Elucidation of the metabolic pathway due to 50% dietary restriction carried out in this study. Seventy male and female wealning rats, weighed $43{\pm}2g$ were divided into seven groups, 10 rats each. Twenty rats, ten males and ten females were sacrificed every three weeks after 50% dietary restriction for whole length of the experiment, nine weeks. Pair-feeding was employed in this study. According to the increment of the dietary restricted period, the body and organ weights were decreased. Especially liver and spleen were mostly shrinked in their weights, and brain was the most stable organ in account of dietary restricted effect. In comparison nitrogen retention between restricted and unrestricted groups, the former showed lower than the later but tubulated into the rate of Nitrogen retention per gram of body weight, reverse was true in this respect. In regardness of the experimental organs, spleen revealed the most fast change and the brain the most slow change their content of RNA and DNA in account of the 30% dietary restriction. Hematological investigation did not show any anemic conditions in both restricted and unrestricted groups. Also serum albumin contents A/G ratio, did not effect due to 50% dietary restrictions.

To determine the effects of dietary restriction on obese type 2 diabetes we measured body weight, blood glucose and serum lipid level in dietary restricted Otsuka Long Evans Tokushima Fatty (OLETF) rats. OLETF rats (obese diabetic rats) and LETO rats (control rats) were grouped into 3 groups; control (free feed) group, 20% dietary restricted (20% DR) group and 40% dietary restricted (40% DR) group. Body weight of rats was measured every weeks and the level of glucose, triglyceride (TG), total cholesterol (TC) and HDL-cholesterol in blood of rats were also determined at 12 weeks after dietary restriction. Body weight of control, 20% DR and 40% DR groups were increased by 41%, 20% and 10%, respectively in LETO rats and by 24%, 10% and -2%, respectively in OLETF rats. Blood glucose level of LETO rats were decreased by 12% on 40% DR compared to control group but the differences between control group and 20% DR group was not observed. The blood glucose level of OLETF rats were decreased by 20% in 40% DR group and by 15% in 20% DR group. The levels of blood triglyceride in 20% DR and 40% DR group were decreased by 20%, 15% in LETO rats and by 37%, 32% in OLETF rats, respectively Total cholesterol revel was not changed by dietary restriction in LETO rats, but significant changes were observed in OLETF rats by both 20% and 40% dietary restriction. HDL-cholesterol levels were also increased by dietary restriction in both LETO and OLETF rats. These results suggested that body weight and blood glucose, serum triglyceride and total cholesterol levels were decreased by dietary restriction and these changes are more susceptive in diabetic rats than non-diabetic animals.
A quantitative restriction of maternal diet was given to the Sprague Dawley rats during the third week of gestation and during the lactation Dams of deficient groups were fed 20g of a stock diet a day and compared with the control group fed ad libitum throughout the experimental Period Dietary restriction started from birth in deficient 1 group and from the 14th day of gestation in deficient 2 group. Brain serotonin, norepinephrinet dopamine, and tyrosine of offsprings were determined at several intervals. Brain serotonin of offsprings of deficient groups 1 and 2 were significantly lower than the control and significant differences between the deficient greeds were noticed Brain catecholamines and tyrosine of offsprings of deficient groups were significantly lower than the control and there was no difference between the deficient groups. Maternal dietary restriction during the gestation and the lactation may have retarded the synthesis of serotonin and catecholamines in the brain.
A quantitative restriction of maternal diet without changes in quality of diet was given to the Sprague Dawley rats during the third week of gestation and lactation. Half the normal average daily intake of control group was given to deficient groups in this period. Female rats of control group were fed a commercial diet ad libitum throughout the experimental period. Dietary restriction started from birth to weaning in deficient I group and from the 15th day of gestation to weaning in deficient II group. Body and brain weight of offsprings of deficient groups were significantly lower than control group, but the ratios of brain weight to body weight in deficient groups were higher than the control group. Significant difference between deficient groups (I and II) was noticed at weaning. Brain DNA, RNA and total protein of offsprings of deficient groups were significantly lower than control group, but RNA/DNA, brain weight/DNA, and total protein/DNA show that cell number were more affected than the cell size by the maternal dietary restriction during the third week of gestation and lactation. Between the deficient groups, there was a significant difference in brain DNA and RNA, but no significant difference in total brain protein. Data are limited on biological mechanisms underlying the associations of sleep insufficiency with obesity and dyslipidemia. To explore these mechanisms, we investigated appetite-regulating hormones, stress-related hormones, and cardiometabolic indicators in association with sleep fragmentation, which is a type of sleep disorder. In an experimental study, we randomly allocated 40 Wistar male rats aged 7 weeks into four groups; rats with ad libitum sleep and ad libitum intake (Control), those exposed to sleep fragmentation (SF), those with diet restriction (DR), and those exposed to sleep fragmentation and diet restriction (SF+DR). Amongst them, 13-day chronic sleep fragmentation was applied to the SF and SF+DR groups while 50% reduction in chow intake was applied to the DR and SF+DR groups for 13 days. After these experiments, blood lipid and lipoprotein profiles, leptin, ghrelin, adiponectin, cortisol, epinephrine, and norepinephrine levels were compared among the four groups. In the results, the SF group showed the highest levels of serum ghrelin (P<0.001) and the lowest levels of serum adiponectin (P<0.01). All experimental groups showed higher levels of low density lipoprotein-cholesterol (LDL-C) than the Control (P<0.001). LDL-C levels and the ratio of LDL-C and high density lipoprotein-cholesterol were positively correlated with ghrelin levels (P<0.05) in the SF group, but not in the DR and SF+DR groups. In the SF group, the highest levels of serum free fatty acids were also observed and correlated with lower levels of serum adiponectin, which reflects insulin resistance (P<0.05). Based on these findings, we suggest that chronic sleep fragmentation may induce disturbances in lipid metabolism and appetite-regulating hormones independent of food intake, and these metabolic disturbances may be worse due to insulin resistance related to overeating, which is indicated by elevated ghrelin levels in sleep fragmentation. For persons with sleep insufficiency, anti-atherogenic dietary interventions may be recommended to prevent cardiovascular disease.
A quantitative restriction of maternal diet was given to the Sprague Dawley rats during the lactation. The control group were fed a commercial diet ad libitum throughout the experimental period. Dietary restriction started from birth to weaning in deficient group. After weaning at 21 days, all offsprings were fed the same diet ad libitum for 4 weeks of rehabilitation. They were analyzed for body weight, muscle protein, muscle 3-methylhistidine and serum protein level of offsprings at 0, 1, 2, 3, 5, 7 weeks. Body weight of offsprings of deficient group were significantly lower than the control group, but after rehabilitation there were no significant differences between two groups. Maternal dietary restriction caused a decrease in the 3-methylhistidine, protein content in muscle and total protein level in serum, and rehabilitation after weaning induced a marked increase. These results suggest that there is a quantitative reduction in 3-methylhistidine metabolism in the undernourished rats and these reductions can be nearly recovered by 4 weeks of dietary rehabilitation after weaning.
Thirty female and six male rats aged fourty days were divided into two groups in order to feed them by pairfeeding for 50% dietary restriction in the pair group two weeks interval. Each group contains 15 female and three male rats matched each rat between two groups in consideration of body weight. Two female groups, one fed by 50% restricted diet and other Ad Libitum were divided into four groups each by the duration of dietary restriction during pregnancy: First ten days dietary restriction at 50% level, Last ten days dietary restriction at 50% level, Dietary restriction at 50% level for full period, And dietary unrestriction for full period Urinary total nitrogen and creatinine were determined. The birth weights of offsprings were decreased partial and full period dietary restriction of pregnant rats. There was no significant difference in the litter size of progeny due to the maternal diets. The growth was stunted in offsprings from the mothers fed restricted diet at 50% level for full period of pregnancy. No effect in the body weight gain of offsprings was observed in account of partial period of maternal dietary restriction. The urinary nitrogen of offsprings from eight different groups did not show any statistically significant difference.

