Plant Early Growth and Chlorophyll Fluorescence of Seven Varieties of Italian Ryegrass Under Various Salt Stress Conditions (다양한 염 스트레스 조건에서 이탈리안 라이그라스 7 품종의 초기 생장 및 엽록소 형광 반응)

  • This study evaluated the salt tolerance of Italian ryegrass varieties under various salt stress conditions. Seven varieties, including Kowineary, Greencall, Greenfarm, Greenfarm 3, IR 603, IR 605, and IR 901, were used in this experiment. Under various salt concentrations, there was no significant decrease compared to the control at salt concentrations of 0.1% to 0.3%. IR 901, Kowineary, and IR 605 had a lower relative reduction rate, but the absolute growth of IR 603 was the highest among the varieties. Among chlorophyll fluorescence parameters, ABS/RC, TRo/RC, ETo/RC, and DIo/RC values (activities expressed per reaction center) showed a significantly negative correlation with growth rates. In the experiment where the salt concentration was drastically increased, the growth decreases in Greenfarm 3 and IR 901 were relatively small compared to other varieties. Among the relative ratio of chlorophyll fluorescence parameters, ABS/RC, TRo/RC, ETo/RC (after 1 hour of salt concentration increase), and PIABS (3 hours, 7 days) showed a significant positive correlation, and DIo/RC (3 hours, 1 day) showed a negative correlation with plant growth. In the experiment where the salt concentration was drastically decreased, the growth increases in Greenfarm, IR 901, and Kowineary were relatively high compared to other varieties. Among the relative ratio of chlorophyll fluorescence parameters, ABS/RC, ETo/RC, DIo/RC (5 hours), and PIABS (1 hour) showed a significant positive correlation with plant growth. In summary, among the seven varieties, Kowineary and IR 901 had superior relative growth (compared to the control) under various salt stress conditions. IR 603 showed a greater decrease in relative growth but had the highest absolute growth amount under salt stress conditions. In addition, some parameters related to chlorophyll fluorescence showed a significant correlation with early growth, which could be used as indicators of the initial growth of IRG under salt stress.

Evaluation on Growth Characteristics of Red Pepper (Capsicum annuum L.) and Soil Chemical Properties by Continuous Application of Food Waste Compost with Manure (음식물류폐기물 혼합 가축분 퇴비 연용에 따른 고추(Capsicum annuum L.) 생육 및 토양 화학적 특성 평가)

  • Food waste compost with high salt content produced by aerobic digestion, but concerns about application of cropland. To address this issue, food waste is being composted by mixing it with livestock manure, which has a lower salt content. Therefore, this study aimed to evaluate the growth characteristics of red pepper and soil chemical properties for continuous application with different amounts of food waste compost with manure (FWC). Treatments were consisted of no fertilizer (NF), inorganic fertilizer (N-N-P2O5-K2O, 19.0-11.2-14.9 kg 10a-1), and inorganic fertilizer + food waste compost with manure (NPKFWC). FWC treatment was applied at three treatment rates based on soil organic matter content: 900 kg 10a-1, 1,800 kg 10a-1, and 2,700 kg 10a-1 ( referred to a s FWC 1, 2, 3 , respectively). As a r esult of the red pepper yield was about 1.8 times higher in NPKFWC 1 and NPKFWC 2 than that in the NF, but decreased in the NPKFWC 3, 300% of the recommended FWC application rate. Yield decreased in all FWC treatments with continuous application for three year and also decreased about 40% from 3,265 kg 10a-1 in the first year to 1,948 kg 10a-1 in the third year. For the soil chemical properties, the content of soil organic matter, available P2O5, and exchangeable cations increased in the FWC treatments, and were higher than the NF. Exchangeable sodium in all treatments was increased slightly compared to the soil before used, and no significant salinization was observed in the FWC. This study confirmed that excessive use of FWC not only reduced nutrient use efficiency, but also decreased the red pepper yield. Therefore, it is concluded that optimum usage of FWC is effective for agroecological impacts.

