방풍(防風)과 해방풍(海防風) 중 뇌경색 연구에 더욱 적합한 약재 선정을 위한 네트워크 약리학적 분석 (Network pharmacoligical analysis for selection between Saposhnikoviae Radix and Glehniae Radix focusing on ischemic stroke)

  • Objectives : Saposhnikoviae Radix (SR) and Glehniae Radix (GR) have been frequently used in traditional medicine to treat diseases related to 'wind' syndrome, but there have been cases where it has been mixed in a state where the plant of origin is not clear. In this study, to select materials for conducting preclinical cerebral infarction research, the network pharmacology analysis method was used to select suitable medicinal materials for the study. Methods : In this study, a Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) based network pharmacology analysis method was used, and oral bioavailability (OB), drug likeness (DL), Caco-2 and BBB permeability were utilized to select compounds with potential activity. For the values of each variable used in this study, OB ≥ 20%, DL ≥ 0.18, Caco-2 ≥ 0, and BBB ≥ -0.3 were applied, then networks of bioactive compounds, target proteins, and target diseases was constructed. STRING database was used to construct a protein-protein interaction network. Results : It was confirmed that SR rather than GR has various target proteins and target diseases based on network pharmacological analysis using TCMSP database. And it was analyzed that the bioactive compounds only in SR act more on neurovascular diseases, and both drugs are expected to be effectively used for cardiovascular diseases. Conclusions : In our future study, SR will be used in an ischemic stroke mouse model, and the mechanism of action will be explored focusing on apoptosis and cell proliferation.

Therapeutic effects of paeoniflorin on irritable bowel syndrome in rats

  • Background: Irritable bowel syndrome (IBS) is a functional bowel disorder (FBD). Objectives: To assess the therapeutic effects of paeoniflorin (PF) on IBS in rats. Method: Sixty male Sprague-Dawley rats were randomly divided into normal, model, positive drug, low-dose PF, medium-dose PF and high-dose PF groups (n = 10). After gavage for 2 consecutive weeks, the effect of PF on abdominal pain symptoms was assessed based on the abdominal withdrawal reflex (AWR) score, fecal water content and pathological changes in colon tissues. D-lactate, interleukin-1β (IL-1β), transforming growth factor-β (TGF-β) and tumor necrosis factor-α (TNF-α) were detected by enzyme-linked immunosorbent assay, and phosphorylated nuclear factor kappa B (p-NF-κB) p65 was detected by Western blotting. The abundance and diversity changes of intestinal flora were explored using 16S ribosomal RNA sequencing. Result: In PF groups, the mucosal morphology of colon tissues was intact, and the glands were arranged neatly and structured clearly, without obvious inflammatory cell infiltration. Compared with the model group, PF groups had significantly elevated pain threshold, and mRNA and protein levels of zonula occludens-1 (ZO-1) and occludin, decreased AWR score at 20 mmHg pressure, fecal water content, mRNA levels of IL-1β, TGF-β, and TNF-α, protein level of p-NF-κB p65 and level of serum D-lactate, and reduced levels of serum IL-1β, TGF-β, and TNF-α (p < 0.05, p < 0.01). PF groups had higher abundance of Lactobacillus, Akkermansia, Alistipes, and Bacteroides, but lower abundance of Desulfovibrio, Parasutterella, and Enterococcus than those of the model group. Conclusions: PF exerts therapeutic effects on IBS in rats probably by regulating the intestinal flora, and then up-regulating the expressions of ZO-1 and occludin in colon tissue while down-regulating the levels of IL-1β, TGF-β, TNF-α, D-lactate and p-NF-κB p65.

