The differentiation of skeletal muscle precursor cells in culture is marked by the transcriptional activation of muscle-specific genes and the morphological differentiation of myoblast into multinucleate myotube. In this study, we examined the effect of TSA (Trichostatin A) on WF-kB DNA binding activity and muscle cell fusion in the process of myogenesis. Under TSA treatment, C2C12 myoblast could not fuse to myotube and its NF-kB DNA binding activity was also blocked. To investigate whether these phenomenons were affected by TSA in direct or not, differentiation media (DM) used to differentiate cells without TSA was concentrated and added to C2C12 myoblast with TSA simultaneously. C2C12 myoblast was fused to myotube and NF-kB DNA binding activity was recovered. These results suggest that TSA affects on the differentiation of myoblast, perhaps through several factors, by inhibiting myoblst fusion and blocking NF-kB DNA binding activity.

In this way, data collection and analysis becomes easy. In such data, personal information including an identifier such as a sensor id, a device number, IP address, or the like may be collected. However, if systematic management is not accompanied by collecting and disposing of large-scale data, violation of relevant laws such as "Personal Data Protection Act". Furthermore, data quality problems can also occur and make incorrect decisions. In this paper, we propose a new data governance maturity model(DGMM) that can identify the personal data contained in the data collected by companies, use it appropriately for the business, protect it, and secure quality. This study is a qualitative study conducted using focus group interviews to examine the mixed offline and online education experience of nursing students in the context of Covid-19. The subjects of this study included 7 students enrolled in the Department of Nursing at J City V University, and focus group interviews were conducted with sufficient explanation and written consent for the study. The contents of the interviews were recorded, and the interview contents were directly transcribed directly after the interview. Research results were derived through content analysis. As a result of the study, 5 domains, 10 categories, and 24 subcategories were derived from the experiences of nursing college students on mixed lectures. The 5 domains included 'mixed lectures,' 'tasks,' 'tests,' 'motivation,' and 'improvement.' The contents of each domain derived from this research result are expected to be used as basic data in the design of the on/offline mixed lectures in the future.

The p53 gene plays a critical role in the transcriptional regulation of cellular response to stress, DNA damage, hypoxia, and tumor development. Keeping in mind the recently discovered manifold physiological functions of p53, its involvement in the regulation of cancer is not surprising. In about 50% of all human cancers, inactivation of p53's protein function occurs either through mutations in the gene itself or defects in the mechanisms that activate it. This disorder plays a crucial role in tumor evolution by allowing the evasion of a p53-dependent response. Many recent studies have focused on directly targeting p53 mutants by identifying selective, small molecular compounds to deplete them or to restore their tumor-suppressive function. These small molecules should effectively regulate various interactions while maintaining good drug-like properties. Among them, the discovery of the key p53-negative regulator, MDM2, has led to the design of new small molecule inhibitors that block the interaction between p53 and MDM2. Some of these small molecule compounds have now moved from proof-of-concept studies into clinical trials, with prospects for further, more personalized anti-carcinogenic medicines. Here, we review the structural and functional consequences of wild type and mutant p53 as well as the development of therapeutic agents that directly target this gene, and compounds that inhibit the interaction between it and MDM2.
To understand seasonal changes it is necessary to understand the relationship between celestial bodies in a three-dimensional space, and to this end, modeling activities in which students directly construct, use, evaluate, and modify three-dimensional models are important. In this study, the process of elementary school students using globes and light bulbs to model Earth's motion in a three-dimensional space as a cause of seasonal changes was analyzed. Seventeen sixth graders participated in the modeling process. After exploring phenomena and concepts related to seasonal change, students constructed models using globes and bulbs and used them to explain seasonal changes. Video data recording students' modeling process, students' activity sheets, and transcripts of post-interview were used as research data, and data triangulation was conducted. The modeling level analysis framework was also developed based on previous studies. In particular, the framework was developed in detail in this study in consideration of the concept of Earth's motion as well as understanding model and implementing modeling. In the final analysis framework, the 3D modeling level was classified from level 1 to level 3, and student performance that may appear at each level was specified. As a result of the study, there were two main levels of modeling using globes for elementary school students to explain seasonal changes. The rotation and tilt of the axis of rotation and revolution of the earth were considered but the level at which empirical evidence was not used (level 2), the level at which empirical evidence was used to explain seasonal chages (level 3). However, even when students use empirical evidence, it did not lead to the construction of a scientific model. In this study, the cause was explored in relation to the characteristics of the tool used for modeling.
This study was carried out to elucidate the relation between expression levels of three melanin synthesis genes (Tyrosinase, Tyrosinase-related protein 1 and Dopachrome tautomerase) according to the Melanocortin-1 receptor genotypes with coat color patterns in Hanwoo cattle, Jeju black cattle and Holsteins. Using real-time semiquantitative reverse transcription-PCR assay (RT-PCR), the expression levels of these three genes were analyzed in skin tissues from four representative coat colored areas: yellowish-brown from MC1R e/e Hanwoo, wild type black from $E^+/E^+$ Jeju black cattle (JBC), and dominant black and white pied regions from $E^D/E^D$ Holstein. The TYR, TYRP1 and DCT genes showed higher expression levels of 4.5, 2.3 and 2.5 times higher in the black skin area of Holsteins than those of from JBC, respectively (p<0.001). In addition, the expression levels of these three genes from JBC were significantly higher than those from Hanwoo cattle (p<0.001). These results show that coat color phenotypes in Hanwoo cattle, JBC and Holsteins is directly correlated with TRY, TYRP1 and DCT transcription levels, which probably reflected involvement with MC1R genotypes; e/e in Hanwoo, $E^+/E^+$ in JBC and $E^D/E^D$ in Holsteins. Consequently, this study suggested that the status of MC1R protein may not only induce the transcription activities of a series of TYR and its related genes responsible for melanin synthesis, but also determine the levels of total melanin contents in bovine skin.

