Characterization of an Amylase-sensitive Bacteriocin DF01 Produced by Lactobacillus brevis DF01 Isolated from Dongchimi, Korean Fermented Vegetable

  • A DF01 strain that inhibits tyramine-producing Lactobacillus curvatus KFRI 166 was isolated from Dongchimi, a traditional Korean fermented vegetable, and identified as Lactobacillus brevis by biochemical analysis and reverse transcriptase sequencing of 16S rRNA. The antimicrobial compound produced by L. brevis DF01 was secreted at a maximum level of 640 AU/mL in late exponential phase in MRS broth, and its activity remained constant during stationary phase. The activity of bacteriocin DF01 was totally inactivated by $\alpha$-chymotrypsin, pronase E, proteinase K, trypsin, and $\alpha$-amylase, but not by catalase, which indicates the compound was glycoprotein in nature. The activity was not affected by pH changes ranging from 2 to 12 or heat treatment (60, 80, and $100^{\circ}C$ for 30 min), but was reduced after autoclaving. Bacteriocin DF01 had bacteriolytic activity and a molecular weight of approximately 8.2 kDa, as shown by tricine-SDS-PAGE analysis. Therefore, bacteriocin DF01 can be used in the manufacture of fermented meat products due to its inhibition of tyramine-producing L. curvatus and non-inhibition of L. sake, which is used as a starter culture for meat fermentation.

  • Chulwha pieces collected from Hakbong-ri site in Keryong mountain were studied whether there is reactions or not in each layer to investigate firing condition of glaze, body, Chulwha, engobe of Buncheong ware in the early Chosun Dynasty, 15th~16th Century. As a result of XRD analysis of a Chulwha piece, a main crystal phase was $\alpha$-Quartz and a second was Mullite. It was assumed the firing temperature would be around $1200^{\circ}C$ because a little amount of Mullite was formed in the body and there was no phase transition $\alpha$-Quartz to cristobalite. As a result of ICRM analysis, Chulwha and glaze didn't react and the melted glazes were sunk into the Chunwha particles. The thin layer of glaze was found on the Chulwha layer. As a result, the color of Chulwha layer always came to vivid black. Moreover, Chulwha painting didn't spread over the Buncheong ware, because Chulwha and engobe didn't react. The boundary interface of engobe and body was not clear because they had similar compositions. This shows engobe was composed of more $Al_2O_3$ than body.

  • The damping property of Fe-12Cr-22Mn-X alloys has been investigated to develop high damping and high strength alloy. Particularly, the effect of the phase of austenite, alpha and epsilon martensite, which constitute the structure of the alloys Fe-12Cr-22Mn-X alloys, on the damping capacity at room temperature has been investigated. Various fraction of these phases were formed depending on the alloy element and cold work degree. The damping capacity is strongly affected by ${\varepsilon}$ martensite while the other phase, such as ${\alpha}'$ martensite, actually exhibit little effect on damping capacity. In case of Fe-12Cr-22Mn-3Co alloy, the large volume fraction of ${\varepsilon}$ martensite formed at about 30% cold rolling, and in case of Fe-12Cr-22Mn-1Ti alloy, formed at about 20% cold rolling and showed the highest damping capacity. Damping capacity showed higher value in Fe-12Cr-22Mn-1Ti alloy than one in Fe-12Cr-22Mn-3Co alloy.

Screening procedure for organochlorinated pesticide residues in meat fat using a matrix solid phase dispersion(MSPD) extraction and GC/ECD (MSPD법과 GC/ECD를 이용한 식육중의 잔류농약 분석)

  • A multiresidue, simple and rapid isolation technique known as matrix-solid phase dispersoin (MSPD) for the extraction and quantitative gas chromatographic/electron capture detection (GC/ECD) determination of 14 organochlorinated pesticides($\alpha$-BHC, ($\beta$-BHC, ($\gamma$-BHC, aldrin, dieldrin, endrin, heptachlor, ($\alpha$-endosulfan, ($\beta$-endosulfan, endosulfan sulfate, p,p'-DDE, o,p'-DDD, p,p'-DDD, p,p'-DDT) from meat fats. The 14 pesticide were fortified into meat fat(0.5g) and blend with 2g $C_{18}$, $C_{18}$meat fat matrix blend and 2g activated florisil comprise an extraction column from which the pesticides are eluted by adding 8ml acetonitrile. Then 2${\mu}\ell$ of the eluate is analyzed by GC/ECD. Unfortified blank controls are tested similarly. The eluate contained all the pesticide analytes and was free of interfering coextractants. Recovery rate(31.3-500ng/g) were ranged from 80$\pm$4% to 97$\pm$4%. Any organochlorinated pesticides were not detected in 120 samples of beef and pork collected from slaughter houses in Kyeonggi province.

Microstructural Feature and Aging Characteristics of Spray-Formed Cu-5Ni-10Sn Alloy (가스분무성형 Cu-5Ni-10Sn 합금의 미세조직 및 시효강화)

  • In this study, Cu-5Ni-10Sn(wt%) spinodal alloy was manufactured by gas atomization spray forming, and the microstructural features and mechanical properties of Cu-5Ni-10Sn alloy have been investigated during homogenization, cold working and age-hardening. The spray formed Cu-5Ni-10Sn alloy consisted of an equiaxed microstructure with a mixture of solid solution ${\alpha}$-(CuNiSn) grains and lamellar-structure grains. Homogenization at $800^{\circ}C$ and subsequent rapid quenching formed a uniform solid solution ${\alpha}$-(CuNiSn) phase. Direct aging at $350^{\circ}C$ from the homogenized Cu-5Ni-10Sn alloy promoted the precipitation of finely distributed ${\gamma}$' or ${\gamma}-(Cu,Ni)_3Sn$ phase throughout the matrix, resulting in a significant increase in microhardness and tensile strength. Cold working prior to aging was effective in strengthening Cu-5Ni-10Sn alloy, which gave rise to a maximum tensile strength of 1165 MPa. Subsequent aging treatment slightly reduced the tensile strength to 1000-1100 MPa due to annealing effects.

