Evaluation of Methods Used in Nutrition Surveys in Korea(1960-1990) (국내 영양조사(1960-1990년)에서 적용된 영양평가 방법의 내용 및 추이분석)

  • The purpose of the study is to assess and evaluate methods employed in nutrition surveys and to give an information on methodological considerations for future studies in Korea. Three hundred thirty six papers of nutritional surveys published from 1960 to 1990 were classified into 9 groups according to the survey subject and analyzed for methods used in survey. The frequency of publications according to the survey subject was as the following order : 'in-fants and preschool children' 'school age children' 'adolescence' 'college students' 'preg-nant and lactating women' 'housewives' 'the elderly' 'workers' and 'patients' Eighty five percents of papers applied methods of dietary assessment. of which weighing method recall method an record method were extensively used in the order. Questionnairs included items related to nutrition such as food habit nutrition knowledge etc. Anthropometric techniques were employed in 53% of papers and in general growth measurement were undertaken for children and body composition was emphasized for adults. biochemical assessment was limited to 32% of papers and assessment of anemia was the major test. Clinical studies were applied to 26% of the studies. of which only 5% employed examination of clinical signs of malnutrition Recently self-reported health status assessed using questionnaires has been adopted frequently. Further researches are required for evaluation of the methods employed in nutrition surveys in order to develop the standard model for nutrtion survey so that obtained data can become more reliable and utilized efficiently.

Growth Characteristics, Bio-chemical Composition and Antioxidant Activities of Benthic Diatom Grammatophora marina from Jeju Coast, Korea

  • Benthic diatoms are known as a good food for shellfish in nature and in commercial hatchery of Jeju Island, Korea. Grammatophora marina is commonly found as dominant benthic micro-algae in Jeju coastal waters throughout the year. To know the best growth conditions of this species, culture was done in terms of three parameters; water temperature, salinity and nutrients. Each parameter was controlled by temperature of 15, 20 and 25°C; salinity of 25, 30 and 35 psu; and nutrient concentrations of 50, 100 and 200%. F/2 media was used with artificial seawater for the culture, which was continued for two weeks with L:D cycle 12:12 by using fluorescent light. Maximum specific growth rate was recorded 1.68 d–1 at temperature of 25°C with salinity of 35 psu and nutrient concentration of 200% on 6th day during the culture period. Maximum biomass was also observed 4.9 × 105 cells mL–1 in the same condition. This species may belong to the euryhaline and eutrophic habitat with warm condition. For nutritional aspects of this species, protein, lipid and carbohydrate were measured. The value of protein, lipid and carbohydrate was 4.96%, 15.82% and 5.65%, respectively. The antioxidant activities of 80% methanolic extract were 46.7%, 23.7% and 23.8% on DPPH (1,1-Diphenyl-2-picrylydrazy) radical, superoxide anion radical and hydrogen peroxide scavenging, respectively. Percentage metal chelating activity was 81.2%. Enzymatic extracts of Alcalase and Ultraflow showed remarkable scavenging activities on DPPH radical (86.5% and 57.2%, respectively), and superoxide anion scavenging activities were 45.3% and 41.4% from Kojizyme and Viscozyme extracts, respectively. Extract of Protomex revealed 24.8% activity on hydrogen peroxide and Neutase showed 30.8% on hydroxyl radical scavenging effects. Celluclast and Viscozyme extracts showed 33.2% and 32.1% activities on nitric oxide scavenging, respectively, while Alcalase showed 61.5% on metal chelating. This species contains higher lipids among the biochemical compounds and higher metal chelating activities from both 80% methanolic and enzymatic extracts.