Triptolide is a compound found in Tripterygium wilfordii and reported to have an anti-inflammatory and anti-oxidant activities. A previous study shows that the dietary supplementation with triptolide increases resistance to environmental stressors, including oxidative stress, heat shock, and ultraviolet irradiation, and extends lifespan in C. elegans. Here, we investigated the underlying mechanisms involved in the lifespan-extending effect of triptolide. The effect of triptolide on age-related diseases, such as diabetes mellitus and Alzheimer's disease, was also examined using animal disease models. The longevity phenotype conferred by triptolide was not observed in the eat-2 mutant, a well-known genetic model of dietary restriction, while there was an additional lifespan extension with triptolide in age-1 and clk-1 mutants. The long lifespan of age-1 mutant is resulted from a reduced insulin/IGF-1-like signaling and the clk-1 mutant lives longer than wild-type due to dysfunction of mitochondrial electron transport chain reaction. The effect of dietary restriction using bacterial dilution on lifespan also overlapped with that of triptolide. The toxicity of high glucose diet or transgenic human amyloid beta gene was significantly suppressed by the supplementation with triptolide. These findings suggest that triptolide can mimic the effect of dietary restriction on lifespan and onset of age-related diseases. We conclude that triptolide can be a strong candidate for the development of dietary restriction mimetics.

Effects of wild plant extract (Lak) based on Korea traditional prescription on maximal exercise performance and endurance were evaluated using calorie-restriction animal model. In acute forced swimming test with 10% body weight attached to tail, dietary Lak supplementation increased exercise performance endurance by increasing concentrations of ATP and insulin-like growth factor-1 (IGF-1) under calorie-restriction condition, and decrement of blood lactic acid concentration and increment of muscle ATP content were observed. These results suggest Lak is very effective for decreasing side-effects of obesity therapy using very low calorie diet.
