Growth Characteristics and Variation in Amino Acids Composition of Quality Protein Maize Lines (고라이신(QPM) 주요 계통과 교잡계의 생육특성 및 아미노산 조성 변이)

  • Maize grain quality can be improved by raising lysine content, which is an essential amino acid present in insufficient quantities in normal maize. Maize varieties with such modifications are known as quality protein maize (QPM). To date, no Korean maize cultivars contain high amounts of lysine. To introduce quality protein maize to Korean cultivars, we crossed QPM CIMMYT maize lines (CML) with KS140, an elite inbred line used as a parent of several cultivars such as 'Gangdaok' and 'Pyeonganok'. We analyzed the phenotypic characteristics of F1 plants as well as the protein contents, amino acids, and fatty acids profiles of the self-pollinated seeds of the F1 hybrids, and evaluated the feasibility of CML as a source of QPM. Days to anthesis of the CML ranged from 78 to 90 days after planting (DAP), whereas a range of 81~87 was recorded for F1 hybrids. The average days to anthesis was 85 for KS140, CML, and the F1 hybrids. The protein content of the CML was measured to be between 9.1 and 12.1%, with the highest and lowest values being recorded in CML153 and 191, respectively. The F1 hybrids had protein contents of 9.1~11.1%, and the highest content was observed in KS141/CML188. The fatty acids profiles were very similar across all analyzed maize samples, and linoleic acid (C18:2) composed the greatest proportion. Glutamic acid made up the largest proportion of amino acids in all maize samples. Lysine composition was highest in CML155 (6.92% of all amino acids), with an average composition of 4.83% across the CML. In contrast, KS140 showed a lysine content of 2.51%. In F1 hybrids, the average lysine composition was 3.46%, and KS140/CML164 (4.18%) and KS140/CML163 (4.99%) contained more lysine than either parent. Taken together, these results indicate that CML could become promising QPM sources to improve grain quality in Korean maize cultivars.

Reconsideration of the taxonomic characteristics of Callicarpa japonica Thunb. and C. dichotoma (Lour.) K. Koch (Verbenaceae) in Korea (한국산 작살나무와 좀작살나무(마편초과)의 분류학적 형질 재검토)

  • To clarify some ambiguous diagnostic characters of Callicarpa japonica Thunb. and Callicarpa dichotoma (Lour.) K. Koch in Korea (Verbenaceae), the external morphology and micromorphology (leaf, anther, pollen, fruit, in particular endocarp surface and structure) of two taxa are studied and described in detail. It is confirmed that the following characteristics (e.g., the stem outline in a cross-section, the bud shape and length, the corolla tube length and the lobe length, the pattern of the anther dehiscence and length, and the endocarp morphology) are useful for distinguishing these two taxa. In particular, following three characteristics are most useful for an identification: (1) the corolla tube length of C. japonica (2.5-4.6 mm) is longer than that of C. dichotoma (0.7-1.0 mm); (2) the anther is dehiscing by an apical pore at the anthesis of C. japonica, while it is opening by a long fissure from the apex to the base in C. dichotoma; and (3) the edge of the endocarp (lateral view) in C. japonica is concave, while in C. dichotoma it is shown to be flat. On the other hand, the teeth state on the leaf margin and the position of inflorescence are not good diagnostic characteristics for identification. Additionally, the keys for all known Callicarpa taxa in Korea are provided.

Studies on the Duration and Rate of Grain Filling in Rice (Oryza sativa L.) II. Difference between the Parts of a Panicle (수도의 등숙기간 및 등숙속도 연구 II. 이삭의 부위별 차이)