Background : IL-8 is a potent chemotactic cytokine that plays an important role in the host defense mechanism against M. tuberculosis by recruiting inflammatory cells to the site of the infection. Lung epithelial cells, as well as alveolar macrophages are known to produce IL-8 in response to M. tuberculosis. IL-8 gene expression is mainly regulated on the level of transcription by NF-${\kappa}B$. This study investigated whether or not A549 cells produce IL-8 in NF-${\kappa}B$ dependent mechanism in response to macrophages phagocytosing M. tuberculosis. Methods : Peripheral blood monocytes that were obtained from healthy donors were cultured for 24 h with M. tuberculosis and a conditioned medium(CoMTB) was obtained. As a negative control, the conditioned medium without M. tuberculosis (CoMCont) was used. A549 cells were stimulated with M. tuberculosis, CoMCont and CoMTB and the IL-8 concentration in the culture media was measured by ELISA. The CoMTB induced IL-8 mRNA expression in the A549 cells was evaluated using RT-PCR, and CoMTB induced $I{\kappa}B{\alpha}$ degradation was measured using western blot analysis. CoMTB induced nuclear translocation and DNA binding of NF-${\kappa}B$ was also examined using an electrophoretic mobility shift assay(EMSA), and the CoMTB induced NF-${\kappa}B$ dependent IL-8 transcriptional activity was measured using a luciferase reporter gene assay. Results : CoMTB induced IL-8 production by A549 cells($46.8{\pm}4.8\;ng/ml$) was higher than with direct stimulation with M. tuberculosis ($6.8{\pm}2.9\;ng/ml$). CoMTB induced IL-8 mRNA expression increased after 2 h of stimulation and was sustained for 24 h. $I{\kappa}B{\alpha}$ was degraded after 10 min of CoMTB stimulation and reappeared by 60 min. CoMTB stimulated the nuclear translocation and DNA binding of NF-${\kappa}B$. The CoMTB induced NF-${\kappa}B$ dependent IL-8 transcriptional activity($13.6{\pm}4.3$ times control) was higher than either CoMCont($2.0{\pm}0.6$ times control) or M. tuberculosis ($1.4{\pm}0.6$ times control). Conclusion : A conditioned medium of peripheral blood monocytes phagocytosing M. tuberculosis stimulates NF-${\kappa}B$ dependent IL-8 production by the lung epithelial cells.

The reductions of troops/human resources, and improvement in combat power have made Korean Department of Defense actively adapt 4th Industrial Revolution technology (Artificial Intelligence, Big Data). The defense information system has been developed in various ways according to the task and the uniqueness of each military. In order to take full advantage of the 4th Industrial Revolution technology, it is necessary to improve the closed defense datamanagement system.However, the establishment and usage of data standards in all information systems for the utilization of defense big data and artificial intelligence has limitations due to security issues, business characteristics of each military, anddifficulty in standardizing large-scale systems. Based on the interworking requirements of each system, data sharing is limited through direct linkage through interoperability agreement between systems. In order to implement smart defense using the 4th Industrial Revolution technology, it is urgent to prepare a system that can share defense data and make good use of it. To technically support the defense, it is critical to develop Multi Repository Meta-Data Management (MRMM) that supports systematic standard management of defense data that manages enterprise standard and standard mapping for each system and promotes data interoperability through linkage between standards which obeys the Defense Interoperability Management Development Guidelines. We introduced MRMM, and implemented by using vocabulary similarity using machine learning and statistical approach. Based on MRMM, We expect to simplify the standardization integration of all military databases using artificial intelligence and bigdata. This will lead to huge reduction of defense budget while increasing combat power for implementing smart defense.

The purpose of this qualitative study was to develop a social skills training program for improving early school-age children's social behaviors and to investigate its effectiveness by observing their experiences on the program with a qualitative method. Data were collected from 7 children using observer's descriptive notes and reflective notes, compliment notes by assistant leader, program leader's weekly journals, children's weekly journals, and video recordings. Four theme categories and 11 sub-lower categories emerged. Theme categories were (1) relationship building, (2) changes in emotional expressions, (3) changes in prosociality, and (4) changes in social skills. This study observed early school-age children's positive changes in social behaviors and emotional expressions through the social skills program.
This study investigated the song-induced autobiographical memory of patients with early Alzheimer's Dementia (AD) by comparing the effects of patient-selected songs (M-AD) versus music therapist-selected songs (M-MT). A total of 19 patients with early AD participated in this study. In the M-AD and M-MT conditions, each participant listened to a song and was instructed to recall memory. The time to recall memory, the specificity of the memory, mood changes, and the type of recalled memory were measured. Perceived familiarity and preference of the used songs and association of the song with the recalled memory were also analyzed. The results of the study showed that the M-AD condition elicited more specific memory and positive mood change than the M-MT condition. In addition, AD patients reported a higher level of familiarity with and preference of songs in the M-AD condition, compared to the M-MT condition. These results indicate that patient-selected songs, which have a personal meaning to an individual, could be effectively used for intervening with memory of this population, which would support music therapists to make better decision with regard to song selection. Further studies would be needed to deepen the understanding of autobiographical memory in older population with cognitive impairment and to propose more effective music therapy strategies for intervening with memory.
