Antioxidant Activity of Methanol Extracts from the Genus Lespedeza (싸리속 식물 메탄올 추출물의 항산화 활성)

  • The genus Lespedeza belonging to Leguminosae is an annual or perennial herb, which has been used as a Chinese herbal medicine. Antioxidant activity on DPPH ((1,1)-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis(3-ethylenebenzothiazoline-6-sulfonic acid) radicals and the contents of total polyphenol and flavonoid were analyzed with 12 methanol extracts from six Lespedeza genus plants with different plant parts. The highest content of total polyphenol was detected in the leaves of Lespedeza$\times$robusta (194.6 mg GAE/g), while the highest content of total flavonoid existed in the aerial part of Lespedeza thunbergii var. intermedia (47.9 mg QE/g). Generally, the amounts of polyphenol and flavonoid compounds were higher in the leaves than in the stems. The $SC_{50}$ value of DPPH radical scavenging activity ranged from 10.16 to 90.94 ppm, of which the strongest activity was determined in the leaves of L. robusta. Most ABTS radical scavenging activities from the investigated methanol extracts were higher than that of L-ascorbic acid, implying the excellent antioxidant activity. The radical scavenging activity in this study showed high correlation with the amount of total polyphenol rather than that of total flavonoid. These data suggest that the methanol extracts from these Lespedeza spp. could be potential candidates for natural antioxidants.

Characterization of Transgenic Lettuce (Lactuca sativa L.) Using a BL1 Gene Encoding Bromelain Isolated from Pneapple (제주산 파인애플 유래 Bromelain관련 유전자 (BL1)를 이용반 형질전환 상추의 특성)

  • To clarify the roles of bromelain in plants, we isolated BL1 gene encoding bromelain from pineapple stem tissues and sequenced. The full length cDNA is 933 bp and encodes a polypeptide of 311 amino acid residues. The cDNA is most similar 94% at the amino acid level to bromelain previously isolated from pineapple (BAA21929). Explants of Lactuca sativa were co-cultivated with Agrobacterium tume-faciences LBA 4404 strains containing nptII and BL1 gene for transformation. Through initial selection of regenerated explants by culturing on a kanamycin and carbenicillin containing MS medium, multiple shoots were obtained after 2 months of culture. For a complementary step of selection, putative transgenic shoots were transferred to 1/2 Ms basal medium supplemented with 100 mg/L kanamycin and 500 mg/L carbenicillin. The selected shoots were obtained T1 generation seeds with emasculation, and tested with PCR analysis using 35S promoter and BL1 specific primers whether BL1 gene was introduced to genome of the plants. These results confirmed that produced the specific PCR bands in the putative transgenic lines. Additionally the Northern blot and endo protease activity showed that transcripts of BL1 gene were detected in transgenic lines. Theses results suggest that BL1 gene be successfully integrated and transcripted in the transgenic lettuce plants.

Evaluation of Acute Toxicity of Plant Extracts, Lavender, Lemon Eucalyptus and Cassia Essential Oil (식물 추출물 라벤더, 레몬 유칼립투스 및 계피 오일의 급성독성평가)

  • The acute toxicity test of high bio-active plant essential oils was conducted with Lavender, Lemon eucalyptus and Cassia oils selected to develop environment-friendly insecticides. The results of acute oral toxicity using rats showed that $LD_{50}$ of over 2,000 mg/kg bw for Lavender, Lemon eucalyptus and Cassia oils. The calculated acute dermal $LD_{50}$ value of all testing materials was over 4,000 mg/kg bw. The Skin irritation test indicated that Lavender and Lemon eucalyptus oil have no irritation while Cassia oil has a moderate irritation. For the Eye Irritation test, the result showed no irritation for Lavender and Lemon Eucalyptus oil and irritation for Cassia oils. However, the irritation was not showed for Eye Irritationwashing test of Cassia oil. Consequently, the Lavender and Lemon eucalyptus oils were showed to be low in toxicity whereas Cassia oil indicated to cause a moderate irritation on the skin and eyes.