A Comprehensive Analysis of HLA-A and HLA-DR Allele Frequencies and Haplotype Associations in a Korean Population of 790 Individuals (한국인 790명을 대상으로 한 HLA-A 및 HLA-DR 대립유전자 빈도 및 일배체형 연관성에 대한 종합적 분석)

  • The human leukocyte antigen (HLA) system, which is part of the major histocompatibility complex (MHC) plays a vital role in immune responses by differentiating between itself and foreign cells and antigens. The significant diversity of alleles affects disease susceptibility and immune responses within different populations. Specifically, the HLA-A and HLA-DRB1 alleles are associated with various immune-related diseases, and understanding the frequency and haplotype associations of these alleles is vital for genetic and immunological research. To investigate the distribution of these characteristics in Koreans, we isolated peripheral blood mononuclear cells (PBMCs) from blood samples donated by volunteers at the Seoul Central Blood Bank and performed HLA typing on 790 samples. Our study found that the HLA-A and HLA-DRB1 alleles are widely distributed within the Korean population, with HLA-A*24:02 (21.7%) and HLA-DRB1*09:01 (9.9%) being the most frequent. Significant haplotype associations between specific HLA-A and HLA-DRB1 alleles were identified using the Chi-square test, suggesting that certain genetic combinations may influence disease onset. This insight could contribute to the development of predictive and preventative strategies for various diseases. The unique genetic characteristics of the Korean population highlight the importance of studying the HLA allele and the haplotype distributions in this group as key indicators for understanding disease susceptibility.

Employment Gap Between Capital and Non-Capital Regions since the Global Financial Crisis in Korea (글로벌 금융위기 이후 수도권과 비수도권 간 일자리 격차)

  • Using Regional Employment Survey, this article analyzes the widening employment gap between Capital and Non-Capital regions since the 2008 global financial crisis through describing the spatial distribution of employment by industry and occupation, calculating the regional downward employment rate of university graduates, and estimating the regional distribution of employment growth in terms of wage distribution, and then statistically analyzes the effect of the declining manufacturing employment in the Non-Capital region since the mid-2010s on total employment change. The results from these analyses are as follows. First, since the mid-2000s, the share of producer services and white-collar jobs has increased in the Capital region. Second, the Non-Capital region has a higher share of non-regular workers relative to wage workers than the Capital region. Third, while the downward employment rate has increased in the Non-Capital region since the mid-2010s, it has risen very modestly or stagnated in the Capital region. Fourth, in terms of wage distribution, the pattern of employment growth since the mid-2010s has been U-shaped, a typical polarized pattern of simultaneous growth in low- and high-wage jobs and decline in middle-wage jobs in the Non-Capital region, while in the Capital region it has been L-shaped with the sides reversed, driven by growth in high-wage jobs. Fifth, this employment polarization in the Non-Capital region since the mid-2010s is associated with employment changes in manufacturing, which accounts for a large share of middle wage jobs. Finally, according to the analysis of the effect of regional manufacturing employment changes on total employment changes since the mid-2010s, declines in manufacturing employment reduce the region's regular employment rate and have a larger negative employment effect in the Non-Capital region. Based on these findings, some policy issues are discussed including the branch plant economy regime based on spatial separation of conception and execution, the mix of geographic and skill mobility, the need to improve employability and jobs through human capital accumulation, and short- and long-term responses to employment fluctuations.

Analysis of Ingredients and DPPH, ABTS Activity for the Development of Cosmetic Raw Materials using 5 Kinds of Plants Native to Mt. Jiri.-III (지리산 자생식물 5종의 화장품 원료개발을 위한 성분 및 DPPH, ABTS 활성분석-III)

  • Five species, including Synurus plamatopinnatifidus (Makino) Kitam, Lindera obtusiloba Blume, Acorus gramineus Sol., Swertia japonica (Schult.) Griseb., and Valeriana fauriei Briq., native to Mt. Jiri, were collected from their habitats from April to October 2022. The collected plants were analyzed for 17 types of polyphenol components in the whole plant, and the antioxidant activities of DPPH and ABTS were measured according to the parts of the leaves and roots, and the contents of total polyphenol and total flavonoid were measured. Based on these results, we hope that they will be used as basic data for the development of antioxidant cosmetic raw materials targeting the five plants native to Mt. Jiri with high antioxidant efficacy. 1. The polyphenol components contained in the five species of plants native to Mt. Jiri were 12 types, including catechin and quercetin. Among them, the total polyphenol content of A. gramineus was 39.5ppm, and the variety with the lowest polyphenol content was S. japonica with 5.9ppm. 2. DPPH radical scavenging activity was confirmed by EC50 value. As a result, the variety with the highest DPPH radical scavenging activity was L. obtusiloba leaves at 35.31㎍/mL. The variety with the lowest scavenging activity was V. fauriei at 262.69㎍/mL for the leaves and 278.99㎍/mL for the roots. 3. ABTS radical scavenging activity was confirmed by EC50 value. The variety with the highest scavenging activity was L. obtusiloba roots at 55.79㎍/mL and 58.85㎍/mL for leaves. The variety with the lowest scavenging activity was S. japonica leaves at 576.5㎍/mL. 4. The variety with the highest total polyphenol content was L. obtusiloba leaves at 158.72mg/g, and the variety with the lowest content was V. fauriei leaves at 0.13mg/g. Therefore, there was a large difference in the total periphenol content by variety. 5. The variety with the highest total flavonoid content was L. obtusiloba leaves at 163.75mg GAE/g, and the other varieties showed low contents at 17.67~41.38mg GAE/g.

