties, treated with various electrolyzed water. Initial physicochemical properties could be kept more than 1 month in electrolyzed oxidizing water(EW-1) of diaphragm type and 15 days in electrolyzed water(EW-2 and EW-3) of non-diaphragm system, there was no significant difference by storage temperature. 4 kinds of microorganism (initial total counts, $10^7\~10^9$ CFU/mL) were sterilized within $0.5\~1$ minutes by electrolyzed water. In fresh sesame leaves, total viable cell count and coliform group in the treatment of electolyzed water were decreased to about $2\~3$ log scale comparing non-treated ones. Especially Bacillus cereus was not detected until 13th day when treated with EW-l. Decaying ratio of sesame leaf appears on day 6 of storage in the untreated but the treatments of electrolyzed water has no sign until day 10 of storage. Change in color difference(${\Delta}E$) during storage was observed the treatments of electrolyzed low-alkaline water(EW-2) and electrolyzed neutral water(EW-3) were very desirable at the level $1\~2$ after day 13 of storage comparative to the untreated Change of Chlorophyll content was biggest decreased to 6.8 $mg\%$ on the untreated and decreased least to 8.35 $mg\%$ on EW-3 treated group on 13th day from initial value of $9.0\~10.3\;mg\%$ The overall sensory evaluation appeared most acceptable in the treatments of EW-2 and EW-3.

Effects of Depth and Duration of Water-logging on Growth and Yield at Transplanting and Flowering Stage in Pepper (Capsicum annuum L.) (고추(Capsicum annuum L.)의 이식기(移植期) 및 개화기(開化期) 침수처리(浸水處理) 따른 생육반응(生育反應))

  • Pepper plants were water-logged at 0, 5, 10 and 15 cm at transplanting and flowering stages under the condition of greenhouse. Treatment of water-logging times were 6, 12, 24, 48 and 120 hours. The results obtained are summarized as follows. At the transplanting stage, plant height, number of leaves, shoot and root fresh weight decreased by water-logging at 0cm for 24 hours and at 5cm or more for 6 hours. Number of fallen leaf was negligible by 12 hours water-logging at 0cm, however, its increased by more increased the water-logging depth and time. Diffusion resistance and chlorophyll content of leaf, and root activity decreased at more than 24 hours of water-logging regardless of the water-logging depth treatments. Photosynthesis and respiration rate diminished by increased the water-logging depth at 120 hours water-logging treatment. Plant diseases, mainly anthracnose(Colletotrichum) occurred in proportion to increase the depth and time of water-logging. It was not possible to control the diseases by fungicides. At the treatment of foliar spray of urea for recovery to water-logging damage, the efficiency was not found on plant height, but the number of leaves. Number of fruit and weight of fruit per plant showed no difference from no water-logging to 24 hours water-logging at 0cm, but its decreased that more than 24 hours water-logging at 0cm and more than 6 hours water-logging at 5cm or more. The averaged weight of a fruit on survival plants increased by more hours and deeper water-logging. There was positive correlation between all the investigated characteristics of growth and yield. There was, however, negative correlation between the characteristics and diffusion resistance of leaf stomata. The correlation between number of fallen leaf and averaged weight of a fruit was not significant. At flowering stage, number of fruit and weight of fruit per plant showed a similar tendency to no water-logging and by 12 hours water-logging at 0cm and 5cm, but significantly decreased at more than 24 hours water-logging from 0 to 5cm, and more than 6 hours water-logging at 10cm or more. The averaged weight of a fruit on survival plants increased by more hours and deeper water-logging except for 120 hours water-logging at all water depths.
