Changes of Fruit Structure and Sugar Contents during the Fruit Growth and Development in 'Yumyeong' Peach [Prunus persica (L.) Batsch] (과실생육시기별 '유명' 복숭아의 과실조직 발달과 당 변화)

  • 'Yumyeong' is one of the most popular peach varieties in Korea. This study was conducted to monitor the developments of cells and tissues, and the changes in sugar contents during fruit growth and development. At bloom, there were two rows of vascular tissues, and the number and the position of internal vascular bundles were consistent during the fruit growth, however, the number of vascular tissues were increased and the distribution was irregular in the flesh tissues. The tissues between the inner integument and the internal vascular bundles showed different development characteristics from other parenchyma cells which were consisted of small and dense cells containing tannins. Such observation suggested that the stone of peach was consisted of inner epidermis and cells in the internal vascular tissues. The outer epidermis consisted of single layer cells at bloom differrentiated into 1-2 layers by horizontal cell divisions on 14 days after full bloom. On 30 days after full bloom, the epidermis was consisted of 5-6 layers by vertical cell divisions. The cell layers consisting the outer epidermis were gradually decreased to 1-2 layers at maturity. The observations on the changes in the epidermis confirmed that some of the cells consisting the hypodermis of peach fruit were originated from the cells of outer epidermis. Tylosis was observed from 35 days after full bloom and the size and number of tylosis were increased until full fruit maturity. The sucrose content showed the tendency of sharp increase from 50 days to 120 days after full bloom then decrease slightly. After when stone hardening ended, other solids showed a gradual decrease tendency from 80 days after full bloom.

Calyx Abscission in Pear (Pyrus spp.) Cultivars and Its Inheritance (배 품종별 꽃받침 탈리와 유전 양식)

  • During pear fruit development, calyx can abscise from fruitlet following petal fall. The calyx abscission varies with pear cultivars. The presence of calyx on pear fruit makes the fruit shape calyx end protruded. In the present study, the degrees of the calyx abscission were examined in 120 Southern-type Asian pear (Pyrus pyrifolia), 52 Nothern-type Asian pear (P. ussuriensis), and 34 European pear (P. communis) cultivars, and its inheritance was investigated using cross combinations between the Southern-type Asian pear cultivars showing different degrees of calyx abscission. Majority of the cultivars produced < 10% or > 90% calyx-perpetual fruit, but the cultivars producing both calyx-perpetual and -deciduous fruit were in minor frequency. The cultivars producing < 10% calyx-perpetual fruit were in higher frequency in Southern-type Asian, Nothern-type Asian, and European pears in that order, while those producing calyx-perpetual fruit were in higher frequency in European, Nothern-type Asian, and Southern-type Asian pears in that order. In the cross between the parents producing < 10% calyx-perpetual fruit, most of the $F_1$ seedlings also produced < 10% calyx-perpetual fruit. In the cross between the parents producing > 90% calyx-perpetual fruit, on the contrary, most of the $F_1$ seedlings also produced > 90% calyx-perpetual fruit. When the paternal parent produced < 10% calyx-perpetual fruit, most of the $F_1$ seedlings also produced < 10% calyx-perpetual fruit regardless of the degree of calyx abscission in the maternal parent. When the cross was between the maternal parent producing < 10% calyx-perpetual fruit and the paternal parent showing different degrees of calyx abscission, the $F_1$ seedlings showed similar degrees of the calyx abscission to those in the paternal parent. These results suggest that the characteristics of the calyx abscission is influenced more greatly by the paternal parent than by the maternal parent, and the calyx abscission in Southern-type Asian pears is a qualitative trait which is governed by dominant gene(s).

