Sophorae radix extract (SRE) has been registered as an environment-friendly organic material that is widely used in the cultivation of crops in Korea. Matrine, the active ingredient in SRE, was reported as a toxic substance in the nervous system in mice. However, no information is available on its toxic effects in other organisms. Therefore, antimutagenicity and two kinds of genotoxicity tests (bacterial reverse mutation and chromosome aberration test) of two samples of SRE were investigated in this study. Antimutagenicity test was experimented by using bacterial reverse mutation test. In the reverse mutation test, Salmonella Typhimurim TA98, TA1535 and TA1537 were used to evaluate the mutagenic potential of SRE. Bacterial reverse mutation test was also performed on positive and negative control groups in the presence of the metabolic activation system (with S-9 mix) and metabolic non-activation system (without S-9 mix). In the chromosome aberration test, Chinese hamster lung cells were exposed to SRE for 6 or 24 hours without S-9 mix, or for 6 hours with S-9 mix. Negative and positive control groups were experimented for chromosome aberration test. As a result, the number of mutated colonies induced by 4-NQO were reduced by SRE treatment in all strains, indicating that SRE may have antimutagenic effects. Reverse mutation was not shown at all concentrations of SRE, regardless of application of the metabolic activation system. In the chromosomal aberration test, one of the SRE sample gave a suspicious positive result at 250 ${\mu}g/ml$ in the presence of S-9 mix. For the more adequate evaluation of the genotoxic potential of SRE samples, other in vivo genotoxicity study is needed.

This study was conducted to elucidate the relationship between light and coloring and to obtain basic results for promoting redness expression in 'Kalle' (Pyrus communis L.) pear skin. It was investigated in location of anthocyanin layer by microscopic observation and differences in skin color expression of 'Kalle' bagged with paper bag which has different light transmittance rate and inside temperature. However, there was no anthocyanin layer in the brown skin and golden yellow color, anthocyanin layer was distributed in epidermins or hyperdermis of red skin pear and apple. Dark red colored 'Kalle' had more anthocyanin content, $29.8mg{\cdot}100g^{-1}$ FW than light red colored apple 'Hongro'. Light transmittance rate of physical characteristics used paper bags was the highest in white paper bag, 42.2% and it also had more light quantity, $8.9{\mu}mol$ than any other tested paper bags in specific wave length 650-655 nm. The maximum temperature of inner bag was higher about $3^{\circ}C$ in yellow paper bag. The red coloration and anthocyanin contents in no bagged fruits were higher than in any other bagged fruit. However, red color expression among the bagged fruits was higher in white paper bag than in double layered black paper bag and yellow paper bag. Also, chromaticity value seemd to be a good index to explain variation of fruit skin color, because anthocyanin content and chromaticity value were higher. Based on these results, it is desirable to cultivate 'Kalle' without bag for stable redness expression but bagging is essential for decreasing damage by insect in Korea. Further examination to find suitable time of removing paperbag for redness expression and decreasing insect damage. In addition, it is required to develop paperbag whose transmittance rate is high in specific light wavelength or temperature of inner bags is low. In this study, the annual growth of roots and their branching forms of Norway spruce(Picea abies [L.] Karst.) were analysed to compare the development of their root in the differently acidified forest soils. And there was no significant difference among the stands for the modified roots and the non-modified roots depending on the root branch forms, and however in the most acidified Weidenbrunnen stand, the modified roots, the abnormal root branch form such as fork, gun and rake types were appeared. As a result of the ring of the root, the annual horizontal root growth were 6.3cm for Weidenbrunnen and Barbis stands and 9.5cm for Eberg$\ddot{o}$tzen stand. The average annual vertical root growth was 4.4cm, 5.4cm, and 6.7cm for Weidenbrunnen stand, Barbis stand, and Eberg$\ddot{o}$tzen stand, respectively. The cross section area by root distribution at 80cm deep showed that the thick and thin roots were evenly distributed in of Eberg$\ddot{o}$tzen stand and the sum of root cross section area was $32.6cm^2$. In Barbis stand, the thick roots were distributed in the center while the thin roots were comparatively rare. And the sum of root cross section area was $29.2cm^2$. In Weidenbrunnen stand, only a few thin roots were found, and the total root cross section area was $10.9cm^2$. The stability coefficient of roots were in the order of Eberg$\ddot{o}$tzen stand(1.04), Barbis stand (0.3), and Weidenbrunnen(0.08) stand. Among the investigated Norway spruce stands, the modified abnormal root branching form and the low root growth appeared in the Weidenbrunnen stand could be attributed by the soil acidification etc.
