  • In order to utilize the agricultural waste products for animal feeds, two high xylanase producing mold strains were selected from various sources of samples. The optimum conditions of xylanase production and the characteristics of the mold enzyme were investigated,and summarized as follows. 1. Two Aspergillus niger strains (experimental No. 1701 and 430) showed the high xylanase activity. 2. The highest xylanase production was obtained at pH 5.0-6.0 in two days. 3. Xylanase production in strain 1701 was increase with the addition of carboxy methyl cellulose, $NH_4H_2PO_4$ and corn steep liquor as carbon sources and natural nutrients, as respectively, while the other carbon, nitrogen, phosphate sources, natural nutrients and minerals gave no remarkable effect. In the strain 430, the enzyme procuction was not effected with the above substrate sources. 4. Maximum xylan hydrolysis reaction with the crude enzyme extract (33.3% v/v) was obtained in the 2% substrate concentration at pH 5.0 and $60^{\circ}C$ in three hours in both strains. 5. Maximum xylan hydrolysis rate was 95% at the optimum conditions for xylanase activity.

Effects of Asparagus cochinchinensis (Lour.) Merr. on the Stimulation of Osteoblast Differentiation and Inhibition of Osteoclast Generation (천문동 추출물에 의한 조골세포 분화 촉진 및 파골세포 생성 억제효과)

  • Bone mass in adults decreases with age because of the imbalance between the rate of bone formation and resorption. We performed this study to investigate whether Asparagus cochinchinensis (Lour.) Merr. (ACAE) plays a role in osteoblasts differentiation and osteoclasts formation. Ethanol extract of ACAE showed increase in the differentiation and alkaline phosphatase activity of osteoblasts. Also, it decreased the number of tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells (OCLs) and TRAP activity. Therefore, ACAE has the potential to prevent bone-related diseases such as osteoporosis by increasing the differentiation of osteoblasts and reducing both the number and activity of osteoclasts.

  • The production of xylanase and $\beta$-xylosidase was investigated in submerged culture of Aspergillus niger NRC 107. The maximum production occurred when the pH was controlled at 6.0 during the fermentation. Among the various carbon sources investigated, corn-cob xylan (1.5%, w/v) yielded maximal production of the enzymes. The $NaNO_{3}$ was the most favorable nitrogen source for enzyme production and $KH_2P0_4$ concentration at 0.3%(w/v) was found to be optimum. Incorporation of wheat bran to the culture medium improved xylanase production. Addition of L( -) sorbose to the culture medium promoted the secretion of $\beta$-xylosidase. It was possible to increase the production of xylanase (39.43 units/ml) and that of $\beta$-xylosidase (4.2 unitslml) by submerged culturing the A. niger NRC 107 in the modified medium.

  • 담낭용종의 잘 알려진 위험요소는 대사증후군, 연령, 이상지질혈증 등이다. 제주도는 한반도에서 가장 큰 섬으로 한국의 본토에서 약 80km 떨어져 있으며 행정구역상 제주시와 서귀포시로 나뉘어 있지만 기업들과 편의시설들, 학교들 때문에 자연스럽게 시내와 시외로 더 나누어진다. 많은 이주민들과 젊은 성인들은 상업 활동이 활발한 시내 지역에 사는 경향이 있다. 고령 성인들은 어업이나 농업에 종사하기 때문에 시외 지역에 사는 경향이 있다. 고령의 성인들은 전통적인 식이를 유지하고 있고, 그들 중 다수는 힘든 노동으로 인해서 고위험 알코올 음주자의 비율이 높다. 이러한 요인들로 인해 두 지역간의 담낭용종 유병률에도 차이가 있을 것이다. 따라서 이 연구의 목적은 제주도 두 지역간에 담낭용종의 유병률을 알아보고 그 유병률과 관련된 다양한 요인들을 비교해 보고자 하는 것이다. 이 연구는 2009년 1월부터 2019년 12월까지 제주대학교병원 단일 건강검진센터에서 건강검진을 시행한 21,734명을 대상으로 하였다. 시내 지역과 시외 지역 거주민들의 담낭용종 유병률은 각각 10.1%와 8,7% (P = 0.006)로 의미있게 차이가 있었다. 평균 총콜레스테롤 수치가 시내 지역 거주민들에서 시외 지역 거주민들보다 높았고, 음주와 관련된 평균 알카라인 인산분해효소와 감마-글루타밀전이효소 수치들과 평균연령은 시외 지역 거주민들이 시내 지역 거주민들보다 더 높았다. 나이, 알코올 섭취와 평균 콜레스테롤 수치가 이러한 두 지역 간에 담낭용종 유병률의 차이를 만들었을 것이다.