  • Carrot (Daucus carota L. var. sativa) is one of the most widely used crops in the world and is nutritionally important crop. However, seed-hair which is generated in epidermal cell of seeds causes the difficulty of the seedling process, because of the seed germination and absorption inhibitions. For these reasons, carrot seeds are commercialized after mechanical hair removal process. However, in this process, various damage and seed loss occur and breeding of hairless-seed carrot cultivar is needed to overcome these various weaknesses and additional seed production costs. In this study, cDNA libraries using 2 combinations, which were composed of short-hair seed CT-ATR 615 OP 666-13 & long-hair seed CT-ATR 615 OP 671-9, and short-hair seed CT-SMR 616 OP 659-1 & long-hair seed CT-SMR 616 OP 677-14, were constructed and EST sequences of each individuals were analyzed to reveal carrot seed-hair characteristics. Firstly, analyzed EST sequences were classified into FunCat functional categories. As a result, significant differences have been identified in metabolism category, protein folding and stabilization, protein binding, C-compound binding category from both of two combinations. Secondly, several candidate EST sequences related to seed trichome differentiation and cellulose biosynthetic process were selected based on GO data of EST sequences. These differences based on FunCat categories and candidate EST obtained by GO data analysis are thought to be involved in the formation of carrot seed hair. Finally, 741 SSR sites and 33 SNP sites were identified from analyzed EST sequences of two combinations. Then we designed SNP and SSR primer sets to develop molecular markers. These molecular markers will be used for classification of carrot cultivars and study seed-hair characteristic.

Monitoring of Biosafety of Agricultural Products from Urban Community Gardens and Roof Gardens in Korea (도시텃밭과 옥상정원에서 재배되는 농산물의 생물학적 안전성 조사)

  • Urban farming supplies emotional stability and fresh vegetables to participating persons, however, no information regarding the biosafety of agricultural products from urban farming is available. Here, we collected 260 samples of Chinese cabbages and lettuce from 4 urban community gardens and 6 roof gardens in Seoul from September through October 2012, and monitored the microbiological and parasitological contamination quantitatively and/or qualitatively. The mean counts of total aerobic bacteria and coliforms were $6.1{\pm}0.8\;log\;CFU{\cdot}g^{-1}$ ( range, $5.4{\pm}0.6{\sim}7.1{\pm}0.8\;log\;CFU{\cdot}g^{-1}$) and $4.0{\pm}0.7\;log\;CFU{\cdot}g^{-1}$ (range, $2.3{\pm}0.6{\sim}6.1{\pm}0.9\;log\;CFU{\cdot}g^{-1}$), respectively. Coliforms were detected on 59.6% among 260 vegetable samples. There were no significant differences in the contamination levels of total aerobic bacteria and coliforms between the Chinese cabbages and lettuce, whereas both levels of vegetables from urban community gardens were higher than those of roof gardens (p > 0.05). Escherichia coli was isolated at 3.1% among whole vegetables, and contamination level was $1.5{\pm}0.2\;log\;CFU{\cdot}g^{-1}$. Among foodborne pathogens, Staphylococcus aureus was detected in 1.5%, however, Salmonella spp., Listeria monocytogenes and E. coli O157:H7 were not detected on any of the vegetable samples. We also found undefined parasite eggs from two samples of Chinese cabbages (0.8% of total vegetables we tested). From these study, we found the presence of microbial contamination of agricultural products from urban farming, thus we need further concern to improve the biosafety during production of agricultural products.

  • A food list representing the typical diet of Koreans was developed for a total diet study based on the 687 food items which appeared in the dietary intake data collected during the 2008 & 2009 Korea National Health & Nutrition Examination Survey. Food items included in 90% of the cumulative intake, items with a consumption frequency of 5% or higher and items contributing to 80% of cumulative fat intake were considered first. From the resulting 165 food items, 15 items were removed to avoid similar foods, while 10 distinct items were added to increase the efficiency of 'best-fit' mapping. Consequently, 160 items were selected as representative foods, which cover 90.93% of intake amount, 91.36% of energy intake and 89.05% of fat intake. Given that every food consumed by Koreans can't be analyzed for contaminants of interest, mapping other foods to the previously mentioned representative 160 foods is warranted because it enables a more comprehensive dietary exposure assessment that is close to reality.

