Analysis of the orf 282 Gene and Its Function in Rhodobacter sphaeroide 2.4.1 (R. sphaeroides 에서의 orf282 유전자의 분석과 이들의 기능)

  • The orf282 gene of Rhodobacter sphaeroides is located between the ccoNOQP operon encoding $cbb_3$ terminal oxidase and the fnrL gene encoding an anaerobic activator, FnrL. Its function remains unknown. In an attempt to reveal the function of the orf282 gene, we disrupted the gene by deleting a portion of the orf282 gene and constructed an orf282-knockout mutant. Two FnrL binding sites were found to be located upstream of orf282, and it was demonstrated that orf282 is positively regulated by FnrL. The orf282 gene is not involved in the regulation of spectral complex formation. The $cbb_3$ oxidase activity detected in the orf282 mutant was comparable to that in the wild-type sample, indicating that the orf282 gene is not involved in the regulation of the ccoNOQP operon and the biosynthesis of the cbb3 cytochrome c oxidase. The elevated promoter activity of the nifH and nifA genes, which are the structural genes of nitrogenase and its regulator, respectively, in the orf282 mutant, suggests that the orf282 gene product acts as a negative effector for nifH and nifA expression.

  • A bridge resistance deviation-to-frequency (BRD-to-F) converter is presented for interfacing resistive sensor bridges. It consists of a linear operational transconductance amplifier (LOTA), a current-controlled oscillator (CCO). The prototype converter was simulated using commercially available discrete components. The result shows that the converter has a conversion sensitivity amounting to 16.90 kHz/${\Omega}$ and a linearity error less than ${\pm}$0.03 %.

Artificial Microparticle Diets for Culturing Rotifer, Brachionus plicatilis

  • Rotifer culture fed on five types of artificial microparticle diets were evaluated to substitute the natural diets such as Chlorella or w-yeast. These microparticle diets including solidified blood using squid oil (SBSO), solidified blood using soybean oil (SBSB), nylon protein walled particle (NPW) simple coacervation oil capsule (SCO), complex coacervation oil capsule(CCO), were tested for the evaluation of feeding efficiency. The prepared micro particle diets had diameters ranging from 3 to 30 Jim. Rotifer culturing experiments were carried out in 3-liter beakers for 13-16 days. The initial inoculum density of rotifers was 10 ind./ml. The rotifers fed on Chlorella or $\omega-yeast$ showed maximal densities of 2,000 ind./ml in 9 days or 500 ind./ml in 7 days, respectively. Those fed on SBSO, SBSB or NPW showed maximal densities of 1568 ind./ml, 586 ind./ml or 503 ind./ml, respectively and the reproductive rates for those diets were equivalent to or better than w-yeast. However, the coacervated oil capsule showed lower maximal densities of 400 ind./ml for SCO and less than 100 ind./ml for CCO due to the unbalanced diet formulation and indigestibility.

A Study to Select and Apply Description Elements for the Integrated Search of Archival Objects in University Archives (대학기록관 박물의 통합검색을 위한 기술 요소의 선정 및 적용에 관한 연구)

  • The purpose of this study is to select and apply description elements for the integrated search of archival objects in university archives. For this purpose, the act of public archives management and the research literature were reviewed, and a survey was conducted on related institutions. Moreover, this paper made a comparison by using the archival description of ISAD(G), DACS, RDA, and CCO and referred to the descriptions that are being used at university archives. Based on these analyses, we suggested the description elements for the integrated search of archival objects in university archives and presented the practical application method for it.

3D Structure of Bacillus halodurans O-Methyltransferase, a Novel Bacterial O-Methyltransferase by Comparative Homology Modeling

  • Bacillus halodurans O-methyltransferase (BhOMT) is a S-adenosylmethionine (SAM or AdoMet) dependent methyltransferase. Three dimensional structure of the BhOMT bound to S-adenosyl-L-homocysteine (SAH or AdoHcy) has been determined by comparative homology modeling. BhOMT has 40% sequence identity with caffeoyl-CoA 3-O-methyltransferase (CCoAOMT) from alfalfa. Based on x-ray structure of CCoAOMT, three dimensional structure of BhOMT was determined using MODELLER. The substrate binding sites of these two proteins showed slight differences, but these differences were important to characterize the substrate of BhOMT. Automated docking study showed that four flavonoids, quercetin, fisetin, myricetin, and luteolin which have two hydroxyl groups simultaneously at 3'- and 4'-position in the B-ring and structural rigidity of Cring resulting from the double bond characters between C2 and C3, were well docked as ligands of BhOMT. These flavonoids form stable hydrogen bondings with K211, R170, and hydroxyl group at 3'-position in the Bring has stable electrostatic interaction with Ca2+ ion in BhOMT. This study will be helpful to understand the biochemical function of BhOMT as an O-methyltransferase for flavonoids.