A Bibliographical Study of Golden Manuscript Baegcheonin-dalanigyeong, Manufactured from Chunglyeol-King & Wonseong-Princess in 1284 (1284년 충렬왕(忠烈王), 원성공주(元成公主) 발원 금자대장(金字大藏) 『백천인다라니경(百千印陁羅尼經)(합부(合部))』의 서지적 연구)

  • This study attempted to introduce and analyze Baegcheonin-dalanigyeong(百千印陀羅尼經) which were recently confirmed among the golden manuscript in the Koryo Dynasty. The summary is as follows. (1) This is a book written in golden manuscript and it was manufactured by King Chunglyeol & Jegukdaejang-Princess in the 10th year of King Chongryeol's reign (1284). (2) This golden manuscript is written down on the blue paper(紺紙) with gold line border, has a scroll-form with a crystal axis on each end of it, and belongs to the part of the Scripture Tripitaka with the sign of "景". The text were formally written down in 17 letters in a line, however few 16 and 18 letters in one line were discovered. (3) The contents are consisted of following four scriptures in order; Baeg-cheon-in-dalanigyeong(百千印陀羅尼經), Gu-myeon-yeon-agwi-dalani-sinjugyeong (救面然餓鬼陁羅尼神呪經), Bul-seol- gam-lo-dalaniju (佛說甘露陁羅尼呪) 7 line and Jang-eom-wang-dalani-jugyeong (莊嚴王陁羅尼呪經), Hyang-wang-bosal-dalani-jugyeong (香王菩薩陁羅尼呪經). (4) There are unprecedented red signature and a stamp on the reverse side of the front page. The signature belongs to the King Chungryeol and it is the second name of him. The stamp shows 'Wonseongjeon(元成殿)' which is the Princess Jegukdaejang's original temple, and it could be the answer of the controversial debates about the original palace owner. (5) On the cover, there are a figure and floral pattern which seems to be made in the 13th century and the craftsman's name is '鄭(Zheng)[益迂(ig-u)]'. The figure's expression is similar to the other gold and silver manuscript version, but the postures and object hold in a hand are different. (6) Records of publications are similar to the other golden manuscripts, but showed some differences. In addition, the person who wrote this sutra is Choi jeong(崔楨), but the person is not confirmed in the historical data. Yet, it is presumed that he were in lower classes but had outstanding writing skills to be mobilized for the Scripture work. (7) The contents of '景' and four kinds of scriptures (including gamlosudalani(甘露水陁羅尼)) are included in the '景' of the 9th book sinjibjang-gyeong-eum-uisuhamlog (新集藏經音義隨函錄) and Bangsan-seoggyeong(房山石經). It is presumed that the narrative sequence of the scriptures corresponds to the order of The Khitan Tripitaka(契丹藏). In addition, these four types of scriptures are included in the "羔" of joseong-geumjang(趙成金藏), chojo Tripitaka(初雕大藏經), and jaejo Tripitaka(再雕大藏經) but the order of the scripture is recorded in reverse. In addition, this golden manuscript is a valuable data as it holds a lot of differentiator character and various sobriquet of Buddhist literature that can be utilized frequently in researches about Buddhist literature.