Effects of Shipping Temperature on Quality Changes of Cucumber, Eggplant, Melon, and Cherry-tomato Fruit during Simulated Export and Marketing (오이, 가지, 멜론 및 방울토마토 과실의 수송온도가 모의 수출 및 유통중 품질에 미치는 영향)

  • Quality changes of fruit vegetables were investigated during simulated export and marketing to find out the optimum shipping temperature. Fruit vegetables were loaded into a small refrigerated-container and kept for four days at various temperatures, and fruit quality was assayed immediately after harvest, 4 days after storage and 4 days after marketing at ambient temperature. In 'Back Seong Ilho' cucumber fruits, fresh weight loss was further reduced at $13^{\circ}C$ and $11^{\circ}C$ than at $15^{\circ}C$ and room temperature. Soluble solid contents remained at relatively lower levels when cucumbers were stored at $13^{\circ}C$ and $11^{\circ}C$. In 'Chun Ryang' eggplant fruits, fresh weight loss was greatly increased at all the temperatures (room, $12^{\circ}C,\;9^{\circ}C,\;and\;6^{\circ}C$). However, flesh browning, a primary quality factor of eggplant fruit, was most effectively inhibited at $9^{\circ}C$, whereas chilling injury occurred in fruit flesh at $6^{\circ}C$. Water loss of 'Eals Seinu' melon fruits was most inhibited and soluble solid contents at harvest were maintained for the longest period at $4^{\circ}C$. In 'Pe Pe' cherry tomatoes, storage at $10^{\circ}C$ and $7^{\circ}C$ seemed to more effectively inhibit metabolic changes and the incidence of cracking, the severest disorder than room temperature. But the fruits stored at $10^{\circ}C$ contained higher level of soluble solids than those at $7^{\circ}C$. The overall results suggest that the optimum shipping temperature range is 11 to $13^{\circ}C$ for cucumbers, around $9^{\circ}C$ for eggplant fruit, $4^{\circ}C$ for melons, and $10^{\circ}C$ for cherry tomatoes.

Effect of Medium, Soil, and Irrigation Water Contaminated with Escherichia coli and Bacillus cereus on the Microbiological Safety of Lettuce (Escherichia coli 와 Bacillus cereus에 오염된 상토, 토양 및 관개용수가 상추의 미생물 안전에 미치는 영향)

  • Many outbreaks of food-borne illnesses have been associated with the consumption of fresh vegetables and fruits contaminated with food-borne pathogens. Contaminated medium, manure and irrigation water are probable vehicles for the pathogen in many outbreaks. The aim of this study was to determine the potential transfer of Escherichia coli and Bacillus cereus from medium and soil fertilized with contaminated compost or irrigation with contaminated water to the edible parts of lettuce. Moreover, survivals of the two pathogens on lettuce contaminated medium, soil and irrigation water were estimated. Lettuce seeds were planted in medium contaminated with 7.5 log colony forming unit (CFU)/g of E. coli and B. cereus. Seedlings grown in the contaminated medium were transplanted in soil fertilized with contaminated pig manure compost or uncontaminated soil. Contaminated irrigation water with E. coli and B. cereus at 8.0 log CFU/mL was applied only once on the plant by sprinkle irrigation and surface irrigation. Although E. coli and B. cereus in medium and sprouted lettuce after planting seeds were reduced as time passed, these pathogens survived in seedling raising stage for extended periods. The numbers of E. coli and B. cereus in lettuce grown on contaminated soil were detected over 4.0 log CFU/g for 21 days. The numbers of E. coli and B. cereus in lettuce applied by sprinkle irrigation were higher than those of surface irrigation by 5.0 log CFU/g. Our results indicated that contaminated medium, soil and irrigation water can play an important role in the presence of food-borne pathogens on vegetables.

Analyses of CO2 Concentration and Balance in a Closed Production System for King Oyster Mushroom and Lettuce (밀폐형 식물생산시스템 내 새송이 버섯과 상추의 혼합 재배 비율에 따른 CO2 농도 변화 및 균형 분석)