  • Varietal differences of grain filling duration and rate are generally recognized in rice (Oryza sativa L.). But the differences of grain filling characteristics between the parts within a panicle might be present since it takes 6-8 days to finish anthesis in a panicle. To elucidate this, 6 Japonica and 6 Japonica/Indica varieties were grown under three nitrogen levels at the Experimental Farm of Chungbuk National University in 1986, and grain filling characteristics of top, middle and bottom parts of a panicle were analyzed. Rice grain filling duration could be divided into 3 phases, i.e., Lag phase(LP) of 5 days after heading. Linear increasing period (LIP) of 5-20 days after heading and Late filling period (LFP) thereafter. Grain weight difference between the panicle parts was greater during LIP in all the varieties and was decreased during LFP. The difference was greater in Japonica/Indica varieties. Samgangbyeo and Seomjinbyeo had the shortest and the longest grain filling durations with 22.6 and 37.1 days, respectively. In average, grain filling duration of the bottom part of a panicle was 1-2 days shorter than that of top or middle part. The differences were negligible in Daecheongbyeo, Taebaegbyeo and Milyang 23, while it was greater in Tongil with 4.9 days. Grain filling rates were ranged from 0.612 to 1.097 mgㆍgrain$^{-1}$ㆍday$^{-1}$ㆍMilyang 23 showed the lowest rate with 0.612-0.682 mgㆍgrain$^{-1}$ㆍday$^{-1}$, while the rates of Baegyangbyeo, Yongmoonbyeo, Samgangbyeo, Nongbaeg and Daeseongbyeo were about 1 mgㆍgrain$^{-1}$ㆍday$^{-1}$ㆍGrain filling rate of bottom part of a panicle was lower than that of top or middle part. The difference was samll in Nongbaeg and Daeseongbyeo, while it was the greatest in Tongil with 0.222 mgㆍgrain$^{-1}$ㆍday$^{-1}$. Grain filling duration and rate were not afftected by nitrogen level, but varietal and spatial differences of grain filling characteristics were highly significant. However, the spatial difference of grain filling rates within a variety was not significant. Grain filling characteristics of each part of a panicle were significantly correlated (r=0.7207-0.9879) with those of a whole panicle.

Fruit Growth, Sugar, and Acid Characteristic in Leafy and Leafless Fruits of Satsuma Mandarin (온주밀감에서 유엽과와 직과의 생장과 당산 특성)

  • The study was conducted to investigate the effect of fruit bearing type with leafy (LY) and leafless (LS) fruits on fruit growth, sugar, and acid characteristics in satsuma mandarin. Fruit growth including fruit length, diameter, shape index (diameter/length), weight, and size distribution was not different between LY and LS fruits at ripening time. Total soluble solids (TSS) concentration of the fruit juice increased and acidity decreased continuously and then TSS:acidity ratio increased with fruit development from 100 days after anthesis to ripening time. Soluble sugar was continuously increased, whereas organic acid decreased. Theses tendencies were related to the increase of sucrose and decline of citric acid, respectively. However, there was no effect of fruit bearing type on TSS, acidity, and TSS:acidity ratio, and soluble sugar and organic acid composition in fruit juice. Also, photosynthetic rate and chlorophyll index with SPAD value were not different between leaves adjacent to LY and LS fruits. The results indicated that fruit growth and sugar and acid characteristics were not affected by fruit bearing type with leafy and leafless fruits in satsuma mandarin.

Development of Female Gametophyte of Six Species of Corydalis (Fumariaceae) (현호색속(현호색과) 6종의 자성배우자체 발달과정)

  • The development of the female gametophyte of Corydalis albipetala, C. ambigua, C. filistipes, C. nobilis, C. solida, C. ophiocarpa have been comparatively investigated using laser scanning confocal microscope (LSCM) and light microscope. An archesporium was originated from one of the outmost parietal cells beneath the one-layered epidermis of protuberant nucellus, and acted directly as a megaspore mother cell (MMC). These species had linear tetrads after successional meiotic division during the megasprogenesis. A functional megasprore developed from one of the tetrad in the chalazal end, and the rest three being degenerated. The developmental type of the female gametophyte was monosporic in accordance with the Polygonum type. Prior to anthesis the female gametophyte was organized. So mature embryo sac was comprised a three-celled egg apparatus, three large antipodals were developed from the apex of each antipodal cell, and extended toward micropylar end to be contacted with egg apparatus. Two synergids were usually observed as degenerated condition, and in this time the apices of antipodal haustoria were connected with the degenerated synergids. The developmental characteristics of seven-nucleate female gametophytes were common in all the species investigated. But the shape of mature embryo sac was ovoidal in C. albipetala, C. filistipes, C. ophiocarpa and C. solida, reflexed in C. ambigua, and rather flattened ovoidal in C. nobilis. Also, the type of megasporangium was anatropous in all the species except C. ambigua with campylotropous ovule.