Review of Functional Volatile Component in Essential Oil of Medicinal and Aromatic Plants (자원식물의 기능성 정유성분 이용 고찰)

  • The number of natural products obtained from plants has now reached over 100,000 and new chemical compounds are being discovered ever year. Medicinal and Aromatic plants and their extracts have been used for centuries to relieve pain, aid healing, kill bacteria and insects are important as the antifungal and anti-herbivore agents with further compounds being involved in the symbiotic associations. Although their functions in plants have not been fully established, it is Known that some substances have growth regulatory properties while others are involved in pollination and seed dispersal. The complex nature of these chemicals are usually produced in various types of secretory structures which is an important character of a plant family and also influenced and controlled by genetic and ecological factors. Detailed anatomical description of these structures ave relevant to the market value of the plants, the verification of authenticity of a given species and for the detection of substitution or adulteration. Volatile oils are used for their therapeutic action for flavoring of lemon, in perfumery of rose or as starting materials for the synthesis of other compounds of turpentine. For therapeutic purposes they are administered as inhalations of eucalyptus oil, peppermint oil, as gargles and mouthwashes of thymol and transdermally many essential oils including those of lavender, etc. With these current trend for using volatile components in essential oil will be increasing in the future in Korea and in the world as well.

Comparative Study on the Essential Oil Components of Panax Species (인삼속(Pauax species) 식물의 정유성분 조성 비교)

  • This study was carried out to determine the differences of essential oil components among Korean, Chinese and Japanese red ginseng, and Korean white ginseng (Panax ginseng C.A Mayer) , American and Canadian ginseng (P. Quinquefolium), and sanchi ginseng (P notoginseng). The steam distilled oils of these ginsengs were analyzed by GC and GC-MS, and 22 sesquiterpenes, 8 sesquiterpene alcohols, 8 monoterpenes, 5 aldehydes, 4 esters, 3 acids, 2 alcohols and 5 miscellaneous components were identified. The major oil components of Korean, Chinese and Japanese red ginseng were $\beta$-panasinsene, $\beta$-caryophyllene, $\alpha$-panasinsene, $\alpha$-neoclovene, selina-4,11-diane, bicyclo-ger-macrene and spathulenol. The contents of $\beta$-panasinsene, $\alpha$-neoclovene, $\alpha$-basabolene and spathulenol were higher in Korean red ginseng than Chinese and Japanese red ginseng. The contents of $\alpha$-cubebene, selina-4,11-diene and ledol were higher in Chinese red ginseng than Korean and Japanese red ginseng, but those of selina-4,11-diene and spathulenol were lower in Japanese red ginseng than Korean or Chinese red ginseng. On the other hand, the GC patterns of the oils from American, Canadian and sanchi ginseng were different from that of Korean white ginseng.

Settling Velocity of Phytoplankton in the Nakdong-River (낙동강 수계의 식물플랑크톤 침강속도)

  • Settling velocity is one of major parameters determining algal biomass in water quality modeling. In this study, the settling velocity of phytoplankton was measured in reservoir and stream sites of the Nakdong River, Korea. Settling velocities of various phytoplankton species were determined by measuring algal cell biomass settled in a sedimentation cylinder. Mean settling velocities were $0.22m\;day^{-1}$ in reservoir sites and $0.33m\;day^{-1}$ in stream sites, which were relatively higher compared with other default values suggested by water quality models (e.g. $0.1m\;day^{-1}$ in CE-QUAL-W2). The lower settling velocity in reservoirs than in stream implies the adaptation of phytoplakton to low turbulence in lentic environments. Cyanobacteria showed lower settling velocity ($0.2m\;day^{-1}$) than diatoms ($0.3m\;day^{-1}$), and this phenomenon may have resulted from buoyancy mechanisms of cyanobacteria. Cell volume did not show a significant correlation with settling velocity in this study, implying that conformation factors of colonies or other factors had large effects on settling velocity of algal cells as well as cell size. The result of this study may suggest proper coefficients of settling velocity of phytoplankton in the calibration of water quality model.