Dengue Virus 2 NS2B Targets MAVS and IKKε to Evade the Antiviral Innate Immune Response

  • Dengue virus (DENV) is a widespread arbovirus. To efficiently establish infection, DENV evolves multiple strategies to hijack the host innate immune response. Herein, we examined the inhibitory effects of DENV serotype 2 (DENV2) nonstructural proteins on RIG-I-directed antiviral immune response. We found that DENV2 NS2A, NS2B, NS4A, and NS4B significantly inhibited RIG-I-mediated IFN-β promoter activation. The roles of NS2B in RIG-I-directed antiviral immune response are unknown. Our study further showed that NS2B could dose-dependently suppress RIG-I/MAVS-induced activation of IFN-β promoter. Consistently, NS2B significantly decreased RIG-I- and MAVS-induced transcription of IFNB1, ISG15, and ISG56. Mechanistically, NS2B was found to interact with MAVS and IKKε to impair RIG-I-directed antiviral response. Our findings demonstrated a previously uncharacterized function of NS2B in RIG-I-mediated antiviral response, making it a promising drug target for anti-DENV treatments.

형개 추출물의 시스템 약리학적 분석과 비소세포폐암세포에 대한 증식 억제효과 (Systems Pharmacological Approach to Identification of Schizonepeta teunifolia Extract via Active Ingredients Analysis and Cytotoxicity Effect on A549 Cell Lines)

  • Objectives : This study aimed to predict the effectiveness and potential of Schizonepeta tenuifolia as an anticancer treatment for non-small cell lung cancer through network-based pharmacology and cellular experiment. Methods : To identify the major bioactive compounds in Schizonepeta tenuifolia, we used the Traditional Chinese Medicine Systems. The target genes for the cancer treatment were selected using the UniProt database and the networked using Cytoscape. We performed functional enrichment analysis based on the Gene Ontology Biological Process and Kyoto Encyclopedia of Genes and Genomes Pathways to predict the mechanisms. To investigate the effect of Schizonepeta tenuifolia on lung cancer cell growth, we treated A549 cells, a lung cancer cell line, with different concentrations of the drug and used the MTT assay for cell viability. Results : Research has shown that the most effective mechanism of active compounds from Schizonepeta tenuifolia is through the pathway of cancer. The results of the network pharmacology analysis indicate that Schizonepeta tenuifolia has potential medicinal value as an adjuvant in anticancer treatment. The concentration-dependent inhibition of cell viability was observed on A549 cells. Furthermore, synergistic anticancer activity with Doxorubicin was also observed. Conclusions : Through a network pharmacological approach, Schizonepeta tenuifolia was predicted to have potential as an anticancer agent, and its efficacy was experimentally demonstrated using A549 cells. These findings suggest that Schizonepeta tenuifolia is a promising candidate for future research.

배추김치의 활성성분인 3-(4-hydroxyl-33,53-dimethoxyphenyl)propionic acid의 고지혈증 치료 효과 (The Effect of 3-(4-hydroxyl -33,53-dimethoxyphenyl)propionic Acid in Chinese Cabbage Kimchi on Lowering Hypercholesterolemia)

  • 배추김치의 생리활성물질인 3-(4-hydroxyl-3',5'-dimethoxyphenyl)propionic acid의 고지혈증 치료효과를 살펴보기 위하여 고콜레스테롤 혈증을 유발시킨 토끼에게 1차 실험에서는 정상식이를 공급하고 2차 실험에서는 고콜레스테롤 식이를 공급하면서 각각 16일간 정맥으로 투여하여 혈장지질 농도 변화를 살펴보았고, 그 효과는 고지혈증 치료제로 사용되고 있는 Simvastatin과 비교하였다. 정상식이 공급시 Kimchi군 및 Simvastatin군에 의한 총콜레스테롤 감소는 대조군에 비해 18.65%와 29.67%이었고 고콜레스테롤 식이 공급시에는 각각 33.79%및 21.81%이었다. LDL-C의 저하현상은 정상식이 공급시에는 미미하게 관찰되었고, 고콜레스테롤 식이 공급시에는 Kimchi군 및 Simvastatin군에서 약 130%정도의 감소 효과가 관찰되었다. 중성지방의 감소효과는 정상식이시에는 거의 관찰되지 않았고, 고콜레스테롤 식이시에는 Kimchi군에서 약 105%, 그리고 Simvastatin군에서 약 62%정도의 감소가 관찰되었다. HDL-C의 변화는 모든 군에서 관찰되지 않았다. 고콜레스테롤 식이를 섭취한 토끼간의 HMG-CoA reductase 활성을 살펴보았을 때 Kimchi군 및 Simvastatin군의 활성이 48.44% 그리고 139.06% 증가하였다. 본 연구결과 Kimchi군은 고지혈증을 치료하는 효과가 있음이 관찰되었고, 그 효과는 Simvastatin군과 유사하였다. Kimchi군 및 Simvastatin군에서 관찰된 HMG-CoA reductase 활성의 증가는 지질저하 현상에 대한 보상 작용으로 생체 항상성 유지를 위한 현상으로 사료된다 3-(4-hydroxyl-3',5'-dimethoxyphenyl)propionic acid의 고지혈증 치료기전은 Simvastatin과 유사할 것으로 예상된다.

