mPW1PW91 Calculated Structures and IR Spectra of the Conformational Stereoisomers of C-Cyanophenyl Pyrogallol[4]arene

  • Molecular structures of the various conformational stereoisomers of 2,8,14,20-cyanophenyl pyrogallol[4]arenes 1 were optimized using the mPW1PW91 (hybrid Hartree-Fock density functional) calculation method. The total electronic and Gibbs free energies and the normal vibrational frequencies of the different structures from three major conformations (CHAIR, TABLE, and 1,2-Alternate) of the four stereoisomers [1(rccc), 1(rcct), 1(rctt), and 1(rtct)] were analyzed. The mPW1PW91/6-31G(d,p) calculations suggested that $1(rcct)_{1,2-A}$, 1(rctt)CHAIR, and $1(rtct)_{CHAIR}$ were the more stable conformations of the respective stereoisomers. Hydrogen bonding is the primary factor for the relative stabilities of the various conformational isomers, and maximizing the ${\pi}-{\pi}$ interaction between the cyanophenyl rings is the secondary factor. The calculated IR spectra of the more stable conformers [$1(rctt)_{CHAIR}$, $1(rcct)_{1,2-A}$, $1(rtct)_{CHAIR}$] were compared with the experimental IR spectrum of $1(rtct)_{CHAIR}$.

Transcriptional activation of pref-1 by E2F1 in 3T3 L1 cells

  • The E2F gene family appears to regulate the proliferation and differentiation of events that are required for adipogenesis. Pref-1 is a transmembrane protein that inhibits adipocyte differentiation in 3T3-L1 cells. In this study, we found that the expression of pref-1 is regulated by the transcription factor E2F1. The expression of pref-1 and E2F1 was strongly induced in preadipocytes and at the late differentiation stage. Using luciferase reporter assay, ChIP assay and EMSA, we found that the -211/-194 region of the pref-1 promoter is essential for the binding of E2F1 as well as E2F1-dependent transcriptional activation. Knockdown of E2F1 reduced both pref-1 promoter activity and the level of pref-1 mRNA. Taken together, our data suggest that transcriptional activation of pref-1 is stimulated by E2F1 protein in adipocytes.

  • Glioma-Associated Oncogene Homolog1 (Gli1) is known to be activated in malignant glioma; however, its downstream pathway has not been fully explained. The aim of this study was to explore the role of Gli1-Aquaporin1 (AQP1) signal pathway in glioma cell survival. Our data suggests that both Gli1 and AQP1 are upregulated in glioma tissues, as in comparison to in normal tissues. These up-regulation phenomena were also observed in glioma U251 and U87 cells. It was demonstrated that Gli1 positively regulated the AQP1 expression. By luciferase reporter gene and ChIP assay, we observed that this modulation process was realized by combination of Gli1 with AQP1 promotor. In addition, knock down of Gli1 by siRNA interference reduced the viability of glioma cells as well as suppressed cell metastasis. Also, the inhibitory effects of cell survival by silenced Gli1 were abrogated by AQP1 overexpression. In summary, glioma cell survival is a regulatory process and can be mediated by Gli1-AQP1 pathway.

Epigenetic regulation of long noncoding RNA UCA1 by SATB1 in breast cancer

  • Special AT-rich sequence binding protein 1 (SATB1) is a nuclear matrix-associated DNA-binding protein that functions as a chromatin organizer. SATB1 is highly expressed in aggressive breast cancer cells and promotes growth and metastasis by reprograming gene expression. Through genome-wide cross-examination of gene expression and histone methylation, we identified SATB1 target genes for which expression is associated with altered epigenetic marks. Among the identified genes, long noncoding RNA urothelial carcinoma-associated 1 (UCA1) was upregulated by SATB1 depletion. Upregulation of UCA1 coincided with increased H3K4 trimethylation (H3K4me3) levels and decreased H3K27 trimethylation (H3K27me3) levels. Our study showed that SATB1 binds to the upstream region of UCA1 in vivo, and that its promoter activity increases with SATB1 depletion. Furthermore, simultaneous depletion of SATB1 and UCA1 potentiated suppression of tumor growth and cell survival. Thus, SATB1 repressed the expression of oncogenic UCA1, suppressing growth and survival of breast cancer cells.

Novel AGLP-1 albumin fusion protein as a long-lasting agent for type 2 diabetes

  • Glucagon like peptide-1 (GLP-1) regulates glucose mediated-insulin secretion, nutrient accumulation, and ${\beta}$-cell growth. Despite the potential therapeutic usage for type 2 diabetes (T2D), GLP-1 has a short half-life in vivo ($t_{1/2}$ <2 min). In an attempt to prolong half-life, GLP-1 fusion proteins were genetically engineered: GLP-1 human serum albumin fusion (GLP-1/HSA), AGLP-1/HSA which has an additional alanine at the N-terminus of GLP-1, and AGLP-1-L/HSA, in which a peptide linker is inserted between AGLP-1 and HSA. Recombinant fusion proteins secreted from the Chinese Hamster Ovary-K1 (CHO-K1) cell line were purified with high purity (>96%). AGLP-1 fusion protein was resistant against the dipeptidyl peptidase-IV (DPP-IV). The fusion proteins activated cAMP-mediated signaling in rat insulinoma INS-1 cells. Furthermore, a C57BL/6N mice pharmacodynamics study exhibited that AGLP-1-L/HSA effectively reduced blood glucose level compared to AGLP-1/HSA.