Nutritional Evaluation of Lentil (Lens culinaris) Straw and Urea Treated Wheat Straw in Goats and Lactating Buffaloes

  • The nutritive value of lentil straw (Lens culinaris) relative to 4% urea treated wheat straw (w/w) was assessed in adult bucks and milking buffaloes on-station and on-farm, respectively. A digestion-cum-nitrogen balance experiment was conducted with three bucks (24.35 kg) fed ad libitum lentil straw (LS), urea treated wheat straw (UTS) and a 1:1 (w/w DM basis) mixture of LS and UTS (LS:UTS) in a 3$\times$3 Latin square design. Though daily dry matter intake (g kg $W^{0.75}$ or % LW) of bucks fed LS and UTS did not differ significantly, that of LS:UTS was significantly (p<0.05) low. Similarly, digestibility of nutrients (DM, OM and CP) and nitrogen balance (g/day) did not differ significantly for bucks given LS or UTS although a significant positive improvement was evident in these parameters for LS:UTS. The digestibility of fibre fractions (NDF and ADF) was significantly (p<0.05) lower in LS as compared to UTS or LS:UTS. Except for the significantly (p<0.05) lower intake of DCP (g kg$W^{0.75}$), the bucks fed LS had statistically similar nutrient intake as compared to UTS. Feeding of LS:UTS resulted in significantly higher nutrient (DCP, TDN) intake by bucks. The important blood-biochemical parameters (haemoglobin,packed cell volume, glucose, urea nitrogen, total protein) and activity of serum enzymes (alanine aminotransferase and aspartate aminotransferase) did not differ significantly in goats irrespective of dietary treatment. In the onfarm study, eighteen multiparous milking buffaloes were randomly assigned into three equal groups to assess the relative effect of feeding untreated wheat straw (WS), UTS and LS:UTS ad libitum as basal feed with a supplement. The study continued for 3 months and revealed that daily dry matter intake and average milk production (liters/day) by buffaloes offered LS:UTS and UTS was statistically similar but it was significantly higher compare to WS. However, milk composition did not differ significantly among the dietary treatments. It may be concluded that the nutritive value of lentil straw appeared to be no different to UTS, however, a positive synergistic effect was evident by feeding a mixture of LS and UTS on performance of goats and lactating buffaloes.

Production and Characterization of Thermostable Protease from Bacillus licheniformis Isolated from Korean Traditional Soybean Paste (재래식 된장에서 분리된 Bacillus licheniformis의 내열성 Protease 특성과 생산성)

  • Among 63 Bacillus strains grown at $60^{\circ}C$ from sixteen samples of homemade Korean soybean paste, one strain was selected for producing the thermostable protease. The isolate has been identified as Bacillus licheniformis on the basis of its 16S rDNA sequence, morphology and biochemical properties. Culture filtrate of the isolate showed maximal protease activity at the reaction condition of $60-65^{\circ}C$ and pH 11. The culture filtrate retained more than 87% of initial protease activity after incubation for 30 min at $60^{\circ}C$ without substrate. In order to develop the medium composition, effects of ingredients including nitrogen sources, carbon sources, metal ions and phosphate were examined for protease production of the isolate. Lactose and soytone peptone were the most effective carbon and nitrogen source for the enzyme production. After the late logarithmic growth phase the isolate began to produce the protease, and the maximum protease productivity was reached to 550 unit/ml in the optimized medium consisting of lactose (3%), soytone peptone (1.5%), $MgSO_4$ (0.1%), $K_2HPO_4$ (0.03%), and $KH_2PO_4$ (0.03%) at 28 h of incubation.

Food Intake and Clinical Blood Indices of Female College Students by Body Mass Index (체질량지수에 따른 여대생의 식이섭취 실태와 혈액성상에 관한 연구)