For study on the variation of natural stand, three populations of Pinus densiflora S. et Z. were selected at samsanri Yongogmyun Myongjugun Kangwondo (4), Hawonri seomyun Uljingun Kyongbuk (5), and Emogdong Suweon Kyongkido (6) successibely after the selection of three population in 1974. Twenty individual trees were chosen from each population and the morphological characteristics of trees, needle and wood properties were investigated on the trees. The results are summerized as follows; 1. Serration density, resin canal number in needle did not show significant differences, however stomata row number in the both sides of needle showed highly significant differences among 3 populations. But significant differences were calculated among individual trees in a population regarding any character of needles. 2. Ail population had high correlation on the stomata row between abaxial and adaxial side of needle. 3. The Myongjungun population showed the highest value of resin duct index, which means the population had the highest degree of hybrid character. 4. The ring segment width and summerwood percentage in the wood properties had significant differences, and yet specific gravity and tracheid length had not significant differences statistically among 3 populations. But all the values were significant statistically among the ring segments within population. 5. The ring segment width decreased rapidly with increasing tree age but summerwood percentage, specific gravity, tracheid length increased slowly to the middle age of tree and then decreased slowly after the age. But the patterns of decrease or increase were some different by population. 6. The values of Uljingun population were generally high in the coefficient of variation on all the needle characters. And the values of Suweon population were always the highest and those of Myongjugun population the lowest in the coefficient of variation on all the wood properties.
This study was initiated to find out the possible insect vector and other means of transmission of jujube witches'-broom disease. 1. Eight species of leafhoppers were found to feed on both healthy and diseased jujube trees. Among these, rhombic marked leafhoppers (Hishimonus sellatus Uhler) were most abundant throughout the growing season of jujube trees in all localities surveyed. H. sellatus was far more abundant on diseased trees than healthy ones. 2. Jujube witches'-broom mycoplasma(JWM) was transmitted to jujube seedlings by Hishimonus sellatus. Jujube seedlings inoculated with H. sellatus which had been fed for 14-21 days on diseased jujube plants, developed smaller, chlorotic leaves 40-60 days after inoculation. Electron microscopy of midveins and petioles of the infected jujube seedlings revealed the presence of numerous mycoplasmalike organisms in phloem tissues. 3. Jujube witches'-broom mycoplasma was also transmitted to Vinca rosea plants by H. sellatus. Infected Vinca rosea plants developed vein clearing and marginal chlorosis of upper leaves 25-38 days after inoculation and followed by stunting and ultimate wilting and death of plant. Electron microscopy of petioles and midveins of infected Vinca rosea plant revealed the presence of numerous mycoplasmalike organisms in phloem tissues. 4. H. sellatus survived more than 30 days on jujube, Vinca rosea, carrot, celery, eggplant, hop, Calystegia japonica, Humulus japonicus, Astragalus sinicus, white clover, red clover, and radino clover. Many second generation nymphs of H. sellatus were also abserved on these plants but clovers. 5. Jujube seeds collected from witches'-broom diseased jujube trees yielded healthy seedlings.
In order to contribute to the Korean tea-plant culture and tea industry by means of increasing the production of tea-plants, I have performed the tissue culture of the organs of the anther, leaf and stem. As for the culture-material, I have used the anther of tea (Thea sinensis) at the tetrad uninucleate microspore stage and used medium of modified Murashige and Skoog as the basal medium supplemented with the growth regulators of NAA and 2, 4-D, yeast, kinetin and others at various concentrations. As for the handling of material, I have followed the common methods of sterilization and microtoming and paraffine imbedding method and observed systematically periodic changes of the microspores in culture. I have divided the leaf, stem and root into segments and sterilized them and used the modified Murashige and Skoog as the basal medium and observed the differentiation of roots and callus and the results are as follows. 1. In case of anther, I have found 2n callus was found in 30 out of 100 segments in M2 medium. 2. The differentiation of roots appeared in 24.5% of total leaf segments cultured and in 50.5% of stem and in 43.9% of root. 3. When the differentiation of stem in different parts was observed, the most frequent differentiation was found in the second part of all the 4 parts. 4. The most frequent formation of callus was noticed from the anther-walls in case of anther culture and from the veins in case of leaf culture. It is concluded that the seedlings of tea-plant could be multiplied most by means of tissue culture of the second part of the tea-plant stem and reduction in the expenditures of tea-plant propagation was possible through tissue culture.