Effects of Artificial Shading on Flowering and Growth of Maesa japonica Seedlings (차광 처리가 빌레나무(Maesa japonica)의 개화 및 생장에 미치는 영향)

  • Maesa japonica (Thunb.) Moritzi & Zoll. is a rare evergreen shrub that occurs in west Gotjawal, Jeju island, Korea. This study was conducted to investigate effects of an artificial shading on flowering and growth characteristics of M. japonica seedlings. The level of shading had been set to be 35%, 55% and 75% using shading net. The stage of flowering and flowering ratio, seedling height, the number of leaves and stems, leaf area were measured. Flowering timings were delayed by shading. The number of flowering seedlings per plot and inflorescences per seedling were the most in 55% shading level. Tendency of decreased seedling height with increased shading level was shown. The number of leaves and stems were the fewest values in 75% shading level. The increased biomass with decreased shading level was statistically significant. Aboveground and underground biomass were 2.1 and 1.7 times higher in shading 35% than in shading 75%, respectively. Meanwhile, all seedlings in non-shading plots were dead in winter. Such might be speculated as results of the light stress. As a result, we conclude that M. japonica vitally demands the shading for growth and the optimal shading levels for growth and flowering are 35% and 55%, respectively.

Analysis of Morphological Characteristics Among Super Sweet Corn Inbred Lines (초당옥수수 자식계통들에 대한 형태적 특성 연구)

  • We evaluated the morphological characteristics in 100 super sweet corn inbred lines, which were developed to breeding super sweet corn variety at Gyeongsangbuk-do Agricultural Research and Extension Services, by examining six quantitative and two qualitative characteristics. On the result of evaluation of two qualitative traits, most of inbred lines showed yellow (91 inbred lines) at seed color (QL1) and weak (68 inbred lines) at seedling vigor (QL2). In the survey of six quantitative traits, the average value for each trait indicated as follows: days of tasseling (QN1, 41.0 to 55.0 days), days of silking (QN2, 44.0 to 59.0 days), anthesis-silking interval (QN3, 2.0 to 7.0 days), tillering (QN4, 0.0 to 2.0), plant height (QN5, 96.0 to 187.0 cm) and ear height (QN6, 30.0 to 86.0 cm). In PCAs (principal component analysis) for 8 morphological characteristics, seedling vigor (QL2) and tillering (QN4) greatly contributed in negative direction and the days of tasseling (QN1) and days of silking (QN2) greatly contributed in positive direction on the first principal component. While, ear height (QN6) and plant height (QN5) contributed in positive direction on the second principal component. Thus these morphological traits, which were greatly contributed in the first and second principal components, might be considered to be useful for discrimination in 100 super sweet corn inbred lines. In our study, the results of morphological variation and PCAs for 100 super sweet corn inbred lines will be helpful for super sweet corn breeding programs such activities as planning crosses for hybrid and line development.

Taxonomy of the infraspecific taxa of Adonis amurensis Regel et Radde (Ranunculaceae) (복수초(미나리아재비과)의 종내분류군에 대한 분류)