HPLC-UVD/MS를 이용한 농산물 중 fomesafen의 분석 (Determination of Fomesafen Residue in Agricultural Commodities Using HPLC-UVD/MS)

  • 광엽 제초제로 사용되는 fomesafen은 미국 및 중국 등에서 두류 및 과수의 광엽 제초제로 사용되고 있으나, 국내에서는 미사용 농약이며, 잔류허용기준 및 분석법이 확립되어 있지 않다. 최근 FTA 등으로 외국에서 수입되는 농산물 중 fomesafen에 대한 안전성 검토가 필요하나, 국내 식품공전 상에는 fomesafen의 분석법이 확립되어 있지 않다. 본 연구는 HPLC-UVD/MS를 이용하여 농산물 중 fomesafen의 잔류 분석법을 확립하고자 하였으며, 대상 농산물은 현미, 콩, 사과, 배추 및 고추를 선정하였다. 농산물 시료에 acetone을 가하여 추출된 fomesafen 성분을 dichloromethane 액-액 분배법과 Florisil 흡착크로마토그래피법으로 정제하여 HPLC-UVD/MS 분석대상 시료로 하였다. Fomesafen의 정량적 분석을 위한 최적 HPLC 분석 조건을 확립하였으며, 정량한계(LOQ)는 0.04 mg/kg 이었다. 각 대표 농산물에 대해 정량한계, 정량한계의 10 및 50배 수준에서 회수율을 검토한 결과 모든 처리농도에서 87.5~102.5% 수준을 나타내었으며, 반복 간 변이계수(CV)는 최대 7.7%를 나타내어 잔류분석 기준인 회수율 70~120% 및 분석오차 10% 이내를 충족시키는 만족한 결과를 도출하였으며, LC/MS SIM을 이용하여 실제 농산물 시료에 적용하여 재확인 하였다. 이상의 결과로 신규 fomesafen의 HPLC-UVD/MS 분석법은 검출한계, 회수율 및 분석오차 면에서 국제적 분석기준을 만족하는 신뢰성이 확보된 정량 분석법으로 사용가능할 것이다.

Filtration과 Integrated Cell Culture/Real-Time Reverse Transcription PCR 기법을 이용한 채소류에서 Human Rotavirus 신속 검출 (Rapid Detection Method for Human Rotavirus from Vegetables by a Combination of Filtration and Integrated Cell Culture/Real-Time Reverse Transcription PCR)