Characteristics and classification of paddy soils on the Gimje-Mangyeong plains (김제만경평야(金堤萬頃平野)의 답토양특성(沓土壤特性)과 그 분류(分類)에 관(關)한 연구(硏究))

  • This study, designed to establish a classification system of paddy soils and suitability groups on productivity and management of paddy land based on soil characteristics, has been made for the paddy soils on the Gimje-Mangyeong plains. The morphological, physical and chemical properties of the 15 paddy soil series found on these plains are briefly as follows: Ten soil series (Baeggu, Bongnam, Buyong, Gimje, Gongdeog, Honam, Jeonbug, Jisan, Mangyeong and Suam) have a B horizon (cambic B), two soil series (Geugrag and Hwadong) have a Bt horizon (argillic B), and three soil series (Gwanghwal, Hwagye and Sindab) have no B or Bt horizons. Uniquely, both the Bongnam and Gongdeog series contain a muck layer in the lower part of subsoil. Four soil series (Baeggu, Gongdeog, Gwanghwal and Sindab) generally are bluish gray and dark gray, and eight soil series (Bongnam, Buyong, Gimje, Honam, Jeonbug, Jisan, Mangyeong and Suam) are either gray or grayish brown. Three soil series (Geugrag, Hwadong and Hwagye), however, are partially gleyed in the surface and subsurface, but have a yellowish brown to brown subsoil or substrata. Seven soil series (Bongnam, Buyong, Geugrag, Gimje, Gongdeog, Honam and Hwadong) are of fine clayey texture, three soil series (Baeggu, Jeonbug and Jisan) belong to fine loamy and fine silty, three soil series (Gwanghwal, Mangyeong and Suam) to coarse loamy and coarse silty, and two soil series (Hwagye and Sindab) to sandy and sandy skeletal texture classes. The carbon content of the surface soil ranges from 0.29 to 2.18 percent, mostly 1.0 to 2.0 percent. The total nitrogen content of the surface soil ranges from 0.03 to 0.25 percent, showing a tendency to decrease irregularly with depth. The C/N ratio in the surface soil ranges from 4.6 to 15.5, dominantly from 8 to 10. The C/N ratio in the subsoil and substrata, however, has a wide range from 3.0 to 20.25. The soil reaction ranges from 4.5 to 8.0. All soil series except the Gwanghwal and Mangyeong series belong to the acid reaction class. The cation exchange cpacity in the surface soil ranges from 5 to 13 milliequivalents per 100 grams of soil, and in all the subsoil and substrata except those of a sandy texture, from 10 to 20 milliequivalents per 100 grams of soil. The base saturation of the soil series except Baeggu and Gongdeog is more than 60 percent. The active iron content of the surface soil ranges from 0.45 to 1.81 ppm, easily-reduceable manganese from 15 to 148 ppm, and available silica from 36 to 366 ppm. The iron and manganese are generally accumulated in a similar position (10 to 70cm. depth), and silica occurs in the same horizon with that of iron and manganese, or in the deeper horizons in the soil profile. The properties of each soil series extending from the sea shore towards the continental plains change with distance and they are related with distance (x) as follows: y(surface soil, clay content) = $$-0.2491x^2+6.0388x-1.1251$$ y(subsoil or subsurface soil, clay content) = $$-0.31646x^2+7.84818x-2.50008$$ y(surface soil, organic carbon content) = $$-0.0089x^2+0.2192x+0.1366$$ y(subsoil or subsurface soil, pH) = $$-0.0178x^2-0.04534x+8.3531$$ Soil profile development, soil color, depositional and organic layers, soil texture and soil reaction etc. are thought to be the major items that should be considered in a paddy soil classification. It was found that most of the soils belonging to the moderately well, somewhat poorly and poorly drained fine and medium textured soils and moderately deep fine textured soils over coarse materials, produce higher paddy yields in excess of 3,750 kg/ha. and most of the soils belonging to the coarse textured soils, well drained fine textured soils, moderately deep medium textured soils over coarse materials and saline soils, produce yields less than 3,750kg/ha. Soil texture of the profile, available soil depth, salinity and gleying of the surface and subsurface soils etc. seem to be the major factors determining rice yields, and these factors are considered when establishing suitability groups for paddy land. The great group, group, subgroup, family and series are proposed for the classification categories of paddy soils. The soil series is the basic category of the classification. The argillic horizon (Bt horizon) and cambic horizon (B horizon) are proposed as two diagnostic horizons of great group level for the determination of the morphological properties of soils in the classification. The specific soil characteristics considered in the group and subgroup levels are soil color of the profile (bluish gray, gray or yellowish brown), salinity (salic), depositonal (fluvic) and muck layers (mucky), and gleying of surface and subsurface soils (gleyic). The family levels are classified on the basis of soil reaction, soil texture and gravel content of the profile. The definitions are given on each classification category, diagnostic horizons and specific soil characteristics respectively. The soils on these plains are classified in eight subgroups and examined under the existing classification system. Further, the suitability group, can be divided into two major categories, suitability class and subclass. The soils within a suitability class are similar in potential productivity and limitation on use and management. Class 1 through 4 are distinguished from each other by combination of soil characteristics. Subclasses are divided from classes that have the same kind of dominant limitations such as slope(e), wettness(w), sandy(s), gravels(g), salinity(t) and non-gleying of the surface and subsurface soils(n). The above suitability classes and subclasses are examined, and the definitions are given. Seven subclasses are found on these plains for paddy soils. The classification and suitability group of 15 paddy soil series on the Gimje-Mangyeong plains may now be tabulated as follows.

