The $\beta$-lactamase gene was cloned into E. coli DH5$\alpha$ from Bacillus sp. J105 with strong resistance against $\beta$-lactam antibiotics. The chromosomal DNA was partially digested with Sau3AI and ligated to BamHI digested pLAFR3. $\beta$-Lactamase positive clones were obtained by using in vitro packaging kit. The pKL11-${\Delta}4.6$ with $\beta$-lactamase activity was obtained by subcloning of the recombinant plasmid ($\beta$-lac +). The 6.5 kb fragment in the subcloned plasmid was sequenced. The DNA fragment that contains the $\beta$-lactamase gene encodes 309 amino acids. The 0.17 kb upstream region was similar to those of B. thuringinesis and B. cereus with 97% identity. The deduced amino acids sequence was also similar to those of $\beta$-lactamase from B. thuringinesis and B. cereus with 97% and 94% identity, respectively. The phylogenetic tree also showed the relationships of the $\beta$-lactamase gene of Bacillus sp. J105 to genetically related that of other Bacillus strains. Analysis of expression pattern of the pKL11-${\Delta}4.6$ in E. coli, revealed that the secretion efficiency of $\beta$-lactamase was $4{\sim}5%$ and the molecular weight was as same as that of original $\beta$-lactamase (31 kDa) from Bacillus sp. J105.
Natural products has been occasionally used in place of growth regulators to control lateral root formation and growth of soybean sprouts. The study was done to measure the effects of treatment concentration of hard rubber tree (Eucommia ulmoides Oliver) leaf extract on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into water, its 5 and 10% solutions immediately before 6 day culture. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyl lengths; >7 cm (A),4 to 7 cm (B),<4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Rate of A was decreased with increased concentrations, while that of C showed reverse result compared to A. Regardless of concentrations, the former rate was less in cv. Junjery than in cv. Pungsannamulkong and Sowonkong than in cv. Junjery although the latter rate was truly opposite to the former one. In 10% treatment concentration, lateral roots were less formed in cv. Junjery although nearly formed in cv. Pungsannamulkong and Sowonkong. In all the 3 cultivars, lateral roots per sprouts were decreased with increased treatment concentration. shorter and thicker sprout was observed in cv. Junjery than in the other cultivars, and in increased concentrations compared to relatively lower ones. Total fresh weight was the greatest in cv. Sowonkong due to mainly increment of its hypocotyl, but was reduced with increased concentration due to mainly decrement of hypocotyl in all the cultivars.
This study was performed to investigate the effect of dried powder of chestnut on lipid metabolism, anti-thrombotic effect in rats. Thirty 5-week-old male Sprague Dawley (SD) rats were randomly allocated into five groups and used for experiment. We examined the lipid metabolism and antithrombotic capacity of SD rats administered for 5 weeks with 0.16 g/kg, 0.5 g/kg chestnut flesh powder and 0.16 g/kg, 0.5 g/kg chestnut inner shell and flesh powder mixture, respectively. Food intake, body weight gain and food efficiency ratio were also checked. The levels of serum triglyceride and tree fatty acid were not statistically significant between the all experimental groups. However, the antithrombotic capacity and total lipid levels of the treatment groups were significantly lower than those of the negative control group. These results suggest that the supplementation of chestnut on diet lower the total lipid level in SD rats.
This experiment was carried out to elucidate the behavior of nutrition absorption and the acid tolerance, on varying soil pH levels, of Pinus thunbergii, Pinus densiflora, Pinus rigida, PInus koraiensis, Larix leptolepis and Abies holophylla which are 6 month old seedlings through examinations of the absorption behavior, the acid and alkali tolerance of N,P,K, Ca and Fe in the sand culture media with artificially controlled pH. 1. The increase rate of dry matter conditioned by the behavior of nutrition absorption in a strong acidic condition (pH3) was found in the following decreasing order of Pinus rigida, Pinus thunbergii. This order, therefore, can also be said to apply to the case of acid tolerance. 2. The increase rate of dry matter in a strong alkaline condition (pH9) was found in the following decreasing order of Pinus thunbergii, Pinus rigida, Pinus densiflora, Pinus koraiensis, Abies hoplohylla, Larix leptolepis. This order, therefore can be said to apply to the case of alkali tolerance. 3. Considering the absorption behavior of K and Ca, it is possible to decide the tolerance of acid from the absorption behavior and it can be said that if the absorption quantity is K>Ca the acid tolerance is strong and vice versa. 4. Considering the ratio of K:Fe and Ca:Fe in this experiment, a strong acid tolerance shows a higher ratio of K/Fe and vice versa. And also a strong acid tolerance shows a lower ratio of Ca/Fe and vice versa. It is therefore possible to decide the acid tolerance from the absorption behavior of K, Ca and Fe. 5. Of the tree species subjected to this experiment, Pinus thunbergii was the strongest in both acid and alkali tolerance. 6. In spite of that in all species the dry weight in the root is smailer than that in the top, the quantity of Fe contained in the root was larger than that in the top.