  • 본 연구는 human rotavirus (HRV)의 검출법을 최적화하기 위해 real-time RT-PCR과 세포 배양법을 이용하여 여러 가지 탈리 농축법을 비교 및 평가하는 것을 목적으로 하였다. 채소류 중 배추, 상추, 깻잎을 선정하여 바이러스 희석액을 접종하고 탈리액 비교를 위하여 buffer A (100 mM Tris-HCl, 50 mM glycine, 3% beef extract, pH 9.5)와 buffer B (250 mM Threonine, 300 mM NaCl, pH 9.5)를 이용하여 탈리하였고, 농축방법을 비교하기 위하여 PEG (polyethylene glycol) 침전법 또는 filtration [Nanoceram filter$^{(R)}$ (Argonide corporation)]을 이용하여 농축하였다. 또한 바이러스의 감염성 평가를 위하여 MA-104 cell을 배양하여 탈리, 농축 방법을 거쳐 회수된 HRV를 접종하고 1, 48, 72, 96, 120, 144, 168시간 후 세포를 수거하여 real-time RT-PCR을 시행하고 세포병변을 관찰하였다. 탈리 용액은 buffer A가 회수율 29.54%로 buffer B의 18.32%보다 더 뛰어난 탈리효과를 보였으며 농축방법을 비교했을 때 filtration 방법이 회수율 51.89%를 나타내며 PEG 침전법에 비해서 바이러스의 농축에 효과적이었으며 검출 소요시간이나 간단한 과정 면에서 효율적이었다. ICC/real-time RT-PCR을 시행하였을 때 세포병변 72시간 후부터 나타나기 시작했지만 Ct value는 48시간부터 감소하기 시작하여 더 빠른 시간 내에 감염성을 평가할 수 있었다. 따라서, filtration과 integrated/cell culture real-time RT-PCR을 이용하면 기존의 검출방법보다 빠른 시간 내에 바이러스 검출이 가능할 것으로 여겨진다.

홍화 추출물이 치주인대세포, 조골세포 활성도에 미치는 영향 (The biologic effects of safflower(Carthamus tinctorius $Linn\acute{e}$) extract and Dipsasi Radix extract on periodontal ligament cells and osteoblastic cells)

  • Safflower(Carthamus tinctorius $Linn\acute{e}$ has been traditionally used for the treatment of blood stasis, and Dipsasi Radix has been used as a drug for fracture in Chinese medicine. The purpose of present study was to examine the biologic effects of safflower extract and Disasi radix extracts on the periodontal. ligament cells and osteoblastic cells and on the wound healing of rat calvarial defect. The ethanolic extract of safflower blossom, safflower seed and Dipsasi Radix(125, 250, and 500 ${\mu}g/ml$) were prepared as test group, and PDGF-BB(lOng/ml) and unsafonifiable fraction of Zea Mays L.(125, 250, and 500 ${\mu}g/ml$) were employed as positive control. The effects of each agents on the growth and survival, ALPase activity, expression of PDGF-BB receptor, chemotactic response of PDL cell and ATCC human osteosarcoma MG63 cells in vitro were examined. The tissue regenerative effect of each extracts was evaluated by histomorphometric measuring of newly formed bone on the 8mm defect in rat calvaria after oral administration of 3 different dosages groups : 0.02, 0.1 and 0.35g/kg, per day. It was also employed the same dosages of unsaponifiable fraction of Zea Mays L. as positive controls. Safflower blossom extract, safflower seed extract, and Dipsasi Radix extract stimulate the cellular activity of MG63 cells in concentration range of $125-500{\mu}g/ml$, and safflower bolssom extract and safflower seed extract stimulate also the cellular activity of periodontal ligament cells in concentration range of $250-500{\mu}g/ml$. In activity of ALPase, $250-500{\mu}g/ml$ of safflower blossom extracts showed significant stimulating effects on MG63 cells, and the same concentration range of safflower seed extracts showed significant effect on periodontal ligament cells. In the recovery on PDGF-BB receptor expression which was depressed by $IL-1{\beta}$, $125-250{\mu}g/ml$ of safflower blossom extracts and $250-500{\mu}g/ml$ of safflower seed extracts showed significant increasing effect on MG63 cells, and $500{\mu}g/ml$ of safflower blossom extract and $250-500{\mu}g/ml$ of safflower seed extracts showed significant effect on periodontal ligament cells. In chemotactic response, among all tested group, safflower seed extracts only were chemotactic to MG63 cells and periodontal ligament cells in concentration range of $125-500{\mu}g/ml$. Also in the view of bone regeneration in rat calvarial defect model, the only group that was orally administrated 0.35g/kg, day of safflower seed extract showed significant new bone formation. These results suggested that safflower extracts might have a potential possibilities as an useful drug for adjunct to treatment for regeneration of periodontal defect.