  • This study was conducted to investigate the relationships among body composition, dietary intake, and clinical blood indices in college students by body mass index (BMI). Their body compositions were determined by means of BIA (Bioelectrical Impedance Analysis) method. Their dietary intake was determined using a 3-day record method and their hematological indices were determined by semi-automated microcell counter (Sysmex F-520). Their serum lipid levels were measured using biochemical analyzer (Spotchem). Subjects were classified as underweight, normal or over-weight groups according to their BMI. The subjects were 69 healthy college students aged 20 to 26 years. The average age, height, weight, and BMI was 21.3 years, 162.6 cm, 54.4 kg, and $20.6cm/m^2$, respectively. Their average consumption of energy was 1693 kcal, 84.7% of RDA and their mean ratio of carbohydrate: protein: fat were 54.5 : 16.4 : 29.0. There was no significant difference in nutrient intake among the groups except ${\beta}$-carotene and vitamin C. The ${\beta}$-carotene intake was significantly higher in the underweight group. Vitamin C intake was significantly higher in the overweight group. The mean intakes of Ca, Fe, Zn and folate of subjects were 74.8% to 83.2% of RDA. Especially, intakes of Ca, Fe, Zn and folate were lower in the abnormal weight groups. The overall mean values of the hematological indices in female college students were within the normal range and there was no significant difference among the groups. However, anemic subjects with hemoglobin (< 12 g/dl) and hematocrit (< 36 g/dl) accounted for about 11% of the subjects. The everall mean values of the serum lipid levels were within the normal range and there was no significant difference among the groups. But serum HDL-cholesterol level of the overweight group was lower than that of the other groups. LDL-/HDL-cholesterol ratio and AI index were significantly higher in the overweight group compared to the other groups. Based upon this study, it is necessary for college women to be educated regarding consuming more Ca, Fe, Zn, folate and less fat and cholesterol in order to have better health promotion.

A Study on the Consciousness Survey for Visitors and Physical Properties of Refuse in Summer Resort - Focused on Beach and Valley in Gangwon province - (피서지 쓰레기의 물리적 특성 및 피서객 의식조사 연구 - 강원도 해수욕장.산간계곡 중심으로 -)

  • In order to making resources for the organic wastes, it is very important to understand for the life cycle of wastes before the physicochemical analysis and engineering technology. Therefore we try to fine the solution through the interdisciplinary consilience between natural science and social science for the management of refuge in summer resort. Summer visitors of beach answered that fly tipping of refuse was 65.56% and insufficient separation was 17.78% about the survey. But insufficient separation was 42.5% and fly tipping was 37.5% in valley. The survey for the effective methods at reducing refuse was represented that campaign and teaching was 47.78%, fine was 23.33% and using the standard bags was 18.89% in beach. Campaign and teaching was 37.5%, using the standard bags was 37.5% and fine was 15% in valley. Bulk density of refuse in gyeongpo beach was measured in $74kg/m^{3}$. This value was three times as much low than municipal solid wastes. Moreover, the composition of refuses in beach showed that combustible materials was 81.1% and incombustible materials was 18.9%. Moisture, ash and combustibles were analyzed 19.0%, 9.2% and 91.8% respectively.

Identification of Alkalophilic Bacillus sp. S-1013 Producing Non-Cariogenicity Sugar Fuc($1{\to}4$)gaINAc($2{\to}6$)NeuAc and Optimization of Culture Condition for Its Production (비우식성 당 Fuc($1{\to}4$)gaINAc($2{\to}6$)NeuAc를 생산하는 호알칼리성 Bacillus sp. S-1013의 동정 및 생산조건의 최적화)

  • The study was performed to identification of producing microbe Non-Cariogenicity Sugar (NCS; Fuc($1{\to}4$) gaINAc($2{\to}6$)NeuAc) with anti-caries activity, and to optimization of production condition. A typical strain which produced the NCS was identified alkalophilic Bacillus sp. S-1013 through the results of morphological, biochemical and chemotaxonomic characteristics and 16S rDNA sequencing. The optimal medium composition for the maximal production of the NCS from alkalophilic Bacillus sp. S-1013 was as follow: soluble starch 30 g, dextrin 15 g, yeast extract 5 g, peptone 10 g, $K_{2}HPO_4$ 2 g in a liter of distilled water. Optimal temperature and pH were 25 and 11.0, respectively. The highest production of NCS was shown 60 hrs cultivation using the optimal medium, and then NCS productivity and dry cell weight of culture broth increased 4.24 and 2.67 time than initial medium, respectively.