A Survey of Nutritional Status on Pre-School Children in Korea (학영기전아동(學齡期前兒童)의 영양실태조사(營養實態調査))

  • The primary purpose of this study is to evaluate the correct nutritional status on pre-school children in Korea. Furthermore, it made an attempt to find and define nutrional problems, and assist in establishment on their nutritional improvement plan. For this, food intake and health condition (physical, clinical, biochemical and parasitological) survey on 109 Pre-school children in both sexes, randomly selected from Yang-Gu area in Gang-Won province and Rea-ju area in Kyong-gy Province, were conducted by means of three-day records, during the two periods of Spring and Fall season in 1975. The results obtained are summerized as follows: 1. The food intake; Average food intake of the subjects per day were $508{\sim}647g$ ($83{\sim}91%$ in vegetable foods and $5.5{\sim}11.7%$ in animal foods) in Yang-gu area, and $587{\sim}698g$ ($88{\sim}89%$ in vegetable foods and $6.3{\sim}7.6%$ in animal foods) in Rea-ju area. 2. The intake of energy and nutrients; a) Calory intake. Average energy intake of subjects per day in Yang-gu area$(1120{\sim}1415kcal)$ were all lower than the Korean Recommended Dietary Allowances (RDA) in either Spring and Fall survey, whereas the subjects in Rea-ju area were lower intake $(1213{\sim}1418kcal)$ than the RDA in the Spring but higher intake$(1516{\sim}1755kcal)$ than the RDA in the Fall, and the average intake were similar level with that of RDA. b) Protein intake. Average protein intake of the subjects per day in Yang-gu area $(33{\sim}43g)$ girl subjects in Rea-ju area $(35{\sim}39g)$ were lower than the RDA in either Spring and Fall survey, whereas the boy subjects in Rea-ju area$(36{\sim}38g)$ were lower in Spring and higher $(49{\sim}57g)$ in the Fall than that of the RDA, but the average $(43{\sim}47g)$ were similar level with the RDA. The protein intake from animal sources in all subjects were much lower $(5.5{\sim}11.7\;of\;total\;protein)$ than the RDA. c) Fat intake. Average fat intake were very lower in all subjects of both area $(14{\sim}24g\;in\;Yang-gu,\;10{\sim}12g\;in\;Rea-ju)$ than that of RDA which is recommended $12{\sim}14%$ of total energy to be supplied from fat. d) Calcium intake. Average calcium intake were very low in all subjects of both area $(264{\sim}355mg\;in\;Yang-gu\;and\;283{\sim}429mg\;in\;Rea-ju)$, especially, these in Spring were about a half level of the RDA, and it was much increased in the Fall due to increased intake of milk, but it was still not enough than the RDA. e) Vitamin A intake. Average intake of V.A ($703{\sim}1465\;IU$ in Ynag-gu and $750{\sim}1521\;IU$ in Rea-ju) were also lower than the RDA, moreover their V-A sources were mainly vegetable, so that the V-A supply might be critical one for the subjected. f) Riboflavin intake. Average riboflavin intake on all subjects in both area except boys in Rea-ju area in Fall, were very lower than the RDA. 3. The physical status; a) Average weight and height of boys aged 4 and 5 in Yang-gu area and girls of aged 5 in Rea-ju area were lower than those of Korean Standard of 1967 report, but those by age of girls in Yang-gu area and boys in Rea-ju area were a little heigher than the Korean Standard. It is, hower, present Korean standard of physical status might be somehow heigher than the 1967, since the socio-economical situation has been much improved during past a decade. So that, if one considered on this sense, the physical status of the subjects on this survey might be somehow lower than those of present Korean standard. b) Average upper arm circumference in both area were no difference each other, and their mean values of age 4, 5 and 6 in boy and girl were 15.6, 16.5, 16.4 and 15.5, 16.5, 16.4cm respectively. c) Average chest girth of boys were similar to those of Korean standard whereas the girls were smaller than the Korea standard. The average head circumference also showed similar tendency with the chest girth. 4. The clinical findings; The most popular clinical signs were angular stomatitis and dental caries, and boys had more heigher incidence then the girls. 5. The biochemical findings; a) Hemoglobin and anemia Average Hb value of boys and girls were 11.4 and 10.9g per 100 ml of blood respectively. The incidence of anemia (Hb value below 11 g/100 ml, by WHO) was increased by age, and girls had more heigher incidence than the boy (34% : 48%). The incidence of anemia in age of 4,5, and 6 in boys and girls were 28%, 41% 34%, and 33%, 50%, 49% respectively. The degree of the anemia was not severe, and the anemia of there subjects may be caused mainly low intake of better quality protein and low iron intake as well. b) Hematocrit. Average Ht value of whole subject were $39.9{\sim}41.6%$. c) Blood plasma protein. Average blood plasma protein contents of whole subjects were $6.6{\sim}7.4gm$ per 100 ml. The incidence of deficient range (<6.0g%, by ICNND) was only one girl of age 4 in yang Gu area. 6. Parasitological findgs; The most popular parasitism were asicris lumbicoides and trichocephalus trichiura, and about 2/3 of the whole subjects were suffering one or more of these parasitism.