This study was carried out to evaluate the fruit and growth characteristics of selecting Vitis amurensis through functional material analysis and sensory evaluation in V. amurensis collected in Gangwondo. For evaluation of growth characteristics in V. amurensis, experiments were carried out by compared with the two grape cultivars 'Campbell Early' and 'Kyoho'. The full bloom and verasion time in V. amurensis was investigated faster than those of examined cultivar grapes, while harvest time was investigated latter than those of examined cultivar grapes, but agronomic characteristics was not thought significantly difference between cultivar grapes and V. amurensis. For evaluation of shoot growth phase, the growth curve was very similar to cultivar grapes. The berry size in V. amurensis showed that increases rapidly between 3 and 4 days after full bloom time, and approximately doubles between the second growth period and harvest time, and the berry development phase investigated that consist of two sigmoid growth periods separated by a lag phase. The berry weight and soluble solids in V. amurensis increased with the tree age, but acidity and total sugar contents decreased, and showed a special quality and stable growth according to vine age. To investigation of functional materials, the anthocyanin content in V. amurensis ranged from 16.6 to 50.2 mg/100 g, and the resveratrol content ranged from 0.143 to $0.236{\mu}g/100\;g$ which was higher than those of cultivar grapes. These result indicated that V. amurensis tended to have the useful material larger than cultivar grapes. Therefore, other edibility factors of V. amurensis collected in Gangwondo may contribute to breeding studies in Vitis. spp.

This study was wanted to select a value evaluation item to maintain the authenticity of the Planting Landscape in Hahoe and Yangdong villages. Above all, after checking the suitability of the 43 selected items by the Focus Group Interview, the weight was calculated in the Analytic Hierarchy Process expert questionnaire to establish the importance of the indicators for the development of the assessment items. The expert analysis analyzed the importance of 2 sections, 6 divisions and 11 detailed categories, and summarized the results of the study as follows : First, the comparative importance of each category and selection of the assessment items for the stabilization of the Planting Landscape of Hahoe and Yangdong indicated that cultural values are more important than biological values. In particular, details of the biological values of trees were derived with relatively low estimates, except for the specific characteristics of species and items of type of tree. Second, as a result of verification of the suitability of the 43 items selected by the Focus Group Interview, the source diameter, the collection width, reception, flushing, supersonality, records, memorials, 11 items were selected, Third, the results of the importance evaluation of the value properties for maintaining the resistance of vegetation through the Analytic Hierarchy Process were : specific in biological values (0.187), steady (0.094), and water pipe width (2007). There was relatively little difference in the highest weights, the width of the pipe except for the lowest value received, and the source diameter flushing. Fourth, the results of an evaluation of the importance of a value property aimed at historical values were whether or not the cultural asset was designated (0.134), the record value (0.092), the time (0.088), and the monument (0.063). In terms of the importance of evaluating the historical values of Planting sites consisting of Hahoe and Yangdong, the importance of designation of cultural properties was considered to be relative to the maintenance of the Planting Landscape including culture and history. Based on the assessment items and weighted values of the Planting Landscape of Yangdong Village and the World Cultural Heritage below and below, this study's Analytic Hierarchy Process can be applied to actual criteria for the assessment of the authenticity of trees in the village. Based on the assessment items and weighted values of the Planting Landscape in Hahoe and Yangdong villages, a follow-up study on the assessment standards for the authenticity of trees in the village will be left as a future task.

The present study investigated pine trees, which forms a major plantation species in Korea, with the objective of improving the survival rate of pine trees after planting. Growth responses and characteristics were assessed by controlling the level of fertilizer application, which is a basic controlling the growth of pine seedlings, to identify the optimal fertilization treatment. Pine tree seedlings were grown in 104 containers and were examined 8 weeks after planting. Stem height and were measured at 4-week intervals. In terms of fertilization treatment for 1-0 pine seedlings, the treatment group with gradually-increasing fertilizer concentration ($500{\rightarrow}1000{\rightarrow}1000{\rightarrow}1000mg{\cdot}L^{-1}$) had the biggest increase in stem height and diameter at the root. The survey results indicated that the increased concentration treatment group and the gradually-increasing concentration treatment group had more growth compared with that in the fixed concentration treatment group. The gradually-increasing concentration treatment group ($500{\rightarrow}1000{\rightarrow}1000{\rightarrow}1000mg{\cdot}L^{-1}$) had the highest total dry matter production. Nine weeks after fertilization, the tips of the pine leaves turned yellow in the fixed concentration treatment group ($3000mg{\cdot}L^{-1}$). The same phenomenon was observed in the treatment group in which the concentration was increased to $2000mg{\cdot}L^{-1}$, and in the gradually-increasing concentration treatment group, when the concentration was raised up to $2000mg{\cdot}L^{-1}$. We concluded that the optimal fertilization conditions for producing healthy pine 1-0 seedlings involve fertilizing once a week with Multifeed 19 at $500mg{\cdot}L^{-1}$ during the seedling period, Multifeed 19 at $1000mg{\cdot}L^{-1}$ during the rapid growth period, and Multifeed 32 at $1000mg{\cdot}L^{-1}$ during the maturation period.

