Addition of plant growth hormones [0.01 mg/L NAA and 0.2mg/L benzyladenine (BA)] to a woody plant medium stimulated the adventitious bud formation of poplar explants during culture. Endogenous IAA content increased rapidly at the initial culture stage and then decreased, being followed by rapid increment again at the late culture. But the content of trans-zeatin riboside (t-ZR) increased continuously during the culture. Cytoplasmic soluble proteins were analyzed by one- and two-dimensional SDS-PAGE. Increased amount of 40 kD band was detected by one-dimensional electrophoresis using Coomassie Blue staining during the culture and two distinctive proteins whose mol wt is 40,000 were detected by two-dimensional electrophoresis using autoradiography and these proteins were synthesized continuously prior to the adventitious bud formation. When the midvein segments were transferred to the actinomycin D-containing medium, the spots of adventitious bud-related proteins(ABRPs) did not disappeared but weakened in intensity. So, it is concluded that genes coding for the ABRPs are regulated to some degree at the transcriptional level. Also, they were not observed in BA-free medium, suggesting that these proteins be regulated by cytokinin, which made then possible to form the adventitious bud.
This study was investigated to establish a regeneration system of plant via adventitious bud formation from leaf explant cultures of strawberry (Fragaria ananassa Duch). Effects of plant growth regulators (2,4-D, BAP), agar sucrose and myo-inositol on adventitious bud formation were investigated. When the leaf explants were cultured on MS medium supplemented with 0.1 mg/L 2,4-D and 3 mg/L BAP, the adventitious bud formation was most promoted. The adventitious bud formation was not induced from leaf explants cultured on MS medium containing 2,4-D alone. Adventitious bud formation was enhanced to almost 3 times on medium with low level of agar concentration (0.4%) in comparison with those on the medium with high level of agar (1%), but almost of shoot was vitrificated on the medium. Therefore, the normal adventitious bud formation from leaf explants was most effective on the medium containing 0.05 mg/L 2,4-D,1 mg/L BAP, 0.8% agar, 30 g/L sucrose and 100 mg/L myo-inositol. Thus, the mass propagation of healthy strawberry could be established using leaf explants.
To establish a regeneration system of mulberry through the adventitious bud formation of winter bud leaflet, medium conditions were examined. Adventitious bud formation rate of mulberry winter bud leaflet on the medium containing different concentrations of BA and TDZ as the growth regulators was high on 5mM BA and 0.5mM TDZ. The effect of preculture on the medium containing BA or TDZ was affected not only by the concentration of growth regulators but the duration of preculture. Fructose as carbon source was effective on adventitious bud formation.
To improve new propagation method, I try to propagate wild polygonatum odoratum roots which formate adventitious bud without bud. The obtained results of size of roots and adventitious bud to formate for multiple propagation of wild polygonatum odoratum as the follow. One year grown wild polygonatum odoratum root in 5cm diameter formate 66.5% adventitious buds. One year grown wild polygonatum odoratum root in 15cm in diameter germinate 113% adventitious bud. Two years grown root in 15cm diameter formate adventitious bud three times more then one year root. Wild polygonatum odoratum in 15cm diameter without any bud in experiment plot makes 4 times of propagation. Sand and Perlite media propagaton plot was increased 3.1∼3.3times more than common culture medium. The best result of mass propagation wild polygonatum odoratum is 10mm diameter root and 5cm cut in length to plant sand media plot.
Cotyledon explants of Korean ginseng (Panax ginseng C.A. Meyer), a perennial medicinal plant, produced direct somatic embryos at a high frequency on MS medium without growth regulators. Cytokinin highly suppressed the somatic embryogenesis but stimulated direct fomlation of adventitious buds. BAP was more effective than kinetin for the formation of adventitious bud. IBA combination with cytokinin enhanced the frequency of adventitious bud formation. The highest frequency of adventitious bud formation were $40\%$ at 0.05 mg/l IBA and 5 mg/l BAP. Adventitious buds were mainly formed near the distal portion of cotyledon, while somatic embryos were only formed near the proximal portion of cotyledon. Adventitious buds were covered with sheath similar to axillary buds in the zygotic embryos, and then leaf-like epicotyls were developed.
This study was conducted to determine the effect of growth regulators, IBA, GA, and BA, on the adventitious bud formation, shoot differentiation, and inflorescence development in embryo culture of Korean ginseng. The adventitious bud formation and shoot differentiation were significantly promoted by application of a combination of 1 ma/l IBA and 5 mg/l GA. The adventitious buds had the primordial shoots and were differentiated as to plantlets. About 5 to 10 adventitious buds developed around the basal axis of the epicotyle of the ginseng embryo, and development of inflorescence was possible only after shoot differentiation. The MS medium supplemented with a combination of 3 mal 1 each of IBA, GA, and BA was most effective for in vitro inflorescence development, and the ratio of inflorescence formation was 18.4%.
We have investigated several factors influencing adventitious shoot induction from the Populus deltodies bartr. leaf segments. To obtain in vitro materials, at first, stem segments from ex vivro were tested for axillary bud breaking on the five macronutrients levels of two differenr media. WPM was better both in bud breaking and leaf expansion than MS medium. The amount of NH$_{4}$NO$_{3}$ added in the medium did not seem to affect axillary bud breaking, significantly and subsequent shoot elongation from the stem segments of P. deltodies. However, other components in WPM might have played important roles in axillary bud breaking and shoot elongation. Regenerability from three sections (the distal, the middle and the base) of leaves cultured on WPM supplemented with TDZ and NAA combinations appeared to be different; middle and basal sections of leaves produced more organogenic sites than those of top section on WPM and those sesctions produced the highest ogranogenic sites on the same medium supplemented with 0.01 mg/l TDZ and 0.02mg.l NAA. Among 3 carbon sources tested for adventitious shoot elongation, fructose seemed to be stimulating the elongation of adventitious shoots. Sucrose and glucose added in the medium resulted in the necrosis which caused dying of adventitious shoots, eventually.
This study was conducted to examine the effect of BA and NAA on adventitious bud induction from in vitro germinants E. pellita. The capacity of adventitious bud formation greatly depends on juvenility and explants origin; the more juvenile materials are the better ability to form adventitious buds even in in vitro raised plantlets. In case of in vitro germinants, 7 day old plantlets showed a better morphological response than did 14 day old ones in the induction of adventitious buds. The capacity to show morphological response was in decreasing order : cotyledons> petioles> roots. Ho adventitious buds formed when root segments were used as culture material. And optimum medium appeared to be MS + 0.5 mg/L BA and 0.2 mg/L NAA. Adventitious buds could be developed into multiple shoots and regenerated normal plantlets on DKW medium plus 0.2 mg/L BA and 0.01 mg/L NAA.

Adventitious buds were obtained from isolated cotyledons cultured on MS medium with various concentrations of 6-benzylamino purine (BA) and thidiazuron (TDZ). The highest numbers of adventitious buds were obtained on MS medium supplemented with 0.2 mg/L BA. Experimental culturing with half the petiole portion and half with the terminal segments were grown on MS medium contained with 0.2 mg/L BA. Frequency of the adventitious bud induction was variable accordingly to the type of cultured explants. Explants with the half petiole showed the highest adventitious bud induction rate (80%) compared to explants of half with terminal segment (20%). An elongated shoot from the buds and growth of advent roots were both possible on the 1/2 MS medium without a plant growth regulator. These results offer an effective way in which clonal propagation can be accomplished.