Occurrence and Chemical Composition of White Mica from Zhenzigou Pb-Zn Deposit, China (중국 Zhenzigou 연-아연 광상의 백색운모 산상과 화학조성)

  • The Zhenzigou Pb-Zn deposit, which is one of the largest Pb-Zn deposit in the northeast of China, is located at the Qingchengzi mineral field in Jiao Liao Ji belt. The geology of this deposit consists of Archean granulite, Paleoproterozoinc migmatitic granite, Paleo-Mesoproterozoic sodic granite, Paleoproterozoic Liaohe group, Mesozoic diorite and Mesozoic monzoritic granite. The Zhenzigou deposit which is a strata bound SEDEX or SEDEX type deposit occurs as layer ore and vein ore in Langzishan formation and Dashiqiao formation of the Paleoproterozoic Liaohe group. White mica from this deposit are occured only in layer ore and are classified four type (Type I : weak alteration (clastic dolomitic marble), Type II : strong alteration (dolomitic clastic rock), Type III : layer ore (dolomitic clastic rock), Type IV : layer ore (clastic dolomitic marble)). Type I white mica in weak alteration zone is associated with dolomite that is formed by dolomitization of hydrothermal metasomatism. Type II white mica in strong alteration zone is associated with dolomite, ankerite, quartz and alteration of K-feldspar by hydrothermal metasomatism. Type III white mica in layer ore is associated with dolomite, ankerite, calcite, quartz and alteration of K-feldspar by hydrothermal metasomatism. And type IV white mica in layer ore is associated with dolomite, quartz and alteration of K-feldspar by hydrothermal metasomatism. It indicated that white mica of from the Zhenzigou deposit has less K, Na and Ca, and more Si than theoretical dioctahedral mica. Compositional variations in white mica from the Zhenzigou deposit are caused by phengitic or Tschermark substitution [(Al3+)VI+(Al3+)IV <-> (Fe2+ or Mg2+)VI+(Si4+)IV] substitution. It means that the Fe in white mica exists as Fe2+ and Fe3+, but mainly as Fe2+. Therefore, white mica from layer ore of the Zhenzigou deposit was formed in the process of remelting and re-precipitation of pre-existed minerals by hydrothermal metasomatism origined metamorphism (greenschist facies) associated with Paleoproterozoic intrusion. And compositional variations in white mica from the Zhenzigou deposit are caused by phengitic or Tschermark substitution [(Al3+)VI+(Al3+)IV <-> (Fe2+ or Mg2+)VI+(Si4+)IV] substitution during hydrothermal metasomatism depending on wallrock type, alteration degree and ore/gangue mineral occurrence frequency.