과루인(瓜蔞仁)이 장기간 고용량 corticosteroid 투여 hairless mouse의 피부장벽에 미치는 영향 (Effect of Seed of Trichosanthes kirilowii on the Skin Barrier of High Dose Corticosteroid Injected hairless Mouse)

  • Objective : Prolonged use of systemic corticosteroids negatively impacts skin barrier function. Corticosteroids have a major role in the practical management of many diseases, so it is necessary to find the drug or supplement which could keep the skin healthy during the systemic corticosteroids therapy. Seed of Trichossnthcs kiiilowii was commonly used for pulmonary disease in traditional Chinese medicine. Many studies have investigated and found that seed of Trichosanthes kirilowii has anti-bacterial, anti-HIV, anti-tumor and immunoregulatory functions. This study was performed to investigate the effect of seed of Trichosmthes kirilowii on the Skin Barrier Method : Triamcinolone 0.4mg was injected male hairless mice for 5 weeks ( 2 times a week, totally 10 times), Just before the first injection, hairless mice were divided into 3 groups; Control ( normal saline medicated group ), GroupA( seed of Trichosuuhes kirilowii qd/day medicated group ) and GroupB( seed of Trichosuuhes kiiilowii bid/day medicated group ), Body weight of all the groups were checked during the experiment, After $10^{th}$ injection, TEWL (Transeidermal water loss) of 3 groups were artificially increased by 9 times tape stripping and the changes of TEWL were checked at before stripping, right after, 2h, 4h, 6h, 24h, 48 and 72h later after stripping, AST, ALT and CBC were also checked. Indeed, stratum corneum of 3 groups were also examined and compared with that of normal hairless mouse. Results : 1. GroupB showed significantly lower body weight among three groups 2, There was no statistical difference at AST, ALT and CBC among three groups 3. GroupB was showed significantly lower TEWL than TEWL of Control at 48h later. 4 GroupB recovered it's TEWL of before tape stripping at 24h later, GroupA recovered that at 48h and Control recovered that at 72h later at the same condition 5. Stratum corneum of GroupA and GroupB were looked almost normal and healthy. On the contrary, stratum corneum of Control was looked thin and unhealthy. Conclusions : Seed of Ttichossnthes kirilowii has some effects on skin barrier function, especially TEWL of high dose corticosteroids injected hairless mouse and the efficacy seemed to be related with the dosage.

Allele Frequencies of the Single Nucleotide Polymorphisms Related to the Body Burden of Heavy Metals in the Korean Population and Their Ethnic Differences

  • This study was performed to select single nucleotide polymorphisms (SNPs) related to the body burden of heavy metals in Koreans, to provide Korean allele frequencies of selected SNPs, and to assess the difference in allele frequencies with other ethnicities. The candidate-gene approach method and genome-wide association screening were used to select SNPs related to the body burden of heavy metals. Genotyping analysis of the final 192 SNPs selected was performed on 1,483 subjects using the VeraCode Goldengate assay. Allele frequencies differences and genetic differentiations between the Korean population and Chinese (CHB), Japanese (JPT), Caucasian (CEU), and African (YIR) populations were tested by Fisher's exact test and fixation index ($F_{ST}$), respectively. The Korean population was genetically similar to the CHB and JPT populations ($F_{ST}$ < 0.05, for all SNPs in both populations). However, a significant difference in the allele frequencies between the Korean and CEU and YIR populations were observed in 99 SNPs (60.7%) and 120 SNPs (73.6%), respectively. Ten (6.1%) and 26 (16.0%) SNPs had genetic differentiation ($F_{ST}$ > 0.05) among the Korean-CEU and Korean-YIR comparisons, respectively. The SNP with the largest $F_{ST}$ value between the Korean and African populations was cystathionine-${\beta}$-synthase rs234709 ($F_{ST}$: KOR-YIR, 0.309; KOR-CEU, 0.064). Our study suggests that interethnic differences exist in SNPs associated with heavy metals of Koreans, and it should be considered in future studies that address ethnic differences in heavy-metal concentrations in the body and genetic susceptibility to the body burden of heavy metals.
