  • 어류에서 바이러스성 질병이 중요하다는 인식이 증가하고 있으며 어류바이러스의 연구를 위한 세포주의 발달이 활성화되고 있다. 하지만상대적으로 해산유래의 세포 주 수는 많지 않다. 특히 유럽과 우리나라 남ㆍ서해안에서 고급어종인 감성돔은 림포 시스티스병, 이리도바이러스병 등의 바이러스성 질병이 발생하고 있다. 본 연구는 감성돔에서 유래한 세포주 개발을 목적으로 하였으며, 감성돔의 비장에서 유래한 세포를 초대배양 및 계대배양 하여 새로운 세포주를 확립하였으며, 일련의 실험을 실시하여 확립된 세포주의 특성을 확인하고 바이러스에 대한 감수성을 검사하였다. Effect of Fish Number in Respiratory Chamber on Routine Oxygen Consumption of Black Porgy Acanthopagrus schlegeli Reared in Seawater or Freshwater (해수 및 담수사육 감성돔, Acanthopagrus schlegeli의 호흡측정실내 수용개체수에 따른 산소소비 비교)

  • Comparison of fish number on routine oxygen consumption (OC) of black porgy (Acanthopagrus schlegeli) reared in seawater (SW) versus freshwater (FW) was performed in closed water-recirculating system containing respiratory chamber. Fish acclimated in separate indoor tanks with SW or FW were sampled as two groups (one or three of fish used, $mean\;{\pm}\;S.D.=36.4{\pm}9.8$). OC of fish reared in both SW and FW showed clear temporal rhythms, with higher values in the daytime and lower values at night, in accordance with light (09:00-21:00 h) and dark (21:00-09:00 h) phases of the diel cycle (12L:12D). The OC of the fish increased linearly with the water temperature. The OCs of black porgy reared in SW and FW at 15, 20, $25^{\circ}C$ were 140.0, 174.8, 282.3 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ and 177.5, 307.8, 413.1 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ for the one of fish used, and 200.5, 274.7, 339.1 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ and 118.2, 188.2, 252.8 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ for three of fish used, respectively. Black porgy reared in SW had higher ventilation rates than those in FW. These results indicate that black porgy reared in FW had higher OC than those in SW at the one of fish used, but the fish reared in SW had higher OC than those in FW at the three of fish used.

  • Feeding habits of juvenile Acanthopagrus schlegeli collected from the eelgrass bed in Kwangyang Bay were studied. A. schlegeli (1~7cm SL) was a carnivore which consumed mainly amphipods (gammarid and caprellid amphipods), crabs and shrimps. Its diets included small quantities of gastropods, polychaetes, hydroids, tanaids, copepods and isopods. A. schlegeli showed ontogenetic changes in feeding habits. Small individuals preyed mainly on gammarid amphipods, gastropods and polychaetes. However, crabs and shrimps were heavily selected with increasing fish size.

  • Hybrid of red seabream (Pagrus major) and Japanese red seabream (Pagrus major) grew rapidly from to August (WT $19.7^{\circ}C{\sim}24.0^{\circ}C$) while black seabream (Acanthopagrus major) did from September to October (WT $19.9{\sim}23.0^{\circ}C$). Survival rate of hybrids (85.5%) was a significantly higher than those of the other fish species (20%). Green liver syndrome were observed in black sea bream and Japanease red seabream but did not in hybrids. Microcotyle tai infected 0~90% in hybrid, 20~100% in red seabream, 10% in black sea bream and 20~100% in Japanese red seabream but no significant difference was found among each fish species.

On the Growth of Young Stage Acanthopagrus schlegelii in Kunsan Coast, Korea (군산연안 유어기 감성돔의 성장)

  • To study on the growth of young stage Acanthopagrus schlegelii, samples were collected from the shallow waters off Kunsan, Korea from July to December, 1995. The planctonic larvae of Acanthopagrus schlegelii were collected in early June. The young fish of 12.7~26.6 mm in total length began to appear in the shallow water in late June. They grew up 19.9~51.4 mm in middle July, 24.8~75.0 mm in late July, 63.3~79.6mm in middle August. A. schlegelii disappear from the shallow water after middle August, and began to be collected in the outer bay off Kokunsan Isls. by long back net. This species that had spawned from Kunsan coast in late June to middle August grew up to 100mm in total length in December of that year.