Polyploids are a potentially important germplasm source in seedless citrus breeding program. Seedlessness is one of the most promising traits of commercial mandarin breeds that mandarin triploid hybrids possess permanently. The formation of new constant triploid hybrids can be recovered through diploid species hybridization from the fusion of divalent gametes at low frequencyor intra-and inter-ploidy crosses. However, extensive breeding work based on small $F_1$ hybrid seeds developed is impossible without a very effective aseptic methodology and ploidy event. In this study, in vitro embryo culture was employed to recover natural hybrids from monoembryonic diploid, open-pollinated mandarin. Flow cytometry was used to determine ploidy level. A total of 10,289 seeds were extracted from 792 fruits having approximately 13 seeds per fruit. Average frequency of small seeds developed was 7.1%, while the average frequency of small seeds per fruit were: 8.9% for 'Clementine' 10.2% for 'Harehime' 2.6% for 'Kamja' 3.1% for 'Pyunkyool' 2.8% for 'Sadookam' and 7.0% for 'Wilking' mandarin. Average size of a perfect seed was $49.52{\pm}0.07mm^2$ ('Clementine') while the small seed measured $7.95{\pm}0.04mm^2$ ('Clementine'), which was about 1/6 smaller than the perfect seed. In total, 731 small seeds were obtained and all of them contained only one embryo per seed. The efficiency of 'Clementine' was 14 times higher than 'Wilking' and more than 109 times higher than 'Pyunkyool'. The basic information on spontaneous polyploidy provides for the hybridization of constant triploids and increases the efficiency of conventional cross.
This study was performed to obtain a basic materials for improvement of garlic storage ability. The "Namhae jaerae" of southern strain and the "Dalsung jaerae" of northern strain were harvested by period, their stems were cut at 7cm and 25cm from disk and then placed them in blast drying system 12 hours per day at $40^{\circ}C$ for 4 days, or in natural state. The decrease rate of bulb weight was compared and rooting, sprouting, the rate of decay was investigated between natural dry(conventional method) and hot-air dry (blast drying system). The results obtained were as follows. In the case of the decrease rate of bulb weight after hot-air dry; 7cm plot" "Namhae jaerae" was similiar to one of 13th day of natural dry and 25cm plot 14th day. In "Dalsung jaerae" 7cm plot of early, common and late harvest was respectively similiar to one of 22nd, 18th and 16th day of natural dry, 25cm plot of early, common and late harvest showed the same decrease rate of bulb weight as that of 18th, 16th and 14th day of natural dry respectively. In the case of rooting and sprouting in sand culture at the early period of storage, hot-air dry showed more prolonged tendency than conventional drying method. In the case of clove state in the latter period of storage, number of eatable cloves was more numerous and number of decayed cloves were less in blast drying system than in conventional method.
This study was performed to obtain the basic informations for red dry pepper fruit with more pericarp weight(or in percentage) with a complete diallel cross(excluding reciprocals) using eight cultivars. Heterosis, combining ability and inheritance of the dry red fruit weight and its components(stem, placenta, seed, and pericarp) were evaluated. The results obtained were summarized as follows : Dry weight/fruit and its four antomical components were heavier in the earlier harvest fruit than in that of the later fruit. They showed 1% significance among parents and $F_1s$, and those of $F_1$ were significantly heavier than in parent. All characters in earlier fruit of parent, however, were higher than in later fruit of $F_1$. Dry weight percentage of pericarp to dry weight/fruit was highest followed by seed. Percentage of pericarp in the later fruit was increased while the seed decreased and percentages of stem and placenta were not differed between the earlier and later fruit. $F_1$ hybrids above the higher parent were observed in all characters. Mean heterosis (%) was positive in all characters while mean heterobeltiosis (%) was negative excepting seed and dry weight/fruit. GCA and SCA variances were highly significant, and GCA vaiances were greater than SCA in all characters. The directions of dominance were positive. Partial dominance was shown in stem, complete dominance in placenta, pericarp and dry weight/fruit, and over dominance in seed. The effective genes were estimated as one for stem and placenta, and two for seed, pericarp and dry weight/fruit. Heritabilities in narrow and broad sense were higher.
Four kinds of makgeolli brewed with different proportions of rice and wheat flour were prepared to investigate the effects of the carbohydrates source on the storage properties, and to examine the chemical characteristics and BA (biogenic amine) contents in makgeolli stored over 30 days at 4, 10 and $20^{\circ}C$, respectively. The chemical properties and BA contents were sustained at a consistent level for those makgeolli stored at $4^{\circ}C$ over 30 days. The alcohol content was significantly higher for makgeolli brewed with higher proportions of rice (R) over wheat flour (WF) (R 100: WF 0, R 70: WF 30) when compared with those brewed with a high proportion of wheat flour (R 30: WF 70, R 0: WF 100), and increased with increasing storage temperature and duration. The free amino acid contents of makgeolli brewed with high proportion of rice were significantly lower under the same conditions. Among the 8 BAs examined, tyramine, putrescine, cadaverine and phenylethylamine were detected, while tyramine and putrescine were mainly detected. Histamine was not detected throughout storage. Makgeolli brewed with rice as the only carbohydrate source exhibited very low levels of total BA during 30 days of storing at $20^{\circ}C$. Total BA contents of R100, R70, R30, R0 were 178.6, 682.9, 1186.7 and 1150.4 mg/L, respectively. These results suggest that makgeolli brewed with relatively higher proportions of rice have better storage properties and are safer for consumption.