  • The infraspecific taxa of Adonis amurensis Regel et Radde are currently known to include one subspecies (subsp. nanus Y. Lee), two varieties (var. ramosa Makino, and var. dissectipetalis Y. Lee) and two forms (for. argentatus Y. Lee, and for. viridescensicalyx Y. Lee). Among them, the remaining three taxa except for subsp. nanus and var. dissectipetalis are unified with A. amurensis. Therefore, we examined the morphological characteristics and geographical distribution of A. amurensis in effort to elucidate the taxonomic status of subsp. nanus and var. dissectipetalis. After the subsp. nanus samples had been transplanted from their type locality, i.e. highland at more than 1,000 m above sea level, to a lowland habitat, we measured the samples' morphological characteristics. The range of values we recorded of the plant height and floral diameter from the subsp. nanus samples were within the parameters used to characterize A. amurensis. Accordingly, subsp. nanus has been characterized as an ecophene of A. amurensis. It is considered as a synonym of A. amurensis. Among the populations of A. amurensis (sensu Y.Lee, author), those with highly dissected petals at their apices have previously been described as var. dissectipetalis. However, this characteristic has been observed together with obtuse or rounded petal apices not only in one population but also in a single individual. However, populations with these characteristics belong to A. pseudoamurensis due to the simultaneous development of leaves and flowers, branched stems, acute leaf apices, and petals that are shorter than sepals. Therefore, we suggest that A. amurensis var. dissectipetalis be unified with A. pseudoamurensis. Very useful characteristics for distinguishing A. amurensis from its relative species are anthesis, the presence or absence of branches, the development of scales into normal leaves, the relative length of the sepals and petals, and the position of the stipules. A. amurensis is geographically restricted to E. Asia, including E. Siberia, and especially on the Korean Peninsula, it is distributed throughout the northern provinces of Gyeonggi province and in Gangwon province, Gyeongbuk province (Mt. Bohyeon-san) and Jeonbuk province (Mt Jeoksang-san) along Baekdudaegan mountain.

Effects of Sowing Times and Spacing on Growth and Yield of Coix lachryma-jobi L. var. ma-yuen STAPF (율무의 파종기(播種期)와 재식밀도(栽植密度)에 따른 생육(生育) 및 수량(收量))

  • This study was conducted to evaluate the growth and yield by sowing times and spacing using machine seeder on Coix Lachryma-jobi L. var ma-yuen $S_{TAPF}$. Adlay was seeded at four different dates (April 20, May 5, May 20 and June 5). Planting spaces were controlled by the seed roller of tractor drill seeder attached to tractor at three spacing ($60{\times}15cm,\;70{\times}15cm\;and\;80{\times}15cm$). The results were summarized as follows. Required days to emergence and days to anthesis were shortened as sowing date was late. But days to maturity were prolonged when sowing date was late. The accumulated temperature increased such as required periods increased. In growth characteristics, culm length was significantly different at different sowing times. But all growth characteristics was not affected by different spacings. In yield components, 1,000 grain weight and ripening rate were significantly different at different sowing times, also the number of tillers and branches was significantly affected at different spacings. Grain yield/ha was significantly different at different sowing times. But it was not significantly different at different spacings. Thus, in order to improve the yield of adlay using drill seeder, if it is not frost, sowing should be done as early as possible. As a result, suitable sowing time was April 20 with spacing at $60{\times}15cm$ using machine seeder in Korea.

Study on the Establishment of Wildflower Pastures (야생화초지의 조성에 관한 연구)

  • The purpose of this study was to suggest the basic data for establishment of wildflower pastures. The experimental design includes two treatments: Native wildflower pasture (NWP, Kentucky bluegrass + native wildflower 8 species) and Introduced wildflower pasture (IWP, Kentucky bluegass + introduced wildflower 24 species). The field trials were carried out on the experimental pastures plots at Chungnam National University throughout from Sep. 2006 to Dec. 2007. The results obtained are as fellows: In terms of the flower color in the wildflower pastures, the IWP was found more various than the NWP. But such problems as seasonal distribution and continuity were raised and moreover the time of flowering depended mainly upon the spring season. Even though the flower color in the NWP did not show up its variousness, seasonal distribution and continuity were found comparatively satisfactory. To perennially maintain the botanical composition, the variousness of wildflower species are found important after examining locally the beginning rearing and flowering characteristics of wildflowers as well as the basic turfgrass. Moreover, when the wildflower pastures was constructed in autumn, since most of the introduced wildflower were annuals, it maintained various botanical composition before wintering, but after wintering the botanical composition were found simplified due to the winter tolerance. The NWP did not show any problems in the winter tolerance but it could not make the botanical composition diversified. Accordingly, to maintain the wildflower pasture for many years, diversifying wildflower species based upon perennials including annuals seems to be good.