Evaluation offish Flesh and Profitability of Black Porgy (Acanthopagrus schlegeli) Cultured in Freshwater (담수양식 감성돔(Acanthopagrus schlegeli)의 어육평가 및 수익성 분석)

  • The objective of this study was to evaluate food value and profitability of black porgy (Acanthopagrus schlegeli) cultured in freshwater. In fish flesh, muscular hardness of black porgy reared in freshwater $(9210{\pm}1215g/cm^2)$ was slightly lower than those of fish in seawater $(9987{\pm}6549g/cm^2)$, but there was no significant difference. Also, there was no difference between muscular strength of fish reared in freshwater and seawater. When the flesh qualities of black porgy reared in freshwater was compared with those of fish reared in seawater through the questionnaire, there were no significant differences between fish reared in freshwater and seawater in appearance, texture, taste and flavor. For 10 months of black porgy culture in fresh water, the gross profit in culturing from juvenile (5.5 g) to adult size (100g), and from adult to marketable size (400 g) were 24,000,000 won (30.0%) and 53,870,000 won (36.9%), respectively.

  • The present paper deals with the results of the observations on the characteristics of the eggs being ripen by injection of synahorin and the larvae of the black porgy. Mylio macrocehalus (Basilewsky). The adult fish used in the experiments, during May 11th to June 5th, 1970, were obtained from a fish-farm of the Atsumi Fish Culture Company in Aichi Prefecture, Japan, The results obtained are as follows: 1. The egg is colorless and buoyant, spherical in shape, measuring from $0.81\~0.96\;mm$ in dia-meter with a single oil globule, measuring $0.20\~0.23\;mm$. 2. Hatching took place within about 35 hours and 50 minutes after insemination of eggs at the water temperature $19.6\~22.5^{\circ}C$ (Mean, $20.6^{\circ}$). 3. The newly hatched larvae were 1.57 mm in total length, with the oil globule situated at the posterior part of the elongated oval-shaped yolk sac. Melanophores and xanthophores were distributed on the middle of the caudal region, and were scattered on the surface of the oil globule, but none on the yolk sac. 4. The diameter of the eggs ill each adult is distinguished into two types, large and small. The former is in the range of $0.811\~0.873\;mm$, the latter is $0.874\~0.963\;mm$. 3. The hatching rate of black porgy was in the range of $78.63\~95.19\%$ in large and $68.84\~74.80\%$ in small eggs. The mean hatching rate of total eggs was $79.80\%$. 6. The optimum temperature of hatching black porgy is in the range of $20\~26^{\circ}C$. The highest temperature limit of hatching the black porgy is from $26.71\~28.56^{\circ}C$. 7. The total length of hatched larvae is not maintained pararell to diameter of eggs and total length of the large larvae is from $1.976\~2.225mm$, and that of small larvae is $1.526\~1.90mm$. 8. The diameter of yolk sac of black porgy is in the range of $0.878\~0.979\;mm$ 9. The initial survived larva of the black porgy was $13.71\%$ in minimum, and $79\%$ in maximam. And the mean initial survived rate was $48.3\%$. 10. In conclusion, the biological characteristics of hatched larva of black porgy is seemed to be possible for large scale production of artificial seedlings. But experimental results make initial food the prime object believed to be supplemented.

  • We isolated complementary DNA(cDNA) encoding prolactin receptor(PRLR) from gill of black porgy Acanthopagrus schlegeli. Its PRLR cDNA consists of 1,611 base pairs and encodes the protein of 536 amino acids. To investigate the osmoregulatory abilities of black porgy in different salinities(35, 10 and 0 psu), we examined the expression of PRLR mRNA in osmoregulatory organs(gill, kidney and intestine) using reverse transcription(RT)-PCR. In gill and intestine, PRLR mRNA levels were high in 10 psu, and then decreased in 0 psu, but there is no changes in kidney. Also, plasma osmolality, $Na^+\;and\;Cl^-$ levels decreased during the experimental period. These results suggest that PRLR plays an important role in hormonal regulation in osmoregulatory organs during freshwater acclimation, thereby improving the hyper-osmoregulatory ability of black porgy in hypoosmotic environments.

  • 감성돔(Black porgy, Acanthopagrus schlegeli)은 우리나라 서ㆍ남해안 전역과 동중국해, 북해도 이남의 일본 연안에 광범위하게 분포하는 어종이다. 정착성 어종이나 동중국해로 회유하는 특성도 가지고 있으며, 서식 수심은 100 m 이내의 암반 지역으로 저층 및 중층에 주로 분포하고 있다. 최근 연근해 어족 자원의 감소로 인해 수산자원의 안정적인 공급에 문제가 발생하고 있으며, 이를 해결하기 위해 1998년부터 경남 통영에 바다 목장 사업이 진행되고 있다. (중략)