Wound Healing Effects of Lespedeza cuneata Extract (야관문 추출물의 창상치유 효과)

  • In this study, the anti-inflammatory effects of Lespedeza cuneata extract on macrophages and wound-healing in wound-induced animal experiments were investigated. In an anti-inflammatory test, 0.1 mg/mL of Lespedeza cuneata extract did not affect growth of RAW 264.7 cells, and Lespedeza cuneata extract suppressed nitric oxide (NO) generation from inflammation-induced macrophages in a concentration-dependent manner. Wounds on the skin of rats were treated with vehicle containing Lespedeza cuneata extract (SSP), vehicle (SCO), and commercial ointment (CCO). The wound and scar sizes in the SSP group were significantly reduced in comparison to the SCO and CCO groups (P<0.05). The epidermis and dermis of the SSP group also recovered faster than the SCO group based on Masson's trichrome staining. The gene expression levels of vascular endothelial growth factor (VEGF) decreased and transforming growth factor-beta 1 (TGF-${\beta}1$) increased in wound tissue from the SSP group compared to that from the SCO group. These results show that Lespedeza cuneata extract accelerates wound-healing through anti-inflammatory activity and induction of collagen regeneration as well as reduces the scar area surrounding wounds. Accordingly, Lespedeza cuneata extract could be useful as a cosmeceutical in the cosmetic industry.

A survey of respiratory pathogens in dogs for adoption in Gwangju metropolitan city animal shelter, South Korea (광주광역시동물보호소 입양 대상 유기견의 호흡기 질병 실태 조사)

  • Canine infectious respiratory disease (CIRD), also known as infectious tracheobronchitis or kennel cough occurs in a multiple-dog environment such as a shelter. In this study, we were collected 300 of nasal swab samples from dogs and 145 of environmental samples from a shelter to investigate respiratory pathogens of dogs in the Gwangju metropolitan city animal shelter from February to October, 2019. Bacteria cultures for isolation of Bordetella (B.) bronchiseptica and polymerase chain reaction (PCR) tests were performed for detection of eleven canine respiratory pathogens, namely Mycoplasma (M.) cynos, canine distemper virus (CDV), canine influenza virus (CIV), canine parainfluenza virus (CPIV), canine respiratory coronavirus (CRCoV), alpha-coronavirus (CCoV), canine pneumovirus (CnPnV), canine hepacivirus (CHeV), canine adenovirus type 2 (CAdV-2), canine herpesvirus-1 (CHV-1) and canine bocavirus (CBoV). Among 300 nasal swab samples, 148 samples (49.3%) were positive for at least one pathogens. CHV-1 was the most common pathogen, found in 95/300 (31.7%) samples. Subsequently, M. cynos (22.0%), B. bronchiseptica (2.3%), CPIV (2.0%), CBoV (1.7%), CCoV (0.7%) were detected. The detection rates of M. cynos and CHV-1 according to the duration of stay in the shelter were statistically significant. Among environmental samples, M. cynos, CCoV, CBoV and CHV-1 were detected in 45/145 (31.0%). These results indicated the need for disease control and prevention systems in the shelter.

A Study on Optimal Airspace Design for Continuous Climb Operation (연속상승운항을 위한 최적 공역 설계에 관한 연구)

  • This paper introduces a new airspace design method for continuous climb operation (CCO). The optimization problem is formulated as Mixed-Integer Linear Program (MILP) to maximize the upper limits of altitude on the waypoints to facilitate continuous climb for aircraft. In the proposed method, the interactions with other flight procedures are considered as well as various aircraft flight performances. The proposed method is applied to one of the departure procedures of Incheon International Airport (ICN) to demonstrate its performances.

  • This study presents a comprehensive solution to the collection management, which is based on the model for Cultural Objects (CCO). The developed system manages and spreads the collections that are safeguarded in museums and galleries more easily by using IT. In particular, we present our approach for a non-structured search and recovery of the objects based on the annotation of artwork images. In this methodology, we have introduced a faceted search used as a framework for multi-classification and for exploring/browsing complex information bases in a guided, yet unconstrained way, through a visual interface.

  • The necessity of university's educational change is rising up, along with a social environment that keeps changing. This study deals with a newly educational style by recognizing the necessity of variance for engineering education, according to engineers' diversified roles within a company. The study was conducted by 7 members of middle-sized group, replaced from 5 members of small-sized and each was given his task by his job such as CEO, CMO, CSO, CIO, CCO, CQO or Auditor. On the basis of four theoretical backgrounds, this paper investigated students' level of learning satisfaction shown before and after task undertaking through a questionnaire and appraised the result made before and after task undertaking. This study is expected to improve a teaching method of university engineering education.