  • Natural resource plants have been useful and helpful materials for human to against diseases and Prevention since ancient Egyptian and Roman times. However, according to recent survey, its habitat and indigenous are continuously decreased and almost extinct some valuable species, as Adonis vernalis, Digitalis lanata etc., especially in Eastern Europe and east Asian countries due to radical political and social reforms. ESCOP and FAO have already initiated to protect the endangered species for conservation, protection and propagation technology but it is not easy tasks without global concerning. Most natural resource plants have volatile components in essential oils and its consumption and demanding is increased every year, which contained certain compounds have no side effects and good properties for biological activities. Essential oils of natural resource plants are also well known for medical purposes, such as Central Nervous System (CNS, linalool), anticancer (d-limonene, perillyl alcohol, etc.), microbial and fungicidal activities (menthol, borneol, etc). Essential oil also used for natural therapy to prevent and increase immune systems instead of chemical synthesis drugs. With this, researchers of natural resource plants are ultimately faced to develop and optimal technology for increasing the Korean farmer's income by high added-value herbal products in present and future.

Repellency of the Constituents of Caraway Oil, Carum carvi against, Tetranychus urticae (점박이응애에 대한 Caraway Oil의 기피활성)

  • Essential oils of three plants (caraway oil, hyssop oil and lime oil) were screened for repellency against Tetranychus urticae female adults in the lab condition. Among them, caraway oil (Carum carvi) showed $92.2\%$ repellency against female adults at 1,000 ppm concentration. Through the constituent analysis using GC and GC/MS, we confirmed that the two main constituents were carvone $(73.3\%)$ and limonene (26.7). Limonene showed 87.8 and $83.1\%$ repellency against adult females at 1,000 and 100 ppm concentrations, respectively.

Antibacterial Effects of Major Compounds in Essential Oil against Fish Disease Bacteria (식물 정유 주요 성분의 어병 세균에 대한 항균활성)

  • In this study, the antibacterial activity of major compounds obtained from domestic plant essential oils was investigated against four species of fish pathogenic bacteria, namely, Edwardsiella tarda, Vibrio harveyi, Streptococcus iniae, and Streptococcus parauberis. We observed that α-terpineol and eucalyptol showed high antibacterial activity against S. iniae, whereas α-terpineol and β-linalool showed high antibacterial activity against S. parauberis. The antibacterial activity of eucalyptol and β-linalool was significantly high against E. tarda, and α-terpineol and β-linalool exerted antibacterial activity against V. harveyi. All compounds showing significant antibacterial activity also exhibited high solubility in water (≥1,000 mg/L). In contrast, compounds such as α-limonene and α-pinene with low water solubility showed significantly low antibacterial activity against all bacteria. These findings can be utilized further to estimate the antibacterial activity of compounds isolated from plant essential oils for the prevention of fish disease.

Effects of Organic Matter Applications on General Components and Essential Oils in Codonopsis lanceolata $T_{RAUTV}$ (유기물(有機物) 종류(種類)에 따른 더덕 근(根)의 일반성분(一般成分)과 정유성분(精油成分) 변화(變化))

  • This experiment was conducted to increase aromatics in roots of Condonopsis lanceolata by applications of organic matters. Fresh root wt. was increased by conifer/moss application to 79.1g per plant. Crude protein content was also higher at rice straw application than native soil application and crude saponin content was increased by conifer/moss application, but contents of crude fat, fiber and ash were not different in all treatments. Although contents of K, Ca, and Mg were increased by rice straw application, Fe, Mn, Zn, Na and Cu were not significantly different in all treatments, The highest free amino acid was arginine, it was increased by the application of fallen leaves and the highest yield (0,008%) of essential oils was obtained by conifer/moss application. As a result, to produce C. lanceolata plant showing higher quality and aromatic essential oils, it was considered that the most effective organic matter showing high yield and higher aromatic constituents was conifer/moss application of over 3M/T per 10a.