Analysis of Microwave Permeability and Damping Constant in Amorphous CoFeHfO Thin Film (비정질 CoFeHfO 박막 재료의 마이크로파 투자율 및 감쇠상수 분석)

  • The saturation magnetization and uniaxial anisotropy constant were obtained from magnetization and torque curves measurement in high resistive CoFeHfO thin film. The measured results were used for the analysis of the microwave complex permeability based on Landau-Lifshitz-Gilbert (LLG) theory. The high resistive CoFeHfO thin films showed very low damping constants of ${\alpha}$ = 0.014. The results are interpreted in terms of various magnetic phase with very low damping constant, which were existing inside the CoFeHfO thin film, through the linewidth analysis of the ferromagnetic resonance signal with magnetic field.

Isolation and Identification of Nonpolar Taxane Derivatives from the Plant Cell Culture of Taxus chinensis

  • Nonpolar taxoides extracted from a large-scale cell culture of Taxus chinensis were isolated through the normal and reverse phase column chromatographies, and their compounds were identified via NMR spectroscopy. The complete separation method was systematically established and described. In dichloromethane, dissolved paclitaxel and other taxoids with hexane were precipitated during the purification of paclitaxel from the plant cell culture of T. chinensis through a large-scale process while the relatively nonpolar taxane derivatives remained dissolved in the hexane phase. 13-Deoxy baccatin III (I), baccatin VI (II), taxchinin I (III), $2{\alpha}$, $5{\alpha}$, $10{\beta}$, $14{\beta}$-tetraacetoxy-4(20), 11-taxadiene(IV), 1-deoxy baccatinVI(V), and taxayuntin C (VI) were isolated through column chromatography and identified via NMR spectroscopy. Compounds I and IV were found to the major components, aside from paclitaxel, in the plant cell culture of T. chinensis. The concentrations of I and IV were compared with the that concentration of the paclitaxel in each of plant cell culture. The possible applications of compounds I, II, IV, and V were discussed.

Structural Design and Characterization of a Channel-forming Peptide

  • A 16-residue polypeptide model with the sequence acetyl-YALSLAATLLKEAASL-OH was derived by rational de novo peptide design. The designed sequence consists of amino acid residues with high propensity to adopt an alpha helical conformation, and sequential order was arranged to produce an amphipathic surface. The designed sequence was chemically synthesized using a solid-phase method and the polypeptide was purified by reverse-phase liquid chromatography. Molecular mass analysis by electro-spray ionization mass spectroscopy confirmed the correct designed sequence. Structural characterization by circular dichroism spectroscopy demonstrated that the peptide adopts the expected alpha helical conformation in 50% acetonitrile solution. Liposome binding assay using Small Unilamellar Vesicle (SUV) showed a marked release of entrapped glucose by interaction between the lipid membrane and the tested peptide. The channel-forming activity of the peptide was revealed by a planar lipid bilayer experiment. An analysis of the conducting current at various applied potentials suggested that the peptide forms a cationic ion channel with an intrinsic conductance of 188 pS. These results demonstrate that a simple rational de novo design can be successfully employed to create short peptides with desired structures and functions.

A study on manufacturing formation of the MLV liposomes by the microfludizer (마이크로플루다이저를 사용한 MLV liposome 형성에 관한 연구)

  • The liposomes have been developed in many drugs and cosmetics fields. In this context, it should be mentioned that MLV liposomes can be prepared standing with the main compounds of the intercellular lipids, cholesterol, palmitic acid, cholesteryl ester and lecithin, by swelling reaction. We report properties of the formation of MLV liposomes with use of the lipid base and Microfluidizer. MLV liposomes formed as multilamellar vesicles(MLV). The effect of the gelation of MLV liposomes have been on swelling reaction which have been mixed lipid with polyol and water phase at high temperature(90$\pm$5$^{\circ}C$). MLV liposomes have been prepared in incorporating alpha hydroxy acid ligrediens. Optimum condition of MLV liposomes were passed three times in the microfluidizer, particle size of the vesicles should be within 150-350nm and those confirmed by freeze-fracture electron microscopy.

Influence of the Electrical Parameters on the Fabrication of Oxide Layers on the Surface of Al-1050 by a Plasma Electrolytic Process (플라즈마 전해 산화법에 의한 Al-1050 표면상의 산화막 제조에 미치는 전기적 변수의 영향)

  • Oxide layers were prepared by an environmentally friendly plasma electrolytic oxidation (PEO) process on an Al-1050 substrate. The electrolyte for PEO was an alkali-based solution with $Na_2SiO_3$ (8 g/L) and NaOH (3 g/L). The influence of the electrical parameters on the phase composition, microstructure and properties of the oxide layers formed by PEO were investigated by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The voltage-time responses were recorded during various PEO processes. The oxides are composed of two layers and are mainly made of ${\alpha}$-alumina, ${\gamma}$-alumina and mullite phases. The proportion of each phase depends on various electrical parameters. It was found that the surface of the oxides produced at a higher current density and Ia/Ic ratio shows a more homogeneous morphology than those produced with the electrical parameters of a lower current density and lower Ia/Ic ratio. Also, the oxide layers formed at a higher current density and higher Ia/Ic ratio show high micro-hardness levels.