Microbiological and Enzymological Studies on Takju Brewing (탁주(濁酒) 양조(釀造)에 관(關)한 미생물학적(微生物學的) 및 효소학적(酵素學的) 연구(硏究))

  • 1. In order to investigate on the microflora and enzyme activity of mold wheat 'Nuruk' , the major source of microorganisms for the brewing of Takju (a Korean Sake), two samples of Nuruk, one prepared at the College of Agriculture, Chung Nam University (S) and the other perchased at a market (T), were taken for the study. The molds, aerobic bacteria, lactic acid bacteria, and yeasts were examined and counted. The yeasts were classified by the treatment with TTC (2, 3, 5 triphenyltetrazolium chloride) agar that yields a varied shade of color. The amylase and protease activities of Nuruk were measured. The results were as the followings. a) In the Nuruk S found were: Aspergillus oryzae group, $204{\times}10^5$; Black Aspergilli, $163{\times}10^5$; Rhizogus, $20{\times}10^5$; Penicillia, $134{\times}10^5$; Areobic bacteria, $9{\times}10^6-2{\times}10^7$; Lactic acid bacteria, $3{\times}10^4$ In the Nuruk T found were: Aspergillus oryzae group, $836{\times}10^5$; Black Aspergilli, $286{\times}10^5$; Rhizopus, $623{\times}10^5$; Penicillia, $264{\times}10^5$; Aerobic bacteria, $5{\times}10^6-9{\times}10^6$; Lactic acid bacteria, $3{\times}10^4$ b) Eighty to ninety percent of the aerobic bacteria in Nuruk S appeared to belong to Bacillus subtilis while about 70% of those in Nuruk T seemed to be spherical bacteria. In both Nuruks about 80% of lactic acid bacteria were observed as spherical ones. c) The population of yeasts in 1g. of Nuruk S was about $6{\times}10^5$, 56.5% of which were TTC pink yeasts, 16% of which were TTC red pink yeasts, 8% of which were TTC red yeasts, 19.5% of which were TTC white yeasts. In Nuruk T(1g) the number of yeasts accounted for $14{\times}10^4$ and constituted of 42% TTC pink. 21% TTC red pink 28% TTC red and 9% TTC white. d) The enzyme activity of 1g Nuruk S was: Liquefying type Amylase, $D^{40}/_{30},=256$ W.V. Saccharifying type Amylase, 43.32 A.U. Acid protease, 181 C.F.U. Alkaline protease, 240C.F.U. The enzyme activity of 1g Nuruk T was: Liquefying type Amylase $D^{40}/_{30},=32$ W.V. Saccharifying type amylase $^{30}34.92$ A.U. Acid protease, 138 C.F.U. Alkaline protease 31 C.F.U. 2. During the fermentation of 'Takju' employing the Nuruks S and T the microflora and enzyme activity throughout the brewing were observed in 12 hour intervals. TTC pink and red yeasts considered to be the major yeasts were isolated and cultured. The strains ($1{\times}10^6/ml$) were added to the mashes S and T in which pH was adjusted to 4.2 and the change of microflora was examined during the fermentation. The results were: a) The molds disappeared from each sample plot since 2 to 3 days after mashing while the population of aerobic bacteria was found to be $10{\times}10^7-35{\times}10^7/ml$ inS plots and $8.2{\times}10^7-12{\times}10^7$ in plots. Among them the coccus propagated substantially until some 30 hours elasped in the S and T plots treated with lactic acid but decreased abruptly thereafter. In the plots of SP. SR. TP. and TR the coccus had not appeared from the beginning while the bacillus showed up and down changes in number and diminished by 1/5-1/10 the original at the end stage. b) The lactic acid bacteria observed in the S plot were about $7.4{\times}10^7$ in number per ml of the mash in 24 hours and increased up to around $2{\times}10^8$ until 3-4 days since. After this period the population decreased rapidly and reached about $4{\times}10^5$ at the end, In the plot T the lactic acid becteria found were about $3{\times}10^8$ at the period of 24 fours, about $3{\times}10$ in 3 days and about $2{\times}10^5$ at the end in number. In the plots SP. SR. TP, and TR the lactic acid bacteria observed were as less as $4{\times}10^5$ at the stage of 24 hours and after this period the organisms either remained unchanged in population or ceased to exist. c) The maiority of lactic acid bacteria found in each mash were spherical and the change in number displayed a tendency in accordance with the amount of lactic acid and alcohol produced in the mash. d) The yeasts had showed a marked propagation since the period of 24 hours when the number was about $2{\times}10^8$ ㎖ mash in the plot S. $4{\times}10^8$ in 48 hours and $5-7{\times}10^8$ in the end period were observed. In the plot T the number was $4{\times}10^8$ in 24 hours and thereafter changed up and down maintaining $2-5{\times}10^8$ in the range. e) Over 90% of the yeasts found in the mashes of S and T plots were TTC pink type while both TTC red pink and TTC red types