  • chitosan은 골치유증진 및 골세포의 분화를 촉진하는 것으로 알려진 천연의 생분해성 고분자이다. 이연구에서는 chitosan 및 chitosan/tricalcium phosphate(TCP) 다공성 기질을 제조하여 골이식재 및 조직공학적 골형성을 위한 3차원적 세포배양 지지체로서의 가능성을 평가하고자 하였다. chitosan 용액 및 TCP가 포함된 chitosan 용액을 동결건조함으로써 소공의 크기가 $100-200{\mu}m$인 스폰지형태의 chitosan 및 chitosan/TCP 다공성 기질을 제작하였다. 골이식재로서의 효과를 평가하기 위하여 백서의 두개골 결손부에 제작된 chiosan 및 chitosan/TCP 다공성 기질을 각각 이식하고 2주 및 4주 후에 동물을 희생하여 조직학적으로 치유양상을 관찰하였다. 조직공학적 골형성을 위한 세포배양 지지체로서의 가능성을 평가하기 위하여 백서 태자의 두개골에서 분리된 골아세포를 chitosan 및 chitosan/TCP 다공성 기질에 각각 접종하고 56일간 배양하면서 각 기간 별로 세포수, 염기성 인산효소 활성, 축적된 calcium의 양을 측정하였고 배양된 세포-기질 혼합체를 광학현미경 및 주사전자현 미경하에서 조직학적 관찰을 시행하였다. 백서 두개골결손부에 이식된 chitosan 및 chiosan/TCP 다공성 기질은 별다른 이물반응 없이 자연 분해되면서 신생골조직 내에 매립되었으며 이식하지 않은 대조군에 비해 유의하게 높은 신생골형성 효과를 나타내어 우수한 골전도성이 있음이 확인되었다. 신생골형성 양상이나 형성된 양에 있어서 두 가지 기질간의 유의한 차이는 없었다. 골아세포-기질 혼합체의 배양결과, 접종후 배양 28일 경과 시까지 골아세포수는 지속적으로 증가하다가 이후에는 5 8일까지 성장정도가 둔화되었다. 염기성 인산효소의 활성 및 calcium 축적량은 접종후 배양시간경과에 따라 56일까지 지속적으로 증가하였다. 세포수 및 염기성 인산효소의 활성에서 두 기질간의 유의한 차이는 없었고, calcium 축적량에 있어서는 chitosan/TCP 기질에서 유의하게 높았고 증가속도도 컸다. 배양된 골아세포가 접종된 다공성 기질의 조직학적 관찰결과, 골아세포는 다공성 기질에 잘 부착하여 중층의 형태로 성장하면서 광화된 골기질을 형성함이 관찰되었다. 배양 14일부터 작은 골편형태의 골형성이 기질 표면에 부착되어 관찰되었고, 배양기간이 길어짐에 따라 성장하여 배양 56일째에는 상당한 양의 광화된 골질이 형성됨이 관찰되었다. 배양 56일 경과후의 광화된 골질의 양은 chitosan/TCP 기질에서 더 많았다. 이 연구의 결과, chitosan 및 chitosan/TCP 다공성 기질이 골이식재로서 뿐만 아니라, 조직공학적 골형성에 적용되는 골아세포의 배양을 위한 3차원구조의 세포지지체로 이용되어 골재생술식에 유용한 생체재료로 활용될 수 있음이 확인되었다.