Optimal Culture Conditions on the Keratinase Production by Bacillus sp. SH-517. (Bacillus sp. SH-517에 의한 keratinase의 생성 최적 배양 조건)

  • A strain SH-517 which produce extracellular keratinase, was isolated from the soil of a poultry waste and a poultry factory. An isolate SH-517 was identified as Bacillus sp. based on its morphological and biochemical characteristics. The optimal culture conditions for the production of keratinase by Bacillus sp. SH-517 were investigated. The optimal medium composition for keratinase production was determined to be 2.0% chicken feather as carbon source, 0.5% beef extract as organic nitrogen source, 0.5% $KNO_3$ as inorganic nitrogen source and 0.06% KCl, 0.05% NaCl, 0.04% $KH_2PO_4$, 0.03% $K_2HPO_4$ as mineral source and 0.01% yeast extract as growth factor. The optimal temperature and pH of medium were shown $40^{\circ}C$ and 8.5 with shaking culture (180 rpm/min), respectively. The maximum keratinase production reached maximum of 125 units/ml/min after 42 hr of cultivation under the optimal culturing conditions.

Microbiological Studies and Biochemical Changes in Fermenting Soybean Curd Residue during Fermentation (비지의 발효과정중 발효미생물 및 성분변화)

  • This study was attempted to identify microorganisms in fermenting soybean curd residues (SCR). The changes in contents of free amino acids, nucleosides, reducing sugars, and oligosaccharides were also studied. The fermentation of SCR which was by change inoculation was carried out at $55^{\circ}C$ for 48 hrs. pH increased gradually during fermentation and isolated microorganisms were identified as Bacillus subtilis and Bacillus licheniformis. Moisture content decreased from 80.8% to 58.4% at 48 hrs of fermentation and other proximate composition did not show any changes during fermentation. The content of total free amino acids increased rapidly and the number and quantities of each amino acid analyzed also increased during the course of fermentation. Glutamic acid, phenylalaine, lysine and aspartic acid were rapidly liberated during fermentation. As to the changes of nucleotides, 5'-AMP little changed during the first 36 hrs but subsequently decreased to approximately 1/6 after 48 hrs of fermentation. On the other hand, 5'-IMP plus 5'-GMP did not show almost any change during the first 36 hrs but increased about 3.5 times at 48 hrs of fermentation. However, 5'-XMP was not detected. The reducing sugar level showed rapid and steady increase throughout the fermentation and that of stachyose plus raffinose decreased slightly. From these results, a possible way of utilization of fermented SCR was proposed as a substitute for soybean in meju preparation.

Development of a new lactic acid bacterial inoculant for fresh rice straw silage

  • Objective: Effects of newly isolated Lactobacillus plantarum on the fermentation and chemical composition of fresh rice straw silage was evaluated in this study. Methods: Lactic acid bacteria (LAB) from good crop silage were screened by growing them in MRS broth and a minimal medium with low carbohydrate content. Selected LAB (LAB 1821) were Gram-positive, rods, catalase negative, and were identified to be Lactobacillus plantarum based on their biochemical characteristics and a 16S rRNA analysis. Fresh rice straw was ensiled with two isolated LAB (1821 and 1841), two commercial inoculants (HM/F and P1132) and no additive as a control. Results: After 2 months of storage at ambient temperature, rice straw silages treated with additives were well-preserved, the pH values and butyric and acetic acid contents were lower, and the lactic acid content and lactic/acetic acid ratio were higher than those in the control (p<0.05). Acidity (pH) was lowest, and lactic acid highest, in 1821-treated silage (p<0.05). The $NH_3-N$ content decreased significantly in inoculant-treated silage (p<0.05) and the $NH_3-N$ content in 1821-treated silage was lowest among the treatments. The dry matter (DM) content of the control silage was lower than that of fresh rice straw (p<0.05), while that of the 1841- and p1174-inoculant-treated silages was significantly higher than that of HM/F-treated silage. Microbial additives did not have any significant (p>0.05) effect on acid detergent fiber or neutral detergent fiber contents. Crude protein (CP) content and in vitro DM digestibility (IVDMD) increased after inoculation of LAB 1821 (p<0.05). Conclusion: LAB 1821 increased the CP, IVDMD, lactic acid content and ratio of lactic acid to acetic acid in rice straw silage and decreased the pH, acetic acid, $NH_3-N$, and butyric acid contents. Therefore, adding LAB 1821 improved the fermentation quality and feed value of rice straw silage.