  • The large amount of $CO_2$ emitted from mushrooms during incubation and developmental stages can be utilized in plant production systems as a $CO_2$ source. The objectives of this study were to measure the $CO_2$ emission and absorption rates of mushroom and lettuce, respectively, and to analyze the $CO_2$ concentrations at various ratios of mushroom and lettuce in a closed production system. The $CO_2$ emission rate of king oyster mushrooms (Pleurotus eryngii ( DC.) Qu$\acute{e}$l) and $CO_2$ absorption rate of lettuces (Lactuca sativa L. cv. Asia Heuk Romaine) were measured by using two closed acryl chambers ($1.0m{\times}0.8m{\times}0.5m$) in which indoor temperatures were maintained at $18^{\circ}C$ and $22^{\circ}C$, respectively. The lettuce was grown at a light intensity of PPF $340mol{\cdot}m^{-2}{\cdot}s^{-1}$ and with nutrient solution at EC $1.2dS{\cdot}m^{-1}$. The air was periodically circulated between the two chambers using a diaphragm pump. The $CO_2$ emission rate of the mushroom increased until the $15^{th}$ day after scratching (DAS) and then decreased. The rate also increased with increased indoor temperature. In particular, the $CO_2$ emission rate per fresh weight of fruit body increased by about 3.1 times after thinning compared to before thinning. In terms of $CO_2$ balance, the $CO_2$ emission rates from a bottle (950 mL) of the mushroom at 9, 12, and 14 DAS were equivalent to those of 3, 4.5, and 5.5 lettuce plants at 7, 10, and 12 DAT (days after transplanting), respectively. This work shows that balance in $CO_2$ concentration could be achieved using an appropriate ratio of the two crops in a closed production system.

Suggestions for Settlement Stable Employment Culture of Dental Hygienist (치과위생사의 안정적인 고용문화 정착을 위한 제언)

  • The purpose of this study was to examine the causes of career interruptions among dental hygienists, institutional measures required for their long service and ways of creating a stable employment culture for them in determine how to resolve labor shortage, create stable jobs, and step up the reemployment of idle manpower. In addition, the following suggestions are made for the establishment of a stable employment culture for dental hygienists by analyzing related literature, research materials, and information such as forums for establishing appropriate jobs for female dental workers. First, a system should be set up to prevent career interruption among dental hygienists. The work environment should be improved to prevent career breaks, and the wages, working hours, and working style should be efficiently structured to maintain the tenure of employees. Second, a plan should be devised to make use of idle manpower, and a variety of necessary programs should be developed. With respect to regular working hours, the time conversion system should be used, which reduces the amount of time one would want to work while receiving a national subsidy. Third, dental hygienists working in different occupations for marriage, childbirth, childcare, school and personal hygiene should make a way to return to the dental system immediately when they want. Fourth, the government should take institutional measures and offer down-to-earth support and benefits for women consideration their social characteristics to guarantee a balance between work and childcare.

Study on the Manufacturing techniques & Conservation of Iron Pot from Cheonmachong Ancient Tomb (천마총 출토 철부(鐵釜)의 제작기법 및 보존처리)

  • It's shown how to proceed the study on Manufacturing techniques & Conservation to the Iron Pot from Cheonmachong Ancient Tomb(the 155th Tomb in Hwangnam-dong). In order to investigate manufacturing techniques of the Iron Pot, some parts of the relic were gathered. After mounting, polishing and etching on the relic, analyzing the metal microstructure was conducted. Also it's conducted a SEM-EDS analysis on the nonmetallic inclusion. White iron structure was observed in the metallurgical structure inspection, SEM-EDS analysis. It seems to be dried slowly at room temperature after casting, doesn't look as particular heat treatment to improve brittleness. It is estimated that it's as the handle seam side were verified about 3cm inch wide, 1.5 thick in center of body, so 2 separate half-completed products was cast with width-type mould. The manufacturing techniques Using white cast iron structure, width-type mould are observable to the Iron Pot excavated from Sikrichong Ancient Tomb & Hwangnamdaechong grand Ancient Tomb around those were constructed the same time. It's able to recognize that it's almost identical manufacturing techniques at that time. Conservation is generically following those are survey of pretreatment, foreign material removal, stabilization, restoration and color matching in the order. cleaning & drying were added to the process as occasion demands. The strengthening treatment were difficult with artifact's volume, low concentration Paraloid NAD-10 solution was spread two or three times with a brush, surface hardening also came up with 15wt% Paraloid NAD-10 solution after the conservation was complete. There were connection & restoration for the restoration to the damage after modeling forms that it's similar to damaged parts by using the Fiber Reinforced Plastic resins(POLYCOAT FH-245, mold laminated type). Throughout this research, capitalizing on accumulations of measurements about the production technique of Iron Pot in the time of the fifth and 6th centuries is no less important than the Iron artifact's conservation for a better study in the future.