  • This paper presents characterizations on the independence of the exponential and Pareto distributions by record values. Effects of Mite Antigen and Toxic Shock Syndrome Toxin-1 on the Biological Actvity of Human Fibroblast (Toxic Shock Syndrome Toxin-1 및 Mite 항원이 사람섬유아세포의 생물활성에 미치는 효과)

  • The production of interleukin-1(IL-1)and nitric oxide(NO) by cultured fibroblast cells of human nasal turbinate was revealed by biological assay respectively. The cells were incubated for various periods of time in the presence of staphyloccocal toxic shock syndrome toxin-1(TSST-1) and house dust mite(Dermatophagoides farinae, HDM), and the culture supernatants were harvested. There was a little difference in the activities of IL-1beta and the amount of NO produced by the cells when stimulated with 0.002-0.1$\mu$g/ml of TSSTO-1 and 0.02-1$\mu$g/ml of HDM. The shapes of the time course curves for the production of IL-1beta and NO by the cells were different. Groups stimulated with TSST-1 or HDM produced more IL-beta in 2 h than no exposure group(Control). A certain mixed group(TSST-1, 10ng+mite, 100 ng) continued to produce IL-1beta highly throughout the entire incubation period. The cells stimulated with TSST-1 or HDM produced more NO in 2 h and 6 h than that produced in the end of incubation(48 h). Also, the mixed groups were generally similar. There results suggest that induction of IL-1beta by a certain mixed condition(TSST-1+mite) in fibroblast cell in vivo may play a role in inflammation.

Chromatin-remodeling Factor INI1/hSNF5/BAF47 Is Involved in Activation of the Colony Stimulating Factor 1 Promoter

  • INI1/hSNF5/BAF47 is a core component of the hSWI/ SNF ATP-dependent chromatin remodeling complex, and it has been implicated in regulating gene expression, cell division and tumorigenesis. We investigated whether INI1/hSNF5/BAF47 functions in activation of the colony stimulating factor 1 (CSF1) promoter in HeLa cells. Overexpression of INI1/hSNF5/BAF47 promoted CSF1 transcription, and siRNA targeting INI1/hSNF5/ BAF47 (siINI1) strongly inhibited the activity of the CSF1 promoter. We demonstrated that all conserved domains of INI1/hSNF5/BAF47 are needed for CSF1 transcription. ChIP experiment showed that INI1/ hSNF5/BAF47 is recruited to the region of the CSF1 promoter. Taken together, these results indicate that INI1/hSNF5/BAF47 is involved in activation of the CSF1 promoter.

Effect of 1/f Fluctuation Sound on Comfort Sensibility (1/f 변동리듬 특성을 가지는 음이 쾌적감성에 미치는 영향)

  • 1/f fluctuation characteristics can be seen in various natural phenomena, such as breezes, streams, candle flames and the luminous patterns of fireflies. It is said that the 1/f fluctuation are comfortable for human beings. And they are introduced into many industrial products, such as an air conditioner, music, lighting, etc. This study focused on verifying the effects of 1/f fluctuation sound on comfort sensibility. Stimulus were divided into three groups(Group 1, 2, 3) by sound generation methodology. Fluctuation patterns of group 1, 2 were given by three types of fluctuation, 1/f0, 1/f1, 1/f2, and its of group 3 were given by two types of pure tone, 1/f1. In order to verify the effects, we measured the physiological responses of the subjects such as EEG(Electroencephalogram), ERP(Event-Related Potential), and these physiological responses were compared with subjective assessments, free answers. Consequently, we found that factor which had an effect on comfort sensibility was cognitive factor(for stimulus) rather than 1/f fluctuation sound pattern.

Human HS1BP3 induces cell apoptosis and activates AP-1

  • In the present study, we characterized the function of HS1-binding protein 3 (HS1BP3), which is mutated in essential tremor and may be involved in lymphocyte activation. We found that HS1BP3 localized to the mitochondria and endoplasmic reticulum partially. Overexpression of HS1BP3 induced apoptosis in HEK293T and HeLa cell lines. When these cell lines were transfected with HS1BP3, they exhibited nuclear DNA condensation, externalization of phosphatidylserine (PS), and cleavage of poly ADP ribose polymerase (PARP). Furthermore, suppression of HS1BP3 or HS1 expression attenuates HS1BP3 induced apoptosis. In addition, HS1BP3 enhanced activator protein 1 (AP-1)-mediated transcription in a dose-dependent manner. Therefore, we conclude that HS1BP3 regulates apoptosis via HS1 and stimulates AP-1-mediated transcription.