held range of $2{\times}10-3{\times}10^7$ throughout the entire fermentation. f) The population of TTC pink yeasts in the plot SP was as $5{\times}10^8$ much as that is, twice of that of S plot at the period of 24 hours. The predominance in number continued until the middle and later stages but the order of number became about the same at the end. g) Total number of the yeasts observed in the plot SR showed little difference from that of the plot SP. The TTC red yeasts added appeared considerably in the early stage but days after the change in number was about the same as that of the plot S. In the plot TR the population of TTC red yeasts was predominant over the T plot in the early stage which there was no difference between two plots there after. For this reason even in the plot w hers TTC red yeasts were added TTC pink yeasts were predominant. TTC red yeasts observed in the present experiment showed continuing growth until the later stage but the rate was low. h) In the plot TP TTC pink yeasts were found to be about $5{\times}10^8$ in number at the period of 2 days and inclined to decrease thereafter. Compared with the plot T the number of TTC pink yeasts in the plot TP was predominant until the middle stage but became at the later stage. i) The productivity of alcohol in the mash was measured. The plot where TTC pink yeasts were added showed somewhat better yield in the earely stage but at and after the middle stage the difference between the yeast-added and the intact mashes was not recognizable. And the production of alcohol was not proportional to the total number of yeasts present. j) Activity of the liquefying amylase was the highest until 12 hours after mashing, somewhat lowered once after that, and again increased around 36-48 hours after mashing. Then the activity had decreased continuously. Activity of saccharifying amylase also decreased at the period of 24 hours and then increased until 48 hours when it reached the maximum. Since, the activity had gradually decreased until 72 hours and rapidly so did thereafter. k) Activity of alkaline protease during the fermentation of mash showed a tendency to decrease continusously although somewhat irregular. Activity of acid protease increased until hours at the maximum, then decreased rapidly, and again increased, the vigor of acid protease showed better shape than that of alkaline protease throughout. 3. TTC pink yeasts that were predominant in number, two strains of TTC red pink yeasts that appeared throughout the brewing, and TTC red yeasts were identified and the physiological characters examined. The results were as described below. a) TTC pinkyeasts (B-50P) and two strains of TTC red pink yeasts (B-54 RP & B-60 RP) w ere identified as the type of Saccharomyces cerevisiae and TTC pink red yeasts CB-53 R) were as the type of Hansenula subpelliculosa. b) The fermentability of four strains above mentioned were measured as follows. Two strains of TTC red pink yeasts were the highest, TTC pink yeasts were the lowest in the fermantability. The former three strains were active in the early stage of fermentation and found to be suitable for manufacturing 'Takju' TTC red yeasts were found to play an important role in Takju brewing due to its strong ability to produce esters although its fermentability was low. c) The tolerance against nitrous acid of strains of yeast was marked. That against lactic acid was only 3% in Koji extract, and TTC red yeasts showed somewhat stronger resistance. The tolerance against alcohol of TTC pink and red pink yeasts in the Hayduck solution was 7% while that in the malt extract was 13%. However, that of TTC red yeasts was much weaker than others. Liguefying activity of gelatin by those four strains of yeast was not recognized even in 40 days. 4. Fermentability during Takju brewing was shown in the first two days as much as 70-80% of total fermentation and around 90% of fermentation proceeded in 3-4 days. The main fermentation appeared to be completed during :his period. Productivity of alcohol during Takju brewing was found to be apporximately 65% of the total amount of starch put in mashing. 5. The reason that Saccharomyces coreanuss found be Saito in the mash of Takju was not detected in the present experiment is considered due to the facts that Aspergillus oryzae has been inoculated in the mold wheat (Nuruk) since around 1930 and also that Koji has been used in Takju brewing, consequently causing they complete change in microflora in the Takju brewing. This consideration will be supported by the fact that the original flavor and taste have now been remarkably changed.