Rock mass classification results have a great influence on construction schedule and budget as well as tunnel stability in tunnel design. A total of 3,526 tunnels have been constructed in Korea and the associated techniques in tunnel design and construction have been continuously developed, however, not many studies have been performed on how to assess rock mass quality and grade more accurately. Thus, numerous cases show big differences in the results according to inspectors' experience and judgement. Hence, this study aims to suggest a more reliable rock mass classification (RMR) model using machine learning algorithms, which is surging in availability, through the analyses based on various rock and rock mass information collected from boring investigations. For this, 11 learning parameters (depth, rock type, RQD, electrical resistivity, UCS, Vp, Vs, Young's modulus, unit weight, Poisson's ratio, RMR) from 13 local tunnel cases were selected, 337 learning data sets as well as 60 test data sets were prepared, and 6 machine learning algorithms (DT, SVM, ANN, PCA & ANN, RF, XGBoost) were tested for various hyperparameters for each algorithm. The results show that the mean absolute errors in RMR value from five algorithms except Decision Tree were less than 8 and a Support Vector Machine model is the best model. The applicability of the model, established through this study, was confirmed and this prediction model can be applied for more reliable rock mass classification when additional various data is continuously cumulated.
The giant freshwater prawn (Macrobrachium rosenbergii) suffers economic losses in aquaculture facilities due to problems such as poor body color and carapace weakness. While this species is farmed on an animal-based diet, in the wild it consumes a detritus diet with a high proportion of plant matter. The plant matter they consume in the wild is not only a source of basic nutrients, but also carotenoids, which are responsible for body color. Korean goldenbell (Forsythia koreana) is a flowering tree widely distributed in Korea, and its leaves contain carotenoids comparable to yellow carrots. In this study, we investigated the effects of feeding Forsythia leaves to M. rosenbergii on their body color and health. The experimental diets were 100% commercial feed (control), CON, 80% commercial feed plus 20% Forsythia leaf powder, FP, and 80% commercial feed plus 20% unprocessed Forsythia leaves, FL, and each diet was fed to juvenile prawn with an average weight of 1.1 ± 0.2 g for 10 weeks. In terms of body color, the CON prawn were consistently clear ivory in color, while the FP and FL prawn changed from blue to dark brown as the days of rearing progressed. Survival and growth were not significantly different between CON and FP or FL. Histological comparison of the hepatopancreas revealed that the vacuole size of B cells, the constituent cells of the hepatopancreatic tubule, was significantly larger in FP and FL compared to CON. The vacuoles of B cells are responsible for the absorption and digestion of nutrients, suggesting that the supply of Forsythia leaves may have had a positive effect on the health of the M. rosenbergii. The above results show that the use of Forsythia leaves in the aquaculture of M. rosenbergii can be expected to improve body color and health without inhibiting growth.
A survey on the intestinal atresias was made among 34 members of Korean Association of Pediatric Surgeons. The response rate was 82.4 %. Two hundred and fifteen patients from the January 1, 1994 to December 31, 1996 were analyzed. The lesions were 73 cases of duodenum(DA), 72 cases of jejunum(JA), 71 cases ileum(IA) and 2 cases cecum and sigmoid colon respectively. There were 2 cases of combined anomalies (DA + JA + IA and DA + JA). Male to female ratio was 1:1 in DA, and 1.8:1 in JA. Seventy four cases(34.3 %) were premature babies(DA 35.2 %, JA:48.6 %, IA:19.2 %), and 62 cases(28.7 %) had low birth weight (DA:39.4 %, JA 33.0 %, IA:13.7 %). Antenatal diagnosis was made in 92 cases(43.6 %). However 22 cases (23.9 %) of them were transferred to pediatric surgeon after delivery. Maternal polyhydramnios was observed in 63 cases(28.9 %). Seventy· five cases(34.4 %) were taken only simple abdominal film for diagnostic studies. The associated malformations were observed in 54 aresia and were observed more frequently in DA(35 cases, 47.9 %). Meconium peritonitis due to intrauterine bowel perforation was more frequently associated with IA compared to DA and JA. The overall mortality rate was 30 %.