  • Inflammation is a host defense mechanism that is activated in response to harmful substances or pathogens. However, an excessive inflammatory response is a problem in itself. Macrophages secrete inflammatory mediators such as nitric oxide (NO) or cytokines through various pathways such as the nuclear factor kappa B (NF-${\kappa}B$)-activated pathway after recognizing pathogen-like lipopolysaccharides (LPSs). In this study, anti-inflammatory effects of Eupatorium chinensis var. simplicifolium (EUC) extracts were investigated using LPS-stimulated RAW 264.7 macrophages. The EUC root extract significantly reduced NO production, inducible nitric oxide synthase (iNOS) expression, and cyclooxygenase-2 expression in a concentration-dependent manner. In addition, the EUC root extract reduced phosphorylation of mitogen-activated protein kinases and protein kinase B, which is upstream of NF-${\kappa}B$. The EUC root extract also reduced the degradation of inhibitory kappa B. These results indicate that EUC root extract exerts anti-inflammatory effects, which are mediated by inhibition of iNOS expression and the NF-${\kappa}B$ pathway.

  • The effect of earthworm on the recycling or control of organic P in environment has been investigated. The activity of phytase(myo-inositol hexaposphate phosphohydrolase, EC 3.1.3.8) produced by isolated microoganisms from vermicomposted cow manure was usually higher than that of phytase produced by isolated microorganisms from composted cow manure. However the activity of phytase excreted by seperated earthworm(Eisenia foetida) was not detected. The concentration of total P and available P was revealed 2.88%, 0.22% in composted cow manure and 1.70% 0.14% in vermicomposted cow manure.

  • Single crystals of deoxy-thymidine diphosphate-D-gluxose-4,6-dehydratase(abbreviated as dTDP-D-glucose dehydratase) from Escherichia coli Strain BL21 clone which harbors the gene of dTDP-D-glucose dehydratase in Salmonella typhimurium LT2 have been grown with and whithout substrates by sitting drop vapor diffusion at room temperature. The precipitating agent was 1.6 to 2.0 M Na, K phosphate buffer(pH 8.0). The crystals diffract to at least 2.5Å and belong to the hexagonal space group P61 with cell dimensions a=b=168.54Å, c=81.08Å. The asymmetric unit contains one dimer with a crystal volume per protein mass(VM) of 2.4Å3/Da and solvent content (Vsol) of 64% by volume.

Effect of the Long-term Application of Organic Matters on Microbial Diversity in Upland Soils (유기물 장기 연용이 밭토양 미생물의 다양성에 미치는 영향)

  • To investigate the effect of long term application of organic matter in upland soils, plots for treatments of NPK, NPK+pig manure compost, rape seed cake, rice straw compost, and green manure were set up. Populations of Bacillus and Gram negative bacteria were high in the plot treated with green manure application, but microbial biomass was increased with chemical fertilizer or pig manure compost in upland soils. Activities of phosphomonoesterase and dehydrogenase were high with organic matter application comparing to control. Cluster patterns analysed using phospholipid fatty acid of plots treated with rice straw and or pig manure compost were clearly different comparing with other treatments. Dominant bacteria in upland soils were Bacillus flexus, B. subtilis and B. megaterium. And the strains isolated from upland soils had amylase, protease and lipase activities.

Electrochemical Degradation of Phenol by Using Reticulated Vitreous Carbon Immobilized Horseradish Peroxidase (Horseradish Peroxidase가 고정화된 다공성 탄소 전극을 이용한 페놀의 전기화학적 분해)

  • Horseradish peroxidase, had the phenol degradation rate of 95% in aqueous phase, was covalently immobilized on the surface of reticulated vitreous carbon(RVC) and the degradation of phenol was performed with in situ generated $H_2O_2$-immobilized HRP complex in an electrochemical reactor. The incorporation of carboxylic group on the RVC surface was confirmed by FT/IR spectrometry and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride(EDC) was used for peptide bonds between the carboxylic groups on the RVC surface and amine groups from HRP. The optimal conditions of in situ $H_2O_2$ generation such as concentration($10{\sim}200$ mM) and pH($5.0{\sim}8.0$) of electrolyte, supply of $O_2(10{\sim}50$ mL/min) and applied voltage($-0.2{\sim}-0.8$ volt, vs. Ag/AgCl) from potentiostat/galvanostat were determined by concentration of hydrogen peroxide and current efficiency. It was observed that the RVC immobilized HRP was stable maintaining 89% of the initial activity during 4 weeks. The phenol degradation rate of 86% was attained under the optimal condition of in situ $H_2O_2$ generation.
